| 1 | Modeling study of regional severe hazes over mid-eastern China in January 2013 and its implications on pollution prevention and control显示文摘The Nested Air Quality Prediction Model System(NAQPMS)was used to investigate the temporal and spatial variations of PM2.5over tropospheric central eastern China in January 2013.The impact of regional transport and its implications on pollution prevention and control were also examined.Comparison between simulated and observed PM2.5showed NAQPMS was able to reproduce the evolution of PM2.5during heavy haze episodes.The results indicated that regional transport of PM2.5played an important role in regional haze episodes in the city cluster including Hebei,Beijing and Tianjin(HBT).The cross-city clusters transport outside HBT and transport among cities inside HBT contributed 20%–35%and 26%–35%of PM2.5as compared with local emission,in HBT respectively.To meet the Air Quality Standards for Grade II,90%,90%and65%of emissions would have to be cut down in Hebei,Tianjin and Beijing,if non-control strategy was taken in the surrounding city clusters of HBT.This implicated that control of emissions in one city cluster is not sufficient to reduce regional haze events,and joint efforts among city clusters are essential.Besides regional transports,two-way feedback between boundary-layer evolution and PM2.5also significantly contributed to the formation of heavy hazes,which contributed 30%of monthly average PM2.5concentration in HBT. | WANG ZiFa LI Jie WANG Zhe YANG WenYi TANG Xiao GE BaoZhu YAN PinZhong ZHU LiLi CHEN XueShun CHEN HuanSheng WAND Wei LI JianJun LIU Bing WANG XiaoYan WAND Wei ZHAO YiLin LU Ning SU DeBin | 2014 | Science China Earth Sciences2014,57,1: | 106 |
| 2 | Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. | 3 | Fluid/melt in continental deep subduction zones: Compositions and related geochemical fractionations显示文摘Plate subduction is the most magnificent process in the Earth. Subduction zones are important sites for proceeding matter- and energy- transports between the Earth's surface and the interior, continental crust growth, and crust-mantle interactions. Besides, a number of geological processes in subduction zones are closely related to human beings' daily life, such as volcanic eruptions and earthquakes, formation of mineral deposits. Subduction process thus has long been the centric topic of Earth sciences. The finding in 1980 s that continental crust could be subducted to mantle depths is a revolutionary progress in plate tectonic theory. Compared to oceanic crust, continental crust is colder, drier, lighter, and much more geochemically/isotopically heterogeneous. Hence, continental subduction process would affect the structure, compositions and evolutions of the overlying mantle wedge even more. During continental subduction and subsequent exhumation, fluids and melts can be generated in the(de)hydration process and partial melting process, respectively. These melts/fluids play important roles in crust-mantle interactions, elemental migrations, isotopic fractionations, and mantle metasomatism. By summarizing recent research works on subduction zones in this paper, we present a review on the types, physicochemical conditions and compositions of fluids/melts, as well as the migration behaviors of fluid-related characteristic elements(Nb-Ta-V) and the fractionation behaviors of non-traditional stable isotopes(Li-Mg) in subduction zones. | 4 | All-in-one cellulose based hybrid tribo/piezoelectric nanogenerator显示文摘Aybrid tribo/piezoelectric nanogenerators (HTPENG) have been proven to be highly efficient and versatile as far as the collection and conversion of ambient energy are concerned, and the introduction of flexible and green materials is a key step for their potential applications. Here, we developed a HTPENG by using nitrocellulose nanofibril paper as the triboelectric layer and BaTiCVMWCNT@ bacterial cellulose paper as the piezoelectric layer. The output of the triboelelctric paper has considerable performance as fluorinated ethylene propylene, and the output of piezoelectric paper is more than ten times higher than the BTO/polydimethylsiloxane structure. The integrated outputs of the sandwich structured HTPENG are 18 V and 1.6 pA·cm^-2, which are capable of lighting up three LED bulbs and charging a 1 pF capacitor to 2.5 V in 80 s. In addition, the voltage signal generated by the HTPENG in contact-separation mode can be used for dynamic pressure detection. The linear range of dynamic pressure is from 0.5 to 3 N·cm^-2 with a high sensitivity of 8.276 V·cm^-2·N^-1 and a detection limit of 0.2 N·cm^-2. | 5 | A telomere-to-telomere gap-free reference genome of watermelon and its mutation library provide important resources for gene discovery and breeding显示文摘Watermelon,Citrullus lanatus,is the world's third largest fruit crop.Reference genomes with gaps and a narrow genetic base hinder functional genomics and genetic improvement of watermelon.Here,we report the assembly of a telomere-to-telomere gap-free genome of the elite watermelon inbred line G42 by incorporating high-coverage and accurate long-read sequencing data with multiple assembly strategies.All 11 chromosomes have been assembled into single-contig pseudomolecules without gaps,representing the highest completeness and assembly quality to date.The G42 reference genome is 369321829 bp in length and contains 24205 predicted protein-coding genes,with all 22 telomeres and 11 centromeres characterized.Furthermore,we established a pollen-EMS mutagenesis protocol and obtained over 200000M1 seeds from G42.In a sampling pool,48 monogenic phenotypic mutations,selected from 223M1and 78 M2 mutants with morphological changes,were confirmed.The average mutation density was 1 SNP/1.69Mband1 indel/4.55 Mb per M1 plant and 1SNP/1.08Mb and 1 indel/6.25 Mb per M2 plant.Taking advantage of the gap-free G42 genome,8039 mutations from 32 plants sampled from M1 and M2 families were identified with 100%accuracy,whereas only 25% of the randomly selected mutations identified using the 97103v2 reference genome could be confirmed.Using this library and the gap-free genome,two genes responsible for elongated fruit shape and male sterility(CiMs1)were identified,both caused by a single basechange from G to A.The validated gap-free genome and its EMS mutation library provide invaluable resources for functional genomics and genetic improvement of watermelon. | Yun Deng Shoucheng Liu Yilin Zhang Jingsheng Tan Xiaopeng Li Xiao Chu Binghua Xu Yao Tian Yudong Sun Bosheng Li YunbiXu Xing Wang Deng Hang He Xingping Zhang | 2022 | Molecular Plant2022,15,8: | 9 |

| 7 | Aneuploidy of chromosome 8 in circulating tumor cells correlates with prognosis in patients with advanced gastric cancer显示文摘Objective: Previous work indicated that aneuploidy of chromosome 8 in circulating tumor cells(CTCs)correlated with therapeutic efficacy for advanced gastric cancer(AGC) patients. In this follow-up study performed on the same population of AGC patients, we investigated whether and how aneuploidy of chromosome 8 in CTCs correlates with patients' clinical prognosis.Methods: The prospective study was performed on 31 patients with newly diagnosed AGC. Previously established integrated subtraction enrichment(SE) and immunostaining-fluorescence in situ hybridization(i FISH)platform was applied to identify, enumerate and characterize CTCs. Quantification of CTCs and analysis of their aneuploidy of chromosome 8 were performed on patients before and after therapy.Results: CTCs were measured in 93.5% of AGC patients, and two CTC subtypes with diverse threshold values were identified, multiploid CTCs with the threshold of ≥2 per 7.5 m L and multiploid plus triploid CTCs with the threshold of ≥4, which were found to significantly correlate with poor progression-free survival(PFS) and overall survival(OS). In particular, patients with ≥10% increased multiploid CTCs after an initial 6 weeks of therapy had poor PFS and OS, whereas improved PFS and OS were observed on those who had ≥10% decreased multiploid CTCs. After adjusting for clinically significant factors, ≥10% increased post-therapy multiploid CTCs was the only independent predictor of PFS and OS.Conclusions: Aneuploidy of CTCs correlates with prognosis of AGC patients. | 9 | Combination of a crude oil-degrading bacterial consortium under the guidance of strain tolerance and a pilot-scale degradation test显示文摘Under the guidance of strain tolerance, a new combination method for crude oil-degrading bacterial consortium was studied. Firstly, more than 50 efficient crude oil-degrading and biosurfactant producing bacteria were isolated from petroleum-contaminated soil and water in Tianjin Binhai New Area Oilfield, China. Twenty-four of them were selected for further study. These strains were identified as belonging Pseudomonas aeruginosa, Bacillus subtilis, Brevibacillus brevis, Achromobacter sp., Acinetobacter venetianus, Lysinibacillus macroides, Klebsiella oxytoca, Stenotrophomonas rhizophila, Rhodococcus sp. and Bacillus thuringiensis. A shake-flask degradation test revealed that 12 of these strains could degrade over 50% of 1% crude oil concentration in 7 d. Of these, 8 strains were able to produce biosurfactants. Furthermore, environmental tolerance experiments indicated that the majority of the strains had the ability to adapt to extreme environments including high temperatures, alkaline environments and high salinity environments. A mixed bacterial agent comprising the strains WB2, W2, W3 and HA was developed based on the environmental tolerance tests and subjected to the pilot-scale degradation test indicating that this bacterial agent could degrade 85.2% of 0.8% crude oil concentration in 60 d. | 10 | Pertuzumab in combination with trastuzumab and chemotherapy for Chinese patients with HER2-positive metastatic gastric or gastroesophageal junction cancer:a subpopulation analysis of the JACOB trial显示文摘Background:The JACOB trial(NCT01774786)was a double-blinded,placebo-controlled,randomized,multicenter,international,phase III trial evaluating the efficacy and safety of adding pertuzumab to trastuzumab and chemo-therapy in first-line treatment of human epidermal growth factor receptor 2(HER2)-positive metastatic gastric cancer/gastroesophageal junction cancer(GEJC).The aim of this analysis was to investigate efficacy and safety outcomes in the Chinese subpopulation from the JACOB trial.Methods:This post hoc subpopulation analysis included all patients recruited in China's Mainland(n=163;20.9%)between June 2013 and January 2016.The patients were randomly assigned in a 1:1 ratio to receive pertuzumab plus trastuzumab and chemotherapy(pertuzumab group;n=82)or placebo plus trastuzumab and chemotherapy(con-trol group;n=81).Intravenous pertuzumab(840 mg)and trastuzumab(8 mg/kg loading and 6 mg/kg maintenance doses)were given every 3 weeks until disease progression or unacceptable toxicity.Chemotherapy was given as per standard regimens/doses of capecitabine or 5-fluorouracil plus cisplatin.The primary endpoint was overall survival(OS);secondary efficacy endpoints included progression-free survival(PFS),and overall objective response rate(ORR).Results:The median OS was 18.7 months in the pertuzumab group and 16.1 months in the control group(hazard ratio[HR]0.75;95%confidence interval[CI]0.49 to 1.14).The median PFS was 10.5 and 8.6 months in the pertuzumab and control groups,respectively(HR 0.85;95%CI 0.60 to 1.21),and the median ORRs were 68.9%and 55.7%,respectively.The treatment effect in this Chinese subpopulation showed consistency with that in the global ITT population with numerically lower HR for OS and PFS compared with the control group.The safety profiles of the pertuzumab and control groups in this Chinese subpopulation analysis were generally comparable.The most common grade 3-5adverse events were neutropenia,anemia,and leukopenia.However,due to the nature of being a post hoc subgroup analysis,the results presented here are descriptive only and need to be interpreted with caution.Conclusions:OS and PFS were numerically improved by adding pertuzumab to trastuzumab and chemotherapy as first-line treatment in Chinese HER2-positive gastric cancer/GEJC patients,and this regimen demonstrated an acceptable safety profile. | Tianshu Liu Yanru Qin Jin Li Ruihua Xu Jianming Xu Shujun Yang Shukui Qin Yuxian Bai Changping Wu Yixiang Mao Haiyan Wu Yilin Ge Lin Shen | 2019 | Cancer Communications2019,39,1: | 6 |

| 15 | An implantable microelectrode array for simultaneous L-glutamate and electrophysiological recordings in vivo显示文摘L-glutamate,the most common excitatory neurotransmitter in the mammalian central nervous system(CNS),is associated with a wide range of neurological diseases.Because neurons in CNS communicate with each other both electrically and chemically,dualmode(electric and chemical)analytical techniques with high spatiotemporal resolution are required to better understand glutamate function in vivo.In the present study,a silicon-based implantable microelectrode array(MEA)composed of both platinum electrochemical and electrophysiological microelectrodes was fabricated using micro-electromechanical system.In the MEA probe,the electrophysiological electrodes have a low impedance of 0.018 MΩat 1 kHz,and the electrochemical electrodes show a sensitivity of 56 pAμM^(−1) to glutamate and have a detection limit of 0.5μM.The MEA probe was used to monitor extracellular glutamate levels,spikes and local field potentials(LFPs)in the striatum of anaesthetised rats.To explore the potential of the MEA probe,the rats were administered to KCl via intraperitoneal injection.K+significantly increases extracellular glutamate levels,LFP low-beta range(12–18 Hz)power and spike firing rates with a similar temporal profile,indicating that the MEA probe is capable of detecting dual-mode neuronal signals.It was concluded that the MEA probe can help reveal mechanisms of neural physiology and pathology in vivo. | Wenjing Wei Yilin Song Li Wang Song Zhang Jinping Luo Shengwei Xu Xinxia Cai | 2015 | Microsystems & Nanoengineering2015,1,1: | 4 |

| 17 | Substantial transition to clean household energy mix in rural China显示文摘The household energy mix has significant impacts on human health and climate,as it contributes greatly to many health-and climate-relevant air pollutants.Compared to the well-established urban energy statistical system,the rural household energy statistical system is incomplete and is often associated with high biases.Via a nationwide investigation,this study revealed high contributions to energy supply from coal and biomass fuels in the rural household energy sector,while electricity comprised∼20%.Stacking(the use of multiple sources of energy)is significant,and the average number of energy types was 2.8 per household.Compared to 2012,the consumption of biomass and coals in 2017 decreased by 45%and 12%,respectively,while the gas consumption amount increased by 204%.Increased gas and decreased coal consumptions were mainly in cooking,while decreased biomass was in both cooking(41%)and heating(59%).The time-sharing fraction of electricity and gases(E&G)for daily cooking grew,reaching 69%in 2017,but for space heating,traditional solid fuels were still dominant,with the national average shared fraction of E&G being only 20%.The non-uniform spatial distribution and the non-linear increase in the fraction of E&G indicated challenges to achieving universal access to modern cooking energy by 2030,particularly in less-developed rural and mountainous areas.In some non-typical heating zones,the increased share of E&G for heating was significant and largely driven by income growth,but in typical heating zones,the time-sharing fraction was<5%and was not significantly increased,except in areas with policy intervention.The intervention policy not only led to dramatic increases in the clean energy fraction for heating but also accelerated the clean cooking transition.Higher income,higher education,younger age,less energy/stove stacking and smaller family size positively impacted the clean energy transition. | Guofeng Shen Rui Xiong Yanlin Tian Zhihan Luo Bahabaike Jiangtulu Wenjun Meng Wei Du Jing Meng Yuanchen Chen Bing Xue Bin Wang Yonghong Duan Jia Duo Fenggui Fan Lei Huang Tianzhen Ju Fenggui Liu Shunxin Li Xianli Liu Yungui Li Mu Wang Ying Nan Bo Pan Yanfang Pan Lizhi Wang Eddy Zeng Chao Zhan Yilin Chen Huizhong Shen Hefa Cheng Shu Tao | 2022 | National Science Review2022,9,7: | 4 |
| 18 | Development of a rapid and sensitivity magnetic chemiluminescence immunoassay for DNA methyltransferase 1 in human serum显示文摘DNA methyltransferase 1(DNMT1)is a useful biomarker for lung cancer in early clinical diagnosis.A rapid magnetic chemiluminescence immunoassay(MCLIA)for DNMT1 in human serum has been developed.Horseradish peroxidase(HRP)-second-Ab was used to labeled polyclonal antibodies of anti-DNMT1.DNMT1 in sample integrates with specific immunomagnetic beads and can constitute a supersandwiched immunoreaction.In magnetic field,nonspecific materials can be separated.After luminescent substrate luminol-H2O2-BIP was added,the relative light unit(RLU)of HRP was detected and was discovered to be directly proportional to the content of DNMT1 in sample.The correlative variables involved in the MCLIA value were optimized and the methodological evaluation was carried out.After optimization,in the range of0.5–128 ng/mL,the linear regression equation was y=0.5014 x+1.769(x was logCDNMT1,y was relative luminescence units(RLU)/RLU0),and the limit of detection was 0.01 ng/mL.The RSD of intra-and interassays were 15.8%–16.9%and 14.3%–18.1%,respectively.The recovery was from 70.0%to 106.2%.Furthermore,paralleled with purchasable enzyme-linked immunosorbent assay(ELISA)kits,MCLEIA had lower detection limit,wider linear range and shorter detection time.Therefore,the MCLEIA established in this study could be used for the sensitive detection of DNMT1 in serum sample. | Sitian He Leiliang He Beibei Liu Songcheng Yu Li'e Liu Yongmei Tian Jia Wang Lihua Ding Yilin Wang Lingbo Qu Fei Yu Yongjun Wu | 2019 | Chinese Chemical Letters2019,30,5: | 4 |
| 19 | PG-CODE:Latent Dirichlet Allocation Embedded Policy Knowledge Graph for Government Department Coordination显示文摘Government policy-group integration and policy-chain inference are significant to the execution of strategies in current Chinese society.Specifically,the coordination of hierarchical policies implemented among government departments is one of the key challenges to rural revitalization.In recent years,various well-established quantitative methods have been proposed to evaluate policy coordination,but the majority of these relied on manual analysis,which can lead to subjective results.Thus,in this paper,a novel approach called“policy knowledge graph for the coordination among the government departments”(PG-CODE)is proposed,which incorporates topic modeling into policy knowledge graphs.Similar to a knowledge graph,a policy knowledge graph uses a graph-structured data model to integrate policy discourse.With latent Dirichlet allocation embedding,a policy knowledge graph could capture the underlying topics of the policies.Furthermore,coordination strength and topic diffusion among hierarchical departments could be inferred from the PG-CODE,as it can provide a better representation of coordination within the policy space.We implemented and evaluated the PG-CODE in the field of rural innovation and entrepreneurship policy,and the results effectively demonstrate improved coordination among departments. | Yilin Kang Renwei Ou Yi Zhang Hongling Li Shasha Tian | 2022 | Tsinghua Science and Technology2022,27,4: | 3 |
| 20 | Recent advances of aptamer sensors显示文摘Aptamers are a series of high-affinity and high-specificity oligoneucleotides (single-stranded DNA or RNA) to the target, usually selected by the combinatorial chemistry SELEX technique (systematic evolution of ligands by exponential enrichment). Aptamers have proved to be one kind of novel functional molecules in life science and chemistry. After being labeled by signaling groups, the aptamer probe can conveniently transfer the characteristics of aptamer-target recognition to a form of high-sensitive signal, and the high-affinity, high-specificity measurements of metal ion, organic molecules, nucleic acid, proteins, or cells become possible.
