| 1 | Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. | 3 | LIM domain proteins Pinch1/2 regulate chondrogenesis and bone mass in mice显示文摘The LIM domain-containing proteins Pinch1/2 regulate integrin activation and cell–extracellular matrix interaction and adhesion.Here,we report that deleting Pinch1 in limb mesenchymal stem cells(MSCs)and Pinch2 globally(double knockout;dKO)in mice causes severe chondrodysplasia,while single mutant mice do not display marked defects.Pinch deletion decreases chondrocyte proliferation,accelerates cell differentiation and disrupts column formation.Pinch loss drastically reduces Smad2/3 protein expression in proliferative zone(PZ)chondrocytes and increases Runx2 and Col10a1 expression in both PZ and hypertrophic zone(HZ)chondrocytes.Pinch loss increases sclerostin and Rankl expression in HZ chondrocytes,reduces bone formation,and increases bone resorption,leading to low bone mass.In vitro studies revealed that Pinch1 and Smad2/3 colocalize in the nuclei of chondrocytes.Through its C-terminal region,Pinch1 interacts with Smad2/3 proteins.Pinch loss increases Smad2/3 ubiquitination and degradation in primary bone marrow stromal cells(BMSCs).Pinch loss reduces TGF-β-induced Smad2/3 phosphorylation and nuclear localization in primary BMSCs.Interestingly,compared to those from single mutant mice,BMSCs from dKO mice express dramatically lower protein levels ofβ-catenin and Yap1/Taz and display reduced osteogenic but increased adipogenic differentiation capacity.Finally,ablating Pinch1 in chondrocytes and Pinch2 globally causes severe osteopenia with subtle limb shortening.Collectively,our findings demonstrate critical roles for Pinch1/2 and a functional redundancy of both factors in the control of chondrogenesis and bone mass through distinct mechanisms. | Yiming Lei Xuekun Fu Pengyu Li Sixiong Lin Qinnan Yan Yumei Lai Xin Liu Yishu Wang Xiaochun Bai Chuanju Liu Di Chen Xuenong Zou Xu Cao Huiling Cao Guozhi Xiao | 2020 | Bone Research2020,8,4: | 2 |

| 6 | Increased plasma leptin as a novel predictor for psychopathological depressive symptoms in chronic schizophrenia显示文摘Background Depressive symptoms are often seen in schizophrenia. The overlap in presentation makes it difficult to distinguish depressive symptoms from the negative symptoms of schizophrenia. The adipokine leptin was found to be altered in both depression and schizophrenia. | 12 | Climate Suitability Assessment of Human Settlements for Regions along the Belt and Road显示文摘Climate is one important environmental variable that affects human life.As the regions along the Belt and Road(B&R)encompass vast territories and large populations,it is significant to assess climate suitability for human settlements,which will influence the migration of various surrounding countries.We selected seven regions including 65 countries along the B&R for the research area and adopted the temperature-humidity index(THI),to assess the climate suitability.We analyzed the spatial characteristics of THI and the correlation between population distribution and THI,the results of which enabled us to adjust the THI classification criteria.We finally assessed the climate suitability of each region.The results reveal that outside the Qinghai-Tibet Plateau,the THI values generally tend to decrease from west to east as longitude increases and downward with increasing latitude.The population distribution is significantly correlated with the THI.Regions with relative suitable climate,including high suitability areas(HASs),moderately suitable areas(MSAs)and low suitability areas(LSAs),account for 50.62%of the total area and encompass in excess of 90%of total population.These areas are widely distributed in the southern regions of 45°N latitude,again with the exception of the Qinghai-Tibet Plateau.Critical suitable area includes 19.48%of the entire area and 8.98%of total population.The non-suitable area accounts for less than 30%of total area,concentrated in the cold high-latitude and high-altitude areas. | LIN Yumei LI Peng FENG Zhiming YANG Yanzhao YOU Zhen ZHU Fuxin | 2021 | Chinese Geographical Science2021,31,6: | 1 |

| 14 | Spatial-Temporal Change of Phytoplankton Biomass in the East China Sea with MODIS Data显示文摘A ten-year chlorophyll-a concentration dataset from Moderate Resolution Imaging Spectro-radiometer(MODIS) were used to analyze the variation of phytoplankton biomass and its potential relation with climate in the East China Sea. The result indicated that the phytoplankton biomass generally had a regular pattern every year, and phytoplankton bloom mainly occurred between May and July. The highest phytoplankton biomass appeared near the Yangtze River Estuary. The lowest phytoplankton biomass located near the Taiwan Strait. In general, the starting bloom time was earlier in the south than in the north, and the span time of the former was also longer. During the recent ten years, the phytoplankton biomass around the Yangtze River Estuary decreased obviously. The change of phytoplankton biomass was found to be related with the Ni?o3.4 Index. | 19 | Continuous activation of polymorphonuclear myeloid-derived suppressor cells during pregnancy is critical for fetal development显示文摘The maternal immune system is vital in maintaining immunotolerance to the semiallogeneic fetus for a successful pregnancy.Although studies have shown that myeloid-derived suppressor cells(MDSCs)play an important role in maintaining feto-maternal tolerance,little is known about the role of MDSCs in pregnancies with intrauterine growth retardation(IUGR).Here,we reported that the activation of polymorphonuclear myeloid-derived suppressor cells(PMN-MDSCs)during pregnancy was closely associated with fetal growth.In humans,class E scavenger receptor 1(SR-E1),a distinct marker for human PMN-MDSCs,was used to investigate PMN-MDSC function during pregnancy.Continuous activation of SR-E1+PMN-MDSCs was observed in all stages of pregnancy,accompanied by high cellular levels of ROS and arginase-1 activity,mediated through STAT6 signaling.However,SR-E1+PMN-MDSCs in pregnancies with IUGR showed significantly lower suppressive activity,lower arginase-1 activity and ROS levels,and decreased STAT6 phosphorylation level,which were accompanied by an increase in inflammatory factors,compared with those in normal pregnancies.Moreover,the population of SR-E1+PMN-MDSCs was negatively correlated with the adverse outcomes of newborns from pregnancies with IUGR.In mice,decreases in cell population,suppressive activity,target expression levels,and STAT6 phosphorylation levels were also observed in the pregnancies with IUGR compared with the normal pregnancies,which were rescued by the adoptive transfer of PMN-MDSCs from pregnant mice.Interestingly,the growth-promoting factors(GPFs)secreted by placental PMN-MDSCs in both humans and mice play a vital role in fetal development.These findings collectively support that PMN-MDSCs have another new role in pregnancy,which can improve adverse neonatal outcomes. | Mengyu Shi Ziyang Chen Meiqi Chen Jingping Liu Jing Li Zhe Xing Xiaogang Zhang Shuaijun Lv Xinyao Li Shaowen Zuo Shi Feng Ying Lin Gang Xiao Liping Wang Yumei He | 2021 | Cellular & Molecular Immunology2021,18,7: | 1 |
