1Oncogenic AURKA-enhanced N6-methyladenosine modification increases DROSHA mRNA stability to transactivate STC1 in breast cancer stem-like cells显示文摘RNase III DROSHA is upregulated in multiple cancers and contributes to tumor progression by hitherto unclear mechanisms.Here,we demonstrate that DROSHA interacts withβ-Catenin to transactivate STC1 in an RNA cleavage-independent manner,contributing to breast cancer stem-like cell(BCSC)properties.DROSHA mRNA stability is enhanced by N6-methyladenosine(m^(6)A)modification which is activated by AURKA in BCSCs.AURKA stabilizes METTL14 by inhibiting its ubiquitylation and degradation to promote DROSHA mRNA methylation.Moreover,binding of AURKA to DROSHA transcript further strengthens the binding of the m^(6)A reader IGF2BP2 to stabilize m^(6)A-modified DROSHA.In addition,wild-type DROSHA,but not an m^(6)A methylation-deficient mutant,enhances BCSC stemness maintenance,while inhibition of DROSHA m^(6)A modification attenuates BCSC traits.Our study unveils the AURKA-induced oncogenic m^(6)A modification as a key regulator of DROSHA in breast cancer and identifies a novel DROSHA transcriptional function in promoting the BCSC phenotype.Fei Peng Jie Xu Bai Cui Qilan Liang Sai Zeng Bin He Hong Zou Manman Li Huan Zhao Yuting Meng Jin Chen Bing Liu Shasha Lv Peng Chu Fan An Zifeng Wang Junxiu Huang Yajing Zhan Yuwei Liao Jinxin Lu Lingzhi Xu Jin Zhang Zhaolin Su Zhiguang Li Fangjun Wang Eric W-FLam Quentin Liu 2021Cell Research2021,31,3:9
2DNA N^6-methyladenine demethylase ALKBH1 enhances osteogenic differentiation of human MSCs显示文摘ALKBH1 was recently discovered as a demethylase for DNA N^6-methyladenine(N6-m A), a new epigenetic modification, and interacts with the core transcriptional pluripotency network of embryonic stem cells.However, the role of ALKBH1 and DNA N6-m A in regulating osteogenic differentiation is largely unknown.In this study, we demonstrated that the expression of ALKBH1 in human mesenchymal stem cells(MSCs)was upregulated during osteogenic induction. Knockdown of ALKBH1 increased the genomic DNA N6-m A levels and significantly reduced the expression of osteogenic-related genes, alkaline phosphatase activity, and mineralization. ALKBH1-depleted MSCs also exhibited a restricted capacity for bone formation in vivo.By contrast, the ectopic overexpression of ALKBH1 enhanced osteoblastic differentiation. Mechanically,we found that the depletion of ALKBH1 resulted in the accumulation of N6-m A on the promoter region of ATF4, which subsequently silenced ATF4 transcription. In addition, restoring the expression of ATP by adenovirus-mediated transduction successfully rescued osteogenic differentiation. 3Natural orifice transluminal endoscopic surgery in urology: The Chinese experience显示文摘Objective:To describe the Chinese experience of natural orifice transluminal endoscopic surgery(NOTES)in urology.Methods:From December 2008 to May 2017,35 animal experiments and 305 clinical surgeries of NOTES or natural orifices specimen extractions(NOSE)were performed in China.The animal experiments included five kidney biopsies,24 nephrectomies and six partial nephrectomies.The clinical surgeries included 12 transvaginal NOSE(TV-NOSE),266 hybrid transvaginal NOTES(TV-NOTES)and 27 pure TV-NOTES.The TV-NOSE procedure was performed in five transumbilical laparoendoscopic single-site(U-LESS)nephrectomies,four suprapubic-assisted laparoendoscopic single-site surgery(SA-LESS)nephroureterectomies,and three laparoscopic radical cystectomies.The hybrid TV-NOTES procedure included 210 nephrectomies,31 adrenalectomies,eight nephroureterectomies,13 partial nephrectomies,and four heminephrectomies.The pure TV-NOTES procedure included five renal cyst decortications and 22 nephrectomies.Results:A total of 29 animal experiments were successfully performed.One partial nephrectomy was converted to standard laparoscopic surgery.Two kidney biopsies and two nephrectomies were unsuccessful.A total of 297 clinical surgeries were successfully performed.Six patients who underwent hybrid TV-NOTES were converted to open surgery.Two patients who underwent pure TV-NOTES were converted to SA-LESS.There were 22 major complications,16 occurred intraoperatively and six postoperatively.The mean visual analog score(VAS)of 48 h after the operation was 2.5 points in TV-NOSE,2.3 points in hybrid TV-NOTES and 1.7 points in pure TV-NOTES.The mean follow-up of 50.6(3.0-87.0)months showed that all patients were in good condition.The umbilicus scars were nearly invisible in TV-NOSE and hybrid TV-NOTES.The vaginal incision healed well.Conclusions:TV-NOSE and TV-NOTES are feasible,safe,and effective with little injury,low pain,fast recovery,and good cosmetic outcomes in properly selected patients.They are worth consideration for urological clinical practice.Xiaofeng Zou Guoxi Zhang Tianpeng Xie Yuanhu Yuan Rihai Xiao Gengqing Wu Xiaoning Wang Hui Xu Folin Liu Yuting Wu Yunfeng Liao Quanliang Liu Yinghao Sun Bo Yang Linhui Wang Chuanliang Xu Xiaofeng Gao 2020Asian Journal of Urology2020,7,1:5

5Modulating the optical and electrical properties of MAPbBr_(3) single crystals via voltage regulation engineering and application in memristors显示文摘Defect density is one of the most significant characteristics of perovskite single crystals(PSCs)that determines their optical and electrical properties,but few strategies are available to tune this property.Here,we demonstrate that voltage regulation is an efficient method to tune defect density,as well as the optical and electrical properties of PSCs.A three-step carrier transport model of MAPbBr_(3) PSCs is proposed to explore the defect regulation mechanism and carrier transport dynamics via an applied bias.Dynamic and steady-state photoluminescence measurements subsequently show that the surface defect density,average carrier lifetime,and photoluminescence intensity can be efficiently tuned by the applied bias.In particular,when the regulation voltage is 20 V(electrical poling intensity is 0.167 Vμm^(−1)),the surface defect density of MAPbBr_(3) PSCs is reduced by 24.27%,the carrier lifetime is prolonged by 32.04%,and the PL intensity is increased by 112.96%.Furthermore,a voltage-regulated MAPbBr_(3) PSC memristor device shows an adjustable multiresistance,weak ion migration effect and greatly enhanced device stability.Voltage regulation is a promising engineering technique for developing advanced perovskite optoelectronic devices.Jun Xing Chen Zhao Yuting Zou Wenchi Kong Zhi Yu Yuwei Shan Qingfeng Dong Ding Zhou Weili Yu Chunlei Guo 2020Light(Science & Applications)2020,9,1:3

8SERS study on the synergistic effects of electric field enhancement and charge transfer in an Ag2S quantum dots/plasmonic bowtie nanoantenna composite system显示文摘Localized surface plasmon resonance(LSPR)of nanostructures and the interfacial charge transfer(CT)of semiconductor materials play essential roles in the study of optical and photoelectronic properties.In this paper,a composite substrate of Ag2S quantum dots(QDs)coated plasmonic Au bowtie nanoantenna(BNA)arrays with a metalinsulator-metal(MIM)configuration was built to study the synergistic effect of LSPR and interfacial CT using surface-enhanced Raman scattering(SERS)in the near-infrared(NIR)region.The Au BNA array structure with a large enhancement of the localized electric field(E-field)strongly enhanced the Raman signal of adsorbed p-aminothiophenol(PATP)probe molecules.Meanwhile,the broad enhanced spectral region was achieved owing to the coupling of LSPR The as-prepared Au BNA array structure facilitated enhancements of the excitation as well as the emission of Raman signal simultaneously,which was established by finite-difference time-domain simulation.Moreover,Ag2S semiconductor QDs were introduced into the BNA/PATP system to further enhance Raman signals,which benefited from the interfacial CT resonance in the BNA/Ag2S-QDs/PATP system.As a result,the Raman signals of PATP in the BNA/Ag2S-QDs/PATP system were strongly enhanced under 785 nm laser excitation due to the synergistic effect of E-field enhancement and interfacial CT.Furthermore,the SERS polarization dependence effeas of the BNA/Ag2S-QDs/PATP system were also investigated.The SERS spectra indicated that the polarization dependence of the substrate increased with decreasing polarization angles(θpola)of excitation from p-polarized(θpola=90°)excitation to s-polarized(θpola=0°)excitation.This study provides a strategy using the synergistic effect of interfacial CT and E-field enhancement for SERS applications and provides a guidance for the development of SERS study on semiconductor QD-based plasmonic substrates,and can be farther extended to other material-nanostructure systems for various optoelectronic and sensing applications.BIN WANG CHEN ZHAO HUANYU LU TINGTING ZOU SUBHASH C.SINGH ZHI YU CHAONAN YAO XIN ZHENG JUN XING YUTING ZOU CUNZHU TONG WEILI YU BO ZHAO CHUNLEI GUO 2020Photonics Research2020,8,4:1

11Polarimetric parity-time symmetry in a photonic system显示文摘Parity-time(PT)symmetry has attracted intensive research interest in recent years.PT symmetry is conventionally implemented between two spatially distributed subspaces with identical localized eigenfrequencies and complementary gain and loss coefficients.The implementation is complicated.In this paper,we propose and demonstrate that PT symmetry can be implemented between two subspaces in a single spatial unit based on optical polarimetric diversity.By controlling the polarization states of light in the single spatial unit,the localized eigenfrequencies,gain,loss,and coupling coefficients of two polarimetric loops can be tuned,leading to PT symmetry breaking.As a demonstration,a fiber ring laser based on this concept supporting stable and single-mode lasing without using an ultranarrow bandpass filter is implemented.Lingzhi Li Yuan Cao Yanyan Zhi Jiejun Zhang Yuting Zou Xinhuan Feng Bai-Ou Guan Jianping Yao 2020Light(Science & Applications)2020,9,1:1
12The capacity and mechanisms of various oxidants on regulating the redox function of ZVI显示文摘It is well known that zero-valent iron(ZVI)could catalyze the oxidation of various oxidants to realize the rapid oxidation removal of pollutants.However,in this study,we found that the addition of different oxidants could regulate the redox function of ZVI system.In three different co-treatment systems,the effects of different oxidizers(peroxymonosulfate(PMS),persulfate(PDS),hydrogen peroxide(H_(2)O_(2)))dosages on the ratios of oxidative degradation rate and reductive degradation rate of p-nitrophenol(PNP)were studied.The effect of the H+released from oxidizers and the generated reactive oxygen species(ROS)in ZVI/PMS,ZVI/PDS,ZVI/H_(2)O_(2)systems were detailed discussed.Especially,the contribution of generated ROS for reductive degradation of PNP was quantified in the ZVI/H_(2)O_(2)system.Based on the results of TOC removal,UV-vis absorption spectra,and intermediates concentration curves,it was found that the degradation of PNP changed from reduction to oxidation with the increase of oxidant proportion.When the molar ratio of ZVI to oxidizer equal to 100,PNP was mainly degraded by reduction accompanied by slight oxidation.Combined with the results of SEM-EDS and XPS,it was confirmed that the enhanced degradation of PNP under the addition of oxidant was mainly related to the generated ROS,the additional H+,and the corrosion products of ZVI.Siqi Zou Qun Chen Yang Liu Yuting Pan Gang Yao Zhicheng Pan Bo Lai 2021Chinese Chemical Letters2021,32,6:1
13Glutamate drives local Ca^(2+)release'in cardiac pacemaker cells显示文摘The sinoatrial node(SAN)is the origin of the electrical signals for rhythmic heartbeats in mammals.The spontaneous fring of SAN pacemaker cells(SANPCS)triggers cardiac contraction.'Local Ca^(2+)release'(LCR),a unique cellular activity,acts as the'engine'of the spontaneous fring of SANPCs.However,the mechanism of LCR initiation remains unclear.Here,we report that endogenous glutamate drives LCRs in SANPCs.Using a glutamate sensor,we unraveled a tight correlation between glutamate accumulation and LCR occurrence,indicating a potential relationship between glutamate and LCRs.Intracellular application of glutamate significantly enhanced the LCRs in both intact and permeabilized SANPCs.Mechanitically,we revealed that mitochondrial excitatory amino acid transporter 1(EAAT 1)-dependent mitochondrial glutamate import promoted ROS generation,which in turn led to the oxidation of Ca^(2+)-handling proteins,ultimately resulting in enhanced LCRs.Importantly,EAAT1 depletion reduced both the spontaneous fring rates of isolated SANPCs and the heart rate in vitro and in vivo,suggesting the central role of EAAT1 as a glutamate transporter in the regulation of cardiac autonomic rhythm.In conclusion,our results indicate that glutamate serves as an LCR igniter in SANPCs,adding a potentially important element to the coupled clock theory that explains the origin of spontaneous firing.These findings shed new light on the future prevention and treatment of cardiac pacemaker cell-related arrhythmias.Duanyang Xie Ke Xiong Xuling Su Guanghua Wang Qicheng Zou Luxin Wang Caihong Zhang Yuting Cao Beihua Shao Yixin Zhang Peidong Zhang Dandan Liang Yi Liu Yi-Han Chen 2022Cell Research2022,32,9:0

15Assessment of heavy metal pollution in Laizhou Bay(China)using the ecological risk index and the integrated biomarker response of the goby Acanthogobius ommaturus显示文摘We used the Integrated Biological Responses version 2(IBRv2)method to evaluate the biological eff ects of heavy metals in the sediments in Laizhou Bay,China on the benthic goby Acanthogobius ommaturus.In December 2018,gobies and sediments were collected from 15 stations.We measured the activities of defense enzymes and the contents of malondialdehyde(MDA)and metallothionein(MT)in the goby liver as well as the levels of heavy metals in the sediments and goby muscle tissue.Most of the heavy metal concentrations in sediment at each station were below the Class I criteria set by Chinese Standards for Marine Sediment Quality,and the Håkanson ecological risk index suggested low risk for the heavy metals.We found that A.ommaturus could eff ectively accumulate mercury,cadmium,arsenic,and zinc and that the contents of MT and MDA and the activities of glutathione peroxidase and glutathione reductase were suitable biomarkers of heavy metal pollution in this species.The IBRv2 method integrated these four biomarkers and discriminated stations according to heavy metal pollution.Higher IBRv2 values suggested more adverse eff ects in gobies,corroborating more serious heavy metal contamination.The stations with high IBRv2 values and high contents of heavy metals were mainly distributed in the west and northeast parts of the bay.These results show that the IBRv2 approach is a feasible strategy for assessing heavy metal pollution through biological response and biological status and that it can be implemented for environmental monitoring in Laizhou Bay.Maosheng LIU Hua XU Qiang ZOU Fei FANG Shan SUN Yuting ZHAO Xin HE Yonghui BO Lei YAO Yan FANG 2023Journal of Oceanology and Limnology2023,41,4:0
16CUL4B orchestrates mesenchymal stem cell commitment by epigenetically repressing KLF4 and C/EBPδ显示文摘Dysregulated lineage commitment of mesenchymal stem cells(MSCs)contributes to impaired bone formation and an imbalance between adipogenesis and osteogenesis during skeletal aging and osteoporosis.The intrinsic cellular mechanism that regulates MSC commitment remains unclear.Here,we identified Cullin 4B(CUL4B)as a critical regulator of MSC commitment.CUL4B is expressed in bone marrow MSCs(BMSCs)and downregulated with aging in mice and humans.Conditional knockout of Cul4b in MSCs resulted in impaired postnatal skeletal development with low bone mass and reduced bone formation.Moreover,depletion of CUL4B in MSCs aggravated bone loss and marrow adipose accumulation during natural aging or after ovariectomy.In addition,CUL4B deficiency in MSCs reduced bone strength.Mechanistically,CUL4B promoted osteogenesis and inhibited adipogenesis of MSCs by repressing KLF4 and C/EBPδexpression,respectively.The CUL4B complex directly bound to Klf4 and Cebpd and epigenetically repressed their transcription.Collectively,this study reveals CUL4B-mediated epigenetic regulation of the osteogenic or adipogenic commitment of MSCs,which has therapeutic implications in osteoporosis.Ruiqi Yu Hong Han Shuxian Chu Yijun Ding Shiqi Jin Yufeng Wang Wei Jiang Yuting Liu Yongxin Zou Molin Wang Qiao Liu Gongping Sun Baichun Jiang Yaoqin Gong 2023Bone Research2023,11,2:0
17A neuropsin-based optogenetic tool for precise control of Gq signaling显示文摘G_(q)-coupled receptors regulate numerous physiological processes by activating enzymes and inducing intracellular Ca^(2+)signals.There is a strong need for an optogenetic tool that enables powerful experimental control over G_(q) signaling.Here,we present chicken opsin 5(cOpn5)as the long sought-after,single-component optogenetic tool that mediates ultra-sensitive optical control of intracellular G_(q) signaling with high temporal and spatial resolution.Expressing cOpn5 in HEK 293T cells and primary mouse astrocytes enables blue light-triggered,G_(q)-dependent Ca^(2+) release from intracellular stores and protein kinase C activation.Strong Ca^(2+) transients were evoked by brief light pulses of merely 10 ms duration and at 3 orders lower light intensity of that for common optogenetic tools.Photostimulation of cOpn5-expressing cells at the subcellular and single-cell levels generated fast intracellular Ca^(2+)transition,thus demonstrating the high spatial precision of cOpn5 optogenetics.The cOpn5-mediated optogenetics could also be applied to activate neurons and control animal behavior in a circuit-dependent manner.cOpn5 optogenetics may find broad applications in studying the mechanisms and functional relevance of G_(q) signaling in both non-excitable cells and excitable cells in all major organ systems.Ruicheng Dai Tao Yu Danwei Weng Heng Li Yuting Cui Zhaofa Wu Qingchun Guo Haiyue Zou Wenting Wu Xinwei Gao Zhongyang Qi Yuqi Ren Shu Wang Yulong Li Minmin Luo 2022Science China(Life Sciences)2022,65,7:0
18Seizures and epilepsy secondary to viral infection in the central nervous system显示文摘Viral infection in the central nervous system(CNS)is a common cause of seizures and epilepsy.Acute symptomatic seizures can occur in the context of almost all types of acute CNS viral infection.However,late unprovoked seizures and epilepsy may not be frequent after viral infection of the CNS.The incidence of seizures and epilepsy after CNS viral infection is mainly dependent on the brain region of infection.It remains to be determined whether treatment of CNS viral infection using antiepileptic drugs(AEDs)can prevent seizures and subsequent epilepsy in patients,particularly with regard to the timing,drug choice and dosage,and duration of AEDs.The postoperative outcome of seizures in patients with intractable epilepsy caused by viral encephalitis primarily depends on the epileptogenic zone.In addition,neuroinflammation is known to be widely involved in the generation of seizures during CNS viral infection,and the effects of anti-inflammatory therapies in preventing seizures and epilepsy secondary to CNS viral infection require further studies.In this review,we discuss the incidence,mechanisms,clinical management and prognosis of seizures and epilepsy secondary to CNS viral infection,and summarize common CNS viral infections that cause seizures and epilepsy.Peng Zhang Yuting Yang Jialin Zou Xinrui Yang Qiankun Liu Yangmei Chen 2020Acta Epileptologica2020,2,1:0
19Optimizing a high-sensitivity NanoLuc-based bioluminescence system for in vivo evaluation of antimicrobial treatment显示文摘Focal and systemic infections are serious threats to human health.Preclinical models enable the development of new drugs and therapeutic regimens.In vivo,animal bioluminescence(BL)imaging has been used with bacterial reporter strains to evaluate antimicrobial treatment effects.However,high-sensitivity bioluminescent systems are required because of the limited tissue penetration and low brightness of the BL signals of existing approaches.Here,we report that NanoLuc(Nluc)showed better performance than LuxCDABE in bacteria.However,the retention rate of plasmid constructs in bacteria was low.To construct stable Staphylococcus aureus reporter strains,a partner protein enolase(Eno)was identified by screening of S.aureus strain USA300 for fusion expression of Nluc-based luciferases,including Nluc,Teluc,and Antares2.Different substrates,such as hydrofurimazine(HFZ),furimazine(FUR),and diphenylterazine(DTZ),were used to optimize a stable reporter strain/substrate pair for BL imaging.S.aureus USA300/Eno-Antares2/HFZ produced the highest number of photons of orange-red light in vitro and enabled sensitive BL tracking of S.aureus in vivo,with sensitivities of approximately 10 CFU from mouse skin and 750 CFU from mouse kidneys.USA300/Eno-Antares2/HFZ was a powerful combination based on the longitudinal evaluation of the therapeutic efficacy of antibiotics.The optimized S.aureus Eno-Antares2/HFZ pair provides a technological advancement for the in vivo evaluation of antimicrobial treatment.Weilong Shang Zhen Hu Mengyang Li Yuting Wang Yifan Rao Li Tan Juan Chen Xiaonan Huang Lu Liu He Liu Zuwen Guo Huagang Peng Yi Yang Qiwen Hu Shu Li Xiaomei Hu Jiao Zou Xiancai Rao 2023mLife2023,2,4:0
20Large-scale preparation of fernwort-like single-crystalline superstructures of CuSe as Fenton-like catalysts for dye decolorization显示文摘A simple green hydrothermal template-free method was developed to prepare single-crystalline superstructures of fernwort-like copper selenide(CuSe) in large-scale by using polyvinylpyrrolidone(PVP) as both reductant and surfactant for the first time. Time-dependent morphologic evolution was made in order to explore the formation mechanism of the as-prepared product. The copper selenides with different morphologies, phases and structural forms could be prepared by varying the synthesis parameters, such as precursor molar ratios, precursor combinations, and the molecular weight of PVP. The fernwort-like superstructures of CuSe show excellent Fenton-like catalytic activities in degrading malachite green(MG) and rhodamine B(Rh B). These catalysts play an important role in the degradation process of MG and Rh B solution with the aid of H_2O_2 which can yield highly reactive hydroxyl radicals(HO?).
