| 4 | Characterization and fine mapping of the rice blast resistance gene Pia显示文摘Blast,caused by Magnaporthe oryzae,is one of the most widespread and destructive diseases of rice.Breeding durable resistant cultivars(cvs) can be achieved by pyramiding of various resistance(R) genes.Pia,carried by cv.Aichi Asahi,was evaluated against 612 isolates of M.oryzae collected from 10 Chinese provinces.The Pia gene expresses weak resistance in all the provinces except for Jiangsu.Genomic position-ready marker-based linkage analysis was carried out in a mapping population consisting of 800 F2 plants derived from a cross of Aichi Asahi×Kasalath.The locus was defined in an interval of approximately 90 kb,flanked by markers A16 and A21.Four candidate genes(Pia-1,Pia-2,Pia-3,and Pia-4) ,all having the R gene conserved structure,were predicted in the interval using the cv.Nipponbare genomic sequence.Four candidate resistance gene(CRG) markers(A17,A25,A26,and A27) ,derived from the four candidates,were subjected to genotyping with the recombinants detected at the flanking markers.The first three markers completely co-segregated with the Pia locus,and the fourth was absent in the Aichi Asahi genome and disordered with the Pia locus and its flanking markers,indicating that the fourth candidate gene,Pia-4,could be excluded.Co-segregation marker-based genotyping of the three sets of differentials with known R gene genotypes revealed that the genotype of A26(Pia-3) perfectly matched the R gene genotype of Pia,indicating that Pia-3 is the strongest candidate gene for Pia. | ZENG XiaoShan YANG XianFeng ZHAO ZhengHong LIN Fei WANG Ling PAN QingHua | 2011 | Science China(Life Sciences)2011,54,4: | 13 |

| 6 | Analysis of genetic architecture and favorable allele usage of agronomic traits in a large collection of Chinese rice accessions显示文摘Genotyping and phenotyping large natural populations provide opportunities for population genomic analysis and genome-wide association studies(GWAS). Several rice populations have been re-sequenced in the past decade;however, many major Chinese rice cultivars were not included in these studies. Here, we report large-scale genomic and phenotypic datasets for a collection mainly comprised of 1,275 rice accessions of widely planted cultivars and parental hybrid rice lines from China. The population was divided into three indica/Xian and three japonica/Geng phylogenetic subgroups that correlate strongly with their geographic or breeding origins. We acquired a total of 146 phenotypic datasets for 29 agronomic traits under multi-environments for different subpopulations. With GWAS, we identified a total of 143 significant association loci, including three newly identified candidate genes or alleles that control heading date or amylose content. Our genotypic analysis of agronomically important genes in the population revealed that many favorable alleles are underused in elite accessions, suggesting they may be used to provide improvements in future breeding efforts. | 9 | Heat Aggregation Mechanisms of Hot Dry Rocks Resources in the Gonghe Basin, Northeastern Tibetan Plateau显示文摘Hot dry rock(HDR) is an important geothermal resource and clean energy source that may play an increasingly important role in future energy management. High-temperature HDR resources were recently detected in deep regions of the Gonghe Basin on the northeastern edge of the Tibetan Plateau, which led to a significant breakthrough in HDR resource exploration in China. This research analyzes the deep temperature distribution, radiogenic heat production, heat flow, and crustal thermal structure in the Qiaboqia Valley, Guide Plain, and Zhacanggou area of the Gonghe Basin based on geothermal exploration borehole logging data, rock thermophysical properties, and regional geophysical exploration data. The results are applied to discuss the heat accumulation mechanism of the HDR resources in the Gonghe Basin. The findings suggest that a low-velocity layer in the thickened crust of the Tibetan Plateau provides the most important source of constant intracrustal heat for the formation of HDR resources in the Gonghe Basin, whereas crustal thickening redistributes the concentrated layer of radioactive elements, which compensates for the relatively low heat production of the basal granite and serves as an important supplement to the heat of the HDR resources. The negative effect is that the downward curvature of the lithospheric upper mantle caused by crustal thickening leads to a small mantle heat flow component. As a result, the heat flows in the Qiaboqia Valley and Guide Plain of the Gonghe Basin are 106.2 and 77.6 m W/m2, respectively, in which the crust-mantle heat flow ratio of the former is 3.12:1, indicating a notably anomalous intracrustal thermal structure. In contrast, the crust-mantle heat flow ratio in the Guide Plain is 1.84:1, which reflects a typical hot crust-cold mantle thermal structure. The Guide Plain and Zhacanggou area show the same increasing temperature trend with depth, which reflects that their geothermal backgrounds and deep high-temperature environments are similar. | 10 | Molecular dynamics simulation for surface melting and self-preservation effect of methane hydrate显示文摘The surface melting process of structure sI methane hydrate is simulated at T = 240, 260, 280, and 300 K using NVT molecular dynamics method. The simulation results show that a quasi-liquid layer will be formed during the melting process. The density distribution, translation, orientation, and dynamic properties of water molecules in the quasi-liquid layer are calculated as a function of the distance normal to the interface, which indicates the performance of quasi-liquid layer exhibits a continuous change from crystal-like to liquid-like. The quasi-liquid layer plays as a resistance of mass transfer restraining the diffusion of water and methane molecules during the melting process. The resistance of quasi-liquid layer will restrain methane molecules diffuse from hydrate phase to gas phase and slow the melting process, which can be considered as a possible mechanism of self-preservation effect. The performance of quasi-liquid layer is more crystal-like when the temperature is lower than the melt- ing-point of water, which will exhibit an obvious self-preservation. | 14 | Citron C-05 inhibits both the penetration and colonization of Xanthomonas citri subsp.citri to achieve resistance to citrus canker disease显示文摘Citrus canker,caused by Xanthomonas citri subsp.citri(Xcc),is a serious bacterial disease that affects citrus production worldwide.Citron C-05(Citrus medica)is the only germplasm in the Citrus genus that has been identified to exhibit strong resistance to Xcc.However,it has not been determined when,where,and how Xcc is restricted in the tissues of Citron C-05 during the infection process.In the present study,we investigated the spatiotemporal growth dynamics of an eGFP-labeled virulent Xcc(eGFP-Xcc)strain in Citron C-05 along with five susceptible biotypes(i.e.,lemon,pummelo,sour orange,sweet orange,and ponkan mandarin)upon inoculation via the spraying or leaf infiltration of a bacterial suspension.The results from extensive confocal laser scanning microscopy analyses showed that while Xcc grew rapidly in plants of all five susceptible genotypes,Xcc was severely restricted in the epidermal and mesophyll cell layers of the leaves of Citron C-05 in the early stage of infection.Not surprisingly,resistance against Xcc in Citron C-05 was found to be associated with the production of reactive oxygen species and hypersensitive response-like cell death,as well as greater upregulation of several defense-related genes,including a pathogenesis-related gene(PR1)and a glutathione S-transferase gene(GST1),compared with sweet orange as a susceptible control.Taken together,our results not only provide further valuable details of the spatiotemporal dynamics of the host entry,propagation,and spread of Xcc in both resistant and susceptible citrus plants but also suggest that resistance to Xcc in Citron C-05 may be attributed to the activation of multiple defense mechanisms. | Hongyan Fu Mingming Zhao Jing Xu Limei Tan Jian Han Dazhi Li Meijun Wang Shunyuan Xiao Xianfeng Ma Ziniu Deng | 2020 | Horticulture Research2020,7,1: | 2 |

| 19 | Novel Graphene Biosensor Based on the Functionalization of Multifunctional Nano-bovine Serum Albumin for the Highly Sensitive Detection of Cancer Biomarkers显示文摘A simple,convenient,and highly sensitive bio-interface for graphene field-effect transistors(GFETs) based on multifunctional nano-denatured bovine serum albumin(nano-dBSA) functionalization was developed to target cancer bio-markers.The novel graphene–protein bioelectronic interface was constructed by heating to denature native BSA on the graphene substrate surface.The formed nano-d BSA film served as the cross-linker to immobilize monoclonal antibody against car-cinoembryonic antigen(anti-CEA mAb) on the graphene channel activated by EDC and Sulfo-NHS.The nano-dBSA film worked as a self-protecting layer of graphene to prevent surface contamination by lithographic processing.The improved GFETbiosensor exhibited good specificity and high sensitivity toward the target at an ultralow concentration of 337.58 fg mL-1.The electrical detection of the binding of CEA followed the Hill model for ligand–receptor interaction,indicating the negative binding cooperativity between CEA and anti-CEA mAb with a dissociation constant of 6.82×10-10M.The multifunctional nano-dBSA functionalization can confer a new function to graphene-like 2D nanomaterials and provide a promising bio-functionalization method for clinical application in biosensing,nanomedicine,and drug delivery. | Lin Zhou Kun Wang Hao Sun Simin Zhao Xianfeng Chen Dahong Qian Hongju Mao Jianlong Zhao | 2019 | Nano-Micro Letters2019,11,2: | 1 |
