1Metabolic analyses reveal different mechanisms of leaf color change in two purple-leaf tea plant (Camellia sinensis L.) cultivars显示文摘Purple-leaf tea plants,as anthocyanin-rich cultivars,are valuable materials for manufacturing teas with unique colors or flavors.In this study,a new purple-leaf cultivar“Zixin”(“ZX”)was examined,and its biochemical variation and mechanism of leaf color change were elucidated.The metabolomes of leaves of“ZX”at completely purple,intermediately purple,and completely green stages were analyzed using ultra-performance liquid chromatography quadrupole time of flight mass spectrometry(UPLC-QTOF-MS).Metabolites in the flavonoid biosynthetic pathway remained at high levels in purple leaves,whereas intermediates of porphyrin and chlorophyll metabolism and carotenoid biosynthesis exhibited high levels in green leaves.In addition,fatty acid metabolism was more active in purple leaves,and steroids maintained higher levels in green leaves.Saponin,alcohol,organic acid,and terpenoid-related metabolites also changed significantly during the leaf color change process.Furthermore,the substance changes between“ZX”and“Zijuan”(a thoroughly studied purple-leaf cultivar)were also compared.The leaf color change in“Zijuan”was mainly caused by a decrease in flavonoids/anthocyanins.However,a decrease in flavonoids/anthocyanins,an enhancement of porphyrin,chlorophyll metabolism,carotenoid biosynthesis,and steroids,and a decrease in fatty acids synergistically caused the leaf color change in“ZX”.These findings will facilitate comprehensive research on the regulatory mechanisms of leaf color change in purple-leaf tea cultivars.Jiazhi Shen Zhongwei Zou Xuzhou Zhang Lin Zhou Yuhua Wang Wanping Fang Xujun Zhu 2018Horticulture Research2018,5,1:22
2Repurposing vitamin D for treatment of human malignancies via targeting tumor microenvironment显示文摘Tumor cells along with a small proportion of cancer stem cells exist in a stromal microenvironment consisting of vasculature, cancer-associated fibroblasts, immune cells and extracellular components.Recent epidemiological and clinical studies strongly support that vitamin D supplementation is associated with reduced cancer risk and favorable prognosis. Experimental results suggest that vitamin D not only suppresses cancer cells, but also regulates tumor microenvironment to facilitate tumor repression. In this review, we have outlined the current knowledge on epidemiological studies and clinical trials of vitamin D. Notably, wesummarized and discussed the anticancer action of vitamin D in cancer cells, cancer stem cells and stroma cells in tumor microenvironment, providing a better understanding of the role of vitamin D in cancer. 3Exogenous abscisic acid significantly affects proteome in teaplant (Camellia sinensis) exposed to drought stress显示文摘Tea[Camellia sinensis(L.)O.Kuntze]is an important economic crop,and drought is the most important abiotic stress affecting yield and quality.Abscisic acid(ABA)is an important phytohormone responsible for activating drought resistance.Increased understanding of ABA effects on tea plant under drought stress is essential to develop drought-tolerant tea genotypes,along with crop management practices that can mitigate drought stress.The objective of the present investigation is evaluation of effects of exogenous ABA on the leaf proteome in tea plant exposed to drought stress.Leaf protein patterns of tea plants under simulated drought stress[(polyethylene glycol(PEG)-treated]and exogenous ABA treatment were analyzed in a time-course experiment using two-dimensional electrophoresis(2-DE),followed by matrix-assisted laser desorption/ionization time-of-flight(MALDI-TOF)mass spectrometry(MS).Among the 72 protein spots identified by MALDI-TOF MS,16 proteins were downregulated and two were upregulated by exogenous ABA.The upregulated proteins have roles in glycolysis and photosystem II stabilization.Twenty-one protein spots were responsive to drought stress and most participate in carbohydrate and nitrogen metabolism,control of reactive oxygen species(ROS),defense,signaling or nucleic acid metabolism.The combined treatments of exogenous ABA and drought showed upregulation of 10 protein spots at 12 h and upregulation of 11 proteins at 72 h after initiation of drought stress.The results support the importance of the role that ABA plays in the tea plant during drought stress,by improving protein transport,carbon metabolism and expression of resistance proteins.Lin Zhou Hui Xu Sue Mischke Lyndel W Meinhardt Dapeng Zhang Xujun Zhu Xinghui Li Wanping Fang 2014Horticulture Research2014,1,1:12
4Heat shock protein 90 promotes RNA helicase DDX5 accumulation and exacerbates hepatocellular carcinoma by inhibiting autophagy显示文摘Objective:Hepatocellular carcinoma(HCC),the main type of liver cancer,has a high morbidity and mortality,and a poor prognosis.RNA helicase DDX5,which acts as a transcriptional co-regulator,is overexpressed in most malignant tumors and promotes cancer cell growth.Heat shock protein 90(HSP90)is an important molecular chaperone in the conformational maturation and stabilization of numerous proteins involved in cell growth or survival.Methods:DDX5 m RNA and protein expression in surgically resected HCC tissues from 24 Asian patients were detected by quantitative real-time PCR and Western blot,respectively.The interaction of DDX5-HSP90 was determined by molecular docking,immunoprecipitation,and laser scanning confocal microscopy.The autophagy signal was detected by Western blot.The cell functions and signaling pathways of DDX5 were determined in 2 HCC cell lines.Two different murine HCC xenograft models were used to determine the function of DDX5 and the therapeutic effect of an HSP90 inhibitor.Results:HSP90 interacted directly with DDX5 and inhibited DDX5 protein degradation in the AMPK/ULK1-regulated autophagy pathway.The subsequent accumulation of DDX5 protein induced the malignant phenotype of HCC by activating theβ-catenin signaling pathway.The silencing of DDX5 or treatment with HSP90 inhibitor both blocked in vivo tumor growth in a murine HCC xenograft model.High levels of HSP90 and DDX5 protein were associated with poor prognoses.Conclusions:HSP90 interacted with DDX5 protein and subsequently protected DDX5 protein from AMPK/ULK1-regulated autophagic degradation.DDX5 and HSP90 are therefore potential therapeutic targets for HCC.Ting Zhang Xinrui Yang Wanping Xu Jing Wang Dawei Wu Zhixian Hong Shengxian Yuan Zhen Zeng Xiaodong Jia Shanshan Lu Rifaat Safadi Sen Han Zhihong Yang Leonard M.Neckers Suthat Liangpunsakul Weiping Zhou Yinying Lu 2021Cancer Biology & Medicine2021,18,3:7
5Identification of the varietal origin of processed loose-leaf tea based on analysis of a single leaf by SNP nanofluidic array显示文摘Tea is an important cash crop, representing a $40 billion-a-year global market. Differentiation of the tea market has resulted in increasing demand for tea products that are sustainably and responsibly produced. Tea authentication is important because of growing concerns about fraud involving premium tea products. Analytical technologies are needed for protection and value enhancement of high-quality brands. For loose-leaf teas, the challenge is that the authentication needs to be established on the basis of a single leaf, so that the products can be traced back to the original varieties. A new generation of molecular markers offers an ideal solution for authentication of processed agricultural products. Using a nanofluidic array to identify variant SNP sequences, we tested genetic identities using DNA extracted from single leaves of 14 processed commercial tea products. Based on the profiles of 60 SNP markers, the genetic identity of each tea sample was unambiguously identified by multilocus matching and ordination analysis. Results for repeated samples of multiple tea leaves from the same products(using three independent DNA extractions) showed 100% concordance, showing that the nanofluidic system is a reliable platform for generating tea DNA fingerprints with high accuracy. The method worked well on green, oolong, and black teas, and can handle a large number of samples in a short period of time. 10Recognition for avian influenza virus proteins based on support vector machine and linear discriminant analysis显示文摘Total 200 properties related to structural characteristics were employed to represent structures of 400 HA coded proteins of influenza virus as training samples. Some recognition models for HA proteins of avian influenza virus (AIV) were developed using support vector machine (SVM) and linear discriminant analysis (LDA). The results obtained from LDA are as follows: the identification accuracy (Ria) for training samples is 99.8% and Ria by leave one out cross validation is 99.5%. Both Ria of 99.8% for training samples and Ria of 99.3% by leave one out cross validation are obtained using SVM model, respectively. External 200 HA proteins of influenza virus were used to validate the external predictive power of the resulting model.
