3Effects of AT1 receptor antagonist,Iosartan,on rat hepatic fibrosis induced by CCl_4显示文摘AIM To investigate effect of losartan,an AT1receptor antagonist,on hepatic fibrosis induced byCCl;and to determine whether or not AT1receptors are expressed on hepatic stellate cells,METHODS AND RESULTS Fifty male Sprague-Dawley rats,weighing(180±20)g,wererandomized into five groups(control group,modelgroup,and three losartan treated groups),inwhich all rats were given the subcutaneousinjection of 40% CCl4(every 3 days for 6 weeks)except for rats of control group.Rats of losartan-treated groups were treated with losartan(20 mg/kg,10 mg/kg,5 mg/kg,daily gavage),After 6weeks liver tissue and serum samples of all ratswere examined.Serum hyaluronic acid(HA),procollagen typeⅢ(PCⅢ)were detected byradioimmunoassays,van Giesion collagen stainingwas used to evaluate the extracellular matrix of ratswith liver fibrosis.The expression of AT1receptors,transforming growth factor-beta(TGF-β),and alpha-smooth muscle actin(a-SMA)inliver tissue were determined byimmunohistochemical techniques.Compared withmodel group,serum ALT and AST of losartan-treated groups were significantly reduced(t=4.20,P<0.01 and t=4.57,P<0.01).Serum HAand PCⅢalso had significant differences(t=3.53,P<0.01 and t=2.20,P<0.05).Thedegree of fibrosis was improved by losartan and correlated with the expressions of AT1 receptors,TGF-β,and α-SMA in liver tissue.CONCLUSION AT1 receptor antagonist,losartan,could limit the progression of the hepatic fibrosisinduced by CCl4.The mechanism may be related tothe decrease in the expression of AT1 receptorsand TGF-β,ameliorating the injury of hepatocytes;activation of local renin-angiotensin system mightrelate to hepatic fibrosis;and during progressionof fibrosis,activated hepatic stellate cells mightexpress AT1 receptors.Hong Shan Wei Ding Guo Li Han Ming Lu Yu Tao Zhan Zhi Rong Wang Xin Huang Jing Zhang Ji Lin Cheng Qin Fang Xu Department of Gastroenterology,Xinhua Hospital,Shanghai Second Medical University,Shanghai 200092,China 2000World Journal of Gastroenterology2000,6,4:42
4Resveratrol Induces Apoptosis and Autophagy in T-cell Acute Lymphoblastic Leukemia Cells by Inhibiting Akt/mTOR and Activating p38-MAPK显示文摘Objective To explore the effects of resveratrol-induced apoptosis and autophagy in T-cell acute lymphoblastic leukemia(T-ALL) cells and potential molecular mechanisms. Methods The anti-proliferation effect of resveratrol-induced, apoptosis and autophagy on T-ALL cells were detected by using MTT test, immunofluorescence, electronic microscope, and flow cytometry, respectively. Western blotting was performed for detecting changes of apoptosis-associated proteins, cell cycle regulatory proteins and state of activation of Akt, mTOR, p70S6K, 4E-BP1, and p38-MAPK. Results Resveratrol inhibited the proliferation and induced apoptosis and autophagy in T-ALL cells in a dose and time-dependent manner. It also induced cell cycle arrest at G0/G1 phase via up regulating cyclin-dependent kinase(CDK) inhibitors p21 and p27 and down regulating cyclin A and cyclin D1. Western blotting revealed that resveratrol significantly decreased the expression of antiapoptotic proteins(Mcl-1 and Bcl-2) and increased the expression of proapoptotic proteins(Bax, Bim, and Bad), and induced cleaved-caspase-3 in a time-dependent manner. Significant increase in ratio of LC3-II/LC3-I and Beclin 1 was also detected. Furthermore, resveratrol induced significant dephosphorylation of Akt, mTOR, p70S6K, and 4E-BP1, but enhanced specific phosphorylation of p38-MAPK which could be blocked by SB203580. When autophagy was suppressed by 3-MA, apoptosis in T-ALL cells induced by resveratrol was enhanced.Conclusion Our findings have suggested that resveratrol induces cell cycle arrest,apoptosis,and autophagy in T-ALL cells through inhibiting Akt/mTOR/p70S6K/4E-BP1 and activating p38-MAPK signaling pathways.Autophagy might play a role as a self-defense mechanism in T-ALL cells treated by resveratrol.Therefore,the reasonable inhibition of autophagy in T-ALL cells may serve as a promising strategy for resveratrol induced apoptosis and can be used as adjuvant chemotherapy for T-ALL.GE Jiao LIU Yan LI Qiang GUO Xia GU Ling MA Zhi Gui ZHU Yi Ping 2013Biomedical and Environmental Sciences2013,26,11:38
5The regulatory role of AT 1 receptor on activated HSCs in hepat,c fibrogenesis,effects of RAS inhibitors on hepatic fibrosis induced by CCl_4显示文摘AIM To assess the effect of ACE inhibitor andAng Ⅱ type Ⅰ(AT1)receptor antagonist inpreventing hepatic fibrosis caused by CCl4administration in rats;to investigate whether ornot there are expression of AT 1 receptors onhepatic stellate cells;and to observe the effectof Ang Ⅱ on proliferation and ECM synthesis ofcultured HSCs.METHODS Studies were conducted in maleSprague-Dawley rats.Except for thehepatofibrotic model group and the controlgroup,in three treated groups,either enalapril(5 mg/kg),or Iosartan(10 mg/kg),or enalapril+Iosartan were given to the fibrotic rats bydaily gavage,and saline vehicle was given tomodel and normal control rats.After 6 weeks,liver fibrosis was assessed directly by hepaticmorphometric analysis,which has beenconsidered the gold standard for thequantification of fibrosis.The expressions of AT1 receptors and(α-mooth muscle actin,α-SMA)in liver tissue or isolated hepatic stellate cells(HSCs)were detected by immunohistochemicaltechniques.The effect of Ang Ⅱ on HSCproliferation was determined by MTT method.Effect of Ang Ⅱ on collagen synthesis of HSCswas determined by 3H-proline incorporation.RESULTS Contrasted to the fibrosis in rats ofthe model group,groups of rats treated with either enalapril or Iosartan,or a combination oftwo drugs showed a limited expansion of theinterstitium(4.23±3.70 vs 11.22±4.79,P<0.05),but no difference was observedamong three treated groups(5.38±3.43,4.96±2.96,4.23±2.70,P>0.05).Expression of AT 1receptors was found in fibrotic interstitium offibrotic rats,whereas in normal control rats theywere limited to vasculature only to a very slightdegree.AT 1 receptors were also expressed onactivated HSCs in the culture.At concentrationsfrom 10-9to 10-5mol/L,Ang Ⅱ stimulated HSCproliferation in culture in a dose-dependentmanner.Increasing Ang Ⅱ concentrationsproduced corresponding increases in 3H-prolineincorporation.Differences among groups were significant.CONCLUSION Angiotensin-converting enzyme inhibitors and AT I blocker may slow the progression of hepatic fibrosis; activated HSCs express AT 1 receptors, and Ang Ⅱ can stimulate the proliferation and collagen synthesis of HSCs in a dose-dependent manner; and activation of RAS may be related to hepatic fibrogenesis induced by CCI4.Hong Shan Wei Han Ming Lu Ding Guo Li Yu Tao Zhan Zhi Rong Wang Xin Huang Ji Lin Cheng Qin Fang Xu Department of Gastroenterology,Xinhua Hospital,Shanghai Second Medical University,Shanghai 200092,China 2000World Journal of Gastroenterology2000,6,6:27
6Weapon target assignment problem satisfying expected damage probabilities based on ant colony algorithm显示文摘A weapon target assignment (WTA) model satisfying expected damage probabilities with an ant colony algorithm is proposed. In order to save armament resource and attack the targets effectively, the strategy of the weapon assignment is that the target with greater threat degree has higher priority to be intercepted. The effect of this WTA model is not maximizing the damage probability but satisfying the whole assignment result. Ant colony algorithm has been successfully used in many fields, especially in combination optimization. The ant colony algorithm for this WTA problem is described by analyzing path selection, pheromone update, and tabu table update. 7The crustal structures of the central Longmenshan along and its margins as related to the seismotectonics of the 2008 Wenchuan Earthquake显示文摘In 2010,a 500-km-long wide-angle reflection/refraction seismic profile was completed,running northwest from the central Sichuan Basin.This profile orthogonally crosses the meizoseismal area of great Wenchuan earthquake of 12 May 2008,which occurred in the central part of the Longmenshan.The profile also passes through the northwestern Sichuan Plateau,along which a new deep seismic sounding observation system was set up that was much improved over previous datasets and enabled abundant observations to be recorded.Seismic wave phase records that reflect the structural characteristics of different tectonic blocks,especially the complicated phase features associated with the Wenchuan earthquake,were calculated and analyzed in detail.A 2D crustal P-wave velocity model for the orogenic belt in the central Longmenshan and its margins was determined,and crustal structure differences between the stable Sichuan Basin and the thickened northwestern Sichuan Plateau were characterized.Lithological variations within the upper and lower crust in the interior of the plateau,especially a great velocity decrease and plastic rheological properties associated with strong lithologic weakening in lower crust,were detected.From west to east in the lower crust beneath the orogenic belt lying between the Sichuan Basin and the northwestern Sichuan Plateau,a giant shovel-like upwelling is observed that dips gently in the lower part and at higher angles in the upper part;this is inferred to be related to the fault systems in the central Longmenshan.An upwelling in the upper-middle crust along the eastern margin of the orogenic belt is associated with steeply dipping thrusts that strongly uplift the upper crust and crystalline basement beneath a central fault system in the Longmenshan.The data,combined with an understanding of the regional tectonic stress field and previous geological results,enable a discussion of basin-and-range coupling,orogenic tectonics,the crustal fault system,and the seismogenic tectonic environment of the central Longmenshan along the eastern margin of the Qinghai-Tibet Plateau.JIA ShiXu LIU BaoJin XU ZhaoFan LIU Zhi FENG ShaoYing ZHANG JianShi LIN JieYan TIAN XiaoFeng LIU QiaoXia GUO WenBin 2014Science China Earth Sciences2014,57,4:26
8Three-dimensional crustal velocity structure model of the middle-eastern north China Craton (HBCrust1.0)显示文摘Lithosphere thinning and destruction in the middle-eastern North China Craton(NCC), a region susceptible to strong earthquakes, is one of the research hotspots in solid earth science. All 42 seismic wide-angle reflection/refraction profiles have been completed in the middle-eastern NCC. We collect all the 2-D profiling results and perform gridding of the velocity and interface depth data, building a 3-D crustal velocity structure model for the middle-eastern NCC, named HBCrust1.0, by using the Kriging interpolation method. Our result shows that the first-arrival times calculated by HBCust1.0 fit well with the observations. The result demonstrates that the upper crust is the main seismogenic layer, and the brittle-ductile transition occurs at depths near interface C(the interface between upper and lower crust). The depth of interface Moho varies beneath the source area of the Tangshan earthquake, and a low-velocity structure is found to extend from the source area to the lower crust. Based on these observations, it can be inferred that stress accumulation responsible for the Tangshan earthquake may have been closely related to the migration and deformation of the mantle materials. Comparisons of the average velocities of the whole crust, the upper and the lower crust show that the average velocity of the lower crust under the central part of the North China Basin(NCB) in the east of the craton is obviously higher than the regional average. 9Clinical and experimental study on regional administration of phosphorus32 glassmicrospheres in treating hepatic carcinoma显示文摘AIM To study the therapeutical effectiveness, dosage range and toxic adverse effects of domestic phosphorus 32 glass microsphere and evaluate its clinical significance. METHODS Ⅰ. Fifty two BALB/*!c tumor bearing male nude mice were allocated into treatment group( n =38) and control group( n =14). In the former group different doses of 32 P GMS were injected into the tumor mass, while in the latter 31 P GMS or no treatment was given. The experimental animals were sacrificed in batches, and then the tumors and their nearby tissues were examined by light and electron microscopy. Ⅱ. Through selective catheterization of hepatic artery, 32 P GMS was infused to 5 healthy domestic pigs in a dosage equivalent to the therapeutic dose for human being, and 31 P GMS was infused to another 5 healthy domestic pigs. Two pigs infused with contrast medium served as whole course blank controls. One pig from each group was surrendered to euthanasia at week 1, 4, 8 and 16 respectively. The ultrastructural histopath ological changes in liver tissues taken from different sites were evaluated semiquan titatively. Ⅲ. One hundred and twenty seven times of 32 P GMS intrahepatic artery interventional therapies were performed on 93 patients with hepatic carcinoma, including 79 cases of primary hepatic carcinoma and 14 cases of secondary hepatic carcinoma. 32 P GMS ( n =30), and group B, 32 P GMS and half dose of trans hepatic artery embolization (TAE) ( n =49) , and 18 patients with HCC by TAE only as control group C. Fourteen patients with secondary hepatic carcinoma were treated in the same way as group B or C. RESULTS Ⅰ. Comparing with the control group, the treatment group of tumor bearing nude mice attained the tumor inhibition rates of 59 7%-93 7% ( F =579 62, P <0 01) at 14*!d . At an absorbed dose of 7320Gy, the tumor cells were completely destroyed. When the absorbed doses ranged from 1830Gy to 3660Gy, most of the tumor cells showed the evidences of injury or necrosis, but there appeared some well differentiated tumor cells and enhanced effect of the autoimmunocytes. At an absorbed dose of 366Gy or less, some tumor cells still remained active proliferative ability. The definite anticancer effect appeared as early as 3d after intratumoral injection of 32 P GMS. Ⅱ. The cumulative amount of 32 P GMS in the target tissue after trans hepatic artery instillation attained more than 90% of the total dose administrated. Semiquantitative analysis of ultrastructral morphology in the experimental group showed no statistical difference between the nuclear abnormality (N abn ) and mitochondrial variability (M var ) at week 1 or 2, but revealed prominent difference (χ 2=6 70-9 68, P <0 01 , χ 2=65 09-115 09, P <0 001 ) as compared with those in the other groups. In the experimental group the N abn in tissues showed no significant difference between week 8 and week 16. No apparent changes were found in the stomach, spleen, kidney and lung tissues of the experimental pigs. Ⅲ. The therapeutical results of HCC patients in group A were closely approximated to those of group C, no hematological toxic side effects were noted, and the systemic reaction was mild. In some patients 2*!mos - 3*!mos after treatment some secondary foci appeared around the periphery of the primary lesion. In general better effectiveness was obtained in patients with small lesion. After analyzing by RIDIT method, the therapeutic result in group B was significantly better than that in group C, and secondary foci around the original lesion were rarely seen at 3*!mos after treatment. In group C the collateral circulation was reestablished along the periphery of primary foci and the secondary foci appeared more frequently, and required to undergo several courses of treatment. 10Effects of aminoguanidine on nitric oxide production induced by inflammatory cytokines and endotoxin in cultured rat hepatocytes显示文摘AIM To study the effects of aminoguanidine(AG) and two L-arginine analogues Nω-nitro-L-arginine methyl ester (L-NAME) and Nω-nitro-L-arginine (L-NNA) on nitric oxide (NO) productioninduced by cytokines (TNF-α, IL-11β, and IFN-γ)and bacterial lipopolysaccharide (LPS) mixture(CM) in the cultured rat hepatocytes, andexamine their mechanisms action.METHODS Rat hepatocytes were incubatedwith AG, L-NAME, L-NNA, Actinomycin D (ActD)and dexamethasene in a medium containing CM(LPS plus TNF-α, IL-1β, and IFN-γ) for 24 h. NOproduction in the cultured supernatant wasmeasured with the Griese reaction. IntracellularcGMP level was detected with radioimmunoasey.RESULTS NO production was markedlyblocked by AG and L-NAME in a dose-dependentmanner under inflammatory stimuli conditiontriggered by CM in vitro. The rate of themaximum inhibitory effects of L-NAME (38.9%)was less potent than that obtained with AG(53.7%, P<0.05). There was no significantdifference between the inhibitory effects of AGand two L-arginine analogues on intracellularcGMP accumulation in rat cultured hepatocytes.Non-specific NOS expression inhibitordexamethasone ( DEX ) and iNOS mRNAtranscriptional inhibitor ActD also significantlyinhibited CM-induced NO production. 11Effects of Short-Term Forest Bathing on Human Health in a Broad-Leaved Evergreen Forest in Zhejiang Province,China显示文摘Objective To investigate the effects of short-term forest bathing on human health.Methods Twenty healthy male university students participated as subjects and were randomly divided into two groups of 10.One group was sent on a two-night trip to a broad-leaved evergreen forest,and the other was sent to a city area.Serum cytokine levels reflecting inflammatory and stress response,indicators reflecting oxidative stress,the distribution of leukocyte subsets,and plasma endothelin-1 (ET-1) concentrations were measured before and after the experiment to evaluate the positive health effects of forest environments.A profile of mood states (POMS) evaluation was used to assess changes in mood states.Results No significant differences in the baseline values of the indicators were observed between the two groups before the experiment.Subjects exposed to the forest environment showed reduced oxidative stress and pro-inflammatory level,as evidenced by decreased malondialdehyde,interleukin-6,and tumor necrosis factor α levels compared with the urban group.Serum cortisol levels were also lower than in the urban group.Notably,the concentration of plasma ET-1 was much lower in subjects exposed to the forest environment.The POMS evaluation showed that after exposure to the forest environment,subjects had lower scores in the negative subscales,and the score for vigor was increased.Conclusion Forest bathing is beneficial to human health,perhaps through preventive effects related to several pathological factors.MAO Gen Xiang LAN Xiao Guang CAO Yong Bao CHEN Zhuo Mei HE Zhi Huat LV Yuan Dong WANG Ya Zhen HU Xi Lian WANG Guo Fu YAN Jing 2012Biomedical and Environmental Sciences2012,25,3:20

13Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. 14Movement and strain conditions of active blocks in the Chinese mainland显示文摘The definition of active block is given from the angles of crustal deformation and strain. The movement and strain parameters of active blocks are estimated according to the unified velocity field composed of the velocities at 1598 GPS stations obtained from GPS measurements carried out in the past years in the Chinese mainland and the surrounding areas. The movement and strain conditions of the blocks are analyzed. The active blocks in the Chinese mainland have a consistent E-trending movement component, but its N and S components are not consistent. The blocks in the western part have a consistent N-trending movement and the blocks in the eastern part have a consistent S-trending movement. In the area to the east of 90°E, that is the area from Himalayas block towards NE, the movement direction of the blocks rotates clockwisely and the movement rates of the blocks are different. Generally, the movement rate is large in the west and south and small in the east and north with a difference of 3 to 4 times between the rates in the west and east. The distributions of principal compressive strain directions of the blocks are also different. The principal strain of the blocks located to the west of 90oE is basically in the SN direction, the principal compressive strain of the blocks in the northeastern part of Qingzang plateau is roughly in the NE direction and the direction of principal compressive strain of the blocks in the southeastern part of Qingzang plateau rounds clockwisely the east end of Himalayas structure. In addition, the principal strain and shear strain rates of the blocks are also different. The Himalayas and Tianshan blocks have the largest principal compressive strain and the maximum shear strain rate. Then, Lhasa, Qiangtang, Southwest Yunnan (SW Yunnan), Qilian and Sichuan-Yunan (Chuan-Dian) blocks followed. The strain rate of the blocks in the eastern part is smaller. The estimation based on the stain condition indicates that Himalayas block is still the area with the most intensive tectonic activity and it shortens in the NS direction at the rate of 15.2±1.5 mm/a. Tianshan block ranks the second and it shortens in the NS direction at the rate of 10.1±0.9 mm/a. At present, the two blocks are still uprising. It can be seen from superficial strain that the Chinese mainland is predominated by superficial expansion. Almost the total area in the eastern part of the Chinese mainland is expanded, while in the western part, the superficial compression and expansion are alternatively distributed from the south to the north. In the Chinese mainland, most EW-trending or proximate EW-trending faults have the left-lateral or left-lateral strike-slip relative movements along both sides, and most NS-trending faults have the right-lateral or right-lateral strike-slip relative movements along both sides. According to the data from GPS measurements the left-lateral strike-slip rate is 4.8±1.3 mm/a in the central part of Altun fault and 9.8±2.2 mm/a on Xianshuihe fault. The movement of the fault along the block boundary has provided the condition for block movement, so the movements of the block and its boundary are consistent, but the movement levels of the blocks are different. The statistic results indicate that the relative movement between most blocks is quite significant, which proves that active blocks exist. Himalayas, Tianshan, Qiangtang and SW Yunnan blocks have the most intensive movement; China-Mongolia, China-Korea (China-Korea), Alxa and South China blocks are rather stable. The mutual action of India, Pacific and Philippine Sea plates versus Eurasia plate is the principal driving force to the block movement in the Chinese mainland. Under the NNE-trending intensive press from India plate, the crustal matter of Qingzang plateau moves to the NNE and NE directions, then is hindered by the blocks located in the northern, northeastern and eastern parts. 19Grape Seed Proanthocyanidin Extract Alleviates Arsenic-induced Oxidative Reproductive Toxicity in Male Mice显示文摘Objective To determine the ability of grape seed proanthocyanidin extract(GSPE) in alleviating arsenic-induced reproductive toxicity. Methods Sixty male Kunming mice received the following treatments by gavage: normal saline solution(control); arsenic trioxide(ATO; 4 mg/kg); GSPE(400 mg/kg); ATO+GSPE(100 mg/kg); ATO+GSPE(200 mg/kg) and ATO+GSPE(400 mg/kg). Thereafter, the mice were sacrificed and weighed, and the testis was examined for pathological changes. Nuclear factor(erythroid-derived 2)-like 2(Nrf2), heme oxygenase 1(HO1), glutathione S-transferase(GST), NAD(P)H dehydrogenase, and quinone 1(NQO1) expression in the testis was detected by real-time PCR. Superoxide dismutase(SOD), glutathione(GSH), total antioxidative capability(T-AOC), malondialdehyde(MDA), 8-hydroxydeoxyguanosine(8-OHd G), and reproductive indexes were analyzed. Results ATO-treated mice showed a significantly decreased sperm count and testis somatic index and activity levels of SOD, GSH, and T-AOC than control group. Compared to the ATO-treated group, ATO +GSPE group showed recovery of the measured parameters. Mice treated with ATO+high-dose GSPE showed the highest level of m RNA expression of Nrf2, HO, NQO1, and GST.
