1Inhibition of SARS-CoV-2 (previously 2019-nCoV) infection by a highly potent pan-coronavirus fusion inhibitor targeting its spike protein that harbors a high capacity to mediate membrane fusion显示文摘The recent outbreak of coronavirus disease(COVID-19)caused by SARS-CoV-2 infection in Wuhan,China has posed a serious threat to global public health.To develop specific anti-coronavirus therapeutics and prophylactics,the molecular mechanism that underlies viral infection must first be defined.Therefore,we herein established a SARS-CoV-2 spike(S)protein-mediated cell-cell fusion assay and found that SARS-CoV-2 showed a superior plasma membrane fusion capacity compared to that of SARS-CoV.We solved the X-ray crystal structure of six-helical bundle(6-HB)core of the HR1 and HR2 domains in the SARS-CoV-2 S protein S2 subunit revealing that several mutated amino acid residues in the HR1 domain may be associated with enhanced interactions with the HR2 domain.We previously developed a pan-coronavirus fusion inhibitor,EK1,which targeted the HR!domain and could inhibit infection by divergent human coronaviruses tested,including SARS-CoV and MERS-CoV.Here we generated a series of lipopeptides derived from EK1 and found that EK1C4 was the most potent fusion inhibitor against SARS-CoV-2 S protein-mediated membrane fusion and pseudovirus infection with IC50s of 1.3 and 15.8 nM,about 241-and 149-fold more potent than the original EK1 peptide,respectively.EK1C4 was also highly effective against membrane fusion and infection of other human coronavirus pseudoviruses tested,including SARS-CoV and MERS-CoV,as well as SARSr-CoVs,and potently inhibited the replication of 5 live human coronaviruses examined,including SARS-CoV-2.Intranasal application of EK1C4 before or after challenge with HCoV-OC43 protected mice from infection,suggesting that EK1C4 could be used for prevention and treatment of infection by the currently circulating SARS-CoV-2 and other emerging SARSr-CoVs.Shuai Xia Meiqin Liu Chao Wang Wei Xu Qiaoshuai Lan Siliang Feng Feifei Qi Linlin Bao Lanying Du Shuwen Liu Chuan Qin Fei Sun Zhengli Shi Yun Zhu Shibo Jiang Lu Lu 2020Cell Research2020,30,4:81
2A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 3Fast-track rehabilitation program vs conventional care after colorectal resection:A randomized clinical trial显示文摘AIM:To compare the fast-track rehabilitation program and conventional care for patients after resection of colorectal cancer.METHODS:One hundred and six consecutive patients who underwent fast-track rehabilitation program were encouraged to have early oral feeding and movement for early discharge,while 104 consecutive patients underwent conventional care after resection of colorectal cancer.Their gastrointestinal functions,postoperative complications and hospital stay time were recorded.RESULTS:The restoration time of gastrointestinal functions in the patients was significantly faster after fasttrack rehabilitation program than after conventional care(2.1 d vs 3.2 d,P < 0.01).The percentage of patients who developed complications was significantly lower 30 d after fast-track rehabilitation program than after conventional care(13.2% vs 26.9%,P < 0.05).Also,the percentage of patients who had general complications was significantly lower 30 d after fast-track rehabilitation program than after conventional care(6.6% vs 15.4%,P < 0.05).The postoperative hospital stay time of the patients was shorter after fast-track rehabilitation program than after conventional care(5 d vs 7 d,P < 0.01).No significant difference was observed in the readmission rate 30 d after fast-track rehabilitation program and conventional care(3.8% vs 8.7%).CONCLUSION:The fast-track rehabilitation program can significantly decrease the complications and shorten the time of postoperative hospital stay of patients after resection colorectal cancer.Gang Wang Zhi-Wei Jiang Jing Xu Jian-Feng Gong Yang Bao Li-Fei Xie Jie-Shou Li 2011World Journal of Gastroenterology2011,17,5:43
4The State of the Art of Data Science and Engineering in Structural Health Monitoring显示文摘Structural health monitoring (SHM) is a multi-discipline field that involves the automatic sensing of structural loads and response by means of a large number of sensors and instruments, followed by a diagnosis of the structural health based on the collected data. Because an SHM system implemented into a structure automatically senses, evaluates, and warns about structural conditions in real time, massive data are a significant feature of SHM. The techniques related to massive data are referred to as data science and engineering, and include acquisition techniques, transition techniques, management techniques, and processing and mining algorithms for massive data. This paper provides a brief review of the state of the art of data science and engineering in SHM as investigated by these authors, and covers the compressive sampling-based data-acquisition algorithm, the anomaly data diagnosis approach using a deep learning algorithm, crack identification approaches using computer vision techniques, and condition assessment approaches for bridges using machine learning algorithms. 5Fibroblast growth factor-4 and hepatocyte growth factor induce differentiation of human umbilical cord blood-derived mesenchymal stem cells into hepatocytes显示文摘AIM: To investigate the differentiation of human umbilical cord blood (HUCB)-derived mesenchymal stem cells (MSCs) into hepatocytes by induction of fibroblast growth factor-4 (FGF-4) and hepatocyte growth factor (HGF), and to find a new source of cell types for therapies of hepatic diseases.METHODS: vSCs were isolated by combining gradient density centrifugation with plastic adherence. When HUCB-derived MSCs reached 70% confluence, they were cultured in Iscove modified Dulbecco medium (IMDM) supplemented with 10 mL/L FBS, 20 ng/mL HGF and 10 ng/mL FGF-4. The medium was changed every 4 d and stored for albumin, alpha-fetoprotein (AFP) and urea assay. Expression of CK-18 was detected by immunocytochemistry. Glycogen storage in hepatocytes was determined by PAS staining.RESULTS: By combining gradient density centrifugation with plastic adherence, we could isolate MSCs from 25.6% of human umbilical cord blood. When MSCs were cultured with FGF-4 and HGF, approximately 63.6% of cells became small, round and epithelioid on d 28 by morphology. Compared with the control, the level of AFP increased significantly from d 12 to 18.20±1.16 μg/L (t = 2.884, P<0.05) in MSCs cultured with FGF-4 and HGF, and was higher (54.28±3.11 μg/L) on d 28 (t = 13.493, P<0.01). Albumin increased significantly on d 16 (t = 6.68, P<0.01) to 1.02±0.15 μg/mL, and to 3.63±0.30 μg/mL on d 28 (t = 11.748, P<0.01). Urea(4.72±1.03 μmol/L) was detected on d 20 (t = 4.272,P<0.01), and continued to increase to 10.28±1.06 μmol/L on d 28 (t = 9.276, P<0.01). Cells expressed CK-18 on d 16. Glycogen storage was observed on d 24. CONCLUSION: HUCB-derived MSCs can differentiate into hepatocytes by induction of FGF-4 and HGF. 6New vacuum solar telescope and observations with high resolution显示文摘The New Vacuum Solar Telescope(NVST) is a one meter vacuum solar telescope that aims to observe fine structures on the Sun. The main goals of NVST are high resolution imaging and spectral observations, including measurements of the solar magnetic field. NVST is the primary ground-based facility used by the Chinese solar research community in this solar cycle. It is located by Fuxian Lake in southwest China, where the seeing is good enough to perform high resolution observations. We first introduce the general conditions at the Fuxian Solar Observatory and the primary science cases of NVST. Then, the basic structures of this telescope and instruments are described in detail. 7Anisotropic coding metamaterials and their powerful manipulation of differently polarized terahertz waves显示文摘Metamaterials based on effective media can be used to produce a number of unusual physical properties(for example,negative refraction and invisibility cloaking)because they can be tailored with effective medium parameters that do not occur in nature.Recently,the use of coding metamaterials has been suggested for the control of electromagnetic waves through the design of coding sequences using digital elements‘0’and‘1,'which possess opposite phase responses.Here we propose the concept of an anisotropic coding metamaterial in which the coding behaviors in different directions are dependent on the polarization status of the electromagnetic waves.We experimentally demonstrate an ultrathin and flexible polarization-controlled anisotropic coding metasurface that functions in the terahertz regime using specially designed coding elements.By encoding the elements with elaborately designed coding sequences(both 1-bit and 2-bit sequences),the x-and y-polarized waves can be anomalously reflected or independently diffused in three dimensions.The simulated far-field scattering patterns and near-field distributions are presented to illustrate the dual-functional performance of the encoded metasurface,and the results are consistent with the measured results.We further demonstrate the ability of the anisotropic coding metasurfaces to generate a beam splitter and realize simultaneous anomalous reflections and polarization conversions,thus providing powerful control of differently polarized electromagnetic waves.The proposed method enables versatile beam behaviors under orthogonal polarizations using a single metasurface and has the potential for use in the development of interesting terahertz devices.Shuo Liu Tie Jun Cui Quan Xu Di Bao Liangliang Du Xiang Wan Wen Xuan Tang Chunmei Ouyang Xiao Yang Zhou Hao Yuan Hui Feng Ma Wei Xiang Jiang Jiaguang Han Weili Zhang Qiang Cheng 2016Light(Science & Applications)2016,5,1:36

9New chaotic system and its hyperchaos generation显示文摘To seek for lower-dimensional chaotic systems that have complex topological attractor structure with simple algebraic system structure, a new chaotic system of three-dimensional autonomous ordinary differential equations is presented. The new system has simple algebraic structure, and can display a 2-scroll attractor with complex topological structure, which is different from the Lorenz's, Chen's and Lu¨'s attractors. By introducing a linear state feedback controller, the system can be controlled to generate a hyperchaotic attractor. 11Research on preservation and enrichment mechanisms of organic matter in muddy sediment and mudstone显示文摘Using multidiscipline methodologies, the differences in preservation and enrichment mechanisms of organic matter (OM) in muddy sediment and mudstone are investigated. In clay fractions, concentra- tions of TOC and chloroform bitumen “A” are significantly higher than those in coarser fractions. This indicates that clay minerals (CM) play an important role in enriching OM. The content of chloroform bitumen “A” increases obviously in the clay fraction, which reveals that dissolvable OM is the main composition of coalesce with clay minerals. Furthermore, TG and DTA data show that OM enrichment mechanisms and preservation forms have multiplicity. Several exothermic peaks in the DTA curves demonstrate that muddy sediment and mudstone contain a number of bioclasts and amorphous OM besides dissolvable OM. Through analyzing with XRD and DTA after mudstone samples were pretreated, the conclusions can be arrived at. Firstly, CM interlayer space of XRD curves and exothermic peaks of DTA curves both change as temperature increases. Secondly, the changes of CM interlayer space and exothermic peaks are concordant and stable around 350℃. All these are the features that OM enters CM interlayers to form stable organo-clay complexes. Therefore, the combination format of OM with CM is not only surface adsorption, partial OM enters CM interlayers to form stable organo-clay complexes. 12Adakite-type sodium-rich rocks in Awulale Mountain of west Tianshan:Significance for the vertical growth of continental crust显示文摘The sodium-rich dacites and albite porphyries of Permian in the Awulale Mountain of west Tianshan have unique chemical and trace element signatures identical to adakite. These intermediate-acidic igneous rocks are characterized by high Na2O, Al2O3 and Sr contents and high Sr/Y and La/Y ratios (>40 and >20, respectively), and low Y and Yb contents, and strong depletion in HREE, and positive Eu anomaly. The (143Nd/144Nd)i is in the range from0.51236 to 0.51248 and the εNd(t) is positive value (+0.79-+3.11); the (87Sr/86Sr)i is in the range from 0.7052 to 0.7054. These Nd and Sr isotopic composition features indicate that the source rocks of these adakite-type rocks are from a weakly depleted mantle, or a depleted mantle, but was contaminated by the crustal materials. 13DNA cytosine methylation in plant development显示文摘Cytosine bases of the nuclear genome in higher plants are often extensively methylated.Cytosine methylation has been implicated in the silencing of both transposable elements (TEs) and endogenous genes,and loss of methylation may have severe functional consequences.The recent methylation profiling of the entire Arabidopsis genome has provided novel insights into the extent and pattern of cytosine methylation and its relationships with gene activity.In addition,the fresh studies also revealed the more dynamic nature of this epigenetic modification across plant development than previously believed.Cytosine methylation of gene promoter regions usually inhibits transcription,but methylation in coding regions (gene-body methylation) does not generally affect gene expression.Active demethylation (though probably act synergistically with passive loss of methylation) of promoters by the 5-methyl cytosine DNA glycosylase or DEMETER (DME) is required for the uni-parental expression of imprinting genes in endosperm,which is essential for seed viability.The opinion that cytosine methylation is indispensible for normal plant development has been reinforced by using single or combinations of diverse loss-of-function mutants for DNA methyltransferases,DNA glycosylases,components involved in siRNA biogenesis and chromatin remodeling factors.Patterns of cytosine methylation in plants are usually faithfully maintained across organismal generations by the concerted action of epigenetic inheritance and progressive correction of strayed patterns.However,some variant methylation patterns may escape from being corrected and hence produce novel epialleles in the affected somatic cells.This,coupled with the unique property of plants to produce germline cells late during development,may enable the newly acquired epialleles to be inherited to future generations,which if visible to selection may contribute to adaptation and evolution.Meishan Zhang Josphert N.Kimatu Kezhang Xu Bao Liu 2010Journal of Genetics and Genomics2010,37,1:26

15Full-colour nanoprint-hologram synchronous metasurface with arbitrary hue-saturationbrightness control显示文摘The colour gamut,a two-dimensional(2D)colour space primarily comprising hue and saturation(HS),lays the most important foundation for the colour display and printing industries.Recently,the metasurface has been considered a promising paradigm for nanoprinting and holographic imaging,demonstrating a subwavelength image resolution,a flat profile,high durability,and multi-functionalities.Much effort has been devoted to broaden the 2D HS plane,also known as the CIE map.However,the brightness(B),as the carrier of chiaroscuro information,has long been neglected in metasurface-based nanoprinting or holograms due to the challenge in realising arbitrary and simultaneous control of full-colour HSB tuning in a passive device.Here,we report a dielectric metasurface made of crystal silicon nanoblocks,which achieves not only tailorable coverage of the primary colours red,green and blue(RGB)but also intensity control of the individual colours.The colour gamut is hence extruded from the 2D CIE to a complete 3D HSB space.Moreover,thanks to the independent control of the RGB intensity and phase,we further show that a singlelayer silicon metasurface could simultaneously exhibit arbitrary HSB colour nanoprinting and a full-colour hologram image.Our findings open up possibilities for high-resolution and high-fidelity optical security devices as well as advanced cryptographic approaches.Yanjun Bao Ying Yu Haofei Xu Chao Guo Juntao Li Shang Sun Zhang-Kai Zhou Cheng-Wei Qiu Xue-Hua Wang 2019Light(Science & Applications)2019,8,1:25
16Factors Affecting Sensory Quality of Cooked japonica Rice显示文摘The sensory quality of cooked rice is an important factor in determining its market price, as well as consumer acceptance and breeding efforts aimed at improvement of rice grain quality. In this study,the sensory quality and physicochemical properties of three japonica rice varieties harvested in two different growing locations(Xiangshui and Hangzhou of China) were compared to determine the most important factors affecting the sensory quality. All the three varieties had higher scores for overall sensory quality in Xiangshui than in Hangzhou, indicating that the growing location is a key factor in determining the sensory quality of cooked japonica rice. In addition to growing location, variety(genotype) also had an important effect. Longdao 18 scored the highest for overall sensory quality in the two locations, whereas Longdao 30 had the lowest score in Xiangshui, and both Longdao 20 and Longdao 30 had the lowest scores in Hangzhou. Many physicochemical properties, such as apparent amylose content, protein content, thermal properties and free amino acid contents, showed significant differences between the two locations. Correlation analysis showed that apparent amylose content and protein content had contrasting effects on all the sensory attributes. The overall sensory quality was negatively correlated with protein content(r =-0.89, P < 0.01) and positively correlated with gel hardness(r = 0.91, P < 0.01),indicating that the protein content and hardness are important physicochemical properties for predicting the sensory quality of japonica rice. 18Observation of the effect of targeted therapy of 64-slice spiral CT combined with cryoablation for liver cancer显示文摘AIM To observe the effect of targeted therapy with 64-slice spiral computed tomography(CT) combined with cryoablation for liver cancer.METHODS A total of 124 patients(142 tumors) were enrolled into this study. According to the use of dual-slice spiral CT or 64-slice spiral CT as a guide technology, patients were divided into two groups: dual-slice group(n = 56, 65 tumors) and 64-slice group(n = 8, 77 tumors). All patients were accepted and received targetedtherapy by an argon-helium superconducting surgery system. The guided scan times of the two groups was recorded and compared. In the two groups, the lesion ice coverage in diameter of ≥ 3 cm and < 3 cm were recorded, and freezing effective rate was compared. Hepatic perfusion values [hepatic artery perfusion(HAp), portal vein perfusion(p Vp), and the hepatic arterial perfusion index(HAp I)] of tumor tissues, adjacent tissues and normal liver tissues at preoperative and postoperative four weeks in the two groups were compared. Local tumor changes were recorded and efficiency was compared at four weeks post-operation. Adverse events were recorded and compared between the two groups, including fever, pain, frostbite, nausea, vomiting, pleural effusion and abdominal bleeding.RESULTS Guided scan times in the dual-slice group was longer than that in the 64-slice group(t = 11.445, p = 0.000). The freezing effective rate for tumors < 3 cm in diameter in the dual-slice group(81.58%) was lower than that in the 64-slice group(92.86%)(χ2 = 5.707, p = 0.017). The HAp and HAp I of tumor tissues were lower at four weeks post-treatment than at pretreatment in both groups(all p < 0.05), and those in the 64-slice group were lower than that in the dualslice group(all p < 0.05). HAp and p Vp were lower and HAp I was higher in tumor adjacent tissues at post-treatment than at pre-treatment(all p < 0.05). Furthermore, the treatment effect and therapeutic efficacy in the dual-slice group were lower than the 64-slice group at four weeks post-treatment(all p < 0.05). Moreover, pleural effusion and intraperitoneal hemorrhage occurred in patients in the dual-slice group, while no complications occurred in the 64-slice group(all p < 0.05).
