1Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 2A bioaugmentation failure caused by phage infection and weak biofilm formation ability显示文摘Two biological aerated filters (BAF) were setup for ammonia removal treatment of the circulation water in a marine aquaculture. One of the BAFs was bioaugmented with a heterotrophic nitrifying bacterium, Lutimonas sp. H10, where the ammonia removal was not improved and the massive inoculation was even followed by a nitrification breakdown from day 9 to 18. The nitrification was remained stable in control BAF operated under the same conditions. Fluorescent in situ hybridization (FISH) with rRNA-targeted probes and cultivable method revealed that Lutimonas sp. H10 almost disappeared from the bioaugomented BAF within 3 d, and this was mainly due to the infection of a specific phage as revealed by flask experiment, plaque assay and transmission electron observation. Analyses of 16S rRNA gene libraries showed that bacterial groups from two reactors evolved differently and an overgrowth of protozoa was observed in the bioaugmented BAF. Therefore, phage infection and poor biofilm forming ability of the inoculated strain are the main reasons for bioaugmentation failure. 3Fast integration and accumulation of beneficial breeding alleles through an AB–NAMIC strategy in wheat显示文摘Wheat(Triticum aestivum)is among the most important staple crops for safeguarding the food security of the growing world population.To bridge the gap between genebank diversity and breeding programs,we developed an advanced backcross-nested association mapping plus inter-crossed population(AB-NAMIC)by crossing three popular wheat cultivars as recurrent founders to 20 germplasm lines from a mini core collection.Selective backcrossing combined with selection against undesirable traits and extensive crossing within and between sub-populations created new opportunities to detect unknown genes and increase the frequency of beneficial alleles in the AB-NAMIC population.We performed phenotyping of 590 AB-NAMIC lines and a natural panel of 476 cultivars for six consecutive growing seasons and genotyped these 1066 lines with a 660K SNP array.Genome-wide association studies of both panels for plant development and yield traits demonstrated improved power to detect rare alleles and loci with medium genetic effects in AB-NAMIC.Notably,genome-wide association studies in AB-NAMIC detected the candidate gene TaSWEET6-7B(TraesCS7B03G1216700),which has high homology to the rice SWEET6b gene and exerts strong effects on adaptation and yield traits.The commercial release of two derived AB-NAMIC lines attests to its direct applicability in wheat improvement.Valuable information on genome-wide association studymapping,candidate genes,and their haplotypes for breeding traits are available through WheatGAB.Our research provides an excellent framework for fast-tracking exploration and accumulation of beneficial alleles stored in genebanks.Chengzhi Jiao Chenyang Hao Tian Li Abhishek Bohra Lanfen Wang Jian Hou Hongxia Liu Hong Liu Jing Zhao Yamei Wang Yunchuan Liu Zhiwei Wang Xin Jing Xiue Wang Rajeev K.Varshney Junjie Fu Xueyong Zhang 2023Plant Communications2023,4,3:3
4A Peridynamic Model for Analyzing Fracture Behavior of Functionally Graded Materials Used as an Interlayer显示文摘Functionally graded materials(FGMs)can be used els an interlayer.The interlayer connects two homogeneous materials and slows mutations in material properties,owing to the variability of parameters and microstructure.In this numerical simulation,a bond-based peridynamic(PD)model was used to perform dynamic fracture analysis of a continuous beam and a sandwiched structure with an FGM interlayer.Some simulation results of two convergence studies,which include grid refinement(m-convergence)and the reducing radius of PD horizon(^-convergence),were discussed.Under a four-point bending load,the propagation behaviors of single pre-crack in the continuous beam with the FGM interlayer at different locations are simulated to compare with the experimental results.For the sandwiched structure,the problems of a single crack and double cracks were considered.The numerical results show that the crack can deflect toward areas where the energy release rate is low.Additionally,the crack on the weak side for symmetric cracks only has fracture behavior,and if the double cracks are asymmetrical,all these cracks have fracture behaviors and propagation of the initial crack in the stiff region has a higher sensitivity than in the compliant region.Zhanqi Cheng Zhengqiang Fu Yu Zhang Hongxia Wu 2020Acta Mechanica Solida Sinica2020,33,6:1
5Release of myocardial noradrenaline during acute hibernation in the isolated rat heart显示文摘Objective: To explore the release of myocardial noradrenaline during acute hibernation. Methods: The hearts were gained from rats and set up as modified Langendorf preparations beating isometrically. They were perfused with modified Krebs-Henseleit buffer under controlled pressure. Mechanical measurements and coronary effluent were recorded simultaneously at 30 min intervals for 150 min. Lactate dehydrogenase in coronary effluent was assayed at the beginning, 60 min and 120 min low-flow ischemia. Noradrenaline in coronary effluent was determined at the beginning of low-flow and 120 min of low-flow ischemia and also in control, during hibernation and after 30 min reperfusion during stimulation, myocardial noradrenaline response on tyramine was investigated in absence or presence of desipramine after 30 min reperfusion. Results: In the control, there was nosignificant chant in noradrenaline overflow during 120 min perfusion; In the acute myocardial hibernation group, there was also nosignificant difference in noradrenaline overflow between the beginning and 120 min low-flow ischemia. The electrical field stimulation-induced overflow of noradrenaline during hibernation myocardium was significantly less than preischemia or after reperfusion, but there was nosignificant difference between preischemia and reperfusion group. Tyramine induced significant noradrenaline release in absence of desipramine after 30 min reperfusion, but this increase in noradrenaline release had nosignificant in the presence of desipramine. These studies indicated that there was not significant spontaneous noadrenaline overflow during acute myocardial hibernation in isolated rat hearts, the stimulation-induced noradrenaline overflow decreased during hibernation and restored to the level of preischemia after reperfusion, myocardial noradrenaline response to tyramine remained after 30 min reperfusion. 14Accurate design of spatially separated double active site in Bi_(4)NbO_(8)Cl single crystal to promote Z-Scheme photocatalytic overall water splitting显示文摘The efficiency of photocatalytic overall water splitting was mainly limited by the slow reaction kinetics of water oxidation.How to design effective surface active site to overcome the slow water oxidation reaction was a major challenge.Here,we propose a strategy to accelerate surface water oxidation through the fabrication spatially separated double active sites.FeCoPi/Bi_(4)NbO_(8)Cl-OVs photocatalyst with spatially separated double active site was prepared by hydrogen reduction photoanode deposition method.Due to the high matching of the spatial loading positions of FeCoPi and OVs with the photogenerated charge distribution of Bi_(4)NbO_(8)Cl and corresponding reaction mechanisms of substrate,the FeCoPi and OVs on the(001)and(010)crystal planes of Bi_(4)NbO_(8)Cl photocatalyst provided surface active site for water oxidation reaction and electron shuttle reaction(Fe^(3+)/Fe^(2+)),respectively.Under visible light irradiation,the evolution O_(2)rate of FeCoPi/Bi_(4)NbO_(8)Cl OVs was 16.8μmol h^(-1),as 32.9 times as Bi_(4)NbO_(8)Cl.Furthermore,a hydrogen evolution co-catalyst PtRu@Cr_(2)O_(3)was prepared by sequential photodeposition method.Due to the introduction of Ru,the Schottky barrier between PbTiO_(3)and Pt was effectively reduced,which promoted the transfer of photogenerated electrons to PtRu@Cr_(2)O_(3)thermodynamically,the evolution H_(2)rate on PtRu@Cr_(2)O_(3)/PbTiO_(3)increased to 664.8 times.On based of the synchronous enhancement of the water oxidation performance on FeCoPi/Bi_(4)NbO_(8)Cl-OVs and water reduction performance on PtRu@Cr_(2)O_(3)/PbTiO_(3),a novel Z-Scheme photocatalytic overall water splitting system(FeCoPi/Bi_(4)NbO_(8)Cl-OVs)mediated by Fe^(3+)/Fe^(2+)had successfully constructed.Under visible light irradiation,the evolution rates of H_(2)and O_(2)were 2.5 and 1.3μmol h^(-1),respectively.This work can provide some reference for the design of active site and the controllable synthesis of OVs spatial position.On the other hand,the hydrogen evolution co catalyst(PtRu@Cr_(2)O_(3))and the co catalyst FeCoPi for oxygen evolution contributed to the construction of an overall water splitting system.Kailong Gao Hongxia Guo Yanan Hu Hongbin He Mowen Li Xiaoming Gao Feng Fu 2023Journal of Energy Chemistry2023,,12:0
15Immediate and long-term outcomes after treat-all among people living with HIV in China:an interrupted time series analysis显示文摘Background In 2003,China implemented free antiretroviral therapy(ART)for people living with HIV(PLHIV),establishing an eligibility threshold of CD4<200 cells/μl.Subsequently,the entry criteria were revised in 2012(eligibility threshold:CD4≤350 cells/μl),2014(CD4≤500 cells/μl),and 2016(treat-all).However,the impact of treat-all policy on HIV care and treatment indicators in China is unknown.We aimed to elucidate the immediate and long-term impact of the implementation of treat-all policy in China.Methods Anonymized programmatic data on ART initiation and collection in PLHIV who newly started ART were retrieved between 1 January 2015 and 31 December 2019,from two provincial and municipal Centers for Disease Control and Prevention and ten major infectious disease hospitals specialized in HIV care in China.We used Poisson and quasi-Poisson segmented regression models to estimate the immediate and long-term impact of treat-all on three key indicators:monthly proportion of 30-day ART initiation,mean CD4 counts(cells/μl)at ART initiation,and mean estimated time from infection to diagnosis(year).We built separate models according to gender,age,route of transmission and region.Results Monthly data on ART initiation and collection were available for 75,516 individuals[gender:83.8%males;age:median 39 years,interquartile range(IQR):28-53;region:18.5%Northern China,10.9%Northeastern China,17.5%Southern China,49.2%Southwestern China].In the first month of treat-all,compared with the contemporaneous counterfactual,there was a significant increase in proportion of 30-day ART initiation[+12.6%,incidence rate ratio(IRR)=1.126,95%CI:1.033-1.229;P=0.007]and mean estimated time from infection to diagnosis(+7.0%,IRR=1.070,95%CI:1.021-1.120;P=0.004),while there was no significant change in mean CD4 at ART initiation(IRR=0.990,95%CI:0.956-1.026;P=0.585).By December 2019,the three outcomes were not significantly different from expected levels.In the stratified analysis,compared with the contemporaneous counterfactual,mean CD4 at ART initiation showed significant increases in Northern China(+3.3%,IRR=1.033,95%CI:1.001-1.065;P=0.041)and Northeastern China(+8.0%,IRR=1.080,95%CI:1.003-1.164;P=0.042)in the first month of treat-all;mean estimated time from infection to diagnosis showed significant increases in male(+5.6%,IRR=1.056,95%CI:1.010-1.104;P=0.016),female(+14.8%,IRR=1.148,95%CI:1.062-1.240;P<0.001),aged 26-35(+5.3%,IRR=1.053,95%CI:1.001-1.109;P=0.048)and>50(+7.8%,IRR=1.078,95%CI:1.000-1.161;P=0.046),heterosexual transmission(+12.4%,IRR=1.124,95%CI:1.042-1.213;P=0.002)and Southwestern China(+12.9%,IRR=1.129,95%CI:1.055-1.208;P<0.001)in the first month of treat-all.Conclusions The implementation of treat-all policy in China was associated with a positive effect on HIV care and treatment outcomes.To advance the work of rapid ART,efforts should be made to streamline the testing and ART initiation process,provide comprehensive support services,and address the issue of uneven distribution of medical resources.Xinsheng Wu Guohui Wu Ping Ma Rugang Wang Linghua Li Yinghui Sun Junjie Xu Yuwei Li Tong Zhang Quanmin Li Yuecheng Yang Lijing Wang Xiaoli Xin Ying Qiao Bingxue Fang Zhen Lu Xinyi Zhou Yuanyi Chen Qi Liu Gengfeng Fu Hongxia Wei Xiaojie Huang Bin Su Hui Wang Huachun Zou 2023Infectious Diseases of Poverty2023,12,4:0
16Highly selective separation of propylene/propane mixture on cost-effectively four-carbon linkers based metal-organic frameworks显示文摘The separation of propylene and propane is an important but challenging process,primarily achieved through energy-intensive distillation technology in the petrochemical industry.Here,we reported two natural C4linkers based metal–organic frameworks(MIP-202 and MIP-203)for C_(3)H_(6)/C_(3)H_(8)separation.Adsorption isotherms and selectivity calculations were performed to study the adsorption performance for C_(3)H_(6)/C_(3)H_(8)separation.Results show that C_(3)H_(6)/C_(3)H_(8)uptake ratios(298 K,100 kPa)for MIP-202 and MIP-203 are 2.34 and 7.4,respectively.C_(3)H_(6)/C_(3)H_(8)uptake ratio(303 K,100 k Pa)for MIP-203 is up to50.0.The mechanism for enhanced separation performance of C_(3)H_(6)/C_(3)H_(8)on MIP-203 at higher temperature(303 K)was revealed by the in situ PXRD characterization.The adsorption selectivities of C_(3)H_(6)/C_(3)H_(8)on MIP-202 and MIP-203(298 K,100 k Pa)are 8.8 and 551.4,respectively.The mechanism for the preferential adsorption of C_(3)H_(6)over C_(3)H_(8)in MIP-202 and MIP-203 was revealed by the Monte Carlo simulation.The cost of organic ligands for MIP-202 and MIP-203 was lower than that of organic ligands for those top-performance MOFs.Our work sets a new benchmark for C_(3)H_(6)sorbents with high adsorption selectivities.Daofei Lv Junhao Xu Pingjun Zhou Shi Tu Feng Xu Jian Yan Hongxia Xi Zewei Liu Wenbing Yuan Qiang Fu Xin Chen Qibin Xia 2022Chinese Journal of Chemical Engineering2022,,11:0
17THE EFFECT OF IONIC STRENGTH ON THE UPTAKE OF TAURINE ON A STRONG-BASIC ANION EXCHANGE RESIN显示文摘Studied the effect of ionic strength on the uptake of taurine on a strong-basic anion exchange resin. The batch phase equilibrium experiments of taurine on the anion exchange resin D290 were conducted at different ionic strength, and then the amounts of uptake of taurine on the resin at different pH were determined. The ion exchange mechanisms of taurine on the anion exchange resin at different pH were discussed. 19INFLUENCE OF THE UPSTREAM RESERVOIRS ON THE ADJUSTMENT OF DOWNSTREAM ALLUVIAL CHANNEL显示文摘From the systematic analysis of field data of the existing reservoirs on the main stem of the Yellow River it has been found that different modes of operation of the reservoirs have different effects on the regulation of runoff and sediment load, and consequently, on the self-adjustment of the downstream channel. If a reservoir functions as water impoundment or flood detention, the sediment transport capacity will decrease because of the reduction of flood peaks. Espeially when large amount of sediment load are brought into the river by sediment-laden tributaries downstream of the reservoir, the deposition on the downstream channel will be getting serious, the conveyance capacity will decrease and the channel itself will degenerate. Under the operation of storing the clear flow and releasing the muddy the reservior capacity can be maintained for long-term use and the sediment transport capacity of the downstream channel can be increased. 20Rapid determination of tannin in Danshen and Guanxinning injections using UV spectrophotometry for quality control显示文摘A technique for the determination of tannin content in traditional Chinese medicine injections(TCMI)was developed based on ultraviolet(UV)spectroscopy.Chemometrics were used to construct a mathematical model of absorption spectrum and tannin reference content of Danshen and Guanxinning injections,and the model was veried and applied.The results showed that the established UV-based spectral partial least squares regression(PLS)tannin content model performed well with a correlation coefficient(r)of 0.952,root mean square error of calibration(RMSEC)of 0.476g/ml,root mean square error of validation(RMSEV)of 1.171g/ml,and root mean square error of prediction(RMSEP)of 0.465g/ml.Pattern recognition models using linear discriminant analysis(LDA)and k nearest neighbor(k-NN)classiers based on UV spectrum could successfully classify different types of injections and different manufacturers.The established method to measure tannin content based on UV spectroscopy is simple,rapid and reliable and provides technical support for quality control of tannin in Chinese medicine injections.Hongxia Huang Yuanyuan Lv Xiaoyi Sun Shuangshuang Fu Xuefang Lou Zengmei Liu 2018Journal of Innovative Optical Health Sciences2018,,6:0
