| 1 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 3 | Detection of Citrus yellow vein clearing virus by Quantitative Real-time RT-PCR显示文摘To develop a rapid and reliable detection method for Citrus yellow vein clearing virus(CYVCV), a quantitative real-time reverse transcriptionpolymerase chain reaction(q RT-PCR) system based on SYBR Green I was established by using a pair of specific primers designed from its conserved coat protein gene. The sensitivity, specificity, and applicability of the system were evaluated accordingly. The results showed that amplicons were produced from CYVCV isolates, whereas no amplicons from non-CYVCV citrus virus samples, including Citrus tristeza virus(CTV) and Citrus tatter leaf virus(CTLV), were obtained. The sensitivity of the q RT-PCR was 100-fold higher than that of conventional RT-PCR. An excellent linear correlation(R2= 0.999) was obtained from two standard curves of c RNA, and the amplification efficiency was 102%. | 4 | Preparation,Thermal Stability and Electrochemical Properties of LiODFB显示文摘Lithium oxalyldifluoroborate (LiODFB) was synthesized in dimethyl carbonate solvent and purified by the method of solvent-out crystallization. The structure characterization and thermal stability of LiODFB were performed by Fourier transform infrared (FTIR) spectrometry, nuclear magnetic resonance (NMR) spectrometry and thermogravimetric analyzer (TGA). LiODFB was exposed to 50% humid air at 25 C for different time, then dried at 80 C for 12 h, and the electrochemical properties of the cells using 1 mol/L dried LiODFB in ethylene carbonate + dimethyl carbonate + ethyl(methyl)carbonate were investigated. | 5 | Gut Microbiota Dysbiosis Strengthens Kupffer Cell-mediated Hepatitis B Virus Persistence through Inducing Endotoxemia in Mice显示文摘Background and Aims:Change of gut microbiota com-position is associated with the outcome of hepatitis B virus(HBV)infection,yet the related mechanisms are not fully characterized.The objective of this study was to investigate the immune mechanism associated with HBV persistence induced by gut microbiota dysbiosis.Methods:C57BL/6 mice were sterilized for gut-microbiota by using an antibi-otic(ABX)mixture protocol,and were monitored for their serum endotoxin(lipopolysaccharide[LPS])levels.An HBV-replicating mouse model was established by performing HBV-expressing plasmid pAAV/HBV1.2 hydrodynamic injec-tion(HDI)with or without LPS,and was monitored for se-rum hepatitis B surface antigen,hepatitis B e antigen,HBV DNA,and cytokine levels.Kupffer cells(KCs)were purified from antibiotic-treated mice and HBV-replicating mice and analyzed for IL-10 production and T cell suppression ability.Results:ABX treatment resulted in increased serum LPS levels in mice.The KCs separated from both ABX-treated and LPS-treated HBV-replicating mice showed significantly increased IL-10 production and enhanced ability to suppress IFN-γproduction of TCR-activated T cells than the KCs sep-arated from their counterpart controls.HDI of pAAV/HBV1.2 in combination with LPS in mice led to a delayed HBV clear-ance and early elevation of serum IL-10 levels compared to pAAV/HBV1.2 HDI alone.Moreover,IL-10 function blockade or KC depletion led to accelerated HBV clearance in LPS-treated HBV-replicating mice.Conclusions:Our results suggest that dysbiosis of the gut microbiota in mice leads to endotoxemia,which induces KC IL-10 production and strengthens KC-mediated T cell suppression,and thus fa-cilitates HBV persistence. | Wenqing Zhou Jinzhuo Luo Xiaohong Xie Shangqing Yang Dan Zhu Hongming Huang Dongliang Yang Jia Liu | 2022 | Journal of Clinical and Translational Hepatology2022,10,1: | 4 |
| 6 | Holocene Dune Mobility and Forcing Mechanisms at the Northern Margin of the East Asian Monsoon显示文摘Dune fields at the northern margin of the East Asian monsoon (EAM), are mosaics of mobile and vegetation-stabilized aeolian dunes. These sand dunes are highly sensitive to environmental change, thus the distribution and the timing of their development may provide important clues to past environmental dynamics. Due to the strong wind erosion and dune migration, long and continuous stratigraphic records are seldom preserved. Synthesizing a large body of events, ultimately producing a relatively complete and high-resolution record, may be a proper method to investigate the dune development history and climate change. In this study, we synthesized a large body of luminescence ages for aeolian deposits from the Mu Us, Otindag, Horqin dune fields at the northern margin of the EAM. The results show that these dune fields, as a whole experienced a most extensive mobility during the early Holocene, followed by a widespread shift toward limited mobility and soil development in the mid-Holocene, and widespread reactivation occurred during late Holocene. The dune developments are directly linked to the effective moisture change controlled by the EAM changes, which respond to the low latitude summer insolation variation. The increased subsidence at the margin contrary to the core EAM, the delay from the feedback of the soil-vegetation-air coupled system, the increased evaporation due to the high temperature all play partial role in the lag of the margin EAM effective moisture change to the low latitude summer insolation. The asynchronous end of the wetter mid-Holocene mainly responds to the southeastwardly shift of the precipitation belt, while the regional sensitivity, response speed and internal feedback also contributed. | 8 | Doping Effects on Electronic Conductivity and Electrochemical Performance of LiFePO_4显示文摘Olivine-structured pure LiFePO4 and doped Li(M, Fe)PO4 (M=La, Ce, Nd, Mn, Co, Ni) have been synthesized by a solvothermal method. X-ray diffraction and field emission scanning electron microscopy analyses indicate that the as-prepared LiFePO4 is well-crystallized nanopowders without any detectable impurity phases. The electronic conductivity of LiFePO4 is enhanced by around 1-3 orders by doping. It was found that doping alone is not suffcient for the high-rate performance of LiFePO4 and surface coating with such as carbon should be needed. The best dopant for LiFePO4 is Nd among those studied in the present work.
