2The apricot(Prunus armeniaca L.)genome elucidates Rosaceae evolution and beta-carotenoid synthesis显示文摘Apricots,scientifically known as Prunus armeniaca L,are drupes that resemble and are closely related to peaches or plums.As one of the top consumed fruits,apricots are widely grown worldwide except in Antarctica.A high-quality reference genome for apricot is still unavailable,which has become a handicap that has dramatically limited the elucidation of the associations of phenotypes with the genetic background,evolutionary diversity,and population diversity in apricot.DNA from P.armeniaca was used to generate a standard,size-selected library with an average DNA fragment size of~20 kb.The library was run on Sequel SMRT Cells,generating a total of 16.54 Gb of PacBio subreads(N50=13.55 kb).The high-quality P.armeniaca reference genome presented here was assembled using long-read single-molecule sequencing at approximately 70×coverage and 171×Illumina reads(40.46 Gb),combined with a genetic map for chromosome scaffolding.The assembled genome size was 221.9 Mb,with a contig NG50 size of 1.02 Mb.Scaffolds covering 92.88%of the assembled genome were anchored on eight chromosomes.Benchmarking Universal Single-Copy Orthologs analysis showed 98.0%complete genes.We predicted 30,436 protein-coding genes,and 38.28%of the genome was predicted to be repetitive.We found 981 contracted gene families,1324 expanded gene families and 2300 apricot-specific genes.The differentially expressed gene(DEG)analysis indicated that a change in the expression of the 9-cis-epoxycarotenoid dioxygenase(NCED)gene but not lycopene beta-cyclase(LcyB)gene results in a lowβ-carotenoid content in the white cultivar“Dabaixing”.This complete and highly contiguous P.armeniaca reference genome will be of help for future studies of resistance to plum pox virus(PPV)and the identification and characterization of important agronomic genes and breeding strategies in apricot.Fengchao Jiang Junhuan Zhang Sen Wang Li Yang Yingfeng Luo Shenghan Gao Meiling Zhang Shuangyang Wu Songnian Hu Haoyuan Sun Yuzhu Wang 2019Horticulture Research2019,6,1:9
3Development and validation of an endoscopic images-based deep learning model for detection with nasopharyngeal malignancies显示文摘Background:Due to the occult anatomic location of the nasopharynx and frequent presence of adenoid hyperpla-sia,the positive rate for malignancy identification during biopsy is low,thus leading to delayed or missed diagnosis for nasopharyngeal malignancies upon initial attempt.Here,we aimed to develop an artificial intelligence tool to detect nasopharyngeal malignancies under endoscopic examination based on deep learning.Methods:An endoscopic images-based nasopharyngeal malignancy detection model(eNPM-DM)consisting of a fully convolutional network based on the inception architecture was developed and fine-tuned using separate training and validation sets for both classification and segmentation.Briefly,a total of 28,966 qualified images were collected.Among these images,27,536 biopsy-proven images from 7951 individuals obtained from January 1st,2008,to December 31st,2016,were split into the training,validation and test sets at a ratio of 7:1:2 using simple randomiza-tion.Additionally,1430 images obtained from January 1st,2017,to March 31st,2017,were used as a prospective test set to compare the performance of the established model against oncologist evaluation.The dice similarity coef-ficient(DSC)was used to evaluate the efficiency of eNPM-DM in automatic segmentation of malignant area from the background of nasopharyngeal endoscopic images,by comparing automatic segmentation with manual segmenta-tion performed by the experts.Results:All images were histopathologically confirmed,and included 5713(19.7%)normal control,19,107(66.0%)nasopharyngeal carcinoma(NPC),335(1.2%)NPC and 3811(13.2%)benign diseases.The eNPM-DM attained an overall accuracy of 88.7%(95%confidence interval(CI)87.8%-89.5%)in detecting malignancies in the test set.In the prospective comparison phase,eNPM-DM outperformed the experts:the overall accuracy was 88.0%(95%CI 86.1%-89.6%)vs.80.5%(95%CI 77.0%-84.0%).The eNPM-DM required less time(40 s vs.110.0±5.8 min)and exhibited encouraging performance in automatic segmentation of nasopharyngeal malignant area from the background,with an average DSC of 0.78±0.24 and 0.75±0.26 in the test and prospective test sets,respectively.Conclusions:The eNPM-DM outperformed oncologist evaluation in diagnostic classification of nasopharyngeal mass into benign versus malignant,and realized automatic segmentation of malignant area from the background of nasopharyngeal endoscopic images.Chaofeng Li Bingzhong Jing Liangru Ke Bin Li Weixiong Xia Caisheng He Chaonan Qian Chong Zhao Haiqiang Mai Mingyuan Chen Kajia Cao Haoyuan Mo Ling Guo Qiuyan Chen Linquan Tang Wenze Qiu Yahui Yu Hu Liang Xinjun Huang Guoying Liu Wangzhong Li Lin Wang Rui Sun Xiong Zou Shanshan Guo Peiyu Huang Donghua Luo Fang Qiu Yishan Wu Yijun Hua Kuiyuan Liu Shuhui Lv Jingjing Miao Yanqun Xiang Ying Sun Xiang Guo Xing Lv 2018Cancer Communications2018,38,1:9
4Artificial Intelligence for Edge Service Optimization in Internet of Vehicles:A Survey显示文摘The Internet of Vehicles(IoV)plays a crucial role in providing diversified services because of its powerful capability of collecting real-time information.Generally,collected information is transmitted to a centralized resourceintensive cloud platform for service implementation.Edge Computing(EC)that deploys physical resources near road-side units is involved in IoV to support real-time services for vehicular users.Additionally,many measures are adopted to optimize the performance of EC-enabled IoV,but they hardly help make dynamic decisions according to real-time requests.Artificial Intelligence(AI)is capable of enhancing the learning capacity of edge devices and thus assists in allocating resources dynamically.Although extensive research has employed AI to optimize EC performance,summaries with relative concepts or prospects are quite few.To address this gap,we conduct an exhaustive survey about utilizing AI in edge service optimization in IoV.Firstly,we establish the general condition and relative concepts about IoV,EC,and AI.Secondly,we review the edge service frameworks for IoV and explore the use of AI in edge server placement and service offloading.Finally,we discuss a number of open issues in optimizing edge services with AI.Xiaolong Xu Haoyuan Li Weijie Xu Zhongjian Liu Liang Yao Fei Dai 2022Tsinghua Science and Technology2022,27,2:7
5Famitinib in combination with concurrent chemoradiotherapy in patients with locoregionally advanced nasopharyngeal carcinoma: a phase 1, open-label, dose-escalation Study显示文摘Background:Famitinib is a tyrosine kinase inhibitor against multiple targets,including vascular endothelial growth factor receptor 2/3,platelet-derived growth factor receptor,and stem cell factor receptor(c-kit).Previous studies have demonstrated anti-tumour activities of famitinib against a wide variety of advanced-stage solid cancers.We aimed to determine the safety and efficacy of famitinib with concurrent chemoradiotherapy(CCRT)in patients with locoregionally advanced nasopharyngeal carcinoma(NPC).We also evaluated the feasibility of contrast-enhanced ultrasound(D-CEUS)as a predictor of early tumour response to famitinib and to correlate functional parameters with clinical efficacy.Methods:The trial was conducted in subjects with stage III or IVa-b NPC using a 3+3 design of escalating fami-tinib doses.Briefly,subjects received 2 weeks of famitinib monotherapy followed by 7 weeks of famitinib plus CCRT.D-CEUS of the neck lymph nodes was performed at day 0,8 and 15 after famitinib was administered before starting concurrent chemoradiotherapy.End points included safety,tolerability and anti-tumour activity.Results:Twenty patients were enrolled(six each for 12.5,16.5 and 20 mg and two for 25 mg).Two patients in the 25 mg cohort developed dose-limiting toxicities,including grade 4 thrombocytopenia and grade 3 hypertension.The most common grade 3/4 adverse events were leukopenia,neutropenia and radiation mucositis.D-CEUS tests showed that more than 60%of patients achieved a perfusion parameter response after 2 weeks taking famitinib alone,and the parameter response was associated with disease improvement.In the famitinib monotherapy stage,three patients(15%)showed partial responses.The complete response rate was 65%at the completion of treatment and 95%3 months after the treatment ended.After a median follow-up of 44 months,the 3-year progression-free survival(PFS)and distant metastasis-free survival were 70%and 75%,respectively.Subjects with a decrease of perfusion parameter response,such as peak intensity decreased at least 30%after 1 week of famitinib treatment,had higher 3-year PFS(90.9%vs.44.4%,95%CI 73.7%-100%vs.11.9%-76.9%,P<0.001)than those with an increase or a reduction of less than 30%.Conclusions:The recommended famitinib dose for phase II trial is 20 mg with CCRT for patients with local advanced NPC.D-CEUS is a reliable and early measure of efficacy for famitinib therapies.Further investigation is required to confirm the effects of famitinib plus chemoradiotherapy.Qiuyan Chen Linquan Tang Na Liu Feng Han Ling Guo Shanshan Guo Jianwei Wang Huai Liu Yanfang Ye Lu Zhang Liting Liu Pan Wang Yingqin Li Qingmei He Xiaoqun Yang Qingnan Tang Yang Li YuJing Liang XueSong Sun Chuanmiao Xie Yunxian Mo Ying Guo Rui Sun Haoyuan Mo Kajia Cao Xiang Guo Musheng Zeng Haiqiang Mai Jun Ma 2018Cancer Communications2018,38,1:6
6Study of lithium/polypyrrole secondary batteries with Lithium as cathode and polypyrrole anode显示文摘Lithium/polypyrrole (Li/PPy) batteries were fabricated using lithium sheet as cathode, PPy as anode, microporous membrane polypropylene/polyethylene/polypropylene (PP/PE/PP) composite as separator and LiPF6/ethylene carbonate-dimethyl carbonate-methyl ethyl carbonate (EC-DMC-EMC) as electrolyte. Polypyrrole was prepared by chemical polymerization. Certain fundamental electrochemical performances were investigated. Properties of the batteries were characterized and tested by SEM, galvanostatic charge/discharge tests, cyclic voltammetry (CV), and a.c. impedance spectroscopy. The influences of separator, morphology, and conductivity of PPy anode, cold-molded pressure, and electric current on the performances of the batteries were studied. Using PP/PE/PP membranes as separator, the battery showed good storage stability and cycling property. The conductivity of materials rather than morphology affected the behavior of the battery. The higher the conductivity, the better performances the cells had. Proper cold-molded pressure 20 MPa of the anode pellet would make the properties of the cells good and the fitted charge/discharge current was 0.1 mA. The cells showed excellent performance with 97%-100% coulombic efficiency. 9Theoretical prediction for effective properties and progressive failure of textile composites:a generalized multi-scale approach显示文摘A generalized analytical model is developed to predict progressive failure behavior of several types of textile composites,including plain weave composites,twill weave composites,two-dimensional tri-axially braided composites and warpreinforced 2.5-dimensional braided composites.In this model,the unit cell(UC)of composite is firstly identified and reconstructed into a refined lamina structure with multiple equivalent lamina elements(ELEs)based on apt geometrical approximation and assumptions.Secondly,two-way coupled stress-strain responses within the UC(macro-scale)and ELE(meso-scale)are established through a universal series-parallel model(SPM).Finally,a progressive damage model,which consists of damage initiation criteria and a stiffness evolution strategy,is employed to predict damage behavior of the ELE.The analytical results including mechanical properties and progressive failure process are validated against the existing numerical and experimental ones in literature.The validated analytical model is then used to study the effects of global fiber volume fraction,braided angle,shear failure coefficient and selected failure criteria on stiffness,strength and failure process.The present results demonstrate the efficiency and generic capability of the present analytical model for predicting the mechanical responses of a range of textile composites.Haoyuan Dang Peng Liu Yinxiao Zhang Zhenqiang Zhao Liyong Tong Chao Zhang Yulong Li 2021Acta Mechanica Sinica2021,37,8:3
10Scaled-up synthesis of defect-rich layered double hydroxide monolayers without organic species for efficient oxygen evolution reaction显示文摘The scaled-up synthesis of organic-free monolayer nanomaterials is highly desirable,especially in obtaining green energy by electrocatalysis.In this study,a method for the scaled-up synthesis of the series of monolayer layered double hydroxides(LDHs)without the addition of organic solvents is reported via the separate nucleation and aging steps process.The resulting monolayer LDHs with the thicknesses of less than 1 nm showed a narrow thickness distribution.X-ray absorption fine-structure revealed that monolayer NiFe-LDH nanosheets have a number of oxygen and metal vacancies defects.As a practical application,monolayer NiFe-LDH nanosheets containing defects showed an enhanced electrocatalytic water oxidation activity compared with that of bulk NiFe-LDH.Density functional theory calculations uncovered that excellent catalytic activity is attributed to vacancies defects.The proposed method is an economical and universally applicable strategy for the scaled-up production of monolayer LDHs.Haoyuan Chi Jingwen Dong Tian Li Sha Bai Ling Tan Jikang Wang Tianyang Shen Guihao Liu Lihong Liu Luyi Sun Yufei Zhao Yu-Fei Song 2022Green Energy & Environment2022,7,5:2

13Analysis of the Drug Sensitivity of Escherichia coli Isolated from Sheep in Yangling,Shaanxi Province显示文摘This study was conducted to investigate the drug sensitivity of Escherichia coli isolated from sheep,providing data reference for clinical medication. Fifty-four samples were collected from a certain sheep farm in Yangling,Shaanxi Province for E. coli isolation and identification,and K-B disk diffusion method was used to carry out the dug sensitivity test of 40 E. coli to the eight antimicrobials of spectinomycin,amikacin,tobramycin,enrofloxacin,nalidixic acid,levofloxacin,salafloxacin and ciprofloxacin. The results showed that there were 40 positive E. coli strains and the isolation rate was 74. 7%. The resistance rates of the 40 E. coli to spectinomycin,amikacin,tobramycin and nalidixic acid were 85%,95%,97. 5% and 87. 5%,respectively,while the resistance rate to enrofloxacin,levofloxacin,salafloxacin and ciprofloxacin were relatively low and the resistance rates were 35%,20%,25% and 17. 5%,respectively. The isolates showed different levels of drug resistance to eight antimicrobials and the highest resistance rate was 97. 5%. 14Purification and Identification of Antioxidant Peptides from the Scallop Skirts of Mimachlamys Nobilis显示文摘The processes of scallop meats can produce a large amount of by-products like scallop skirts,which are rich in nutrients. This work aimed to obtain the information about antioxidant peptides derived from scallop skirts of Mimachlamys nobilis. The hydro-lysates obtained by trypsin exhibited the highest antioxidant activity( 75.6%,79.5% and 76.7% using DPPH,hydroxyl and superoxide anion radical scavenging activity assays,respectively). The antioxidant peptides( MNAP) were separated and purified using UF membranes,GFC processes and RP-HPLC,and the antioxidant activity of MNAP was identified to be 85.7% using DPPH radical scavenging activity assay. The peptides( MNAP) were composed of two peptides with their molecular weights2 855.6 Da and 3 007.2 Da using MALDI-TOF-MS,respectively. 15A novel index to evaluate discretization methods: A case study of flood susceptibility assessment based on random forest显示文摘The selection of a suitable discretization method(DM) to discretize spatially continuous variables(SCVs)is critical in ML-based natural hazard susceptibility assessment. However, few studies start to consider the influence due to the selected DMs and how to efficiently select a suitable DM for each SCV. These issues were well addressed in this study. The information loss rate(ILR), an index based on the information entropy, seems can be used to select optimal DM for each SCV. However, the ILR fails to show the actual influence of discretization because such index only considers the total amount of information of the discretized variables departing from the original SCV. Facing this issue, we propose an index, information change rate(ICR), that focuses on the changed amount of information due to the discretization based on each cell, enabling the identification of the optimal DM. We develop a case study with Random Forest(training/testing ratio of 7 : 3) to assess flood susceptibility in Wanan County, China.The area under the curve-based and susceptibility maps-based approaches were presented to compare the ILR and ICR. The results show the ICR-based optimal DMs are more rational than the ILR-based ones in both cases. Moreover, we observed the ILR values are unnaturally small(<1%), whereas the ICR values are obviously more in line with general recognition(usually 10%–30%). The above results all demonstrate the superiority of the ICR. 16Prognostic Role and Potential Mechanisms of N6-methyladenosine-related Long Noncoding RNAs in Hepatocellular Carcinoma显示文摘Background and Aims:Numerous studies have explored the important role of N6-methyladenosine(m^(6)A)in cancer.Nonetheless,the interaction between m^(6)A and long noncoding RNAs(lncRNAs)is poorly investigated.Herein,we systematically analyzed the role and prognostic value of m^(6)A-related lncRNAs in hepatocellular carcinoma(HCC).Methods:The m^(6)A-related lncRNAs were identified based on the correlation coefficients with m^(6)A-related genes in HCC from The Cancer Genome Atlas.Subsequently,a novel risk score model was determined using the least absolute shrinkage and selection operator Cox regression analyses.Univariate and multivariate Cox analyses were used to identify independent prog-nostic factors for overall survival(OS)of HCC;thereafter,a prognostic nomogram was constructed.Results:A total of 259 lncRNAs showed significant correlations with m^(6)A in HCC,while 29 lncRNAs had prognostic significance.Further,six critical m^(6)A-related lncRNAs(NRAV,SNHG3,KDM4A-AS1,AC074117.1,AC025176.1,and AL031985.3)were screened out to construct a novel risk score model which classified HCC patients into high-and low-risk groups.Survival analy-ses revealed that patients in the high-risk group exhibited worse OS,both in the training and validation groups.The risk score was also identified as an independent prognostic factor of OS,and a nomogram was established and verified with superior prediction capacity.Besides,the risk score signifi-cantly correlated with the expression of immune checkpoint genes and immune subtypes.Conclusions:These findings indicated the significant role of m^(6)A-related lncRNAs in HCC and the potential application of the novel risk score model for prognostic prediction.Tianxing Dai Jing Li Linsen Ye Haoyuan Yu Mingbin Deng Wei Liu Hua Li Yang Yang Guoying Wang 2022Journal of Clinical and Translational Hepatology2022,10,2:1

18Experimental and theoretical study of the charge transport property of 4,4′-N,N′-dicarbazole-biphenyl显示文摘The hole and electron mobilities of the amorphous films of the organic semiconductor 4,4′-N,N′-dicarbazole-biphenyl (CBP) at different electric fields were measured through the time of flight (TOF) method. Based on its crystalline structure, the hole and electron mobilities of CBP were calculated. A detailed comparison between experimental and theoretical results is necessary for further understanding its charge transport properties. In order to do this, charge mobilities at zero electric field, μ(0), were deduced from experimental data as a link between experimental and theoretical data. It was found that the electron transport of CBP is less affected by traps compared with its hole transport. This unusual phenomenon can be understood through the distributions of frontier molecular orbitals. 19Characterization of intrinsic hole transport in single-crystal spiro-OMeTAD显示文摘Spiro-OMeTAD remains a prominent hole-transport material in perovskite and solid-state dye-sensitized solar cells.However,an understanding of its intrinsic hole-transport properties is still limited.Here,hole transport in spiro-OMeTAD is systematically characterized on the basis of the recently reported X-ray single-crystal data.An approach combining density functional theory calculations,tight-binding modeling,and kinetic Monte Carlo simulations are exploited to describe the key parameters governing hole transport and to investigate the transport mechanism and hole mobilities in the spiro-OMeTAD single crystal.The results provide insight into:(i)why an anisotropic hole-transport mechanism,with an upper range of intrinsic hole mobilities on the order of~10^(−3)cm^(2)/Vs,can be expected in the single crystal;and(ii)how detrimental factors,related to the presence of the spiro motif and of the 4,4′-dimethoxydiphenylamine substituents,limit the intrinsic hole mobilities of the system.Yuan Li Haoyuan Li Cheng Zhong Gjergji Sini Jean-Luc Brédas 2017npj Flexible Electronics2017,1,1:1
20The first local case of mpox caused by an imported case in the Chinese mainland显示文摘Monkeypox (mpox) is a zoonotic disease caused by the mpox virus (MPXV) that has been primarily limited to Central and West African nations since its discovery. The recent spread of the West African lineage of MPXV in historically unaffected countries has raised concerns for global public health. Despite a significant decrease in global mpox cases, there is still a risk of a global resurgence. This study reports the first local case of mpox caused by an imported case in the Chinese mainland. Polymerase chain reaction (PCR) diagnosed the two cases, and the viral genomes were obtained by next-generation sequencing.
