1Clinical and biochemical indexes from 2019-nCoV infected patients linked to viral loads and lung injury显示文摘The outbreak of the 2019-nCoV infection began in December 2019 in Wuhan,Hubei province,and rapidly spread to many provinces in China as well as other countries.Here we report the epidemiological,clinical,laboratory,and radiological characteristics,as well as potential biomarkers for predicting disease severity in 2019-nCoV-infected patients in Shenzhen,China.All 12 cases of the 2019-nCoV-infected patients developed pneumonia and half of them developed acute respiratory distress syndrome (ARDS).The most common laboratory abnormalities were hypoalbuminemia,lymphopenia,decreased percentage of lymphocytes (LYM) and neutrophils (NEU),elevated C-reactive protein (CRP) and lactate dehydrogenase (LDH),and decreased CD8 count.The viral load of 2019-nCoV detected from patient respiratory tracts was positively linked to lung disease severity.ALB,LYM,LYM (%),LDH,NEU (%),and CRP were highly correlated to the acute lung injury.Age,viral load,lung injury score,and blood biochemistry indexes,albumin (ALB),CRP,LDH,LYM (%),LYM,and NEU (%),may be predictors of disease severity.Moreover,the Angiotensin Ⅱlevel in the plasma sample from 2019-nCoV infected patients was markedly elevated and linearly associated to viral load and lung injury.Our results suggest a number of potential diagnosis biomarkers and angiotensin receptor blocker (ARB) drugs for potential repurposing treatment of 2019-nCoV infection.Yingxia Liu Yang Yang Cong Zhang Fengming Huang Fuxiang Wang Jing Yuan Zhaoqin Wang Jinxiu Li Jianming Li Cheng Feng Zheng Zhang Lifei Wang Ling Peng Li Chen Yuhao Qin Dandan Zhao Shuguang Tan Lu Yin Jun Xu Congzhao Zhou Chengyu Jiang Lei Liu 2020Science China(Life Sciences)2020,63,3:157
2Herbal decoctosome is a novel form of medicine显示文摘Traditionally, herbal medicine is consumed by drinking decoctions produced by boiling herbs with water. The functional components of the decoction are heat stable. Small RNAs(sRNAs) were reported as a new class of functional components in decoctions. However, the mechanisms by which sRNAs survive heat treatment of the decoction and enter cells are unclear.Previous studies showed that plant-derived exosome-like nanoparticles(ELNs), which we call botanosomes, could deliver therapeutic reagents in vivo. Here, we report that heat-stable decoctosomes(ELNs) from decoctions have more therapeutic effects than the decoctions in vitro and demonstrate therapeutic efficacy in vivo. Furthermore, sRNAs, such as HJT-sRNA-m7 and PGY-sRNA-6, in the decoctosome exhibit potent anti-fibrosis and anti-inflammatory effects, respectively. Decoctosome is comprised of lipids, chemical compounds, proteins, and s RNAs. A medical decoctosome mimic is called bencaosome. A single lipid sphinganine(d22:0) identified in the decoctosome was mixed and heated with the synthesized sRNAs to form the simplest bencaosome. This simple bencaosome structure was identified by critical micelle concentration(cmc) assay that sRNAs coassembled with sphinganine(d22:0) to form the lipid layers of vesicles. The heating process facilitates co-assembly of sRNAs and sphinganine(d22:0) until a steady state is reached. The artificially produced sphinganine-HJT-sRNA-m7 and sphinganinePGY-sRNA-6 bencaosomes could ameliorate bleomycin-induced lung fibrosis and poly(I:C)-induced lung inflammation, respectively, following oral administration in mice. 3Large-scale analysis of small RNAs derived from traditional Chinese herbs in human tissues显示文摘Plant-derived microRNAs have recently been reported to function in human blood and tissues. Controversy was immediately raised due to possible contamination and the lack of large sample sizes. Here, we report thousands of unique small RNAs derived from traditional Chinese medicine(TCM) herbs found in human blood cells and mouse lung tissues using a large-scale analysis.We extracted small RNAs from decoctions of 10 TCM plants(Ban Zhi Lian, Chai Hu, Chuan Xin Lian, Di Ding Zi Jin, Huang Qin, Jin Yin Hua, Lian Qiao, Pu Gong Ying, Xia Ku Cao, and Yu Xing Cao) and obtained millions of RNA sequences from each herb. We also obtained RNA-Seq data from the blood cells of humans who consumed herbal decoctions and from the lung tissues of mice administered RNAs from herbal decoctions via oral gavage. We identified thousands of unique small RNA sequences in human blood cells and mouse lung tissues. Some of these identified small RNAs from Chuan Xin Lian and Hong Jing Tian could be mapped to the genomes of the herbs, confirming their TCM plant origin. 4Elevated plasma levels of selective cytokines in COVID-19 patients reflect viral load and lung injury显示文摘A recent outbreak of pneumonia in Wuhan,China was found to be caused by a 2019 novel coronavirus(2019-nCoV or SARS-CoV-2 or HCoV-19).We previously reported the clinical features of 12 patients with2019-n Co V infections in Shenzhen,China.To further understand the pathogenesis of COVID-19 and find better ways to monitor and treat the disease caused by 2019-n Co V,we measured the levels of 48 cytokines in the blood plasma of those 12 COVID-19 patients.Thirty-eight out of the 48 measured cytokines in the plasma of 2019-n Co V-infected patients were significantly elevated compared to healthy individuals.Seventeen cytokines were linked to 2019-nCoV loads.Fifteen cytokines,namely M-CSF,IL-10,IFN-α2,IL-17,IL-4,IP-10,IL-7,IL-1 ra,G-CSF,IL-12,IFN-γ,IL-1α,IL-2,HGF and PDGF-BB,were strongly associated with the lung-injury Murray score and could be used to predict the disease severity of2019-n Co V infections by calculating the area under the curve of the receiver-operating characteristics.Our results suggest that 2019-nCoV infections trigger extensive changes in a wide array of cytokines,some of which could be potential biomarkers of disease severity of 2019-n Co V infections.These findings will likely improve our understanding of the immunopathologic mechanisms of this emerging disease.Our results also suggest that modulators of cytokine responses may play a therapeutic role in combating the disease once the functions of these elevated cytokines have been characterized.Yingxia Liu Cong Zhang Fengming Huang Yang Yang Fuxiang Wang Jing Yuan Zheng Zhang Yuhao Qin Xiaoyun Li Dandan Zhao Shunwang Li Shuguang Tan Zhaoqin Wang Jinxiu Li Chenguang Shen Jianming Li Ling Peng Weibo Wu Mengli Cao Li Xing Zhixiang Xu Li Chen Congzhao Zhou William J Liu Lei Liu Chengyu Jiang 2020National Science Review2020,7,6:6
5Signaling pathways and therapeutic interventions in gastric cancer显示文摘Gastric cancer(GC)ranks fifth in global cancer diagnosis and fourth in cancer-related death.Despite tremendous progress in diagnosis and therapeutic strategies and significant improvements in patient survival,the low malignancy stage is relatively asymptomatic and many GC cases are diagnosed at advanced stages,which leads to unsatisfactory prognosis and high recurrence rates.With the recent advances in genome analysis,biomarkers have been identified that have clinical importance for GC diagnosis,treatment,and prognosis.Modern molecular classifications have uncovered the vital roles that signaling pathways,including EGFR/HER2,p53,PI3K,immune checkpoint pathways,and cell adhesion signaling molecules,play in GC tumorigenesis,progression,metastasis,and therapeutic responsiveness.These biomarkers and molecular classifications open the way for more precise diagnoses and treatments for GC patients.Nevertheless,the relative significance,temporal activation,interaction with GC risk factors,and crosstalk between these signaling pathways in GC are not well understood.Here,we review the regulatory roles of signaling pathways in GC potential biomarkers,and therapeutic targets with an emphasis on recent discoveries.Current therapies,including signaling-based and immunotherapies exploited in the past decade,and the development of treatment for GC,particularly the challenges in developing precision medications,are discussed.These advances provide a direction for the integration of clinical,molecular,and genomic profiles to improve GC diagnosis and treatments.Zi-Ning Lei Qiu-Xu Teng Qin Tian Wei Chen Yuhao Xie Kaiming Wu Qianlin Zeng Leli Zeng Yihang Pan Zhe-Sheng Chen Yulong He 2022Signal Transduction and Targeted Therapy2022,7,11:4
6Carbonized wood with ordered channels decorated by NiCo_(2)O_(4) for lightweight and high-performance microwave absorber显示文摘Wood-derived carbon has a 3D porous framework composed of through channels along the growth direction,which is a suitable matrix for preparing electromagnetic wave(EMW)absorbing materials with low cost,light weight,and environmental friendliness.Herein,the carbonized wood decorated by short cone-like NiCo_(2)O_(4)(NiCo_(2)O_(4)@CW)with highly ordered straight-channel architecture was successfully manufactured through a facile calcination procedure.The horizontal arrangement of the through channels of NiCo_(2)O_(4)@CW(H-NiCo_(2)O_(4)@CW)exhibits a strong reflection loss value of-64.0 dB at 10.72 GHz with a thickness of 3.62 mm and a low filling ratio of 26 wt%(with the density of 0.98 g-cnf3),and the effective absorption bandwidth(EAB)is 8.08 GHz(9.92-18.0 GHz)at the thickness of 3.2 mm.The excellent microwave absorption(MA)property was ascribed to the ordered-channel structure with abundant interfaces and defects from NiCo_(2)O_(4)@CW,which could promote the interfacial polarization and dipole polarization.What is more,this advantageous structure increased the multiple reflections and scattering.Finite element analysis(FEA)simulation is carried out to detect the interaction between the prepared material and EMW when the ordered channels are arranged in different directions.This research provides a low-cost,sustainable,and environmentally friendly strategy for using carbonized wood to fabricate microwave absorbers with strong attenuation capabilities and light weight.Guangyu QIN Xiaoxiao HUANG Xu YAN Yunfei HE Yuhao LIU Long XIA Bo ZHONG 2022Journal of Advanced Ceramics2022,11,1:2
7A comprehensive analysis of the Bencao(herbal)small RNA Atlas reveals novel RNA therapeutics for treating human diseases显示文摘Cross-kingdom herbal mi RNA was first reported in 2012.Using a modified herbal extraction protocol,we obtained 73,677,287sequences by RNA-seq from 245 traditional Chinese Medicine(TCM),of which 20,758,257 were unique sequences.We constructed a Bencao(herbal)small RNA(s RNA)Atlas(http://gffzz4a3adbfa122d4a47h0x6cu6pwkv6n6bw6.ffgz.tsg.suse.edu.cn),annotated the sequences by sequence-based clustering,and created a nomenclature system for Bencao s RNAs.The profiles of 21,757 mi RNAs in the Atlas were highly consistent with those of plant mi RNAs in mi RBase.Using software tools,our results demonstrated that all human genes might be regulated by s RNAs from the Bencao s RNA Atlas,part of the predicted human target genes were experimentally validated,suggesting that Bencao s RNAs might be one of the main bioactive components of herbal medicines.We established roadmaps for oligonucleotide drugs development and optimization of TCM prescriptions.Moreover,the decoctosome,a lipo-nano particle consisting of 0.5%–2.5%of the decoction,demonstrated potent medical effects.We propose a Bencao(herbal)Index,including small-molecule compounds(SM),protein peptides(P),nucleic acid(N),non-nucleic and non-proteinogenic large-molecule compounds(LM)and elements from Mendeleev's periodic table(E),to quantitatively measure the medical effects of botanic medicine.The Bencao s RNA Atlas is a resource for developing gene-targeting oligonucleotide drugs and optimizing botanical medicine,and may provide potential remedies for the theory and practice of one medicine.Yinghao Cao Yexuan Lin Na Sun Xinyi Du Yixin Dong Song Mei Xingyu Deng Xiaobei Li Shaoting Guo Kegong Tang Jiaqi Liu Xiangyu Qiao Dandan Zhao Yuhao Qin Cong Zhang Tianyi Xin Xiaohu Shi Congzhao Zhou Tao Dong De-an Guo Benedikt MKessler Dong Xu Jingyuan Song Fengming Huang Xiaoyue Wang Chengyu Jiang 2023Science China(Life Sciences)2023,66,10:2

10Sulfur vs. tellurium: the heteroatom effects on the nonfullerene acceptors显示文摘The effect of chalcogen heteroatom variation on donor materials has been systematically investigated. However, this effect on acceptors has rarely been explored. Herein, nonfullerene acceptors BFPSP and BFPTP were reported by simply changing the chalcogen atoms from S to Te. The differences between BFPSP and BFPTP in light absorption, energy levels, excited-state lifetimes, energy loss, charge mobilities, morphology, and photovoltaic properties were systematically investigated to understand the heteroatom effects. 11Scalable synthesis of macroscopic porous carbon sheet anode for potassium-ion capacitor显示文摘Carbon materials hold the great promise for application in energy storage devices owing to their low cost, high thermal/chemical stability, and high electrical conductivity. However, it remains challenging to synthesize high-performance carbon electrodes in a simple, scalable and sustainable way. Here, we report a facile method for scalable synthesis of porous carbon anode by using cheap and easily accessible zeolitic imidazolate framework-8 as a template and polyvinylpyrrolidone as an additional carbon source. The obtained porous carbon shows the macroscopic sheet-like morphology, which has the highly disordered structure, expanded interlayer spacing, abundant pore structure, and nitrogen doping properties. This porous carbon anode is demonstrated to have the excellent K^(+) charge storage properties in specific capacity, rate capability, and cycling stability. A potassium-ion capacitor assembled by using this porous carbon as the anode, delivers a maximum energy density of 85.12 Wh/kg and power density of 11860 W/kg as well as long cycle life exceeding 3000 cycles. 12Unidirectionally aligned bright quantum rods films,using T-shape ligands,for LCD application显示文摘Unidirectionally aligned nanorods(NRs)are of great importance for different applications,including displays,lighting,and photodetectors.Recently,many alignment techniques were studied to align quantum rods(QRs).However,the brightness of these films,due to the limited concentration of aligned quantum rods in the film,is not enough for their implementation as brightness enhancement films(BEFs)in displays.This can be ascribed to the poor miscibility of quantum rods in polymer and strong concentration dependence of the polarized emission.The ligands of NR are very important for the alignment and brightness.In this article,we proposed a ligand combination comprising T-shape promesogenic phosphonic acid,which on being photoaligned provides mutually parallel alignment of the quantum rods.The T-shape ligands enable the fabrication of hybrid films with a QRs concentration as high as 10 wt.%–20 wt.%retaining high brightness and luminescence polarization property.Later,we used these films in the in-plane switching(IPS)display backlight that shows the color gamut up to 121%of national television system committee(NTSC)(CIE1931),liquid-crystal display(LCD)efficiency up to 7.9%,power efficacy 103±2 nits/W,and the high brightness of~550±10 nits.Thus,the proposed ligands can be used for the alignment of a variety of nanorods.Maksym F.Prodanov Chengbin Kang Swadesh K.Gupta Valerii V.Vashchenko Yuhao Li Minchao Qin Xinhui Lu Abhishek K.Srivastava 2022Nano Research2022,15,6:0

14Canopy structure:An intermediate factor regulating grassland diversity-function relationships under human disturbances显示文摘Grasslands are one of the largest coupled human-nature terrestrial ecosystems on Earth,and severe anthropogenic-induced grassland ecosystem function declines have been reported recently.Understanding factors influencing grassland ecosystem functions is critical for making sustainable management policies.Canopy structure is an important factor influencing plant growth through mediating within-canopy microclimate(e.g.,light,water,and wind),and it is found coordinating tightly with plant species diversity to influence forest ecosystem functions.However,the role of canopy structure in regulating grassland ecosystem functions along with plant species diversity has been rarely investigated.Here,we investigated this problem by collecting field data from 170 field plots distributed along an over 2000 km transect across the northern agro-pastoral ecotone of China.Aboveground net primary productivity(ANPP)and resilience,two indicators of grassland ecosystem functions,were measured from field data and satellite remote sensing data.Terrestrial laser scanning data were collected to measure canopy structure(represented by mean height and canopy cover).Our results showed that plant species diversity was positively correlated to canopy structural traits,and negatively correlated to human activity intensity.Canopy structure was a significant indicator for ANPP and resilience,but their correlations were inconsistent under different human activity intensity levels.Compared to plant species diversity,canopy structural traits were better indicators for grassland ecosystem functions,especially for ANPP.Through structure equation modeling analyses,we found that plant species diversity did not have a direct influence on ANPP under human disturbances.Instead,it had a strong indirect effect on ANPP by altering canopy structural traits.As to resilience,plant species diversity had both a direct positive contribution and an indirect contribution through mediating canopy cover.This study highlights that canopy structure is an important intermediate factor regulating grassland diversity-function relationships under human disturbances,which should be included in future grassland monitoring and management.Xiaoxia Zhao Yuhao Feng Kexin Xu Mengqi Cao Shuya Hu Qiuli Yang Xiaoqiang Liu Qin Ma Tianyu Hu Maggi Kelly Qinghua Guo Yanjun Su 2023Fundamental Research2023,3,2:0
15Visually Intracellular Detection of Telomerase Activity Based on DNA Strand Displacement Reaction and Gold Nanoparticle Labeling显示文摘Telomerase,which is regarded as a common biomarker for early cancer diagnostics and a potential target for clinical therapies,has attracted considerable interests concerning its detection and monitoring.Herein,we propose a sensitive method by designing a gold nanoparticle(AuNP)probe for visually intracellular detection of telomerase activity.The AuNPs were functionalized with a telomer-ase substrate primer(SH-prime).A 6-carboxy-fluorescein(FAM)modified strand(FAM-probe)was attached to the surface of AuNP through its complementary stand(SH-attach).In the absence of telomerase,the fluorescence resonance energy transfer(FRET)from FAM to AuNPs results in efficient fluorescence quenching.In the presence of telomerase,SH-primers on AuNPs were extended with the repeat units(TTAGGGG)n.The extension sequence triggered the strand displacement of FAM-probe to restore the fluorescence signals.It is worth mentioning that the proposed strategy does not need to design complex hairpin structure and allows the meas-urement of telomerase in crude cell extracts down to 0.5 HeLa cells/μL in 2 h.In addition,the present sensing platform can be ap-plied to the visually intracellular detection of telomerase activity in living cells.Yajun Zhang Shen Yan Zhaoxin Chen Xin Jiang Shuang Rao Zhuoran Jiang Shanshan Qin Xiang Zhou Yuhao Du 2022Chinese Journal of Chemistry2022,40,6:0
