2Microvessel changes in the gerbil hippocampus after cerebral ischemia and reperfusion by Buyang Huanwu decoction pretreatment显示文摘Buyang Huanwu decoction(BYHWD) is a classic recipe for the prevention and treatment of ischemic cerebrovascular disease.Gerbils were pretreated with BYHWD,and then subjected to cerebral ischemia and reperfusion.Microvascular changes were determined with laser Doppler monitoring,tannic acid-ferric chloride mordant,and electron microscopy.Results showed that BYHWD pretreatment could enhance the function of hippocampal microvessels,prevent injury,and increase microvasular density and microvasular area density.Thus,these results suggest that BYHWD pretreatment could prevent microvascular occlusion,enhance the capacity of microvascular reperfusion,increase cerebral blood flow,and inhibit neuronal damage,and may be an effective therapy against brain ischemic injury.Xiaoguang Wu Yixue Li Haixia Liu Yuhua Yin Shumin Zhao Yuanyuan Guo 2011Neural Regeneration Research2011,6,9:11

4Mammalian ALKBH1 serves as an N^6-mA demethylase of unpairing DNA显示文摘N^6-methyladenine(N^6-mA)of DNA is an emerging epigenetic mark in mammalian genome.Levels of N^6-mA undergo drastic fluctuation during early embryogenesis,indicative of active regulation.Here we show that the 2-oxoglutarate-dependent oxygenase ALKBH1 functions as a nuclear eraser of N^6-mA in unpairing regions(e.g.,SIDD,Stress-Induced DNA Double Helix Destabilization regions)of mammalian genomes.Enzymatic profiling studies revealed that ALKBH1 prefers bubbled or bulged DNAs as substrate,instead of single-stranded(ss-)or double-stranded(ds-)DNAs.Structural studies of ALKBH1 revealed an unexpected“stretch-out”conformation of its“Flip1”motif,a conserved element that usually bends over catalytic center to facilitate substrate base flipping in other DNA demethylases.Thus,lack of a bending“Flip1”explains the observed preference of ALKBH1 for unpairing substrates,in which the flipped N^6-mA is primed for catalysis.Co-crystal structural studies of ALKBH1 bound to a 21-mer bulged DNA explained the need of both flanking duplexes and a flipped base for recognition and catalysis.Key elements(e.g.,an ALKBH1-specificα1 helix)as well as residues contributing to structural integrity and catalytic activity were validated by structurebased mutagenesis studies.Furthermore,ssDNA-seq and DIP-seq analyses revealed significant co-occurrence of base unpairing regions with N^6-mA in mouse genome.Collectively,our biochemical,structural and genomic studies suggest that ALKBH1 is an important DNA demethylase that regulates genome N^6-mA turnover of unpairing regions associated with dynamic chromosome regulation.Min Zhang Shumin Yang Raman Nelakanti Wentao Zhao Gaochao Liu Zheng Li Xiaohui Liu Tao Wu Andrew Xiao Haitao Li 2020Cell Research2020,30,3:6

6Rabdosia serra alleviates dextran sulfate sodium salt-induced colitis in mice through anti-inflammation,regulating Th17/Treg balance,maintaining intestinal barrier integrity,and modulating gut microbiota显示文摘Rabdosia serra(R.serra),an important component of Chinese herbal tea,has traditionally been used to treat hepatitis,jaundice,cholecystitis,and colitis.However,the chemical composition of R.serra and its effect against colitis remain unclear.In this study,the chemical composition of the water extract of R.serra was analyzed using ultra performance liquid chromatography coupled with a hybrid linear ion trap quadrupole-orbitrap mass spectrometer(UPLC-LTQ-Orbitrap-MS).A total of 46 compounds,comprising ent-kaurane diterpenoids,flavonoids,phenolic acids,and steroids,were identified in the water extract of R.serra,and the extract could significantly alleviate dextran sulfate sodium salt-induced colitis by improving colon length,upregulating anti-inflammatory factors,downregulating proinflammatory factors,and restoring the balance of T helper 17/T regulatory cells.R.serra also preserved intestinal barrier function by increasing the level of tight junction proteins(zonula occludens 1 and occludin)in mouse colonic tissue.In addition,R.serra modulated the gut microbiota composition by increasing bacterial richness and diversity,increasing the abundance of beneficial bacteria(Muribaculaceae,Bacteroides,Lactobacillus,and Prevotellaceae_UCG-001),and decreasing the abundance of pathogenic bacteria(Turicibacter,Eubacterium_fissicatena_group,and Eubacterium_xylanophilum_group).Gut microbiota depletion by antibiotics further confirmed that R.serra alleviated colitis in a microbiota-dependent manner.Overall,our findings provide chemical and biological evidence for the potential application of R.serra in the management of colitis.Hongyi Li Yi Wang Shumin Shao Hui Yu Deqin Wang Chuyuan Li Qin Yuan Wen Liu Jiliang Cao Xiaojuan Wang Haibiao Guo Xu Wu Shengpeng Wang 2022Journal of Pharmaceutical Analysis2022,12,6:4
7Fine particulate matter(PM_(2.5)/PM_(1.0)) in Beijing,China: Variations and chemical compositions as well as sources显示文摘Particulate matter(i.e.,PM_(1.0) and PM_(2.5)),considered as the key atmospheric pollutants,exerts negative effects on visibility,global climate,and human health by associated chemical compositions.However,our understanding of PM and its chemical compositions in Beijing under the current atmospheric environment is still not complete after witnessing marked alleviation during 2013–2017.Continuous measurements can be crucial for further air quality improvement by better characterizing PM pollution and chemical compositions in Beijing.Here,we conducted simultaneous measurements on PM in Beijing during 2018–2019.Results indicate that annualmean PM_(1.0) and PM_(2.5) concentrations were 35.49±18.61μg/m^(3) and 66.58±60.17μg/m^(3),showing a positive response to emission controls.The contribution of sulfate,nitrate,and ammonium(SNA)played an enhanced role with elevated PM loading and acted as the main contributors to pollution episodes.Discrepancies observed among chemical species between PM_(1.0) and PM_(2.5) in spring suggest that sand particles trend to accumulate in the range of 1–2.5μm.Pollution episodes occurred accompanied with southerly clusters and high formation of SNA by heterogeneous reactions in summer and winter,respectively.Results from positive matrix factorization(PMF)combined with potential source contribution function(PSCF)models showed that potential areas were seasonal dependent,secondary and vehicular sources became much more important compared with previous studies in Beijing.Our study presented a continuous investigation on PM and sources origins in Beijing,which provides a better understanding for further emission control as well as a reference for other cities in developing countries.Lining Luo Xiaoxuan Bai Shuhan Liu Bobo Wu Wei Liu Yunqian Lv Zhihui Guo Shumin Lin Shuang Zhao Yan Hao Jiming Hao Kai Zhang Aihua Zheng Hezhong Tian 2022Journal of Environmental Sciences2022,34,11:3
8Brain-wide Mapping of Mono-synaptic Afferents to Different Cell Types in the Laterodorsal Tegmentum显示文摘The laterodorsal tegmentum(LDT) is a brain structure involved in distinct behaviors including arousal,reward, and innate fear. How environmental stimuli and top-down control from high-order sensory and limbic cortical areas converge and coordinate in this region to modulate diverse behavioral outputs remains unclear.Using a modified rabies virus, we applied monosynaptic retrograde tracing to the whole brain to examine the LDT cell type specific upstream nuclei. The LDT received very strong midbrain and hindbrain afferents and moderate cortical and hypothalamic innervation but weak connections to the thalamus. The main projection neurons from cortical areas were restricted to the limbic lobe, including the ventral orbital cortex(VO), prelimbic, and cingulate cortices. Although different cell populations received qualitatively similar inputs, primarily via afferents from the periaqueductal gray area, superior colliculus, and the LDT itself, parvalbumin-positive(PV?) GABAergic cells received preferential projections from local LDT neurons.With regard to the different subtypes of GABAergic cells, aconsiderable number of nuclei, including those of the ventral tegmental area, central amygdaloid nucleus, and VO, made significantly greater inputs to somatostatinpositive cells than to PV?cells. 9Effect of integrated Traditional Chinese and Western Medicine on Gout显示文摘OBJECTIVE:To evaluate the curative effect of integrated Traditional Chinese and Western Medicine on gout,and to investigate the therapy timing and exact treatment options of integrated medicine.METHODS:Totally 860 patients were enrolled,including 460 patients with intermittent gout,200 patients with active Traditional Chinese Medicine(TCM)syndrome(TCM syndrome score≥6)and200 patients with stable TCM syndrome(score<6).They were randomly divided into intervention and control groups.The control group was treated according to Western Medicine guidelines.The intervention group was treated with integrated Traditional Chinese and Western Medicine.The efficacy of TCM syndrome,joint pain score,joint swelling score,ESR,C-reactive protein,serum uric acid,liver and kidney function,and the duration of remission of TCM syndrome were compared between the two groups before and after treatments.RESULTS:For the patients with stable TCM syndrome,there was no significant difference in the effective rate and inefficiency between the intervention group and the control group.For the active type,the effective rate of the intervention group is better than the control group significantly.For the stable type,there was no significant difference between the intervention group and the control group in improving the scores of joint pain and swelling,reducing the level of ESR,C-reactive protein,serum uric acid and improving liver and kidney function.For the active type,the differences between the two groups were significant.The stable stage of gout in the intervention group was longer than the control group.CONCLUSION:For the gout patients with stable TCM syndrome in the acute stage of gout,we can use TCM treatment or Western Medicine alternatively;for the patients with active TCM syndrome,the scheme of combination of Traditional Chinese and Western Medicine can be applied,with the better curative effect than any medicine alone.LIU Wei WU Yuanhao XUE Bin JIN Yue ZHANG Shumin LI Peihao XIE Qing WANG Aihua GAO Jingyue CAI Yue ZHANG Bo LIU Xiaoya WANG Yi DUAN Ran 2021Journal of Traditional Chinese Medicine2021,41,5:3
10Molecular cloning and characterization of a gene encoding the proline transporter protein in common bean(Phaseolus vulgaris L.)显示文摘As a typical compatible solute, proline is accumulated in plants under environmental stresses. Proline transporter(Pro T) plays an important role in proline distribution between plant organs. Using a candidate gene approach, we cloned a c DNA sequence for Pro T from common bean(Phaseolus vulgaris L.) and designated the gene Pv Pro T. The deduced amino acid sequence of Pv Pro T showed high similarity to Bet/Pro T proteins from other leguminous plants, and the highest similarity was observed with mothbean(Vigna aconitifolia L.) Vu Pro T.Relative quantification of the m RNA level of Pv Pro T using real-time PCR analysis showed that the Pv Pro T transcript level was higher in leaves than in stems and roots of common bean plants subjected to drought and salt stress. Under 20%(w/w) PEG-6000 treatment,drought-resistant plants expressed a higher level of Pv Pro T transcripts than droughtsensitive plants. Although heterologous expression of Pv Pro T in the Escherichia coli mutant mkh13 showed that Pv Pro T exhibited uptake activities for proline and betaine, no betaine content was detected in the common bean. 11Modulation of Fano resonances in symmetry-broken gold-SiO_2-gold nanotube dimers显示文摘Fano resonances in the symmetry-broken gold-SiO2-gold(BGSG)nanotubes and the associated dimers have been investigated based on the finite element method.In the BGSG nanotube,the symmetry breaking induced the interactions of the inner gold core and outer gold nanoshell plasmons of all multipolar orders and hence the red-shifts of the plasmon resonance modes and the enhanced quadrupole mode peaks were observed.The interference of the quadrupole mode peak with the subradiant dipole mode caused a Fano-dip in the scattering spectrum.By increasing the core offset-value in the BGSG nanotube,the Fano dip with low energy showed a red-shift and became deeper.Unexpectedly the plasmon coupling between a GSG nanotube and a BGSG nanotube can lead to two strong Fano dips in the scattering spectra of the dimer.It was further noted that the thin side of the BGSG nanotube located at two sides of the dimer gap can lead to the strong near-field coupling between two BGSG nanotubes and hence a deeper and broader Fano dip.WU DaJian YU HaiQun JIANG ShuMin WU XueWei LIU XiaoJun 2014Science China(Physics,Mechanics & Astronomy)2014,57,6:2

13Impact of pre-oxidation on the formation of byproducts in algae-laden water disinfection:Insights from fluorescent and molecular weight显示文摘Pre-oxidation has been reported to be an effective way to remove algal cells in water, but the released algal organic matter (AOM) could be oxidized and lead to the increment in disinfection by-product (DBP) formation. The relationship between pre-oxidation and AOM-derived DBP formation needs to be approached more precisely. This study compared the impact of four pre-oxidants, ozone (O), chlorine dioxide (ClO), potassium permanganate(KMnO) and sodium hypochlorite (NaClO), on the formation of nitrogenous (N-) and carbonaceous (C-) DBPs in AOM chlorination. The characterization (fluorescent properties,molecular weight distribution and amino acids concentration) on AOM samples showed that the characterization properties variations after pre-oxidation were highly dependent on the oxidizing ability of oxidants. The disinfection experiments showed that Oincreased DBP formation most significantly, which was consistent with the result of characterization properties variations. Then canonical correspondent analysis (CCA) and Pearson’s correlation analysis were conducted based on the characterization data and DBP formation. CCA indicated that C-DBPs formation was highly dependent on fluorescent data. The formation of haloacetic acids (HAAs) had a positive correlation with aromatic protein-like component while trichloromethane (TCM) had a positive correlation with fulvic acid-like component.Pearson’s correlation analysis showed that low molecular weight fractions were favorable to form N-DBPs. 19Lysosomal Hydrolase Cathepsin D Non-proteolytically Modulates Dendritic Morphology in Drosophila显示文摘The main lysosomal protease cathepsin D(cathD)is essential for maintaining tissue homeostasis via its degradative function,and its loss leads to ceroid accumulation in the mammalian nervous system,which results in progressive neurodegeneration.Increasing evidence implies non-proteolytic roles of cathD in regulating various biological processes such as apoptosis,cell proliferation,and migration.Along these lines,we here showed that cathD is required for modulating dendritic architecture in the nervous system independent of its traditional degradative function.Upon cathD depletion,class I and class III arborization(da)neurons in Drosophila larvae exhibited aberrant dendritic morphology,including overbranching,aberrant turning,and elongation defects.Reintroduction of wild-type cathD or its proteolyticallyinactive mutant dramatically abolished these morphological defects.Moreover,cathD knockdown also led to dendritic defects in the adult mushroom bodies,suggesting that cathD-mediated processes are required in both the peripheral and central nervous systems.Taken together,our results demonstrate a critical role of cathD in shaping dendritic architecture independent of its proteolytic function.Ting Zhang Daxiao Cheng Cunjin Wu Xingyue Wang Qiang Ke Huifang Lou Liya Zhu Xiao-Dong Wang Shumin Duan Yi-Jun Liu 2020Neuroscience Bulletin2020,36,10:1
