| 3 | Single-cell Analysis Technologies for Immuno-oncology Research:from Mechanistic Delineation to Biomarker Discovery显示文摘The successes with immune checkpoint blockade(ICB)and chimeric antigen receptor(CAR)-T-cell therapy in treating multiple cancer types have established immunotherapy as a powerful curative option for patients with advanced cancers.Unfortunately,many patients do not derive benefit or long-term responses,highlighting a pressing need to perform complete investigation of the underlying mechanisms and the immunotherapy-induced tumor regression or rejection.In recent years,a large number of single-cell technologies have leveraged advances in characterizing immune system,profiling tumor microenvironment,and identifying cellular heterogeneity,which establish the foundations for lifting the veil on the comprehensive crosstalk between cancer and immune system during immunotherapies.In this review,we introduce the applications of the most widely used single-cell technologies in furthering our understanding of immunotherapies in terms of underlying mechanisms and their association with therapeutic outcomes.We also discuss how single-cell analyses help to deliver new insights into biomarker discovery to predict patient response rate,monitor acquired resistance,and support prophylactic strategy development for toxicity management.Finally,we provide an overview of applying cutting-edge single-cell spatial-omics to point out the heterogeneity of tumor–immune interactions at higher level that can ultimately guide to the rational design of next-generation immunotherapies. | Zhiliang Bai Graham Su Rong Fan | 2021 | Genomics, Proteomics & Bioinformatics2021,19,2: | 1 |

| 5 | Performance Analysis and Framework Optimization of Open Source Cloud Storage System显示文摘More and more embedded devices, such as mobile phones, tablet PCs and laptops, are used in every field, so huge files need to be stored or backed up into cloud storage. Optimizing the performance of cloud storage is very important for Internet development. This paper presents the performance evaluation of the open source distributed storage system, a highly available, distributed, eventually consistent object/blob store from Open Stack cloud computing components. This paper mainly focuses on the mechanism of cloud storage as well as the optimization methods to process different sized files. This work provides two major contributions through comprehensive performance evaluations. First, it provides different configurations for Open Stack Swift system and an analysis of how every component affects the performance. Second, it presents the detailed optimization methods to improve the performance in processing different sized files. The experimental results show that our method improves the performance and the structure. | 7 | Recognition for avian influenza virus proteins based on support vector machine and linear discriminant analysis显示文摘Total 200 properties related to structural characteristics were employed to represent structures of 400 HA coded proteins of influenza virus as training samples. Some recognition models for HA proteins of avian influenza virus (AIV) were developed using support vector machine (SVM) and linear discriminant analysis (LDA). The results obtained from LDA are as follows: the identification accuracy (Ria) for training samples is 99.8% and Ria by leave one out cross validation is 99.5%. Both Ria of 99.8% for training samples and Ria of 99.3% by leave one out cross validation are obtained using SVM model, respectively. External 200 HA proteins of influenza virus were used to validate the external predictive power of the resulting model. | 8 | Paleoclimatic and provenance implications of magnetic parameters from the Miocene sediments in the Subei Basin显示文摘Thick sediments from foreland basins usually provide valuable information for understanding the relationships between mountain building,rock denudation,and sediment deposition.In this paper,we report environmental magnetic measurements performed on the Miocene sediments in the Subei Basin,northeastern Tibetan Plateau.Our results show two different patterns.First,the bulk susceptibility and SIRM,ARM,and HIRM mainly reflect the absolute-concentration of magnetic minerals;all have increased remarkably since 13.7 Ma,related to provenance change rather than climate change.Second,the ratios of IRM100mT/SIRM,IRM100mT/IRM30mT,and IRM100mT/IRM60mT,together with the redness and S ratio,reflect the relative-concentration of hematite,being climate-dependent.Their vertical changes correlate in general with the long-term Miocene climatic records of marine oxygen isotope variations,marked by the existence of higher ratios between 17 and 14 Ma.This may imply that global climate change,rather than uplift of the Tibetan Plateau,played a dominant role in the long-term climatic evolution of the Subei area from the early to middle Miocene. | YouSheng Li JiMin Sun ZhiLiang Zhang Bai Su ShengChen Tian MengMeng Cao | 2020 | Earth and Planetary Physics2020,4,3: | 0 |
