2Haze observations by simultaneous lidar and WPS in Beijing before and during APEC, 2014显示文摘We present combined Mie lidar, ozone lidar and wide-range particle spectrometer observations which were carried out in Beijing, north China during two periods—one haze period before the Asia-Pacific Economic Cooperation(APEC) meeting and one moderate pollution period during the meeting in 2014. High extinction coefficient, moderate ozone concentration and variable particle number concentration were obtained throughout the first haze observation period. The mean extinction coefficients in the two pollution periods were 0.52 and 0.23 km?1, respectively, at 532 nm. The ozone concentration during the first haze phase was more various with a higher average value of 49 ppb compared with that in the second pollution observations(32 ppb). Compared with the same metrological condition occurring at the end of October, the sharply decreased aerosol extinction coefficient and ozone concentration show the effectiveness of the emission-cutting measures implemented during APEC in November. The comparison of aerosols and ozone in different heights indicate different pollution sources and the complicated ozone process of generation and disappearance. The correlation between the scattering coefficient and particle number concentrations of various diameter depended on the ambient humidity. Especially the particle number concentration(500 nm–1 ?m) contributed most to PM2.5 concentration. 3Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 4Understanding unusually high levels of peroxyacetyl nitrate(PAN) in winter in Urban Jinan,China显示文摘Peroxyacetyl nitrate(PAN), as a major secondary pollutant, has gained increasing worldwide attentions, but relevant studies in China are still quite limited. During winter of 2015 to summer of 2016, the ambient levels of PAN were measured continuously by an automatic gas chromatograph equipped with an electron capture detector(GC–ECD) analyzer at an urban site in Jinan(China), with related parameters including concentrations of O3, NO, NO_2, PM_(2.5), HONO,the photolysis rate constant of NO_2 and meteorological factors observed concurrently. The mean and maximum values of PAN concentration were(1.89 ± 1.42) and 9.61 ppbv respectively in winter, and(2.54 ± 1.44) and 13.47 ppbv respectively in summer. Unusually high levels of PAN were observed during severe haze episodes in winter, and the formation mechanisms of them were emphatically discussed. Study showed that high levels of PAN in winter were mainly caused by local accumulation and strong photochemical reactions during haze episodes, while mass transport played only a minor role. Accelerated photochemical reactions(compared to winter days without haze) during haze episodes were deduced by the higher concentrations but shorter lifetimes of PAN, which was further supported by the sufficient solar radiation in the photolysis band along with the high concentrations of precursors(NO_2, VOCs) and HONO during haze episodes. 5Strong ozone production at a rural site in the North China Plain: Mixed effects of urban plumes and biogenic emissions显示文摘Regional ozone(O_3) pollution has drawn increasing attention in China over the recent decade, but the contributions from urban pollution and biogenic emissions have not been clearly elucidated. To better understand the formation of the regional O_3 problem in the North China Plain(NCP), intensive field measurements of O_3 and related parameters were conducted at a rural site downwind of Ji'nan, the capital city of Shandong province, in the summer of 2013. Markedly severe O_3 pollution was recorded, with the O_3 mixing ratios exceeding the Chinese national ambient air quality standard on 28 days(a frequency of 78%) and with a maximum hourly value of 198 ppbv. Extensive regional transport of wellprocessed urban plumes to the site was identified. An observation-constrained chemical box model was deployed to evaluate in situ photochemical O_3 production on two episodes.The results show that the in situ formation accounted for approximately 46% of the observed O_3 accumulation, while the remainder(~54%) was contributed by regional transport of the O_3-laden urban plumes. The in situ ozone production was in a mixed controlled regime that reducing either NOx or VOCs would lead to a reduction of ozone formation. Biogenic VOCs played an important role in the local ozone formation. 6Sensors deployment optimization in multi-dimensional space based on improved particle swarm optimization algorithm显示文摘Sensors deployment optimization has become one of the most attractive fields in recent years. However, most of the previous work focused on the deployment problem in 2 D space.Compared to the traditional form, sensors deployment in multidimensional space has greater research significance and practical potential to satisfy the detecting needs in complex environment.Aiming at solving this issue, a multi-dimensional space sensor network model is established, and the radar system is selected as an example. Considering the possible working mode of the radar system(e.g., searching and tracking), two distinctive deployment models are proposed based on maximum coverage area and maximum target detection probability in the attack direction respectively. The latter one is usually ignored in the previous literature.For uncovering the optimal deployment of the sensor network, the particle swarm optimization(PSO) algorithm is improved using the proposed weights determination scheme, in which the linear decreasing, the pooling strategy and the cloud theory are combined for weights updating. 7Vertical Distribution Characteristics of PM2.5 Observed by a Mobile Vehicle Lidar in Tianjin, China in 2016显示文摘We present mobile vehicle lidar observations in Tianjin, China during the spring, summer, and winter of 2016. Mobile observations were carried out along the city border road of Tianjin to obtain the vertical distribution characteristics of PM_(2.5). Hygroscopic growth was not considered since relative humidity was less than 60% during the observation experiments. PM_(2.5) profile was obtained with the linear regression equation between the particle extinction coefficient and PM_(2.5) mass concentration. In spring, the vertical distribution of PM_(2.5) exhibited a hierarchical structure. In addition to a layer of particles that gathered near the ground, a portion of particles floated at 0.6–2.5-km height. In summer and winter, the fine particles basically gathered below 1 km near the ground. In spring and summer, the concentration of fine particles in the south was higher than that in the north because of the influence of south wind. In winter, the distribution of fine particles was opposite to that measured during spring and summer. High concentrations of PM_(2.5) were observed in the rural areas of North Tianjin with a maximum of 350 μg m–3 on 13 December2016. It is shown that industrial and ship emissions in spring and summer and coal combustion in winter were the major sources of fine particles that polluted Tianjin. 12The novel ER stress inducer Sec C triggers apoptosis by sulfating ER cysteine residues and degrading YAP via ER stress in pancreatic cancer cells显示文摘Pancreatic adenocarcinoma(PAAD) is one of the most lethal malignancies. Although gemcitabine(GEM) is a standard treatment for PAAD, resistance limits its application and therapy. Secoemestrin C(Sec C) is a natural compound from the endophytic fungus Emericella, and its anticancer activity has not been investigated since it was isolated. Our research is the first to indicate that Sec C is a broad-spectrum anticancer agent and could exhibit potently similar anticancer activity both in GEM-resistant and GEM-sensitive PAAD cells. Interestingly, Sec C exerted a rapid growth-inhibiting effect(80% death at 6 h), which might be beneficial for patients who need rapid tumor shrinkage before surgery. Liquid chromatography/mass spectrometry and N-acetyl-L-cysteine(NAC) reverse assays show that Sec C sulfates cysteines to disrupt disulfide-bonds formation in endoplasmic reticulum(ER) proteins to cause protein misfolding, leading to ER stress and disorder of lipid biosynthesis. Microarray data and subsequent assays show that ER stress-mediated ER-associated degradation(ERAD) ubiquitinates anddownregulates YAP to enhance ER stress via destruction complex(YAP-Axin-GSK-βTr CP), which also elucidates a unique degrading style for YAP. 13Ginger-indirect moxibustion plus acupuncture versus acupuncture alone for chronic fatigue syndrome:a randomized controlled trial显示文摘OBJECTIVE:To assess the efficacy and safety of gingerindirect moxibustion for chronic fatigue syndrome(CFS).METHODS:In this central randomized,controlled trial,290 CFS participants were recruited and randomly allocated to group A(ginger-indirect moxibustion plus acupuncture)or group B(acupuncture alone).The study consisted of a treatment period of 8 weeks with a total of 24 treatments(3 sessions per week,every other day),and a follow-up period of 12 weeks.The outcome was measured by Fatigue Severity Scale(FSS),Psychological Health Report(SPHERE),the Self-rating depression scale(SDS)and the Hamilton anxiety scale(HAMA)at baseline,2,4,6,8,12 and 20 weeks.RESULTS:With the treatment undergoing,the changes of FSS,SPHERE,SDS and HAMA scores in both groups increased gradually,and the effect maintained at the 12th week.Between groups,significantly higher score changes were seen in group A in FSS after 4 weeks treatment(11.94 vs 9.12,95%CI:0.94,4.7)and in SPHERE after 2 weeks treatment(3.7 vs 2.27,95%CI:0.56,2.31).But for SDS and HAMA,the improvement did not differ significantly between groups.No severe adverse events were reported.CONCLUSION:Ginger-indirect moxibustion is a safe and effective intervention to relieve fatigue and accompanying physical symptoms of CFS.MA Tingting WU Jie YANG Lijie FENG Fen YANG Huilin ZHANG Jinhua ZHONG Yanjin NING Qing HUANG Lirong LIN Youbing YAN Jue CHEN Guiquan HOU Tianshu WANG Li REN Yuanfang TAN Jing 2022Journal of Traditional Chinese Medicine2022,42,2:2

20Visualized and cascade-enhanced gene silencing by smart DNAzyme-graphene nanocomplex显示文摘BCL-2 gene as well as its products is recognized as a promising target for the molecular targeted therapy of tumors.However,due to certain defense measures of tumor cells,the therapeutic effect based on the gene silencing of BCL-2 is greatly reduced.Here we fabricate a smart response nucleic acid therapeutic that could silence the gene effectively through a dual-targeted and cascade-enhanced strategy.In brief,nano-graphene oxide(GO),working as a nano-carrier,is loaded with a well-designed DNAzyme,which can target and silence the BCL-2 mRNA.Furthermore,upon binding with the BCL-2 mRNA,the enzymatic activity of the DNAzyme can be initiated,cutting a substrate oligonucleotide to produce an anti-nucleolin aptamer AS1411.Nucleolin,a nucleolar phosphoprotein,is known as a stabilizer of BCL-2 mRNA.Via binding and inactivating the nucleolin,AS1411 can destabilize BCL-2 mRNA.By this means of simultaneously targeting mRNA and its stabilizer in an integrated system,effective silencing of the BCL-2 gene of tumor cells is achieved at both the cellular and in vivo levels.After being dosed with this nucleic acid therapeutic and without any chemotherapeutics,apoptosis of tumor cells at the cellular level and apparent shrinkage of tumors in vivo are observed.By labeling a molecular beacon on the substrate of DNAzyme,visualization of the enzymatic activity as well as the tumor in vivo can be also achieved.Our work presents a pure bio-therapeutic strategy that has positive implications for enhancing tumor treatment and avoiding side effects of chemotherapeutics.Lingjie Ren Xiaoxia Chen Chang Feng Lei Ding Xiaomin Liu Tianshu Chen Fan Zhang Yanli Li Zhongliang Ma Bo Tian Xiaoli Zhu 2020Nano Research2020,13,8:1
