3Precise nanomedicine for intelligent therapy of cancer显示文摘Precise nanomedicine has been extensively explored for efficient cancer imaging and targeted cancer therapy, as evidenced by a few breakthroughs in their preclinical and clinical explorations. Here, we demonstrate the recent advances of intelligent cancer nanomedicine, and discuss the comprehensive understanding of their structure-function relationship for smart and efficient cancer nanomedicine including various imaging and therapeutic applications, as well as nanotoxicity. In particular, a few emerging strategies that have advanced cancer nanomedicine are also highlighted as the emerging focus such as tumor imprisonment, supramolecular chemotherapy, and DNA nanorobot. The challenge and outlook of some scientific and engineering issues are also discussed in future development. 6Spectral Coexistence between LEO and GEO Satellites by Optimizing Direction Normal of Phased Array Antennas显示文摘This paper addresses the spectral coexistence between LEO constellation and GEO belt for global distributed earth stations. A specific method is introduced to mitigate the in-line interference by tilting the direction normal of phased array antennas of LEO satellites, and the optimal direction is found by solving a non-linear programming problem. 7High mobility group box 1 mediates inflammatory response of astrocytes via cyclooxygenase 2/prostaglandin E2 signaling following spinal cord injury显示文摘High mobility group box 1(HMGB1) interacts with pattern-recognition receptors of immune cells to activate the inflammatory response. Astrocytes play a positive role in the inflammatory response of the central nervous system by expressing a broad range of patternrecognition receptors. However, the underlying relationship between HMGB1 and the inflammatory reaction of astrocytes remains unclear. In this study, we established rat models of spinal cord injury via laminectomy at the T8–10 level, and the injured spinal cord was subjected to transcriptome sequencing. Our results showed that the HMGB1/Toll-like receptor 4(TLR4) axis was involved in the activation of astrocyte inflammatory response through regulation of cyclooxygenase 2(COX2)/prostaglandin E2(PGE2) signaling. Both TLR4 and COX2 were distributed in astrocytes and showed elevated protein levels following spinal cord injury. Stimulation of primary astrocytes with recombinant HMGB1 showed that COX2 and microsomal PGE synthase(mPGES)-1, rather than COX1, mPGES-2, or cytosolic PGE synthase, were significantly upregulated. Accordingly, PGE2 production in astrocytes was remarkably increased in response to recombinant HMGB1 challenges. Pharmacologic blockade of TLR2/4 attenuated HMGB1-mediated activation of the COX2/PGE2 pathway. Interestingly, HMGB1 did not impact the production of tumor necrosis factor-α or interleukin-1β in astrocytes. Our results suggest that HMGB1 mediates the astrocyte inflammatory response through regulating the COX2/PGE2 signaling pathway. 8Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 9Combined prednisone and levothyroxine improve treatment of severe thrombocytopenia in hepatitis B with compensatory cirrhosis accompanied by subclinical and overt hypothyroidism显示文摘The aim of the present study was to investigate the relationship between hypothyroidism and thrombocytopenia in hepatitis Brelated compensatory liver cirrhosis and to determine whether treatment with levothyroxine and prednisone is superior in a multicenter, open-label, observational study in China. In total, 125 consecutive hepatitis B-related compensated liver cirrhosis patients with severe thrombocytopenia accompanied by hypothyroidism were included. The patients were divided into four groups according to treatment strategy: a control group(n=29), a prednisone group(n=25), a levothyroxine group(n=32) and a prednisone plus levothyroxine group(n=39). Severe thrombocytopenia was more prevalent in hepatitis B-associated compensatory liver cirrhosis patients with hypothyroidism than in euthyroid patients(29.6% vs. 14.7%, P<0.05). Combination treatment with prednisone and levothyroxine decreased the risk of bleeding and improved platelet recovery compared to control treatment and treatment with either prednisone or levothyroxine alone. The platelet count before therapy, serum thyroid stimulating hormone and combination treatment with prednisone and levothyroxine were associated with bleeding events. Therefore, the present study suggests that hypothyroidism is associated with severe thrombocytopenia in hepatitis B-associated compensatory liver cirrhosis. 10Crystalline Silica Promotes Rat Fibrocyte Differentiation in Vitro,and Fibrocytes Participate in Silicosis in Vivo显示文摘Objective The aim of this study was to investigate the effects of Si O2 on fibrocytes and whether fibrocytes participate in silicosis in vivo. Methods A macrophagocyte(AM)/fibrocyte coculture system was established, and AMs were treated with 100 μg/mL SiO 2. Flow cytometry was used to detect the number of fibrocytes. Real‐time PCR was performed to measure the expression of collagen I, collagen III, and α‐SMA mR NA. The levels of collagen I, collagen III, and TGF‐β1 protein were determined by ELISA. Immunohistochemical staining was performed to measure α‐SMA protein expression. A rat silicosis model was induced by intratracheal instillation of SiO 2. Lung histopathological evaluation was conducted using HE and Masson's trichrome staining after 1 and 9 weeks. The number of fibrocytes in peripheral blood or lung tissue of rat was detected by flow cytometry. Double‐color immunofluorescence was applied to identify fibrocytes in the lung tissue. Results Peripheral blood monocytes were found to differentiate into fibrocytes in vitro in a time‐dependent manner, and exposure to crystalline silica might potentiate fibrocyte differentiation. In addition, fibrocytes were able to migrate from peripheral blood to the lung tissue, and the number of fibrocytes was increased after SiO 2 exposure. 11Phase equilibria for the pseudo-ternary system(NaCl+Na2SO4+H2O)of coal gasification wastewater at T=(268.15 to 373.15)K显示文摘The pseudo-ternary system(Na Cl + Na_2SO_4+ H_2O) of coal gasification wastewater was studied at T =(268.15 to 373.15) K. The solubility and density of the equilibrium liquid phase were determined by the isothermal solution saturation method. The equilibrium solids were also investigated by the Schreinemaker's method of wet residues and X-ray powder diffraction(XRD). According to the experimental data, the phase diagrams were determined. It was found that there was no significant solubility difference on the Na Cl-rich side between the ternary system(Na Cl + Na_2SO_4+ H_2O) in coal gasification wastewater and in pure water. However, the solubility on the Na_2SO_4-rich side of coal gasification wastewater was apparently higher than that of pure water. The increase in the solubility of Na_2SO_4 was most likely caused by the effects of other impurities apart from Na Cl and Na_2SO_4 in coal gasification wastewater. 14Hyperechoic demarcation line between a tumor and the muscularis propria layer as a marker for deciding the endoscopic treatment of gastric submucosal tumor显示文摘Minimally invasive endoscopic resection has been rapidly adopted as a new technique for treating patientswith gastric submucosal tumors (SMTs) originating in the muscularis propria (MP) layer. This study was conducted toevaluate the information obtained from endoscopic ultrasonography (EUS) to determine the appropriate endoscopicdissection method for treating SMTs originating in the MP layer. Between February 2014 and May 2016, a total of 50patients with gastric SMTs originating in the MP layer were enrolled in this study. The clinical features of the patientsand their endoscopic, EUS, and histopathologic findings, as well as their postoperative follow-up data, were analyzedin this retrospective study. The mean age of the patients was (55.0±10.2) years, and the male/female ratio was 17:33.Endoscopic submucosal dissection (ESD) was performed on 43 patients and an endoscopic full-thickness resection(EFR) was performed on seven patients. The most frequent location for an SMT was in the upper body region of thestomach (n=16), and the most common pathological diagnosis was a gastrointestinal stromal tumor (GIST) (n=32).The overall rates for complete resection were 95.3% (41/43) and 100.0% (7/7) when the SMTs were treated by ESDand EFR, respectively. The presence of a complete tumor capsule was significantly associated with a complete re-section (P=0.001). Of the cases treated by ESD, nine patients developed perforation, one of whom required laparo-scopic surgery. The remaining patients were closed with clips or purse-string sutures. The presence of an MP2-typetumor (P=0.018) and a wide connection with the MP layer (P=0.044) were significantly associated with perforation. Apreoperative evaluation of the integrity and the location of a tumor capsule and the length of the tumor connection withthe MP layer by EUS can improve the complete resection rate and reduce the occurrence of intraoperative complica-tions. 15Structural Engineering of Hierarchical Aerogels Comprised of Multi-dimensional Gradient Carbon Nanoarchitectures for Highly Efficient Microwave Absorption显示文摘Recently,multilevel structural carbon aerogels are deemed as attractive candidates for microwave absorbing materials.Nevertheless,excessive stack and agglomeration for low-dimension carbon nanomaterials inducing impedance mismatch are significant challenges.Herein,the delicate“3D helix-2D sheet-1D fiber-0D dot”hierarchical aerogels have been successfully synthesized,for the first time,by sequential processes of hydrothermal self-assembly and in-situ chemical vapor deposition method.Particularly,the graphene sheets are uniformly intercalated by 3D helical carbon nanocoils,which give a feasible solution to the mentioned problem and endows the as-obtained aerogel with abundant porous structures and better dielectric properties.Moreover,by adjusting the content of 0D core-shell structured particles and the parameters for growth of the 1D carbon nanofibers,tunable electromagnetic properties and excellent impedance matching are achieved,which plays a vital role in the microwave absorption performance.As expected,the optimized aerogels harvest excellent performance,including broad effective bandwidth and strong reflection loss at low filling ratio and thin thickness.This work gives valuable guidance and inspiration for the design of hierarchical materials comprised of dimensional gradient structures,which holds great application potential for electromagnetic wave attenuation.Yongpeng Zhao Xueqing Zuo Yuan Guo Hui Huang Hao Zhang Ting Wang Ningxuan Wen Huan Chen Tianze Cong Javid Muhammad Xuan Yang Xinnan Wang Zeng Fan Lujun Pan 2021Nano-Micro Letters2021,13,9:5
16Unraveling a genetic roadmap for improved taste in the domesticated apple显示文摘Although taste is an important aspect of fruit quality, an understanding of its genetic control remains elusive in apple and other fruit crops. In this study, we conducted genomic sequence analysis of 497 Malus accessions and revealed erosion of genetic diversity caused by apple breeding and possible independent domestication events of dessert and cider apples. Signatures of selection for fruit acidity and size, but not for fruit sugar content, were detected during the processes of both domestication and improvement. Furthermore, we found that single mutations in major genes affecting fruit taste, including Ma1, MdTDT, and MdSOT2, dramatically decrease malate, citrate, and sorbitol accumulation, respectively, and correspond to important domestication events. Interestingly, Ma1 was identified to have pleiotropic effects on both organic acid content and sugar:acid ratio, suggesting that it plays a vital role in determining fruit taste. Fruit taste is unlikely to have been negatively affected by linkage drag associated with selection for larger fruit that resulted from the pyramiding of multiple genes with minor effects on fruit size. 20Childhood BMI and Adult Obesity in a Chinese Sample:A 13-Year Follow-up Study显示文摘Objective Obesity is recognized as a significant risk factor for diabetes and hypertension. The present study aimed to examine the associations between adults' obesity risk and childhood and parental obesity. Methods A total of 204 children aged 6-17 years were recruited in 2002 with an average follow-up period of 13.2 years. Height and body weight were measured by trained staffs. Overweight and obesity were defined based on the Chinese standard for children and adults. T-test, analysis of variance, and Chi-square analysis were used for single factor analysis. Multiple linear and logistic regression analyses were used to perform multifactor analysis. Results The percentage of non-obese children who grew up to be non-obese adults was 62.6%, and that of obese children who grew up to be obese adults was 80.0%. There was a significant association between childhood body mass index(BMI) and adulthood BMI with a β regression coefficient of 3.76 [95% confidence interval(CI): 1.36-6.16], and between childhood obesity and adulthood obesity with an odds ratio of 5.76(95% CI: 1.37-24.34). There was no statistical difference between parental obesity at baseline and children's adulthood obesity, after adjustment of confounders. Male participants and those aged 10.0-13.0 years had a higher risk of adulthood obesity with odds ratios of 2.50(95% CI: 1.12-5.26) and 3.62(95% CI: 1.17-11.24), respectively.
