2Abscisic acid and jasmonic acid are involved in drought priming-induced tolerance to drought in wheat显示文摘Drought stress is a limiting factor for wheat production and food security.Drought priming has been shown to increase drought tolerance in wheat.However,the underlying mechanisms are unknown.In the present study,the genes encoding the biosynthesis and metabolism of abscisic acid(ABA)and jasmonic acid(JA),as well as genes involved in the ABA and JA signaling pathways were up-regulated by drought priming.Endogenous concentrations of JA and ABA increased following drought priming.The interplay between JA and ABA in plant responses to drought priming was further investigated using inhibitors of ABA and JA biosynthesis.Application of fluridone(FLU)or nordihydroguaiaretic acid(NDGA)to primed plants resulted in lower chlorophyll-fluorescence parameters and activities of superoxide dismutase and glutathione reductase,and higher cell membrane damage,compared to primed plants(PD)under drought stress.NDGA+ABA,but not FLU+JA,restored priming-induced tolerance,as indicated by a finding of no significant difference from PD under drought stress.Under drought priming,NDGA induced the suppression of ABA accumulation,while FLU did not affect JA accumulation.These results were consistent with the expression of genes involved in the biosynthesis of ABA and JA.They suggest that ABA and JA are required for priming-induced drought tolerance in wheat,with JA acting upstream of ABA.Xiao Wang Qing Li Jingjing Xie Mei Huang Jian Cai Qin Zhou Tingbo Dai Dong Jiang 2021The Crop Journal2021,9,1:5

4The different root apex zones contribute to drought priming induced tolerance to a reoccurring drought stress in wheat显示文摘Drought priming is a promising approach to improve tolerance to further drought in wheat.The root apex plays important roles in drought however,its contribution to drought priming remains unknown.To provide mechanistic insights into this process,the transcriptomes and proteomes at three different zones along the root axis under drought stress were analyzed.Physiological assessment of root growth indicated that priming augmented roots growth in response to drought and also the levels of protective proline and glycine betaine.Scanning across the proximal to the distal zones of the root apex indicated increases the transcription of genes involved in primary and secondary metabolism.Conversely,genes related to translation,transcription,folding,sorting and degradation,replication and repair were increased in the apex compared to the proximal zone.A single drought episode suppressed their expression but prior drought priming served to maintain expression with recurrent drought stress.The differentially primed responses genes were mainly involved in the pathways related to plant hormone signaling,stress defense and cell wall modification.The prediction of regulatory hubs using Cytoscape implicated signaling components such as the ABA receptor PYL4 as influencing antioxidant status and the cell cycle.Based our integrative transcriptomic-proteomic assessments we present a model for drought priming protected plant hormone signaling transduction pathways to drive the cell cycle and cell wall loosening to confer beneficial effects on roots to counter the effects of drought.This model provides a theoretical basis for improvement of drought tolerance in wheat,via an increased understanding of drought priming induced drought tolerance.Xiao Wang Jing Chen Jiakun Ge Mei Huang Jian Cai Qin Zhou Tingbo Dai Luis Alejandro Jose Mur Dong Jiang 2021The Crop Journal2021,9,5:2

8Over expression of TaFer gene from Tamarix androssowii improves iron and drought tolerance in transgenic Populus tomentosa显示文摘Ferritin, a universal intracellular protein, can store large amounts of iron and improve plant resistance to abiotic and biotic stress. In this study, a ferritin gene(TaFer) from Tamarix androssowii Litv. was transferred into Populus tomentosa Carr. cv 'BJR01' via Agrobacterium. Six independent transgenic lines were obtained with a tolerance to kanamycin and three were randomly selected for further analysis. The PCR and RT-PCR results indicate that the TaFer gene had been integrated into the poplar genome. The effect of the gene on abiotic stress tolerance was tested, and the results show that transgenic plants improve growth, had higher chlorophyll and lower MDA contents, and higher relative electrical conductivity,fewer changes of SOD and POD activities, higher iron content, higher root ferric reductase activity and lower levels of ROS accumulation and cell death in response to drought, Fe-insufficient or Fe-excess tolerance. 9POD Nanozyme optimized by charge separation engineering for light/pH activated bacteria catalytic/photodynamic therapy显示文摘The current feasibility of nanocatalysts in clinical anti-infection therapy,especially for drug-resistant bacteria infection is extremely restrained because of the insufficient reactive oxygen generation.Herein,a novel Ag/Bi2MoO6(Ag/BMO)nanozyme optimized by charge separation engineering with photoactivated sustainable peroxidase-mimicking activities and NIR-II photodynamic performance was synthesized by solvothermal reaction and photoreduction.The Ag/BMO nanozyme held satisfactory bactericidal performance against methicillin-resistant Staphylococcus aureus(MRSA)(~99.9%).The excellent antibacterial performance of Ag/BMO NPs was ascribed to the corporation of peroxidase-like activity,NIR-II photodynamic behavior,and acidity-enhanced release of Ag^(+).As revealed by theoretical calculations,the introduction of Ag to BMO made it easier to separate photo-triggered electronhole pairs for ROS production.And the conduction and valence band potentials of Ag/BMO NPs were favorable for the reduction of O_(2) to·O_(2)^(−).Under 1064 nm laser irradiation,the electron transfer to BMO was beneficial to the reversible change of Mo^(5+)/Mo^(6+),further improving the peroxidase-like catalytic activity and NIR-II photodynamic performance based on the Russell mechanism.In vivo,the Ag/BMO NPs exhibited promising therapeutic effects towards MRSA-infected wounds.This study enriches the nanozyme research and proves that nanozymes can be rationally optimized by charge separation engineering strategy.Changyu Cao Tingbo Zhang Nan Yang Xianghong Niu Zhaobo Zhou Jinlan Wang Dongliang Yang Peng Chen Liping Zhong Xiaochen Dong Yongxiang Zhao 2022Signal Transduction and Targeted Therapy2022,7,4:1
10Angiograms of the Abdominal Aorta in a Patient of Middle Aortic Syndrome显示文摘A 42-year old woman of middle aortic syndrome presented with severe hypertension for 20 years, whose abdominal aorta angiography revealed long segment and nearly complete occlusion in the mid portion of abdominal aorta, and extensively enlarged collateral vascular supply to the lower portion of abdominal aorta. The pressures proximal and distal to stenosis were 185/110 and 95/70 mmHg, and the pressure gradient across the stenosis was 90/40 mmHg. 12Numerical solutions of 2-D steady compressible natural convection using high-order flux reconstruction显示文摘Low Mach number flows are common and typical in industrial applications. When simulating these flows, performance of traditional compressible flow solvers can deteriorate in terms of both efficiency and accuracy. In this paper, a new high-order numerical method for two-dimensional(2-D)state low Mach number flows is proposed by combining flux reconstruction(FR)and preconditioning. Firstly, a Couette flow problem is used to assess the efficiency and accuracy of preconditioned FR. It is found that the FR scheme with preconditioning is much more efficient than the original FR scheme. Meanwhile,this improvement still preserves the numerical accuracy. Using this new method and without the Boussinesq assumption,classic natural convection is directly simulated for cases of small and large temperature differences. For the small temperature difference, a p and h refinement study is conducted to verify the grid convergence and accuracy. Then, the influence of the Rayleigh number (Ra) is analyzed. By comparing with the reference results, the numerical results of preconditioned FR is very close to that calculated by incompressible solvers. 14Protective Effects of Six PPTs on Hypoxia/Reoxygenation Induced Cardiomyocyte Injury by Different Treatments显示文摘[Objectives]To detect the protective effects of six protopanaxatriols(PPTs)on hypoxia/reoxygenation(H/R)induced cardiomyocyte injury by different treatments.[Methods]The 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide(MTT)assay was used for detecting the protective effects of six PPTs including ginsenoside Rg1,Re,Rf,Rg2,(R)Rh1 and(S)Rh1 on cell viability reduced by H/R in different treatments.And the adenosine triphosphate(ATP)content and mitochondrial membrane potential(MMP)were used for detecting the mitochondrial function change during PPTs treatment.[Results]Among six PPTs,ginsenoside Rg1,Re,Rf,Rg2 and(R)Rh1 at the concentration of 12.5μM significantly increased the cell survival when treated before and during H/R.These five PPTs also significantly increased the ATP content and MMP reduced by H/R in the same manner.In comparison,only Rg1 significantly increased the cell viability compared with H/R group by pretreating and treating the cells during hypoxia process.[Conclusions]Different treatments affect the protective effects of PPTs.When treated before and during H/R,ginsenoside Rg1,Re,Rf,Rg2 and(R)Rh1 protect the cardiomyocyte against H/R injury mitochondrial function,and only ginsenoside Rg1 has protective effects when treated before and during hypoxia process.Tingbo CHEN Dong HUANG Ziming ZHOU Gengting DONG 2022Medicinal Plant2022,13,5:0
15Translation:Management of Coronavirus Disease 2019(COVID-19):Experience in Zhejiang Province,China显示文摘The current epidemic situation of coronavirus disease 2019(COVID-19)still remains severe.As the National Clinical Research Center for Infectious Diseases,The First Affiliated Hospital of the Zhejiang University School of Medicine is the primary medical care center for COVID-19 in Zhejiang Province.Based on the present expert consensus carried out by the National Health Commission and National Administration of Traditional Chinese Medicine,our team summarized and established an effective treatment strategy centered on“Four-Anti and Two-Balance”for clinical practice.The“Four-Anti and Two-Balance”strategy includes antivirus,anti-shock,antihypoxemia,and anti-secondary infection,and maintaining of water,electrolyte and acid/base balance and microecological balance.Simultaneously,an integrated multidisciplinary personalized treatment is recommended to improve therapeutic effects.The importance of early viral detection,dynamic monitoring of inflammatory indexes,and chest radiographs has been emphasized in clinical decision-making.Sputum was observed with the highest positive rate by reverse transcription-polymerase chain reaction(RTPRC).Viral nucleic acids could be detected in 10%of the patients’blood samples at the acute phase and 50%of patients had positive RT-PCR results in their feces.We also isolated live viral strains from feces,indicating potential infectiousness of feces.Dynamic cytokine detection was necessary to timely identify cytokine storms and for the application of the artificial liver blood purification system.The“Four-Anti and Two-Balance”strategy effectively increased cure rates and reduced mortality.Early antiviral treatment alleviated disease severity and prevented illness progression.We found that lopinavir/ritonavir combined with abidol showed antiviral effects against COVID-19.Shock and hypoxemia were usually caused by cytokine storms.The artificial liver blood purification system was able to rapidly remove inflammatory mediators and block the cytokine storm.Moreover,it also contributed to the balance of fluids,electrolytes,and acids/bases and thus improved treatment efficacy during critical illness.For cases of severe illness,early and also short periods of moderate glucocorticoid administration was supported.Patients with an oxygenation index below 200mmHg were transferred to the intensive care unit.Conservative oxygen therapy was preferred and noninvasive ventilation(NIV)was not recommended.Patients with mechanical ventilation were strictly supervised with cluster ventilator-associated pneumonia prevention strategies.Antimicrobial prophylaxis was prescribed rationally and was not recommended,except for patients with a long course of disease,repeated fever,and elevated procalcitonin,similarly secondary fungal infections were of concern.Some patients with COVID-19 showed intestinal microbial dysbiosis with decreased genus such as Lactobacillus and Bifidobacterium.Nutritional and gastrointestinal function should;therefore,be assessed for all patients.Nutritional support and application of prebiotics or probiotics were suggested to regulate the balance of intestinal microbiota and reduce the risk of secondary infections due to bacterial translocation.Anxiety and fear were common in patients with COVID-19.Therefore,we established a dynamic assessment and warning for psychological crises.We also integrated Chinese medicine in the treatment to promote rehabilitation.We optimized nursing processes for severe patients to promote their rehabilitation.Since viral clearance patterns after severe acute respiratory syndrome coronavirus 2 infections remained unclear,2 weeks quarantine for discharged patients was required,and a regular following-up was also needed.These Zhejiang experiences and suggestions have been implemented in our center and achieved good results.However,since COVID-19 was a newly emerging disease,more work is warranted to further improve strategies of prevention,diagnosis,and treatment for COVID-19.Kaijin Xu Hongliu Cai Yihong Shen Qin Ni Yu Chen Shaohua Hu Jianping Li Huafen Wang Liang Yu He Huang Yunqing Qiu Guoqing Wei Qiang Fang Jianying Zhou Jifang Sheng Tingbo Liang Lanjuan Li 2020Infectious Microbes & Diseases2020,2,2:0
16Ectopic expression of poplar gene PsnERF138 in tobacco confers salt stress tolerance and growth advantages显示文摘The AP2/ERF family is one of the largest plant-specific transcription factors and plays a vital role in plant growth and stress response.In this study,PsnERF138 was cloned from Populus alba×Populus glandulosa and transformed into tobacco using the Agrobacterium-mediated transformation method.PsnERF138 was localized in the nucleus through subcellular localization assay in tobacco.Under normal conditions,the root lengths of PsnERF138 transgenic lines were much longer than those of wild type.Under salt stress,the transgenic tobacco lines over-expressing PsnERF138 showed a significant increase in seed germination rate,plant height,and root length,compared to control plants.In addition,the transgenic tobacco lines displayed some advantages at the physiological level,such as higher superoxide dismutase(SOD)activity,peroxidase(POD)activity,proline content,and lower malondialdehyde(MDA)content,as compared to those in the control plants.Histochemical staining also showed that the transgenic tobacco lines had lower reactive oxygen species(ROS)accumulation,compared to control plants under salt stress.The combined results indicate that poplar PsnERF138 plays a contributing role in augmenting salt tolerance and conferring multiple growth advantages as being overexpressed in tobacco.Lin Liu Zihan Cheng Wenjing Yao Xueyi Wang Fenglin Jia Boru Zhou Tingbo Jiang 2021Forestry Research2021,1,1:0
