1Tongue coating microbiome as a potential biomarker for gastritis including precancerous cascade显示文摘The development of gastritis is associated with an increased risk of gastric cancer. Current invasive gastritis diagnostic methods are not suitable for monitoring progressIn this work based on 78 gastritis patients and 50 healthy individuals, we observed that the variation of tongue-coating microbiota was associated with the occurrenee and development of gastritis. Twenty-one microbial species were identified for differentiating tongue-coating microbiomes of gastritis and healthy individuals. Pathways such as microbial metabolism in diverse environments, biosynthesis of antibiotics and bacterial chemotaxis were up-regulated in gastritis patients. The abundance of Campylobacter concisus was found associated with the gastric precancerous cascade. Furthermore, Campylobacter concisus could be detected in tongue coating and gastric fluid in a validation cohort containing 38 gastritis patients. 2Key residues of the receptor binding motif in the spike protein of SARS-CoV-2 that interact with ACE2 and neutralizing antibodies显示文摘Coronavirus disease 2019(COVID-19),caused by the novel human coronavirus SARS-CoV-2,is currently a major threat to public health worldwide.The viral spike protein binds the host receptor angiotensin-converting enzyme 2(ACE2)via the receptor-binding domain(RBD),and thus is believed to be a major target to block viral entry.Both SARS-CoV-2 and SARS-CoV share this mechanism.Here we functionally analyzed the key amino acid residues located within receptor binding motif of RBD that may interact with human ACE2 and available neutralizing antibodies.The in vivo experiments showed that immunization with either the SARS-CoV RBD or SARS-CoV-2 RBD was able to induce strong clade-specific neutralizing antibodies in mice;however,the cross-neutralizing activity was much weaker,indicating that there are distinct antigenic features in the RBDs of the two viruses.This finding was confirmed with the available neutralizing monoclonal antibodies against SARS-CoV or SARS-CoV-2.It is worth noting that a newly developed SARS-CoV-2 human antibody,HA001,was able to neutralize SARS-CoV-2,but failed to recognize SARS-CoV.Moreover,the potential epitope residues of HA001 were identified as A475 and F486 in the SARS-CoV-2 RBD,representing new binding sites for neutralizing antibodies.Overall,our study has revealed the presence of different key epitopes between SARS-CoV and SARSCoV-2,which indicates the necessity to develop new prophylactic vaccine and antibody drugs for specific control of the COVID-19 pandemic although the available agents obtained from the SARS-CoV study are unneglectable.Chunyan Yi Xiaoyu Sun Jing Ye Longfei Ding Meiqin Liu Zhuo Yang Xiao Lu Yaguang Zhang Liyang Ma Wangpeng Gu Aidong Qu Jianqing Xu Zhengli Shi Zhiyang Ling Bing Sun 2020Cellular & Molecular Immunology2020,17,6:13
3Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12
4Stable Isotopes in Precipitation in Xilin River Basin,Northern China and Their Implications显示文摘Under the increasing pressure of water shortage and steppe degradation,information on the hydrological cycle in steppe region in Inner Mongolia,China is urgently needed.An intensive investigation of the temporal variations ofδD andδ1 8O in precipitation was conducted in 2007-2008 in the Xilin River Basin,Inner Mongolia in the northern China.TheδD andδ1 8O values for 54 precipitation samples range from+1.1‰to-34.7‰and-3.0‰to -269‰,respectively.This wide range indicates that stable isotopes in precipitation are primarily controlled by different condensation mechanisms as a function of air temperature and varying sources of vapor.The relationship between δD andδ1 8 O defined a well constrained line given by δD =7.89 δ18O+9.5,which is nearly identical to the Meteoric Water Line in the northern China.The temperature effect is clearly displayed in this area.The results of backward tra- jectory of each precipitation day show that the vapor of the precipitation in cold season(October to March)mainly originates from the west while the moisture source is more complicated in warm season(April to September).A light precipitation amount effect existes at the precipitation event scale in this area.The vapor source of precipitation with higher d-excesses are mainly from the west wind or neighboring inland area and precipitation with lower d-excesses from a monsoon source from the southeastern China.WU Jinkui DING Yongjian YE Baisheng YANG Qiyue HOU Dianjiong XUE Liyang 2012Chinese Geographical Science2012,22,5:9

6Photo-production of dissolved inorganic carbon from dissolved organic matter in contrasting coastal waters in the southwestern Taiwan Strait, China显示文摘Photo-production of dissolved inorganic carbon (DIC) from chromophoric dissolved organic matter (CDOM) is an important transformation process in marine carbon cycle, but little is known about this process in Chinese coastal systems. This study investigated an estuarine water sample and a coastal seawater sample from the subtropical waters in southeast of China. Water samples were exposed to natural sunlight and the absorption and fluorescence of CDOM as well as the DIC concentration were measured in the summer of 2009. The estuarine water had higher CDOM level, molecular weight and proportion of humic-like fluorescent components than the seawater that exhibited abundant tryptophan-like fluorescent component. After a 3-day irradiation, the CDOM level decreased by 45% in the estuarine water and 20% in the seawater, accompanied with a decrease in the molecular weight and aromaticity of DOM which was inferred from an increase in the absorption spectral slope parameter. The photo-degradation rates of all the five fluorescent components were also notable, in particular two humic-like components (C4 and C5) were removed by 78% and 69% in the estuarine water and by 69% and 56% in the seawater. The estuarine water had a higher photo-production rate of DIC than the seawater (4.4 vs. 2.5 μmol/(L·day)), in part due to its higher CDOM abundance. 7Optical properties of estuarine dissolved organic matter isolated using cross-flow ultrafiltration显示文摘Dissolved organic matter(DOM) from freshwater, mid-salinity, and seawater endmember samples in the Jiulong River Estuary, China were fractionated using cross-flow ultrafiltration with a 10-kDa membrane. The colloidal organic matter(COM; 10 kDa–0.22 μm) retentate, low molecular weight(LMW) DOM(<10 kDa) permeate, and bulk samples were analyzed using absorption spectroscopy and three-dimensional fluorescence excitation-emission-matrix spectroscopy. The UV-visible spectra of COM were very similar to those obtained for permeate and bulk samples, decreasing monotonically with increasing wavelength. Most of the chromophoric DOM(CDOM, expressed as the absorption coefficient a355) occurred in the LMW fraction, while the percentage of CDOM in the colloidal fraction was substantially higher in the freshwater endmember(13.4% of the total) than in the seawater endmember(6.8%). The bulk CDOM showed a conservative mixing behavior in the estuary, while there was removal of the COM fraction and a concurrent addition of the permeate fraction in the mid-salinity sample, implying that part of the colloidal CDOM was transformed into LMW CDOM. Two humic-like components(C1: <250, 325/402 nm; and C2: 265, 360/458 nm) and one protein-like component(C3: 275/334 nm) were identified using parallel factor analysis. The contributions of the C1, C2, and C3 components of the COM fraction to the bulk sample were 2.5%–8.7%, 4.8%–12.6%, and 7.4%–14.7%, respectively, revealing that fluorescent DOM occurred mainly in the LMW fraction in the Jiulong River Estuary. The C1 and C2 components in the retentate and permeate samples showed conservative mixing behavior, but the intensity ratio of C2/C1 was higher in the retentate than in the permeate fractions for all salinity samples, showing that the humic component was more enriched in the COM than the fulvic component. The intensity ratio of C3/(C1+C2) was much higher in the retentate than in the permeate fraction for mid-salinity and seawater samples, revealing that the protein-like component was relatively more enriched in COM than the humic-like component. 8Progress of Jinping Underground laboratory for Nuclear Astrophysics(JUNA)显示文摘Jinping Underground laboratory for Nuclear Astrophysics(JUNA) will take the advantage of the ultra-low background of CJPL lab and high current accelerator based on an ECR source and a highly sensitive detector to directly study for the first time a number of crucial reactions occurring at their relevant stellar energies during the evolution of hydrostatic stars. In its first phase, JUNA aims at the direct measurements of^(25)Mg(p,γ)^(26)Al,^(19)F(p,α)^(16)O,^(13)C(α,n)^(16)O and ^(12)C(α,γ)^(16)O reactions. The experimental setup,which includes an accelerator system with high stability and high intensity, a detector system, and a shielding material with low background, will be established during the above research. 9Cyanobacteria-based self-oxygenated photodynamic therapy for anaerobic infection treatment and tissue repair显示文摘Photodynamic therapy(PDT)is an important technique to deal with drug-resistant bacterial infections in the post-antibiotic era.However,the hypoxic environment in intractable infections such as refractory keratitis and periodontitis,makes PDT more difficult.In this work,spontaneous oxygen-producing cyanobacteria were used as the carrier of photosensitizer(Ce6),and ultrasmall Cu_(5.4)O nanoparticles(Cu_(5.4)O USNPs)with catalase activity for infection and inflammation elimination and rapid tissue repair(CeCycn-Cu_(5.4)O).The loading of Ce6 and Cu_(5.4)O USNPs onto cyanobacteria surface were confirmed by transmission electron microscopy,nano particle size analyzer,scanning electron microscopy.In vitro sterilization and biofilm removal experiments demonstrated that the restriction of hypoxic environment to PDT was significantly alleviated due to the oxygen production of cyanobacteria.Under laser irradiation,the close transfer of energy photons to oxygen produced by cyanobacteria reduced more than 90% of Ce6 dosages(660 nm,200 mW/cm^(2),2 min).It is worth mentioning that both rapid sterilization through PDT and long-term oxidized free radicals elimination were achieved by adjusting the ratio of Ce6 and Cu_(5.4)O USNPs.Both periodontitis and refractory keratitis animal models proved the excellent self-oxygenation enhanced antibacterial property and promotion of tissue repair.Bailiang Wang Liyang Zhou Yishun Guo Hanwen Guo Yiming Zhong Xiaomin Huang Yifan Ge Qingying Wang Xiaoying Chu Yingying Jin Kaiyue Lan Mei Yang Jia Qu 2022Bioactive Materials2022,7,6:3

11Dietary calcium requirements of broilers fed a conventional corn-soybean meal diet from 1 to 21 days of age显示文摘Background:The current calcium(Ca)recommendation for broilers is primarily based on studies conducted more than 30 years ago with birds of markedly different productive potentials from those which exist today.And the response indicators in these studies are mainly growth performance and bone ash percentage.Therefore,the present study was carried out to investigate the effect of dietary Ca level on growth performance,serum parameters,bone characteristics and Ca metabolism-related gene expressions,so as to estimate dietary Ca requirements of broilers fed a conventional corn-soybean meal diet from 1 to 21 days of age.Methods:A total of 4201-day-old Arbor Acres male broilers were randomly assigned to 1 of 7 treatments with 6 replicates(10 birds per cage)and fed the corn-soybean meal diets containing 0.60%,0.70%,0.80%,0.90%,1.00%,1.10%or 1.20%Ca for 21 days.Each diet contained the constant non-phytate phosphorus content of about 0.39%.Results:The average daily gain decreased linearly(P<0.001)as dietary Ca level increased.The serum and tibia alkaline phosphatase(ALP)activities,tibia bone mineral density(BMD),middle toe BMD,tibia ash percentage,tibia breaking strength,and tibia ALP protein expression level were affected(P<0.05)by dietary Ca level,and showed significant quadratic responses(P<0.02)to dietary Ca levels.The estimates of dietary Ca requirements were 0.80 to 1.00%based on the best fitted broken-line or quadratic models(P<0.03)of the above serum and bone parameters,respectively.Conclusions:The results from the present study indicate that the Ca requirements would be about 0.60%to obtain the best growth rate,and 1.00%to meet all of the Ca metabolisms and bone development of broilers fed a conventional corn-soybean meal diet from 1 to 21 days of age.Shiping Bai Yunfeng Yang Xuelian Ma Xiudong Liao Runlian Wang Liyang Zhang Sufen Li Xugang Luo Lin Lu 2022Journal of Animal Science and Biotechnology2022,13,4:2

15Seeding performance of air-assisted centralized seed-metering device for rapeseed显示文摘In order to analyze seed movement characteristics and improve seeding quality of air-assisted centralized metering device for rapeseed,the effects of the model-hole structure on seed feeding performance were investigated using EDEM simulation.Furthermore,the CFD-DEM coupling approach was applied to determine movement trajectories and airflow fields.The impacts of rapeseed varieties and rotational speed on seeding performance were investigated by bench tests.The results showed that the seed feeding quantity increased with the increase of model-hole’s length,depth and section size.Under the model-hole’s depth of 3.0 mm,the type II model-hole and model-hole’s length of 10 mm,both the variation coefficient of seed feeding quantity and hill diameter were lower which meet the seeding quantity requirement of 2 seeds in each hill.It was revealed that the seed population migrated in a large airflow velocity area and the distribution was uniform.The bench tests indicated that rapeseed varieties and rotational speed had a significant effect on the seed feeding quantity in each hill at rotational speed of 10-40 r/min.The variation coefficient of seed feeding quantity in each hill was less than 17.0%for each treatment.The hill diameter,which did not exceed 3.5 cm,tended to reduce with increasing rotational speed.The variation coefficient of seeding quantity in each row and seeding uniformity was less than 6.5%and 32.0%,respectively.Field experiments demonstrated that the seedling was 9-13 plants per meter each row for three rapeseed varieties.The variation coefficient of plants was less than 25.0%for six rows and their yields reached 2761 kg/hm2,which realized the mechanized planting requirements.The results can optimize structure of an air-assisted centralized metering system and improve seeding performance.Xiaolong Lei Hongji Hu Wenhao Yang Liyang Liu Qingxi Liao Wanjun Ren 2021International Journal of Agricultural and Biological Engineering2021,14,5:1

17Responses of potential ammonia oxidation and ammonia oxidizers community to arsenic stress in seven types of soil显示文摘Soil arsenic contamination is of great concern because of its toxicity to human,crops,and soil microorganisms.However,the impacts of arsenic on soil ammonia oxidizers communities remain unclear.Seven types of soil spiked with 0 or 100 mg arsenic per kg soil were incubated for 180 days and sampled at days 1,15,30,90 and 180.The changes in the community composition and abundance of ammonia oxidizing bacteria(AOB)and ammonia oxidizing archaea(AOA)were analyzed by terminal restriction fragment length polymorphism(TRFLP)analysis,clone library sequencing,and quantitative PCR(qPCR)targeting amoA gene.Results revealed considerable variations in the potential ammonia oxidation(PAO)rates in different soils,but soil PAO was not consistently significantly inhibited by arsenic,probably due to the low bioavailable arsenic contents or the existence of functional redundancy between AOB and AOA.The variations in AOB and AOA communities were closely associated with the changes in arsenic fractionations.The amoA gene abundances of AOA increased after arsenic addition,whereas AOB decreased,which corroborated the notion that AOA and AOB might occupy different niches in arsenic-contaminated soils.Phylogenetic analysis of amoA gene-encoded proteins revealed that all AOB clone sequences belonged to the genus Nitrosospira,among which those belonging to Nitrosospira cluster 3a were dominant.The main AOA sequence detected belonged to Thaumarchaeal Group 1.1b,which was considered to have a high ability to adapt to environmental changes.Our results provide new insights into the impacts of arsenic on the soil nitrogen cycling.Yanan Wang Xibai Zeng Yang Zhang Nan Zhang Liyang Xu Cuixia Wu 2023Journal of Environmental Sciences2023,,5:0
18Fabrication of SA/Gel/C scaffold with 3D bioprinting to generate micro-nano porosity structure for skin wound healing: a detailed animal in vivo study显示文摘Bioprinting has exhibited remarkable promises for the fabrication of functional skin substitutes.However,there are some significant challenges for the treatment of full-thickness skin defects in clinical practice.It is necessary to determine bioinks with suitable mechanical properties and desirable biocompatibilities.Additionally,the key for printing skin is to design the skin structure optimally,enabling the function of the skin.In this study,the full-thickness skin scaffolds were prepared with a gradient pore structure constructing the dense layer,epidermis,and dermis by different ratios of bioinks.We hypothesized that the dense layer protects the wound surface and maintains a moist environment on the wound surface.By developing a suitable hydrogel bioink formulation(sodium alginate/gelatin/collagen),to simulate the physiological structure of the skin via 3D printing,the proportion of hydrogels was optimized corresponding to each layer.These results reveal that the scaffold has interconnected macroscopic channels,and sodium alginate/gelatin/collagen scaffolds accelerated wound healing,reduced skin wound contraction,and re-epithelialization in vivo.It is expected to provide a rapid and economical production method of skin scaffolds for future clinical applications.Changmei Niu Liyang Wang Dongdong Ji Mingjun Ren Dongxu Ke Qiang Fu Kaile Zhang Xi Yang 2022Cell Regeneration2022,11,1:0
19Study on the Photosynthetic Characteristics of Six Varieties(Strains)in Chinese Chestnut显示文摘[Objectives]This study was conducted to investigate the differences of photosynthetic physiological characteristics of different varieties(strains),which will provide a theoretical basis for high photosynthesis efficiency breeding and application in Chinese chestnut.[Methods]Six Chinese chestnut varieties of Castanea mollissima‘Yanbao’,C.mollissima‘Yanqiu’,C.mollissima‘Yanchang’,C.mollissima‘Yanjia’,C.mollissima‘Qianxi 37’,and C.mollissima‘Hybrid 22’were used as the materials.Using the portable photosynthesis system Li-6400,we measured the photosynthetic characteristics and diurnal variation of leaf samples of six different chestnut varieties or strains.We fitted the light response curves and photosynthetic parameters using the leaf floating model.Additionally,we determined the chlorophyll content in the leaves using a UV-visible spectrophotometer.[Results]Among the six chestnut varieties or strains,‘Yanqiu’exhibited a significantly higher photosynthetic light saturation point(P LSP)compared to other five varieties,and‘Hybrid 22’ranked second,indicating that these two varieties had the strongest adaptation to high light intensity.The photosynthetic light compensation point(P LCP)of‘Yanchang’was significantly higher than other five varieties,and'Qianxi 37'ranked second,indicating that these two varieties had the strongest adaptation to low light intensity.Additionally,they exhibited higher chlorophyll content and maintained good photosynthetic characteristics even in shaded environments with weak light stress.Varieties‘Yanbao’and‘Yanjia’showed higher P LSP and lower P LCP,indicating that these two varieties have a wider range of adaptation to light intensity.They were capable of efficiently utilizing light across a broader spectrum of intensities.‘Yanqiu’had the highest maximum net photosynthetic rate(P n,max)and the lowest dark respiration rate(R d),along with the highest chlorophyll content.It indicated that‘Yanqiu’has strong photosynthetic capacity and organic matter accumulation ability.It also had the highest P LSP,enabling it to fully utilize the high light environment of the Yanshan Mountains and possessed high light efficiency characteristics.The P n,max of‘Yanqiu’was significantly higher than other varieties.‘Hybrid 22’and‘Yanbao’also exhibited significantly higher P n,max compared with‘Yanjia’and‘Qianxi 37’.‘Yanchang’had the lowest P n,max.The order of P n,max among the six chestnut varieties or strains was as follows:‘Yanqiu’>‘Hybrid 22’>‘Yanbao’>‘Yanjia’>‘Qianxi 37’>‘Yanchang’.[Conclusions]Lizhi FU Yong YANG Xiaoxiao KANG Meng WANG Jing LIU Xuan WANG Dongsheng WANG Liyang YU Hai’e ZHANG 2024Agricultural Biotechnology2024,13,1:0
20Reverse engineering a predictive signature characterized by proliferation,DNA damage,and immune escape from stage I lung adenocarcinoma recurrence显示文摘Identifying early-stage cancer patients at risk for progression is a major goal of biomarker research.This report describes a novel 19-gene signature(19-GCS)that predicts stage I lung adenocarcinoma(LAC)recurrence and responseto therapy and performs comparably in pancreatic adenocarcinoma(PAC),which shares LAC molecular traits.Kaplan-Meier,Cox regression,and cross-validation analyses were used to build the signature from training,test,and validation sets comprising 831 stage I LAC transcriptomes from multiple independent data sets.A statistical analysis was performed using the R Ianguage.Pathway and gene set enrichment were used to identify underlying mechanisms.19-GCS strongly predicts overall survival and recurrence-free survival in stage I LAC(P=0.002 and P<0.001,respectively)and in stage l-ll PAC(P<0.0001 and Pv0.0005,respectively).A multivariate cox regression analysis demonstrated the independence of 19-GCS from significant clinical factors.Pathway analyses revealed that 19-GCS high-risk LAC and PAC tumors are characterized by increased proliferation,enhanced sternness,DNA repair deficiency,and compromised MHC class I and II antigen presentation along with decreased immune infiltration.Importantly,high-risk LAC patients do not appear to benefit from adjuvant cisplatin while PAC patients derive additi onal ben efit from FOLFIRINOX compared with gemcitabi ne-based regimens.Whe n validated prospectively,this proof-of-c on cept biomarker may con tribute to tailoring treatment,recurrence reduction,and survival improvements in early-stage lung and pancreatic cancers.Jiannan Yao Xinying Xue Dongfeng Qu C.Benedikt Westphalen Yang Ge Liyang Zhang Manyu Li Tianbo Gao Parthasarathy Chandrakesan Kenneth J.Vega Jun Peng Guangyu An Nathaniel Weygant 2020Acta Biochimica et Biophysica Sinica2020,52,6:0
