3Preparation,characterization and biological properties of a novel bone block composed of platelet rich fibrin and a deproteinized bovine bone mineral显示文摘Alveolar bone defects caused by tooth loss often lead to challenges in implant dentistry,with a need for development of optimal bone biomaterials to predictably rebuild these tissues.To address this problem,we fabricated a novel bone block using platelet-rich fibrin(PRF)and Deproteinized Bovine Bone Mineral(DBBM),and characterized their mechanical and biological properties.The bone block was prepared by mixing DBBM,Liquid-PRF,and Solid-PRF fragments in various combinations as follows:(1)BLOCK-1 made with Solid-PRF fragments+DBBM,(2)BLOCK-2 made with Liquid-PRF+DBBM,(3)BLOCK-3 made with Solid-PRF fragments+Liquid-PRF+DBBM.The time for solidification and the degradation properties were subsequently recorded.Scanning electron microscopy(SEM)and tensile tests were carried out to investigate the microstructure and mechanical properties of each block.The bioactivity of the three groups towards osteoblast differentiation was also evaluated by culturing cells with the conditioned medium from each of the three groups including cell proliferation assay,cell migration assay,alkaline phosphatase(ALP)staining,and alizarin red staining(ARS),as well as by real-time PCR for genes encoding runt-related transcription factor 2(RUNX2),ALP,collagen type I alphal(COLIv41)and osteocalcin(OCN).BLOCK-3 made with Solid-PRF fragments+Liquid-PRF+DBBM had by far the fastest solidification period(over a 10-fold increase)as well as the most resistance to degradation.SEM and tensile tests also revealed that the mechanical properties of BLOCK-3 were superior in strength when compared to all other groups and further induced the highest osteoblast migration and osteogenic differentiation confirmed by ALP,ARS and real-time PCR.PRF bone blocks made through the combination of Solid-PRF fragments+Liquid-PRF+DBBM had the greatest mechanical and biological properties when compared to either used alone.Future clinical studies are warranted to further support the clinical application of PRF bone blocks in bone regeneration procedures.Mengge Feng Yulan Wang Yan Wei Xiaoxin Zhang Leyi Xiao Zijjian Gong Masako Fujioka-Kobayashi Anton Sculean Richard J Miron Scott Froum Yufeng Zhang 2022Fundamental Research2022,2,2:3

5Microstructure characteristics of 12Cr ferritic/martensitic steels with various yttrium显示文摘12Cr ferritic/martensitic steels with 0, 0.1 wt%, 0.2 wt% and 0.3 wt% theoretical yttrium(Y) additions were fabricated by vacuum inducting melting and casting method. Solubilities of Y in the 12Cr steels are0.027, 0.078 and 0.17 for 12Cr-0.1 Y, 12Cr-0.2 Y and 12Cr-0.3 Y, respectively. Phase transformations and microstructure characteristics under different heat-treatment schedules were investigated. The starting temperature of ferrite-to-austenite transformation A^(c1) are maintained about 850℃, but the finishing temperature of ferrite-to-austenite transformation A^(c3) are about 950, 970, 980 and 1000℃ for 12Cr-0 Y,12Cr-0.1 Y, 12Cr-0.2 Y and 12Cr-0.3 Y, respectively, which indicates that A^(c3) increases gradually with the addition of Y. Martensite accompanied with a few δ-ferrite is the dominant structure in all the steels. The amount of δ-ferrite shows a strong dependence with the Y content and austenitizing temperature. Area fraction of δ-ferrite increases with the content of Y, which is the ferrite favouring element. The minimum amount of δ-ferrite are achieved at 950℃ for 12Cr-0 Y, 12Cr-0.1 Y, 12Cr-0.2 Y and 1000℃ for 12Cr-0.3 Y.Besides, more carbides precipitate along the martensite laths and grain boundaries in the Y-bearing steel due to the redistribution of carbon between austenite and ferrite resulting from the ferrite favouring element of Y.Yingxue Chen Feifei Zhang Qingzhi Yan Xiaoxin Zhang Zhiyuan Hong 2019Journal of Rare Earths2019,37,5:3
6Beta-amyloid precursor protein cleavage enzyme-1 expression in adult rat retinal neurons in the early period after lead exposure显示文摘Previous studies have reported that non-human primates and rodents exposed to lead during brain development may become dependent on the deposition of pre-determined β-amyloid protein (Aβ),and exhibit upregulation of β-site amyloid precursor protein expression in old age.However,further evidence is required to elucidate the precise relationship and molecular mechanisms underlying the effects of early lead exposure on excessive Aβ production in adult mammals.The present study investigated the effects of lead exposure on expression of β-amyloid precursor protein cleavage enzyme-1 (BACE-1) in the rat retina and the production of Aβ in early development,using the retina as a window for studying Alzheimer's disease.Adult rats were intraocularly injected with different doses of lead acetate (10μmol/L,100μmol/L,1 mmol/L,10 mmol/L and 100 mmol/L).The results revealed that retinal lead concentration,BACE-1 and its cleavage products β-C-terminal fragment and retina Aβ1-40 were all significantly increased in almost all of the lead exposure groups 48 hours later in a dose-dependent manner.The only exception was the 10μmol/L group.The distribution of BACE-1 in the retina did not exhibit obvious changes,and no distinctive increase in the activation of retinal microglia was apparent.Similarly,retinal synaptophysin expression did not exhibit any clear changes.These data suggest that lead exposure can result in the upregulation of retinal neuron BACE-1 expression in the early period of development and further increase the overproduction of Aβ1-40 in the retina.Our results provided novel insight into the molecular mechanisms underlying environmentally-induced Alzheimer's disease.Jufang Huang Kai Huang Lei Shang Hui Wang Xiaoxin Yan Kun Xiong 2011Neural Regeneration Research2011,6,14:3

8REMOTE SENSING OF ALGAL BLOOMS USING A TURBIDITY-FREE FUNCTION FOR NEAR-INFRARED ANDRED SIGNALS显示文摘This article presents a method for real- time mapping of algal blooms in turbid coastal waters using the remote sensing reflectance of red band (Channel 1, 580-680 nm) and near-infrared band (Channel 2, 720-1100 nm) of the AVHRR sensor on the NOAA series satellites. A turbidity-free function for near-infrared and red signals, α0 = (bb(1)/bb(2))(a(2)/ a(1)) based on the first order bb/(a+bb) model deduc-ing equation Rrs(2)?1 = α0 Rrs(1) ?1+ g?1 (1?α0), were se-lected as a chlorophyll-a related index for detecting algal blooms, and the algal blooms with chlorophyll-a concentration of 64-256 mg/L could be defined by window of 1.6 < α0 < 5.2 and 0.01< Rrs(2)/g < 0.2. Such turbidity-free two-band method is supported by both sea-truth data and remote sensing experiment for an algal blooms event on the near-shore water off the Minjiang estuary of southeastern China during early June of 2003. 14The Effect of Solar Cycle on Climate of Northeast Asia显示文摘The impact of solar activity on climate system is spatiotemporally selective and usually more significant on the regional scale. Using statistical methods and solar radio flux(SRF) data, this paper investigates the impact of the solar11-yr cycle on regional climate of Northeast Asia in recent decades. Significant differences in winter temperature,precipitation, and the atmospheric circulation over Northeast Asia are found between peak and valley solar activity years. In peak years, temperature is higher over vast areas of the Eurasian continent in middle and high latitudes, and prone to producing anomalous high pressure there. Northeast Asia is located to the south of the anomalous high pressure, where the easterlies prevail and transport moisture from the western Pacific Ocean to the inland of East Asia and intensify precipitation there. In valley years, temperature is lower over the Eurasian continent and northern Pacific Ocean in middle and high latitudes, and there maintain anomalous low pressure systems in the two regions. Over the Northeast Asian continent, north winds prevail, which transport cold and dry air mass from the high latitude to Northeast Asia and reduce precipitation there. The correlation coefficient of winter precipitation in Northeast China and SRF reaches 0.4, and is statistically significant at the 99% confidence level based on the Student's t-test. 15Functional evaluation of iodoacetic acid induced photoreceptor degeneration in the cat显示文摘Iodoacetic acid(IAA) has been applied to different species to acutely induce photoreceptor degeneration.The purpose of the present study was to use this toxin to thoroughly eliminate photoreceptors and induce complete blindness in the cat.IAA was delivered by single ear vein injection(20 mgkg-1).Six months after the IAA treatment,functional evaluations including pupillary light reflex(PLR),electroretinogram(ERG),visual behavior tests were performed.Morphological examinations were carried out after the functional evaluation.The present result shows that,six months after the IAA application,animals lost visual functions and became completely blind.High dose IAA application via ear vein delivery created an acute and reliable complete photoreceptor degeneration model in the cat.This model can be applied to genetic and cellular therapies for visual function restoration.NAN Yan ZHANG Qin REN ChaoRan HUANG Xin GAO Jie LI XiaoXin PU MingLiang 2013Science China(Life Sciences)2013,56,6:0
16Catalytic ozonation in advanced treatment of kitchen wastewater:multi-scale simulation and pilot-scale study显示文摘Catalytic ozonation is regarded as a promising technology in the advanced treatment of refractory organic wastewater.Packed-bed reactors are widely used in practical applications due to simple structures,installation and operation.However,mass transfer of packed-bed reactors is relatively restrained and amplified deviations usually occurred in scale-up application.Herein,a multi-scale packed-bed model of catalytic ozonation was established to guide pilot tests.First,a laboratory-scale test was conducted to obtain kinetic parameters needed for modeling.Then,a multi-scale packed-bed model was developed to research the effects of water distribution structure,catalyst particle size,and hydraulic retention time(HRT)on catalytic ozonation.It was found that the performance of packed bed reactor was increased with evenly distributed water inlet,HRT of 60 min,and catalyst diameter of about 3-7 mm.Last,an optimized reactor was manufactured and a pilot-scale test was conducted to treat kitchen wastewater using catalytic ozonation process.In the pilot-scale test with an ozone dosage of 50 mg/L and HRT of 60 min,the packed-bed reactor filled with catalysts I was able to reduce chemical oxygen demand(COD)from 117 to 59 mg/L.The performance of the catalytic ozonation process in the packed-bed reactor for the advanced treatment of actual kitchen wastewater was investigated via both multi-scale simulation and pilot-scale tests in this study,which provided a practical method for optimizing the reactors of treating refractory organic wastewater.Zuoyong Zhou Ni Yan Mengxi Yin Tengfei Ren Shuning Chen Kechao Lu Xiaoxin Cao Xia Huang Xiaoyuan Zhang 2023Frontiers of Environmental Science & Engineering2023,17,12:0
17Characterizing and engineering promoters for metabolic engineering of Ogataea polymorpha显示文摘Bio-manufacturing via microbial cell factory requires large promoter library for fine-tuned metabolic engi-neering.Ogataea polymorpha,one of the methylotrophic yeasts,possesses advantages in broad substrate spec-trum,thermal-tolerance,and capacity to achieve high-density fermentation.However,a limited number of available promoters hinders the engineering of O.polymorpha for bio-productions.Here,we systematically characterized native promoters in O.polymorpha by both GFP fluorescence and fatty alcohol biosynthesis.Ten constitutive promoters(P_(PDH),P_(PYK),P_(FBA),P_(PGM),P_(GLK),P_(TRI),P(GPI),P_(ADH1),P_(TEF1) and P_(GCW14))were obtained with the activity range of 13%–130% of the common promoter P_(GAP)(the promoter of glyceraldehyde-3-phosphate de-hydrogenase),among which P_(PDH) and P_(GCW14) were further verified by biosynthesis of fatty alcohol.Furthermore,the inducible promoters,including ethanol-induced P_(ICL1),rhamnose-induced P_(LRA3) and P_( LRA4),and a bidirectional promoter(P_(Mal)-P_(Per))that is strongly induced by sucrose,further expanded the promoter toolbox in O.polymorpha.Finally,a series of hybrid promoters were constructed via engineering upstream activation sequence(UAS),which increased the activity of native promoter P LRA3 by 4.7–10.4 times without obvious leakage expression.Therefore,this study provided a group of constitutive,inducible,and hybrid promoters for metabolic engineering of O.polymorpha,and also a feasible strategy for rationally regulating the promoter strength.Chunxiao Yan Wei Yu Xiaoxin Zhai Lun Yao Xiaoyu Guo Jiaoqi Gao Yongjin J.Zhou 2022Synthetic and Systems Biotechnology2022,7,1:0

19Metabotropic glutamate receptors(mGluRs)in epileptogenesis:an update on abnormal mGluRs signaling and its therapeutic implications显示文摘Epilepsy is a neurological disorder characterized by high morbidity,high recurrence,and drug resistance.Enhanced signaling through the excitatory neurotransmitter glutamate is intricately associated with epilepsy.Metabotropic glutamate receptors(mGluRs)are G protein-coupled receptors activated by glutamate and are key regulators of neuronal and synaptic plasticity.Dysregulated mGluR signaling has been associated with various neurological disorders,and numerous studies have shown a close relationship between mGluRs expression/activity and the development of epilepsy.In this review,we first introduce the three groups of mGluRs and their associated signaling pathways.Then,we detail how these receptors influence epilepsy by describing the signaling cascades triggered by their activation and their neuroprotective or detrimental roles in epileptogenesis.In addition,strategies for pharmacological manipulation of these receptors during the treatment of epilepsy in experimental studies is also summarized.We hope that this review will provide a foundation for future studies on the development of mGluR-targeted antiepileptic drugs.Leyi Huang Wenjie Xiao Yan Wang Juan Li Jiaoe Gong Ewen Tu Lili Long Bo Xiao Xiaoxin Yan Lily Wan 2024Neural Regeneration Research2024,19,2:0
