1PD-L1 is a direct target of cancer-FOXP3 in pancreatic ductal adenocarcinoma(PDAC),and combined immunotherapy with antibodies against PD-L1 and CCL5 is effective in the treatment of PDAC显示文摘High expression of PD-L1 marks the poor prognosis of pancreatic ductal adenocarcinomas(PDAC).However,the regulatory mechanism of PD-L1 remains elusive.We recently reported that cancer Forkhead box protein 3(Cancer-FOXP3 or C-FOXP3)promoted immune evasion of PDAC by recruiting Treg cells into PDAC via upregulation of CCL5.In this study,we confirmed that PD-L1 was overexpressed in PDAC samples from two independent cohorts of patients with radical resection.Moreover,C-FOXP3 was colocalized and correlated with the expression of PD-L1 in tumor cells at the mRNA and protein levels,and this finding was confirmed by the The Cancer Genome Atlas(TCGA)database.Chromatin immunoprecipitation(ChIP)revealed that C-FOXP3 directly bound to the promoter region of PD-L1 in pancreatic cancer cells.Furthermore,overexpression of C-FOXP3 activated the luciferase reporter gene under the control of the PD-L1 promoter.However,mutation of the binding motif-a completely reversed the luciferase activity.In addition,C-FOXP3-induced upregulation of PD-L1 effectively inhibited the activity of CD8+T cells.Based on our recent finding that the CCL-5 antibody achieved a better response to PDAC models with high C-FOXP3 levels,we further demonstrated that the PD-L1 antibody strengthened the antitumor effect of CCL-5 blockade in xenograft and orthotopic mouse models with high C-FOXP3 levels.In conclusion,C-FOXP3 directly activates PD-L1 and represents a core transcription factor that mediates the immune escape of PDAC.Combined blockade of PD-L1 and CCL-5 may provide an effective therapy for patients with PDAC that have high C-FOXP3 levels.Xiuchao Wang Xin Li Xunbin Wei Haiping Jiang Chungen Lan Shengyu Yang Han Wang Yanhui Yang Caijuan Tian Zanmei Xu Jiangyan Zhang Jihui Hao He Ren 2020Signal Transduction and Targeted Therapy2020,5,1:11
2肠系膜上静脉血管鞘内入路在右半结肠切除术中的临床研究(英文)显示文摘Objective: The aim of the study was to investigate the clinical value of superior mesenteric vascular intrathecal approach in right hemicolectomy. Methods: We retrospectively studied the clinical data of 132 patients who had right hemicolectomy from June 2007 to June 2010, including 68 cases with superior mesenteric vascular intrathecal approach to resect specimen, and compared the operation time, blood loss, hospital stay and the number of dissected lymph nodes with patients treated with conventional surgery. Results: Compared the vascular intrathecal approach with conventional approach, the operation time and blood loss were decreased significantly, the number of Dukes C No. 3 lymph node dissection was increased, while the incidence of postoperative complications and hospital stay were equivalent to traditional surgery group. 3Understanding the structural evolution of Au/WO_(2.7) compounds in hydrogen atmosphere by atomic scale in situ environmental TEM显示文摘Hydrogen energy is a resuscitated clean energy source and its sensitive detection in air is crucial due to its very low explosive limit.Metal oxide decorated with noble metal nanoparticles has been used for the enhancement of gas detection and exhibits superior sensitivity.Understanding the intrinsic mechanism of the detection and the enhancement mechanism is thus becoming a fundamental issue for the further development of novel metal/oxide compound gas-sensing materials.However,the correlation between the microstructural evolution,the charge transport and the complex sensing process has not yet been directly revealed and its atomic mechanism is still debatable.In this study,an Au/WO_(2.7) compound was synthesized and exhibited a strongly enhanced gas sensitivity to many reductive gases,especially H2.Aberration-corrected environmental transmission electron microscopy was used to investigate the atomic-scale microstructural evolution in situ during the reaction between H_(2) and Au/WO_(2.7) compound.Swing and sintering processes of the Au particles on the WO_(2.7) surface were observed under heating and gaseous environments,and no injection of hydrogen atoms was suggested.First principle calculations verified the swing and sintering processes,and they can be explained by the enhancement of H2 sensitivity.Fei Hui Chong Li Yanhui Chen Chunhui Wang Jingping Huang Ang Li Wei Li Jin Zou Xiaodong Han 2020Nano Research2020,13,11:6
4miR-23b promotes cutaneous wound healing through inhibition of the inflammatory responses by targeting ASK1显示文摘Wound healing is a complicated event that develops in three overlapping phases:inflammatory, proliferative,and remodeling.MicroRNAs (miRNAs)have been proved to play an important role in the healing process of skin trauma,and alteration of specific miRNA expression during different phases may be associated with abnormal wound healing.In this study,we determined the variation of miR-23b expression after trauma in normal mice and in cultured cells exposed to lipopolysaccharide.We further demonstrated that excessive miR-23b could significantly accelerate wound healing in vivo.Up-regulation of miR-23b decreases infiltration of inflammatory cells,as evidenced by pathologic staining.Meanwhile,miR-23b could significantly inhibit the expression of pro-inflammatory cytokines,including TNF-α,IL-1β,IL-6,and Ccl2,and significantly increase antiinflammatory factor IL-10.Furthermore,miR-23b could also promote α-SMA expression in a fiber pattern and increase the expression of Col1a1 and Col3a1.Importantly,we also showed that miR- 23b could inhibit inflammation to promote wound healing by targeting apoptotic signal-regulating kinase 1 (ASK1).Notably,knockdown of ASK1 could reduce inflammation factor expression in vitro.Together,our data reveal that miR-23b is a potent therapeutic agent for cutaneous wound healing that shortens the period of inflammatory responses and promotes keratinocyte migration for the re-epithelialization of wound sites.Hongzhi Li Xiao Han Kuiyang Zuo Li Li Jieting Liu Xiaohuan Yuan Yongchao Shen Minglong Shao Daxin Pang Yanhui Chu Binghai Zhao 2018Acta Biochimica et Biophysica Sinica2018,50,11:5
5Deep origin of Cenozoic volcanoes in Northeast China revealed by 3-D electrical structure显示文摘The three-dimensional(3-D)electrical structure of the upper-mantle was used to examine the deep origins of and relationship among the Cenozoic volcanoes located in Northeast China(NEC).High-quality,long-period magnetotelluric(LMT)full-impedance tensor data were collected in NEC and subjected to 3-D Gauss-Newton inversion in order to construct a resistivity model.The resulting model reveals the presence of multiple localized low-resistivity anomalies(LRAs)within the high resistivity lithosphere beneath NEC.These LRAs partially coincide with Cenozoic volcanoes on the surface.Three LRAs that form a larger,annular LRA were observed in the deep upper mantle beneath the Songliao Basin,whereas vein-like LRAs were found in the asthenosphere that connect the lithosphere and deep upper mantle.Petrophysical analyses suggest that the LRAs may have been caused by fluid-induced melting.Based on our electrical model,we propose that,following dehydration of the subducted Western Pacific slab into the mantle transition zone(MTZ)beneath NEC,the released water migrated upward and caused partial melting at the top of the MTZ beneath the Songliao Basin.Under the effect of buoyancy,the melted mantle formed a thermal upwelling that caused melting of asthenosphere before diapiring at the base of the dry lithosphere.The magma then penetrated structural boundaries(such as thinner,weaker,or activated suture zones)and finally reached the Earth's surface.This melting and upwelling of hot mantle materials may have resulted in large-scale volcanism in the region throughout the Cenozoic,including the eruption of Changbai Mountain and Halaha Volcanoes.Our results suggest that the Cenozoic NEC volcanoes may all share a similar mode of genesis,and probably originated from the annular LRA in the deep upper mantle.Shiwen LI Aihua WENG Jianping LI Xuanlong SHAN Jiangtao HAN Yu TANG Yanhui ZHANG Xueqiu WANG 2020Science China Earth Sciences2020,63,4:4
6Expression of hippocampal corticosteroid receptors,as well as corticotrophin-releasing hormone and vasopressin in the hypothalamic paraventricular nucleus,in fornix transected rats显示文摘BACKGROUND:The hippocampus regulates the hypothalamic-pituitary-adrenal axis through negative feedback.The hypothalamic paraventricular nucleus receives neuronal input from the hippocampus via the fornix. OBJECTIVE:To explore whether the negative feedback effect of the hippocampus on the hypothalamic-pituitary-adrenal axis is contributed to the inhibitory effect of mineralocorticoid receptor (MR) and glucocorticoid receptor(GR) in the hippocampus on the paraventricular nucleus via the fornix. DESIGN,TIME AND SETTING:Randomized,controlled,animal experiment.The study was performed at the Department of Histology and Embryology,China Medical University between September 2006 and September 2008. MATERIALS:Rabbit anti-rat anti-MR and rabbit anti-rat anti-GR antibodies were purchased from Santa Cruz Biotechnology,USA.Rabbit anti-rat anti-corticotrophin releasing hormone(CRH) and rabbit anti-rat anti-arginine vasopressin antibodies were purchased from Wuhan Boster. METHODS:A total of 90 male,Wistar rats were randomly divided into model and sham-surgery groups(n=45).Fornix transection was performed in the model group,while the sham-surgery group underwent surgery,but no fornix transection. MAIN OUTCOME MEASURES:Immunohistochemistry was used to examine MR and GR expression in the hippocampus,as well as CRH and anti-arginine vasopressin in the paraventricular nucleus.Western blot was used to measure alterations in MR,GR,and CRH protein expression following fomix transection. RESULTS:Compared with the sham-surgery group,there were no obvious changes in MR and GR expression in the hippocampus,or CRH and anti-arginine vasopressin expression in the paraventricular nucleus within 4 days of fornix transection.However,after 7-10 days,significantly decreased MR and GR expression in the hippocampus,and increased CRH and anti-arginine vasopressin expression in the paraventricular nucleus were observed(P<0.05-0.01). 8Verification of two-dimensional LBM-DEM coupling approach and its application in modeling episodic sand production in borehole显示文摘The lattice Boltzmann method(LBM)is implemented in the Particle Flow Code(PFC)as a pore-scale CFD module and coupled with the particulate discrete element assemblage in PFC using an immersed boundary scheme.The implementation of LBM and LBM-PFC coupling is validated with the analytical solutions in a couple of hydrodynamics and fluid-particle interaction problems,i.e.,the accuracy of LBM as a CFD solver is verified by solving channel flow driven by a pressure gradient for which the closed-form solution is also derived;the accuracy of LBM-PFC coupling is validated by solving flow across a cylinder,Taylor-Couette flow,Karman vortex street,and fluid flow through a cylinder array.To demonstrate potential applications of this coupling code,a perforation cavity subjected to axial fluid flush is then tested,showing that the collapse and reconstruction of sand arch in the perforation cavity can be reproduced in this coupling system.The developed system is ready for exploring more complicated physical issues involved in sand production.Yanhui Han Peter Cundall 2017Petroleum2017,3,2:4
9Synthesis of the superfine high-entropy zirconate nanopowders by polymerized complex method显示文摘The high-purity and superfine high-entropy zirconate nanopowders,namely(Y_(0.25)La_(0.25)Sm_(0.25)Eu_(0.25))_(2)Zr_(2)O_(7)nanopowders,without agglomeration,were successfully synthesized via polymerized complex method at low temperatures for the first time.The results showed that the crystallinity degree,lattice strain,and particle size of the as-synthesized powders were gradually enhanced with the increase of the synthesis temperature from 800 to 1300℃.The as-synthesized powders involved fluorite phase in the range of 800-1200℃while they underwent the phase evolution from fluorite to pyrochlore at 1300℃.It is worth mentioning that the as-synthesized powders at 900℃are of the highest quality among all the as-synthesized powders,which is due to the fact that they not only possess the particle size of 11 nm without agglomeration,but also show high purity and good compositional uniformity.Yangjie HAN Renwang YU Honghua LIU Yanhui CHU 2022Journal of Advanced Ceramics2022,11,1:2
10Research progress of semiconductive shielding layer of HVDC cable显示文摘As an indispensable part of high-voltage direct current(HVDC)cable,the semiconductive shielding layer plays the role of uniforming electric field in the cable.However,cable shielding materials>35 kV mainly rely on foreign imports in China,which belongs to the technical weak issues in the field of electrical materials.At present,there are few systematic reports on semiconductive shielding material of HVDC cable.In the work,the mechanisms of charge conduction and thermal conduction of semiconductive material have been introduced.Effect of raw material,carbon black content and the second conductive filler on the resistance characteristics of the semiconductive layer,the charge accumulation characteristics of the insulating layer and the interface characteristics have been studied.A kind of semiconductive layer as a high voltage terminal charge emission method has been proposed to study charge emission from the semiconducting layer to the insulation layer.This work can provide theoretical guidance for the research of semiconductive shielding materials.Yanhui Wei Wang Han Guochang Li Qingquan Lei Mingli Fu Chuncheng Hao Guanjun Zhang 2020High Voltage2020,5,1:2
11The Notch pathway attenuates burn-induced acute lung injury in rats by repressing reactive oxygen species显示文摘Background:Acute lung injury(ALI)is a common complication following severe burns.The underlying mechanisms of ALI are incompletely understood;thus,available treatments are not sufficient to repair the lung tissue after ALI.Methods:To investigate the relationship between the Notch pathway and burn-induced lung injury,we established a rat burn injury model by scalding and verified lung injury via lung injury evaluations,including hematoxylin and eosin(H&E)staining,lung injury scoring,bronchoalveolar lavage fluid and wet/dry ratio analyses,myeloperoxidase immunohistochemical staining and reac-tive oxygen species(ROS)accumulation analysis.To explore whether burn injury affects Notch1 expression,we detected the expression of Notch1 and Hes1 after burn injury.Then,we extracted pulmonary microvascular endothelial cells(PMVECs)and conducted Notch pathway inhibition and activation experiments,via aγ-secretase inhibitor(GSI)and OP9-DLL1 coculture,respectively,to verify the regulatory effect of the Notch pathway on ROS accumulation and apoptosis in burn-serum-stimulated PMVECs.To investigate the regulatory effect of the Notch pathway on ROS accumulation,we detected the expression of oxidative-stress-related molecules such as superoxide dismutase,nicotinamide adenine dinucleotide phosphate(NADPH)oxidase(NOX)2,NOX4 and cleaved caspase-3.NOX4-specific small interfering RNA(siRNA)and the inhibitor GKT137831 were used to verify the regulatory effect of the Notch pathway on ROS via NOX4.Results:We successfully established a burn model and revealed that lung injury,excessive ROS accumulation and an inflammatory response occurred.Notch1 detection showed that the expression of Notch1 was significantly increased after burn injury.In PMVECs challenged with burn serum,ROS and cell death were elevated.Moreover,when the Notch pathway was suppressed by GSI,ROS and cell apoptosis levels were significantly increased.Conversely,these parameters were reduced when the Notch pathway was activated by OP9-DLL1.Mechanistically,the inhibition of NOX4 by siRNA and GKT137831 showed that the Notch pathway reduced ROS production and cell apoptosis by downregulating the expression of NOX4 in PMVECs.Conclusions:The Notch pathway reduced ROS production and apoptosis by downregulating the expression of NOX4 in burn-stimulated PMVECs.The Notch-NOX4 pathway may be a novel therapeutic target to treat burn-induced ALI.Weixia Cai Kuo Shen Peng Ji Yanhui Jia Shichao Han Wanfu Zhang Xiaolong Hu Xuekang Yang Juntao Han Dahai Hu 2022Burns & Trauma2022,10,1:2

13The Effect of Nitrogen Deposition on Soil Microorganisms in the Woodland-Grassland Border of Tibet显示文摘Nitrogen deposition was simulated from July 2014 to August 2015 in the grassland, woodland, and woodland-grassland border in Zhuqudeng Village, Bujiu Township, Linzhi County,(CK, 0 kg·hm^2·a^(-1); LN, 25 kg·hm^2·a^(-1), MN, 50 kg·hm^2·a^(-1); HN, 150 kg·hm^2·a^(-1)). NH_4NO_3 was used as nitrogen source to analyze the number of microorganisms in soil layers of 0–20 cm and 20–40 cm and explore the effect of different degrees of nitrogen deposition on soil microorganisms in grassland, woodland, and woodlandgrassland border. 16Water Holding Characteristics of Coarse Woody Debris of Abies fabri Forest on the East Slope of Tibet Sygera Mountain显示文摘Coarse woody debris( CWD) has an important role in hydrological function and water and soil conservation function of forest ecosystem especially mountain forest ecosystem. To understand water holding capacity of CWD in subalpine forest of southeast Tibet,CWD reserve of primitive A. fabri forest in Sygera Mountain was investigated by sample plot method,and water absorption and storage capacity of CWD was studied by using soaking method. The results showed that decay grade was positively related to water storage potential,that is to say,the higher the decay grade,the larger the water storage potential. Saturated water holding capacity,saturated water holding rate,effective water holding capacity and effective water holding rate of A. fabri CWD all rose with decay grade increased,and the order was Ⅴ>Ⅳ>Ⅲ>Ⅱ>Ⅰ. 17Short-term Effects of Nitrogen Deposition on Soil Physical and Chemical Properties of Tibetan Forest-Grassland Landscape Boundary显示文摘[Objective] The paper was to study the effects of nitrogen deposition on soil nutrients and soil dissolved organic carbon in forest-grass-land landscape in Linzhi, Tibet, and to provide scientific basis and basic data for understanding and assessing the effect of atmospheric nitrogen de-position on soil nutrients and soil dissolved organic carbon. [Method] From July 2014 to August 2015, in situ nitrogen deposition(CK 0 kg·hm^2/a, LN25 kg·hm^2/a, MN 50 kg·hm^2/a, HN 150 kg·hm^2/a) was simulated in the forest-grassland boundary of Zhuqudeng village, Bujiu Township, Linzhi County, Tibet. The soil samples were collected for analyzing nutrient and dissolved contents in the soil layer of 0-20 and 20-40 cm. The effects of different nitrogen deposition levels on soil nutrients and dissolved organic carbon(DOC) were studied. [Result] Nitrogen deposition had significant impacts on soil organic matter, total N, total P, total K, available N, available P, available K, exchangeable Ca, exchangeable Mg, p H, and DOC( P<0.05).(2) With the deepening of nitrogen deposition from CK, LN, MN to HN in the 0-20 cm boundary soil, the contents of organic matter, total N,total P, available P, exchangeable Ca, exchangeable Mg and DOC kept decreasing, and the content of total K and available N increased continuous-ly. The p H increased in LN treatment and decreased in HN treatment, while the available K content was decreased in LN and HN treatment, but increased in MN treatment.(3) The contents of organic matter, total N, total P, available N, available P, exchangeable Ca, exchangeable Mg and DOC all decreased at the soil layer of 20-40 cm under the same nitrogen deposition. The p H increased in LN treatment, but decreased in HN treat-ment; the content of total K decreased in LN treatment and increased in MN and HN treatments; the content of available K decreased in LN and HN treatments, but increased in MN treatment.(4) With the deepening of boundary soil layer(0-20 to 20-40 cm), the organic matter, total N, total P, available P, available K, exchangeable Ca, exchangeable Mg, DOC showed the same response to simulated nitrogen deposition, while the avail-able N and total K responded differently.
