| 3 | MK-0626,a selective DPP-4 inhibitor,attenuates hepatic steatosis in ob/ob mice显示文摘AIM:To investigate the mechanism and in vivo effects of MK-0626,a dipeptidyl peptidase-4 inhibitor,on hepatic steatosis using ob/ob mice.METHODS:We analyzed obese(ob/ob)8-wk-old male mice that had been randomly divided into two groups of ob/ob mice(n=16 each)and were treated with1.5 or 3 mg/kg MK-0626 and two control groups of untreated ob/ob mice and lean littermates(n=16 each).All mice were fed a normal chow diet with or without MK-0626 for either four or eight weeks.Blood samples were collected,and total hepatectomy was performed.RESULTS:The administration of dietary MK-0626 ameliorated the hepatic lipid accumulation in ob/ob micetreated with 3 mg/kg MK-0626(3 MK),P<0.05,vs untreated ob/ob mice(ob/ob).The MK-0626 treatment reduced the serum alanine aminotransferase levels(both treatment groups,P<0.05 vs ob/ob)and glucoses/insulin levels/calculated HOMA scores(1.5 MK,P<0.05vs ob/ob;3 MK,P<0.01 vs ob/ob)and increased the serum adiponectin levels(3 MK,P<0.05 vs ob/ob)in a dose-dependent manner.The MK-0626 treatment increased the m RNA expression of peroxisome proliferator-activated receptorα/microsomal triglyceride transfer protein(1.5 MK,P<0.05 vs ob/ob;3 MK,P<0.01vs ob/ob)but reduced the sterol regulatory element binding transcription factor-1c/fatty acid synthase/stearoyl-Co A desaturase-1(both treatment groups,P<0.01 vs ob/ob).The MK-0626 treatment increased the activity of AMP-activated protein kinase(AMPK)(both treatment groups,P<0.01 vs ob/ob).CONCLUSION:MK-0626 could attenuate hepatic steatosis through enhancing AMPK activity,inhibiting hepatic lipogenic gene expression,enhancing triglyceride secretion from liver and increasing serum adiponectin levels. | Tatsuya Ohyama Ken Sato Yuichi Yamazaki Hiroaki Hashizume Norio Horiguchi Satoru Kakizaki Masatomo Mori Motoyasu Kusano Masanobu Yamada | 2014 | World Journal of Gastroenterology2014，20，43： | 4 |

| 5 | TTF-1 is useful for primary site determination in duodenal metastasis显示文摘We report here on a case of duodenal metastasis from primary lung adenocarcinoma. A 69-year old man was diagnosed with primary lung adenocarcinoma. Four courses of combined chemotherapy with carboplatin and paclitaxel associated with irradiation of 60 Gy shrunk the lung tumor. However, soon after,the para-aortic lymph node became swollen. Esophagogastroduodenoscopy revealed three duodenal tumors. Differential diagnosis between malignant lymphoma and metastatic duodenal cancer was endoscopically difficult. The histology of biopsied specimens was poorly differentiated adenocarcinoma. Immunohistochemical analysis revealed a positive reaction for thyroid transcription factor-1 (TTF-1). Thus, we concluded that these were metastatic duodenal tumors from lung adenocarcinoma. Two courses of gemcitabine led to a complete remission in this duodenal metastasisand para-aortic lymph node swelling with only scarring remaining in computed tomography. He is now on the continuous generalized chemotherapy. In conclusion, duodenal metastasis from primary lung adenocarcinoma is rare and hard to diagnose. | 6 | The deep-space multi-object orbit determination system and its application to Hayabusa2’s asteroid proximity operations显示文摘The deep-space multi-object orbit determination system(DMOODS)and its application in the asteroid proximity operation of the Hayabusa2 mission are described.DMOODS was developed by the Japan Aerospace Exploration Agency(JAXA)for the primary purpose of determining the trajectory of deep-space spacecraft for JAXA’s planetary missions.The weighted least-squares batch filter is used for the orbit estimator of DMOODS.The orbit estimator supports more than 10 data types,some of which are used for relative trajectory measurements between multiple space objects including natural satellites and small bodies.This system consists of a set of computer programs running on Linux-based consumer PCs on the ground,which are used for orbit determination and the generation of radiometric tracking data,such as delta differential one-way ranging and doppler tracking data.During the asteroid proximity phase of Hayabusa2,this system played an essential role in operations that had very strict navigation requirements or operations in which few optical data were obtained owing to special constraints on the spacecraft attitude or distance from the asteroid.One example is orbit determination during the solar conjunction phase,in which the navigation accuracy is degraded by the effect of the solar corona.The large range bias caused by the solar corona was accurately estimated with DMOODS by combining light detection and ranging(LIDAR)and ranging measurements in the superior solar conjunction phase of Hayabusa2.For the orbiting operations of target markers and the MINERVA-II2 rover,the simultaneous estimation of six trajectories of four artificial objects and a natural object was made by DMOODS.This type of simultaneous orbit determination of multi-artificial objects in deep-space has never been accomplished before. | Hiroshi Takeuchi Kent Yoshikawa Yuto Takei Yusuke Oki Shota Kikuchi Hitoshi Ikeda Stefania Soldini Naoko Ogawa Yuya Mimasu Go Ono Fuyuto Terui Naoya Sakatani Manabu Yamada Toru Kouyama Shingo Kameda Takanao Saiki Yuichi Tsuda | 2020 | Astrodynamics2020，4，4： | 2 |
| 7 | Combination therapy with daclatasvir and asunaprevir for dialysis patients infected with hepatitis C virus显示文摘The standard antiviral therapy for dialysis patients infected with hepatitis C virus(HCV) is(pegylated) interferon monotherapy, but its efficacy is insufficient. Oral direct-acting antiviral agents(DAAs) have recently been developed for chronic hepatitis C patients. However, some DAAs have contraindications for chronic renal failure(CRF). Daclatasvir and asunaprevir are metabolized largely in the liver and are not contraindicated in CRF. Combination therapy with daclatasvir and asunaprevir was used for 4 dialysis patients infected with genotype 1b HCV. One patient had viral breakthrough, and the 3 others had sustained virological response 12. One patient was admitted for heart failure and percutaneous coronary intervention due to concomitant ischemic disease. Heart failure was unlikely to be caused by the combination therapy, as it was probably due to water overload. The patient continued to receive the combination therapy after the remission of the heart failure. | 8 | Non-invasive prediction of non-alcoholic steatohepatitis in Japanese patients with morbid obesity by artificial intelligence using rule extraction technology显示文摘AIM To construct a non-invasive prediction algorithm for predicting non-alcoholic steatohepatitis(NASH), we investigated Japanese morbidly obese patients using artificial intelligence with rule extraction technology.METHODS Consecutive patients who required bariatric surgery underwent a liver biopsy during the operation. Standard clinical, anthropometric, biochemical measurements were used as parameters to predict NASH and were analyzed using rule extraction technology. One hundred and two patients, including 79 NASH and 23 non-NASH patients were analyzed in order to create the predictionmodel, another cohort with 77 patients including 65 NASH and 12 non-NASH patients were analyzed to validate the algorithm.RESULTS Alanine aminotransferase, C-reactive protein, homeostasis model assessment insulin resistance, albumin were extracted as predictors of NASH using a recursive-rule extraction algorithm. When we adopted the extracted rules for the validation cohort using a highly accurate rule extraction algorithm, the predictive accuracy was 79.2%. | 11 | Probable case of drug reaction with eosinophilia and systemic symptom syndrome due to combination therapy with daclatasvir and asunaprevir显示文摘A 66-year-old, interferon-ineligible, treatment-naive man who was diagnosed with chronic hepatitis C due to hepatitis C virus genotype 1b began combination therapy with daclatasvir and asunaprevir. On day 14 of treatment, hepatic reserve and renal function deterioration was observed, while his transaminase levels were normal. Both daclatasvir and asunaprevir were discontinued on day 18 of treatment, because the patient complained of dark urine and a rash on his trunk and four limbs. After discontinuing antiviral therapy, the abnormal laboratory finding and clinical manifestations gradually improved, without recurrence. Our case fulfilled the diagnostic criteria of probable drug reaction with eosinophilia and systemic symptom(DRESS) syndrome. Despite the 18-d treatment, sustained virological response 12 was achieved. Based on the clinical course, we concluded that there was a clear cause-and-effect relationship between the treatment and adverse events. To our knowledge, this patient represents the first case of probable DRESS syndrome that includes concomitant deterioration of hepatic reserve and renal function due to combination therapy with daclatasvir and asunaprevir, regardless of normalization of transaminase levels.
