1Exercise-induced irisin in bone and systemic irisin administration reveal new regulatory mechanisms of bone metabolism显示文摘Irisin is a polypeptide hormone derived from the proteolytic cleavage of fibronectin-type III domaincontaining 5(FNDC5) protein. Once released to circulation upon exercise or cold exposure, irisin stimulates browning of white adipose tissue(WAT) and uncoupling protein 1(UCP1) expression, leading to an increase in total body energy expenditure by augmented UCP1-mediated thermogenesis. It is currently unknown whether irisin is secreted by bone upon exercise or whether it regulates bone metabolism in vivo. In this study, we found that 2 weeks of voluntary wheel-running exercise induced high levels of FNDC5 messenger RNA as well as FNDC5/irisin protein expression in murine bone tissues. Increased immunoreactivity due to exercise-induced FNDC5/irisin expression was detected in different regions of exercised femoral bones,including growth plate, trabecular bone, cortical bone, articular cartilage, and bone–tendon interface. Exercise also increased expression of osteogenic markers in bone and that of UCP1 in WAT, and led to bodyweight loss. Irisin intraperitoneal(IP) administration resulted in increased trabecular and cortical bone thickness and osteoblasts numbers, and concurrently induced UCP1 expression in subcutaneous WAT. Lentiviral FNDC5 IP administration increased cortical bone thickness. In vitro studies in bone cells revealed irisin increases osteoblastogenesis and mineralization, and inhibits receptor activator of nuclear factor-k B ligand(RANKL)-induced osteoclastogenesis. 2Potential influence of water level changes on energy flows in a lake food web显示文摘Large seasonal water-level fluctuations may influence isotopic signatures of primary producers and the types and amounts of these potential food sources accessible to aquatic fauna of Poyang Lake,the largest freshwater lake in China.In this study,the isotopic signatures of primary producers and consumers were determined,stable carbon and nitrogen isotope analysis and mixing models were combined to investigate the influence of water levels on the diet and isotopic composition of Poyang Lake fish and invertebrates.Five potential food sources (seston,benthic organic matter,aquatic macrophytes,attached algae,and terrestrial plants),4 species of invertebrates,and 10 species of fish were collected from the lake area during dry and wet seasons between January 2009 and April 2010.The δ 13C values of invertebrates and most fish were within the range of δ 13C values of the potential food sources for both seasons.The δ 13C values of invertebrates and most fish were lower in the dry season than in the wet season,whereas the δ 15N values exhibited different patterns for different species.Mixing models indicated that the most important food sources for common lake fauna were seston in the dry season and aquatic macrophytes and terrestrial plants in the wet season.The fauna were more omnivorous in the wet season than in the dry season.The food web dynamics of Poyang Lake are strongly influenced by changes in the abundance and accessibility of different basal food sources that occur because of seasonal flood pulses.The trophic links within the aquatic communities of Poyang Lake are modified by water-level fluctuations.WANG YuYu YU XiuBo LI WenHua XU Jun CHEN YuWei FAN Na 2011Chinese Science Bulletin2011,56,26:11

4Immunopotentiator Thymosin Alpha-1 Promotes Neurogenesis and Cognition in the Developing Mouse via a Systemic Th1 Bias显示文摘In early life, the immune system plays an essential role in brain development. In our study, the immunopotentiator thymosin alpha-1(Ta1) was peripherally administered to neonatal mice to explore whether the peripheral immunopotentiator affects neurodevelopment and cognition, and to further investigate the relevant mechanism. Compared with the control group, the Ta1 mice displayed better cognitive abilities in early life. The numbers of 5-bromodeoxyuridine(Brd U)+, nestin+,T-box transcription factor 2(Tbr2)+, Brd U+/doublecortin(DCX)+, Brd U+/ionized calcium-binding adaptor molecule 1(Iba1)+, and Brd U+/neuronal nuclei(Neu N)+ cells in the hippocampus were increased in the Ta1 group,accompanied by increased interleukin-4(IL-4), interferon-gamma, brain-derived neurotrophic factor, nerve growth factor, and insulin-like growth factor-1 as well as decreased IL-6 and tumor necrosis factor-a. Furthermore, the Ta1-group showed a Th1-polarized immune response, and the neurotrophic factors were positively associated with the Th1/Th2 ratio. More importantly, administration of Ta1 blocked lipopolysaccharide-induced impairment of hippocampal neurogenesis in early life. 5Virus-induced Phytoene Desaturase(PDS) Gene Silencing Using Tobacco Rattle Virus in Lilium × formolongi显示文摘Gene function analysis is challenging for Lilium due to the lack of an efficient method for stable genetic transformation. Virus-induced gene silencing(VIGS) is an attractive tool for determining gene function in plants. This study reported that Tobacco rattle virus(TRV)-based VIGS of a PHYTOENE DESATURASE(PDS) ortholog(LhPDS) can be achieved in Lilium × formolongi seedlings, with a survival rate of 92%, using the inoculation method of rubbing plus injection. Compared with untreated and mock-treated seedlings, the photobleached leaf phenotype of silenced plants significantly correlates with down-regulation of endogenous LhPDS(P ≤ 0.05). In addition, the silencing phenomenon can be observed in the growing points of plants, indicating that systemic viral infection was achieved using the protocol. The results indicate that TRV-based VIGS can be used to characterize gene function in Lilium × formolongi. 6Synergistic effects of magnesium ions and simvastatin on attenuation of high-fat diet-induced bone loss显示文摘Introduction:Magnesium(Mg)has a prophylactic potential against the onset of hyperlipidemia.Similar to statin,Mg is recommended as lipid-lowering medication for hypercholesterolemia and concomitantly exhibits an association with increased bone mass.The combination of statin with Mg ions(Mg^(2+))may be able to alleviate the high-fat diet(HFD)-induced bone loss and reduce the side-effects of statin.This study aimed to explore the feasibility of combined Mg^(2+)with simvastatin(SIM)for treating HFD-induced bone loss in mice and the involving mechanisms.Materials and methods:C57BL/6 male mice were fed with a HFD or a normal-fat diet(NFD).Mice were intraperitoneally injected SIM and/or orally received water with additional Mg^(2+)until sacrificed.Enzyme-linked immunosorbent assay was performed to measure cytokines and cholesterol in serum and liver lysates.Bone mineral density(BMD)and microarchitecture were assessed by micro-computed tomography(μCT)in different groups.The adipogenesis in palmitate pre-treated HepG2 cells was performed under various treatments.Results:μCT analysis showed that the trabecular bone mass was significantly lower in the HFD-fed group than that in NFD-fed group since week 8.The cortical thickness in HFD-fed group had a significant decrease at week 24,as compared with NFD-fed group.The combination of Mg^(2+)and SIM significantly attenuated the trabecular bone loss in HFD-fed mice via arresting the osteoclast formation and bone resorption.Besides,such combination also reduced the hepatocytic synthesis of cholesterol and inhibited matrix metallopeptidase 13(Mmp13)mRNA expression in pre-osteoclasts.Conclusions:The combination of Mg^(2+)and SIM shows a synergistic effect on attenuating the HFD-induced bone loss.Our current formulation may be a cost-effective alternative treatment to be indicated for obesity-related bone loss.Bingyang Dai Xu Li Jiankun Xu Yuwei Zhu Le Huang Wenxue Tong Hao Yao Dick Ho-kiu Chow Ling Qin 2021Bioactive Materials2021,6,8:4
7SARS-CoV-2 ORF10 suppresses the antiviral innate immune response by degrading MAVS through mitophagy显示文摘The global coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has caused severe morbidity and mortality in humans.It is urgent to understand the function of viral genes.However,the function of open reading frame 10(ORF10),which is uniquely expressed by SARS-CoV-2,remains unclear.In this study,we showed that overexpression of ORF10 markedly suppressed the expression of type I interferon(IFN-I)genes and IFN-stimulated genes.Then,mitochondrial antiviral signaling protein(MAVS)was identified as the target via which ORF10 suppresses the IFN-I signaling pathway,and MAVS was found to be degraded through the ORF10-induced autophagy pathway.Furthermore,overexpression of ORF10 promoted the accumulation of LC3 in mitochondria and induced mitophagy.Mechanistically,ORF10 was translocated to mitochondria by interacting with the mitophagy receptor Nip3-like protein X(NIX)and induced mitophagy through its interaction with both NIX and LC3B.Moreover,knockdown of NIX expression blocked mitophagy activation,MAVS degradation,and IFN-I signaling pathway inhibition by ORF10.Consistent with our observations,in the context of SARS-CoV-2 infection,ORF10 inhibited MAVS expression and facilitated viral replication.In brief,our results reveal a novel mechanism by which SARS-CoV-2 inhibits the innate immune response;that is,ORF10 induces mitophagy-mediated MAVS degradation by binding to NIX.Xingyu Li Peili Hou Wenqing Ma Xuefeng Wang Hongmei Wang Zhangping Yu Huasong Chang Tiecheng Wang Song Jin Xue Wang Wenqi Wang Yudong Zhao Yong Zhao Chunqing Xu Xiaomei Ma Yuwei Gao Hongbin He 2022Cellular & Molecular Immunology2022,19,1:4
8Recent development of perylene diimide-based small molecular non-fullerene acceptors in organic solar cells显示文摘Recently, perylene diimide(PDI) derivatives were attractive as the electron-deficient acceptor materials in non-fullerene organic solar cells since Tang first used a single PDI compound as the n-type semiconductor to fabricate photovoltaic devices in 1986, which achieved a power conversion efficiency of1%. Beside the monomeric PDIs, the linear and three dimensional(3D) PDI-based small molecular acceptors have also made great achievements with the power conversion efficiencies over 9.0% in singlejunction polymer solar cells, and over 10.0% in tandem solar cells. The excellent device performance can be realized by forming suitable twisted structure, developing suitable donor materials and optimizing device technologies. 10An accurate indoor map matching algorithm based on activity detection and crowdsourced Wi-Fi显示文摘Map matching has been widely investigated in indoor pedestrian navigation to improve positioning accuracy and robustness.This paper proposes an accurate map matching algorithm based on activity detection and crowdsourced Wi-Fi(AiFiMatch).Firstly, by taking indoor road segments between activity-related locations as nodes, and the activity type from one road segment to another as directed edge, the indoor floor plan is abstracted as a directed graph. Secondly, the smartphone’s motion sensors are utilized to detect different activities based on a decision tree and then the pedestrian’s walking trajectory is divided into subtrajectory sequence according to location-related activities. Finally, the sub-trajectory sequence is matched to the directed graph of indoor floor plan to position the pedestrian by using a Hidden Markov Model(HMM). Simultaneously, Wi-Fi fingerprints are bound to road segments based on timestamp. Through crowdsourcing, a radio map of indoor road segments is constructed. The radio map in turn inversely promotes the HMM based map matching algorithm. AiFiMatch is evaluated by the experiments using smartphones in a teaching building. 11Auxin response factor 6A regulates photosynthesis,sugar accumulation,and fruit development in tomato显示文摘Auxin response factors(ARFs)are involved in auxin-mediated transcriptional regulation in plants.In this study,we performed functional characterization of SlARF6A in tomato.SlARF6A is located in the nucleus and exhibits transcriptional activator activity.Overexpression of SlARF6A increased chlorophyll contents in the fruits and leaves of tomato plants,whereas downregulation of SlARF6A decreased chlorophyll contents compared with those of wild-type(WT)plants.Analysis of chloroplasts using transmission electron microscopy indicated increased sizes of chloroplasts in SlARF6A-overexpressing plants and decreased numbers of chloroplasts in SlARF6A-downregulated plants.Overexpression of SlARF6A increased the photosynthesis rate and accumulation of starch and soluble sugars,whereas knockdown of SlARF6A resulted in opposite phenotypes in tomato leaves and fruits.RNA-sequence analysis showed that regulation of SlARF6A expression altered the expression of genes involved in chlorophyll metabolism,photosynthesis and sugar metabolism.SlARF6A directly bound to the promoters of SlGLK1,CAB,and RbcS genes and positively regulated the expression of these genes.Overexpression of SlARF6A also inhibited fruit ripening and ethylene production,whereas downregulation of SlARF6A increased fruit ripening and ethylene production.SlARF6A directly bound to the SAMS1 promoter and negatively regulated SAMS1 expression.Taken together,these results expand our understanding of ARFs with regard to photosynthesis,sugar accumulation and fruit development and provide a potential target for genetic engineering to improve fruit nutrition in horticulture crops.Yujin Yuan Xin Xu Zehao Gong Yuwei Tang Mengbo Wu Fang Yan Xiaolan Zhang Qian Zhang Fengqing Yang Xiaowei Hu Qichen Yang Yingqing Luo Lihua Mei Wenfa Zhang Cai-Zhong Jiang Wangjin Lu Zhengguo Li Wei Deng 2019Horticulture Research2019,6,1:3
12Integrated bioinformatics identifies the dysregulation induced by aberrant gene methylation in colorectal carcinoma显示文摘Colorectal carcinoma(CRC)is one of the most common cancers,and is associated with a poor dlinical outcome.The key genes and potential prognostic markers in colorectal carcinoma remain to be identfed and explored for clinical application.DNA expression/methyl-ation profles were downloaded from the Gene Expression Omnibus(GEO)database to identify differentially expressed/methylated genes(DEGs and DEMs).A total of 255 genes and 372 genes were identified as being up-regulated and down-regulated,respectively,in GSE113513,GSE81558,and GSE89076.There were a total of 3350 hypermethylated genes and 443 hypomethylated genes identifed in GSE 48684.Twenty genes were found to be hypermethylated as well as down-regulated,and a functional enrichment analysis revealed that these genes were mainly involved in cancer-related pathways.Among these 20 genes,GPM6A,HAND2 and C2orf40 were related to poor outcomes in cancer patients based on a survival analysis.Concurrent decreases of GPM6A,HAND2 and C2orf40 protein expression were observed in highly-dif ferentiated colorectal carcinoma tissues,and higher expression levels were found in undifferentiated or minimally-differentiated colorectal carcinoma tissues.In conclusion,20 genes were found to be downregulated and hypermethylated in CRC,among which GPM6A,HAND2 and C2orf40 were explored for their potential prognostic value.Zhenyu Ye Yecheng Li Jiaming Xie Zhenyu Feng Xiaodong Yang Yong Wu Yuwei Pu Jiawei Gao Xiangrong Xu Zhaobi Zhu Wei Li Wei Chen Chungen Xing 2021Genes & Diseases2021,8,4:2

14Experimental repetitive quantum channel simulation显示文摘Universal control of quantum systems is a major goal to be achieved for quantum information processing,which demands thorough understanding of fundamental quantum mechanics and promises applications of quantum technologies. So far, most studies concentrate on ideally isolated quantum systems governed by unitary evolutions, while practical quantum systems are open and described by quantum channels due to their inevitable coupling to environment. Here, we experimentally simulate arbitrary quantum channels for an open quantum system, i.e. a single photonic qubit in a superconducting quantum circuit.The arbitrary channel simulation is achieved with minimum resource of only one ancilla qubit and measurement-based adaptive control. By repetitively implementing the quantum channel simulation,we realize an arbitrary Liouvillian for a continuous evolution of an open quantum system for the first time. 15Overpressure Evolution Recorded in Fluid Inclusions in the Dongpu Depression,Bohai Bay Basin,North China显示文摘The Dongpu depression,like other depressions in the Bohai Bay Basin,is characterized by overpressure,however,the published studies have not clearly given the overpressure formation mechanism and paleo-pressure evolution history in the Dongpu depression.In addition,the current researches are mainly focused on the northern Dongpu depression,while rare investigation has been conducted in the southern part.In this context,it is urgent to explore the comprehensive overpressure evolution and formation mechanism in the Dongpu depression in academic and industrial fields.This paper provided a systematic analysis of fluid inclusions,including fluid inclusion petrography,microthermometry,vapor phase filling degree measurement and PVT-x modeling.By using homogenization temperature-burial history projection,the trapping time of the oil inclusions are determined.Four pressure evolution stages are identified in both the northern and the southern Dongpu depression.The results indicate that the pressure accumulation stage is derived by disequilibrium compaction and hydrocarbon generation in the northern part because of the salt rock distribution,while the stage is only derived by hydrocarbon generation in the southern part because of no salt rock distribution.The pressure release stages are mainly caused by the tectonic uplifting movements at the end of Dongying and Minghuazhen Formation.The overpressure in most areas in the northern part has been preserved till now,which is considered as a result of the good plasticity and sealing ability of salt rock that prevent the pressure leakage by smearing on the fault.Areas without salt rock distribution show normal pressure,which means the salt rock is an important factor that contributes to the overpressure preservation.However,there are exceptions that in Baimiao and Qiaokou,the overpressure still exists even without salt rock distribution,which is due to fault closure.Xiuyan Liu Honghan Chen Xuewei Xiao Hongan Zhang Yuwei Wang Tianwu Xu Pei Shang Lingtao Kong 2022Journal of Earth Science2022,33,4:2
16Modulation of Beta Oscillations for Implicit Motor Timing in Primate Sensorimotor Cortex during Movement Preparation显示文摘Motor timing is an important part of sensorimotor control. Previous studies have shown that beta oscillations embody the process of temporal perception in explicit timing tasks. In contrast, studies focusing on beta oscillations in implicit timing tasks are lacking. In this study, we set up an implicit motor timing task and found a modulation pattern of beta oscillations with temporal perception during movement preparation. We trained two macaques in a repetitive visually-guided reach-to-grasp task with different holding intervals. Spikes and local field potentials were recorded from microelectrode arrays in the primary motor cortex, primary somatosensory cortex, and posterior parietal cortex. We analyzed the association between beta oscillations and temporal interval in fixedduration experiments(500 ms as the Short Group and1500 ms as the Long Group) and random-duration experiments(500 ms to 1500 ms). The results showed that the peak beta frequencies in both experiments ranged from15 Hz to 25 Hz. The beta power was higher during the hold period than the movement(reach and grasp) period.Further, in the fixed-duration experiments, the mean poweras well as the maximum rate of change of beta power in the first 300 ms were higher in the Short Group than in the Long Group when aligned with the Center Hit event. In contrast, in the random-duration experiments, the corresponding values showed no statistical differences among groups. The peak latency of beta power was shorter in the Short Group than in the Long Group in the fixed-duration experiments, while no consistent modulation pattern was found in the random-duration experiments. These results indicate that beta oscillations can modulate with temporal interval in their power mode. 18An efficient strategy for generation of transgenic mice by lentiviral transduction of male germline stem cells in vivo显示文摘Background: Male germline stem cells(MGSCs) are a subpopulation of germ cells in the testis tissue. MGSCs are capable of differentiation into spermatozoa and thus are perfect targets for genomic manipulation to generate transgenic animals.Method: The present study was to optimize a protocol of production of transgenic mice through transduction of MGSCs in vivo using lentiviral-based vectors. The recombinant lentiviral vectors with either EF-1 or CMV promoter to drive the expression of enhanced green fluorescent protein(e GFP) transgene were injected into seminiferous tubules or inter-tubular space of 7-day-old and 28-day-old mouse testes. At 5 or 6 wk post-surgery, these pre-founders were mated with wild-type C57BL/6J female mice(1.5 to 2.0-month-old).Results: Sixty-seven percent of F1 generation and 55.56 % of F2 offspring were positive for eG FP transgene under the control of EF-1 promoter via PCR analysis. The transgenic pups were generated in an injection site-and age-independent manner. The expression of transgene was displayed in the progeny derived from lentiviral vector containing CMV promoter to drive transgene, but it was silenced or undetectable in the offspring derived from lentiviral vector with transgene under EF-1 promoter. The methylation level of g DNA in the promoter region of transgene was much higher in the samples derived lentiviral vectors with EF-1 promoter than that with CMV promoter,suggesting e GFP transgene was suppressed by DNA methylation in vivo.Conclusion: This research reported here an effective strategy for generation of transgenic mice through transduction of MGSCs in vivo using lentivirus vectors with specific promoters, and the transgenic offspring were obtained in an injection site-and age-independent manner. 20A truncated protein product of the germline variant of the DUOX2 gene leads to adenomatous polyposis显示文摘Objective:In some patients with adenomatous polyposis,an identifiable pathogenic variant of known associated genes cannot be found.Researchers have studied this for decades;however,few new genes have been identified.Methods:Adenomatous polyposis coli(APC)negative polyposis patients were identified through next-generation sequencing and multiplex ligation-dependent probe amplification.Then,whole-exome sequencing(WES)was used to determine candidate genes harboring pathogenic variants.Functional experiments were performed to explore their effects.Subsequently,using Sanger sequencing,we found other polyposis patients carrying variants of the DUOX2 gene,encoding dual oxidase 2,and analyzed them.Results:From 88 patients with suspected familial adenomatous polyposis,25 unrelated APC negative polyposis patients were identified.Based on the WES results of 3 patients and 2 healthy relatives from a family,the germline nonsense variant(c.1588 A>T;p.K530 X)of the DUOX2 gene was speculated to play a decisive role in the pedigree in relation to adenomatous polyposis.During functional experiments,we observed that the truncated protein,h Duox2 K530,was overexpressed in the adenoma in a carrier of the DUOX2 nonsense variant,causing abnormal cell proliferation through endoplasmic reticulum(ER)retention.In addition,we found two unrelated APC negative patients carrying DUOX2 missense variants(c.3329 G>A,p.R1110 Q;c.4027 C>T,p.L1343 F).Given the results of the in silico analysis,these two missense variants might exert a negative influence on the function of h Duox2.Conclusions:To our knowledge,this is the first study that reports the possible association of DUOX2 germline variants with adenomatous polyposis.With an autosomal dominant inheritance,it causes ER retention,inducing an unfolded protein response.Mengyuan Yang Yingxin Zhao Yuwei Ding Juan Wang Yinuo Tan Dong Xu Ying Yuan 2021Cancer Biology & Medicine2021,18,1:1
