2Young Yum pill inhibits inflammatory mediators and nuclear factor-kappa B signaling in lipopolysaccharide-stimulated RAW 264.7 macrophages显示文摘OBJECTIVE: To investigate the effect of Young Yum pill (YYP) on inflammatory mediators in cultured RAW 264.7 cells and elucidate the nuclear factor- kappa B (NF-κB)-related mechanism behind the action. METHODS: YYP was extracted with 95% ethanol Lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages were used to evaluate the effect of YYP on inflammatory mediators. Production of nitric oxide (NO) and prostaglandin E2 (PGE2) were measured by Griess test and enzyme-linked immunosorbent assay, respectively. The levels of genes and proteins involved in the generation of inflammatory mediators were examined using real-time polymerase chain reaction and Western blotting, respectively. RESULTS: YYP dose-dependently suppressed LPS-induced production of NO, PGE2 and tumor necrosis factor-α(TNF-α), and elevation of mRNA and protein levels of inducible NO synthase and cyclooxygenase- 2 in RAW 264.7 macrophages. These observations were associated with decreased NF-κB p65 phosphorylation and nuclear localization, enhanced Akt (protein kinase B) phosphorylation, as well as reduced inhibitor of κB (IκB)α degradation and IκB kinase α/β phosphorylation. 3Redox responsive nanoparticle encapsulating black phosphorus quantum dots for cancer theranostics显示文摘Effective cancer treatment puts high demands for cancer theranostics.For cancer diagnostics,optical coherence tomography(OCT)technology(including photothermal optical coherence tomography(PT-OCT))has been widely investigated since it induces changes in optical phase transitions in tissue through environmental changes(such as temperature change for PT-OCT).In this report,redox responsive nanoparticle encapsulating black phosphorus quantum dots was developed as a robust PT-OCT agent.Briefly,black phosphorus quantum dots(BPQDs)are incorporated into cysteine-based poly-(disulfide amide)(Cys-PDSA)to form stable and biodegradable nanoagent.The excellent photothermal feature allows BPQD/Cys-PDSA nanoparticles(NPs)as a novel contrast agent for high-resolution PT-OCT bioimaging.The Cys-PDSA can rapidly respond to glutathione and effectively release BPQDs and drugs in vitro and in vivo.And the obtained NPs exhibit excellent near-infrared(NIR)photothermal transduction efficiency and drug delivery capacity that can serve as novel therapeutic platform,with very low chemo drug dosage and side effects.Both of the polymer and BPQD are degradable,indicating this platform is a rare PT-OCT agent that is completely biodegradable.Overall,our research highlights a biodegradable and biocompatible black phosphorus-based nanoagent for both cancer diagnosis and therapy.Haolin Chen Zhiming Liu Bo Wei Jun Huang Xinru You Jingyang Zhang Zhiling Yuan Zhilie Tang Zhouyi Guo Jun Wu 2021Bioactive Materials2021,6,3:1

10Flexible feeding patterns of copepod Centropages tenuiremis in fluctuating conditions: a possible survival strategy to cope with disturbance显示文摘Centropages tenuiremis is a species with a wide distribution range in disturbed coastal waters.However,due to a lack of dietary information,it remains unclear as to how they maintain such dominance in fluctuating conditions.In this study,C.tenuiremis was collected from the Daya Bay Nuclear Power Plant both in inlet and outfall regions at 06:00,12:00 and 18:00 on April 27,2011 and their in situ diet was analyzed using a PCR protocol targeting 18S ribosomal genes.Thirty-four species of prey organisms were identified totally,including Dinophyta,Baciliariophyta,Viridiplantae,Rhizaria,Apicomplexa,Chordata,Mollusca,Arthropoda and Fungi,indicating an obvious omnivorous feeding habit of C.tenuiremis.Centropages tenuiremis obviously exhibited spatial and temporal variations in diet composition.More plant prey(land plants and phytoplankton)were consumed in the morning(~50%),while more animal prey(metazoans and protozoans)were ingested at midday and night(60%–70%).Furthermore,a more diverse diet was detected in the outfall region(10–11 taxa),where the temperatures were relatively higher and more fluctuating,than in the control region(5–10 taxa).This finding indicated that C.tenuiremis could potentially expand its food spectrum under stressful condition.Specifically,C.tenuiremis exhibited phytoplankton preference(58.62%–67.64%)in the outfall region with a lower omnivory index(0.27–0.35)than in the control region(0.51–0.95).However,phytoplankton density was lower than that in the control region,suggesting a possible herbivorous tendency of C.tenuiremis under elevated temperatures to balance the energy acquirement and feeding effort.The flexible food choices of C.tenuiremis observed here could effectively buffer environmental fluctuations and might be an important survival strategy in coastal ecosystems.Cuilian Xu Simin Hu Zhiling Guo Tao Li Hui Huang Leo Lai Chan Sheng Liu 2020Acta Oceanologica Sinica2020,39,2:0

13Geochronology and zircon geochemistry of auriferous intrusions in the Bumo deposit,Hainan Province,China显示文摘Intrusion-related gold deposits(IRGS)are a lowgrade,large-tonnage exploration target.Recently,auriferous magmatic rocks were found in the Bumo deposit of the Gezhen shear zone in Hainan Province,China.However,the geochronology and geochemical characteristics of the intrusions,as well as the mineralization potential,are still unclear.Field and petrographic work show that the sulfidebearing intrusions can be divided into diorite porphyrites,quartz monzodiorites and monzodiorites.Zircon LA–ICP–MS U–Pb dating demonstrates that diorite porphyrites,quartz monzodiorites and monzodiorites were formed at104±1,114±1,114±1 Ma,respectively.In addition,sulfides in Yanshanian intrusion-related gold mineralization haveδ;S values of 0.2–4.4%,lower than those in Hercynian-Indosinian(1.9–9.8%)orogenic deposits(ca.219–378 Ma)in the Gezhen shear zone.In addition,all these intrusions display close correlations between Eu/EuN*with Th/U,consistent with the differentiation of amphibole,apatite and titanite from a hydrous melt.Moreover,zircon Eu/EuN*in the intrusions are higher than0.4,demonstrating that the magmatic rocks have high water contents and oxygen fugacity values,favorable for gold mineralization.Consequently,the Yanshanian magmatic rocks can be a new potential gold exploration target in the Gezhen shear zone.Yan Hai Bingtao Li Teng Deng Deru Xu Li Wang Youzhong Xiong Xiaowen Zhang Zhiling Wang Shaohao Zou Zhengpeng Ding Qian Qian Shichao Guo 2022Acta Geochimica2022,41,2:0
14Novel biomarkers identifying hypertrophic cardiomyopathy and its obstructive variant based on targeted amino acid metabolomics显示文摘Background:Hypertrophic cardiomyopathy(HCM)is an underdiagnosed genetic heart disease worldwide.The management and prognosis of obstructive HCM(HOCM)and non-obstructive HCM(HNCM)are quite different,but it also remains challenging to discriminate these two subtypes.HCM is characterized by dysmetabolism,and myocardial amino acid(AA)metabolism is robustly changed.The present study aimed to delineate plasma AA and derivatives profiles,and identify potential biomarkers for HCM.Methods:Plasma samples from 166 participants,including 57 cases of HOCM,52 cases of HNCM,and 57 normal controls(NCs),who first visited the International Cooperation Center for HCM,Xijing Hospital between December 2019 and September 2020,were collected and analyzed by high-performance liquid chromatography-mass spectrometry based on targeted AA metabolomics.Three separate classification algorithms,including random forest,support vector machine,and logistic regression,were applied for the identification of specific AA and derivatives compositions for HCM and the development of screening models to discriminate HCM from NC as well as HOCM from HNCM.Results:The univariate analysis showed that the serine,glycine,proline,citrulline,glutamine,cystine,creatinine,cysteine,choline,and aminoadipic acid levels in the HCM group were significantly different from those in the NC group.Four AAs and derivatives(Panel A;proline,glycine,cysteine,and choline)were screened out by multiple feature selection algorithms for discriminating HCM patients from NCs.The receiver operating characteristic(ROC)analysis in Panel A yielded an area under the ROC curve(AUC)of 0.83(0.75-0.91)in the training set and 0.79(0.65-0.94)in the validation set.Moreover,among 10 AAs and derivatives(arginine,phenylalanine,tyrosine,proline,alanine,asparagine,creatine,tryptophan,ornithine,and choline)with statistical significance between HOCM and HNCM,3 AAs(Panel B;arginine,proline,and ornithine)were selected to differentiate the two subgroups.The AUC values in the training and validation sets for Panel B were 0.83(0.74-0.93)and 0.82(0.66-0.98),respectively.Conclusions:The plasma AA and derivatives profiles were distinct between the HCM and NC groups.Based on the differential profiles,the two established screening models have potential value in assisting HCM screening and identifying whether it is obstructive.Lanyan Guo Bo Wang Fuyang Zhang Chao Gao Guangyu Hu Mengyao Zhou Rutao Wang Hang Zhao Wenjun Yan Ling Zhang Zhiling Ma Weiping Yang Xiong Guo Chong Huang Zhe Cui Fangfang Sun Dandan Song Liwen Liu Ling Tao 2022Chinese Medical Journal2022,135,16:0
15CHCHD2 Thr61Ile mutation impairs F1F0-ATPase assembly in in vitro and in vivo models of Parkinson's disease显示文摘Mitochondrial dysfunction is a significant pathological alte ration that occurs in Parkinson's disease(PD),and the Thr61lle(T61I)mutation in coiled-coil helix coiled-coil helix domain containing 2(CHCHD2),a crucial mitochondrial protein,has been reported to cause Parkinson's disease.FIFO-ATPase participates in the synthesis of cellular adenosine triphosphate(ATP)and plays a central role in mitochondrial energy metabolism.However,the specific roles of wild-type(WT)CHCHD2 and T611-mutant CHCHD2 in regulating F1FO-ATPase activity in Parkinson's disease,as well as whether CHCHD2 or CHCHD2 T61I affects mitochondrial function through regulating F1FO-ATPase activity,remain unclea r.Therefore,in this study,we expressed WT CHCHD2 and T61l-mutant CHCHD2 in an MPP^(+)-induced SH-SY5Y cell model of PD.We found that CHCHD2 protected mitochondria from developing MPP^(+)-induced dysfunction.Under normal conditions,ove rexpression of WT CHCHD2 promoted F1FO-ATPase assembly,while T61I-mutant CHCHD2 appeared to have lost the ability to regulate F1FO-ATPase assembly.In addition,mass spectrometry and immunoprecipitation showed that there was an interaction between CHCHD2 and F1FO-ATPase.Three weeks after transfection with AAV-CHCHD2 T61I,we intraperitoneally injected 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine into mice to establish an animal model of chronic Parkinson's disease and found that exogenous expression of the mutant protein worsened the behavioral deficits and dopaminergic neurodegeneration seen in this model.These findings suggest that WT CHCHD2 can alleviate mitochondrial dysfunction in PD by maintaining F1F0-ATPase structure and function.Xiang Chen Yuwan Lin Zhiling Zhang Yuting Tang Panghai Ye Wei Dai Wenlong Zhang Hanqun Liu Guoyou Peng Shuxuan Huang Jiewen Qiu Wenyuan Guo Xiaoqin Zhu Zhuohua Wu Yaoyun Kuang Pingyi Xu Miaomiao Zhou 2024Neural Regeneration Research2024,19,1:0
