2Precise nanomedicine for intelligent therapy of cancer显示文摘Precise nanomedicine has been extensively explored for efficient cancer imaging and targeted cancer therapy, as evidenced by a few breakthroughs in their preclinical and clinical explorations. Here, we demonstrate the recent advances of intelligent cancer nanomedicine, and discuss the comprehensive understanding of their structure-function relationship for smart and efficient cancer nanomedicine including various imaging and therapeutic applications, as well as nanotoxicity. In particular, a few emerging strategies that have advanced cancer nanomedicine are also highlighted as the emerging focus such as tumor imprisonment, supramolecular chemotherapy, and DNA nanorobot. The challenge and outlook of some scientific and engineering issues are also discussed in future development. 3Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. 4Erucic acid from Isatis indigotica Fort. suppresses influenza A virus replication and inflammation in vitro and in vivo through modulation of NF-kB and p38 MAPK pathway显示文摘Isatis indigotica Fort.(Ban-Lan-Gen)is an herbal medicine prescribed for influenza treatment.However,its active components and mode of action remain mostly unknown.In the present study,erucic acid was isolated from Isatis indigotica Fort.,and subsequently its underlying mechanism against influenza A virus(IAV)infection was investigated in vitro and in vivo.Our results demonstrated that erucic acid exhibited broad-spectrum antiviral activity against IAV resulting from reduction of viral polymerase transcription activity.Erucic acid was found to exert inhibitory effects on IAV or viral(v)RNA-induced pro-inflam-matory mediators as well as interferons(IFNs).The molecular mechanism by which erucic acid with antiviral and anti-inflammatory properties was attributed to inactivation of NF-kB and p38 MAPK signaling.Furthermore,the NF-kB and p38 MAPK inhibitory effect of erucic acid led to diminishing the transcriptional activity of interferon-stimulated gene factor 3(ISGF-3),and thereby reducing IAV-triggered pro-inflammatory response amplification in IFN-β-sensitized cells.Additionally,IAV-or vRNA-triggered apoptosis of alveolar epithelial A549 cells was prevented by erucic acid.In vivo,erucic acid administration consistently displayed decreased lung viral load and viral antigens expression.Meanwhile,erucic acid markedly reduced CD8+cytotoxic T lymphocyte(CTL)recruitment,pro-apoptotic signaling,hyperactivity of multiple signaling pathways,and exacerbated immune inflammation in the lung,which resulted in decreased lung injury and mortality in mice with a mouse-adapted A/FM/1/47-MA(H1N1)strain infection.Our findings provided a mechanistic basis for the action of erucic acid against IAV-mediated inflammation and injury,suggesting that erucic acid may have a therapeutic potential in the treatment of influenza.Xiaoli Liang Yuan Huang Xiping Pan Yanbing Hao Xiaowei Chen Haiming Jiang Jing Li Beixian Zhou Zifeng Yang 2020Journal of Pharmaceutical Analysis2020,10,2:10
5Unique characteristics of the genetics epidemiology of amyotrophic lateral sclerosis in China显示文摘Continual discoveries of new genes and unraveling the genetic etiology in amyotrophic lateral sclerosis(ALS) have provided greater insight into the underlying pathogenesis in motor neuron degeneration, as well as facilitating the disease modeling and the testing of targeted therapeutics. While, the genetic etiology accounted for two-thirds of FALS and approximately 11% of SALS in Caucasians. However, the contributions of these causative genes to ALS vary among different populations. Furthermore, the prominent difference between Chinese population and other ethnics remains a source of ongoing debate. We systemically reviewed genetics literature of Chinese ALS populations and updated the mutation frequencies of the main ALS-implicated genes aiming to determine the genetic features of ALS in Chinese population. We also reviewed the associations between ALSimplicated single nucleotide polymorphisms(SNPs) and the risk of ALS in Chinese population. A total of 116 studies were included in this analysis(86 gene mutation study articles and 30 SNPs study articles). The results showed that the overall gene mutation rates of ALS-related causative genes were 55.0% in familial ALS(FALS) and 11.7% in sporadic ALS(SALS) in Chinese population. In Chinese FALS, the highest mutation frequency was found in SOD1 gene(25.6%), followed by FUS(5.8%), TARDBP(5.8%), DCTN1(3.6%) and C9 orf 72(3.5%). In Chinese SALS, the highest mutation frequency was also identified in SOD1 gene(1.6%), followed by ANXA11(1.4%), FUS(1.3%), SQSTM1(1.0%), OPTN(0.9%) and CCNF(0.8%).The associations between several SNPs and risk of ALS were also reported in Chinese population. The genetic features of ALS in Chinese population are significantly different from those in Caucasian population, indicating an association between genetic susceptibility and origin of population. 6The Arabidopsis Anaphase-Promoting Complex/Cyclosome Subunit 1 is Critical for Both Female Gametogenesis and Embryogenesis显示文摘Anaphase-promoting complex/cyclosome (APC/C), a multisubunit E3 ligase, plays a critical role in cell cycle control, but the functional characterization of each subunit has not yet been completed. To investigate the function of APC1 in Arabidopsis, we analyzed four mutant alleles of APC1, and found that mutation in APC1 resulted in significantly reduced plant fertility, accumulation of cyclin B, and disrupted auxin distribution in embryos. The three mutant alleles apc1-1, apc1-2 and apc1-3 shared variable defects in female gametogenesis including degradation, abnormal nuclear number, and disrupted polarity of nuclei in the embryo sac as well as in embryogenesis, in which embryos were arrested at multiple stages. All of these defects are similar to those previously identified in apc4. The mutant apc1-4, in which the T-DNA was inserted after the transmembrane domain at the C-terminus, showed much more severe phenotypes; that is, most of the ovules were arrested at the one-nucleate female gametophyte stage (stage FG1). In the apc1 apc4 double mutants, the fertility was further reduced by one-third in apc1-1/+ apc4-1/+, and in some cases no ovules even survived in siliques of apc1-4/+ apc4-1/+. 7Evaluation of efficacy and safety of gandouling plus sodium dimercaptosulphonate in treatment of patients with neurological Wilson's disease from China显示文摘OBJECTIVE: To evaluate the efficacy and safety of gandouling plus sodium dimercaptosulphonate(DMPS) on neurological Wilson's disease(WD) in patients.METHODS: We retrospectively evaluated the clinical records of 125 WD patients with neurological syndromes who were treated with gandouling plus sodium DMPS or DMPS used alone. All patients had a history of neurological deterioration during their diseases courses. The clinical efficacies, adverse reactions, and results of the various hematological and biochemical investigations were recorded for statistical analysis.RESULTS: 92.30%(60 patients) of the WD patients treated with the combined therapy experienced an improved or stable neurological condition paral-leled by a significantly improved GAS score. Meanwhile, the WBC and PLT counts stabilized, liver function and renal function were improved or remained stable. The combined therapy also obviously promoted the 24-h urinary copper excretion. 9Emerging roles of activating transcription factor (ATF) family members in tumourigenesis and immunity: Implications in cancer immunotherapy显示文摘Activating transcription factors, ATFs, are a group of bZIP transcription factors that act as homodimers or heterodimers with a range of other bZIP factors. In general, ATFs respond to extracellular signals, indicating their important roles in maintaining homeostasis. The ATF family includes ATF1, ATF2, ATF3, ATF4, ATF5, ATF6, and ATF7. Consistent with the diversity of cellular processes reported to be regulated by ATFs, the functions of ATFs are also diverse. ATFs play an important role in cell proliferation, apoptosis, differentiation and inflammation-related pathological processes. The expression and phosphorylation status of ATFs are also related to neurodegenerative diseases and polycystic kidney disease. Various miRNAs target ATFs to regulate cancer proliferation, apoptosis, autophagy, sensitivity and resistance to radiotherapy and chemotherapy. Moreover, ATFs are necessary to maintain cell redox homeostasis. Therefore, deepening our understanding of the regulation and function of ATFs will provide insights into the basic regulatory mechanisms that influence how cells integrate extracellular and intracellular signals into genomic responses through transcription factors. Under pathological conditions, especially in cancer biology and response to treatment, the characterization of ATF dysfunction is important for understanding how to therapeutically utilize ATF2 or other pathways controlled by transcription factors. 10H7N9 virus infection triggers lethal cytokine storm by activating gasdermin E-mediated pyroptosis of lung alveolar epithelial cells显示文摘The H7N9 influenza virus emerged in China in 2013,causing more than 1560 human infections,39% of which were fatal.A ‘cytokine storm’ in the lungs of H7N9 patients has been linked to a poor prognosis and death;however,the underlying mechanism that triggers the cytokine storm is unknown.Here,we found that efficient replication of the H7N9 virus in mouse lungs activates gasdermin E(GSDME)-mediated pyroptosis in alveolar epithelial cells,and that the released cytosolic contents then trigger a cytokine storm.Knockout of Gsdme switched the manner of death of A549 and human primary alveolar epithelial cells from pyroptosis to apoptosis upon H7N9 virus infection,and Gsdme knockout mice survived H7N9 virus lethal infection.Our findings reveal that GSDME activation is a key and unique mechanism for the pulmonary cytokine storm and lethal outcome of H7N9 virus infection and thus opens a new door for the development of antivirals against the H7N9 virus.Xiaopeng Wan Jiqing Li Yupeng Wang Xiaofei Yu Xijun He Jianzhong Shi Guohua Deng Xianying Zeng Guobin Tian Yanbing Li Yongping Jiang Yuntao Guan Chengjun Li Feng Shao Hualan Chen 2022National Science Review2022,9,1:3
11Textbook Outcome as a measure of surgical quality assessment and prognosis in gastric neuroendocrine carcinoma:A large multicenter sample analysis显示文摘Objective:Quality assurance is crucial for oncological surgical treatment assessment.For rare diseases,singlequality indicators are not enough.We aim to develop a comprehensive and reproducible measurement,called the'Textbook Outcome'(TO),to assess the quality of surgical treatment and prognosis of gastric neuroendocrine carcinoma(G-NEC)patients.Methods:Data from patients with primary diagnosed G-NEC included in 24 high-volume Chinese hospitals from October 2005 to September 2018 were analyzed.TO included receiving a curative resection,≥15 lymph nodes examined,no severe postoperative complications,hospital stay≤21 d,and no hospital readmission≤30 d after discharge.Hospital variation in TO was analyzed using a case mix-adjusted funnel plot.Prognostic factors of survival and risk factors for non-Textbook Outcome(non-TO)were analyzed using Cox and logistic models,respectively.Results:TO was achieved in 56.6%of 860 G-NEC patients.TO patients had better overall survival(OS),disease-free survival(DFS),and recurrence-free survival(RFS)than non-TO patients(P<0.05).Moreover,TO patients accounted for 60.3%of patients without recurrence.Multivariate Cox analysis revealed non-TO as an independent risk factor for OS,DFS,and RFS of G-NEC patients(P<0.05).Increasing TO rates were associated with improved OS for G-NEC patients,but not hospital volume.Multivariate logistic regression revealed that nonlower tumors,open surgery,and>200 mL blood loss were independent risk factors for non-TO patients(P<0.05).Conclusions:TO is strongly associated with multicenter surgical quality and prognosis for G-NEC patients.Factors predicting non-TO are identified,which may help guide strategies to optimize G-NEC outcomes.Qiyue Chen Zhongliang Ning Zhiyu Liu Yanbing Zhou Qingliang He Yantao Tian Hankun Hao Wei Lin Lixin Jiang Gang Zhao Ping Li Chaohui Zheng Changming Huang on behalf of the Study Group for Gastric Neuroendocrine Tumors 2021Chinese Journal of Cancer Research2021,33,4:2
12Orbital ordering and fluctuations in a kagome superconductor CsV_(3)Sb_(5)显示文摘Recently,competing electronic instabilities,including superconductivity and density-wave-like order,have been discovered in vanadium-based kagome metals AV_(3)Sb_(5)(A=K,Rb,Cs)with a nontrivial band topology.This finding stimulates considerable interest to study the interplay of these competing electronic orders and possible exotic excitations in the superconducting state.Here,we performed51V and133Cs nuclear magnetic resonance(NMR)measurements on a CsV_(3)Sb_(5)single crystal to clarify the nature of density-wave-like transition in these kagome superconductors.A first-order structural transition is unambiguously revealed below T_(s)~94 K by observing the sudden splitting of Knight shift in^(51)V NMR spectrum.Moreover,combined with^(133)Cs NMR spectrum,the present result confirms a three-dimensional structural modulation.By further analyzing the anisotropy of Knight shift and 1/T_(1)T at^(51)V nuclei,we proposed that the orbital order is the primary electronic order induced by the firstorder structural transition,which is supported by further analysis on electric field gradient at^(51)V nuclei.In addition,the evidence for possible orbital fluctuations is also revealed above T_(s).The present work sheds light on a rich orbital physics in kagome superconductors AV_(3)Sb_(5).DianWu Song LiXuan Zheng FangHang Yu Jian Li LinPeng Nie Min Shan Dan Zhao ShunJiao Li BaoLei Kang ZhiMian Wu YanBing Zhou KuangLv Sun Kai Liu XiGang Luo ZhenYu Wang JianJun Ying XianGang Wan Tao Wu XianHui Chen 2022Science China(Physics,Mechanics & Astronomy)2022,65,4:2
13Application and exploration of nanofibrous strategy in electrode design显示文摘The key to develop high specific energy rechargeable batteries is development of new electrode materials.The existing electrode materials still have many problems:the shuttle effect and poor conductivity of the sulfur cathode,the inevitable volume expansion of the silicon anode and the lithium dendrite of the lithium metal anode that cause short circuits,etc.Nanofibers,as active electrical materials,conductive additives and electrode bodies,can play multiple roles in electrode design.More interestingly,nanofibers can be functionalized to obtain better controllable properties(i.e.,electrolyte affinity,pore size distribution and surface electronic structure),thereby further enhancing electrochemical performance.In this article,the latest research progress in electrode design based on nanofibers is reviewed,including processing methods,structure,morphology and electrochemical performance.The key problems affecting the electrochemical performance of the electrode are also discussed,such as the preparation process,atomic structure,electrical conductivity,surface area and pore distribution of nanofibers,to provide reference points for nanofibers in excellent electrode design.Yongliang Li Hua Yuan Yanbing Chen Xiaoyu Wei Kunyan Sui Yeqiang Tan 2021Journal of Materials Science & Technology2021,,15:2

15北方半干旱草原生态系统光合参数的季节和年际变异显示文摘生态系统表观量子效率(α)、最大光合速率(P_(max))和暗呼吸速率(R_(d))不仅反映了生态系统水平光合生理特征,同时也是碳循环模型中光合过程模拟的关键参数。气候和植被因子都会影响光合参数的季节和年际变异,但二者在光合参数调控过程中的相对贡献和作用途径尚不清晰。本研究基于连续12年(2006-2017)的涡度相关观测数据,分析了内蒙古半干旱典型草原光合参数的季节和年际变化规律利用回归分析和结构方程模型(SEM)方法明晰了环境和生理调控的作用途径及相对贡献。结果发现,光合参数(α、P_(max)和R)均表现出单峰的季节变化趋势,并呈现明显的年际波动。温度(T_(a))和土壤含水量(SWC)的变化共同影响光合参数的季节变化,而SWC主导了其年际变异。α和R_(d)的变化主要由T_(a)决定,而P_(max)的变化主要受SWC的影响。SEM模型分析表明,除了直接作用外,环境因子主要通过影响冠层水平气孔导度(gc)对光合参数和碳同化生理过程进行调控。此外,叶面积指数对光合参数特别是Pmax的季节和年际变异起主要调控作用。以上结果明确了环境和植被共同决定了生态系统水平光合参数的季节和年际变异,并强调了在水分受限的草原生态系统中,植被生理调控在光合碳同化能力和碳汇功能评估中的重要作用。Cuihai You Yanbing Wang Xingru Tan Bingwei Zhang Tingting Ren Boyu Chen Mengzhen Xu Shiping Chen 2022Journal of Plant Ecology2022,15,5:1

We generated eight monoclonal antibodies(MAbs) targeted to the hemagglutinin(HA) protein of the index virus A/chicken/Shanxi/2/2006 and found that two representative antigenically drifted clade 7.2 viruses did not react with six of the eight MAbs. The E131 N mutation and insertion of leucine at position 134 in the HA protein of the antigenically drifted strains eliminated the reactivity of the virus with the MAbs. We also found that the amino acid N131 in the H5 HA protein is glycosylated. 18Synthesis and Crystal Structure of a New Lanthanum(III) 4-Cyanobenzoate Complex显示文摘A new dinuclear La(III) complex [La2(4-cba)4[H(4-cba)2]2(phen)2(H2O)4] (4-Hcba = 4-cyanobenzoic acid and phen = 1,10-phenanthroline) 1 has been synthesized by solvothermal reaction in an ethanol/water mixed solution at 100 ℃ and structurally characterized by single- crystal X-ray diffraction. Crystallographic data: C88H58La2N12O20, Mr = 1881.28, triclinic P1, a = 7.518(3), b = 17.033(6), c = 17.551(6), α = 115.333(4), β = 92.9910(10), γ = 99.366(5)°, V = 1985.3(12)3, Z = 1, Dc = 1.574 g/cm3, F(000) = 944 μ = 1.145 mm–1, the final R = 0.0281 and wR = 0.0686 for 6708 reflections with I > 2σ(I). In 1, two nine-coordinated La(III) ions are connected by two 4-cba ligands in a syn-syn bidentate coordination mode, and the other six 4-cba ligands terminate the La(III) ions, resulting in an isolated dinuclear structure. Two different types of intramolecular hydrogen bonds, asymmetrical O-H…O and symmetrical O…H…O, exist in the crystal. The title complex molecules are connected through hydrogen bonds and weak π-π stacking interactions to generate a 2-D layered network. 19Influence of the electric field on carriers recombination zone in bilayer organic electroluminescent device显示文摘The electroluminescence (EL) of bilayer devices (ITO/Polymer/Alqa/AI) made from two PPV derivatives (MN-PPV and MEH-PPV), respectively, and the influence of the organic layers’ thickness on the characteristics of the device are investigated. Different spectra and variations are observed for different thicknesses of Alq3 layer in this bilayer organic light-emitting diodes (LEDs) as increasing applied bias.
