| 1 | Naringenin reduces lung metastasis in a breast cancer resection model显示文摘Metastasis is the main cause of death in cancer patients. To improve the outcomes of patients undergoing a surgery, new adjuvant therapies that can effectively inhibit metastases have to be developed. Studies have shown that flavonoid naringenin, a natural product that is mainly present in grapes and citrus, may contribute to cancer prevention. It has many advantages compared to traditional chemotherapeutic drugs, such as low toxicity. To determine whether naringenin can also inhibit metastases, a breast cancer resection model that mimics clinical situations was established. We found that orally administered naringenin significantly decreased the number of metastatic tumor cells in the lung and extended the life span of tumor resected mice. Flow cytometry analysis revealed that T cells displayed enhanced antitumor activity in naringenin treated mice, with an increased proportion of IFN-γ and IL-2 expressing T cells. In vitro studies further demonstrated that relief of immunosuppression caused by regulatory T cells might be the fundamental mechanism of metastasis inhibition by naringenin. These results indicate that orally administered naringenin can inhibit the outgrowth of metastases after surgery via regulating host immunity. | 4 | Effects of Nutritional Factors on the Growth and Heterotrophic Eicosapentaenoic Acid Production of Diatom Nitzschia laevis显示文摘The effects of several nutritional factors on the growth and eicosapentaenoic acid(EPA) production of diatom Nitzschia laevis were studied. 4 LDM(quadrupled concentration of the nutrient salt) was the optimal concentration of nutrient salt for the growth and EPA production of N. laevis. The growth of N. laevis was inhibited when the glucose concentration was either lower than 10 gL-1 or higher than 15 gL-1. Both sodium nitrate and urea were good nitrogen sources for the growth and EPA production,while ammonium chloride seriously decreased the dry cell weight(DW) and the EPA content. Silicate seriously influenced the growth of N. laevis. The maximum DW of 2.34 gL-1 was obtained in the presence of 150 mgL-1 Na2SiO3· 9H2O. The EPA content remained almost the same when the silicate concentration was lower than 150 mgL-1;however,higher silicate concentrations resulted in a steady de-crease of EPA content. Low medium salinity(≤29) did not seem to influence the DW of N. laevis,and high salinity resulted in a decrease of DW. | 5 | Expression of Pseudorabies Virus gE Core Epitopes in Escherichia coli Strain BL21 and Utilization of Indirect PRV gE-ELISA显示文摘Pseudorabies virus glycoprotein E( PRV gE) has been recognized as a suitable diagnostic antigen for pseudorabies. In order to produce gE antigen in large quantities and at low cost,a gene fragment encoding PRV gE core epitopes was expressed in E. coli BL21 expression system. SDS-PAGE and Western Blotting revealed that the expression product in culture supernatant of E. coli BL21 was a recombinant protein,approximately 51. 8 Kd. At 5 h post-induction,protein concentration assay showed that the expression product amounted to 1. 65 mg /ml,accounting for 24. 17% of total proteins in the culture supernatant. An indirect PRV gE-ELISA was established by using the recombinant expression product as a coating antigen. Cross-reactivity assay showed that this antigen was PRV specific. In addition,the assay was consistently reproducible. | 8 | Rdh13 deficiency weakens carbon tetrachloride-induced liver injury by regulating Spotl4 and Cyp2e1 expression levels显示文摘Mitochondrion-localized retinol dehydrogenase 13 (Rdh13) is a short-chain dehydrogenase/reductase involved in vitamin A metabolism in both humans and mice. We previously generated Rdh13 knockout mice and showed that Rdh13 deficiency causes severe acute retinal light damage. In this study, considering that Rdh13 is highly expressed in mouse liver, we further evaluated the potential effect of Rdh13 on liver injury induced by carbon tetrachloride (CC14). Although Rdh13 deficiency showed no significant effect on liver histology and physiological functions under regular culture, the Rdh13^-/- mice displayed an attenuated response to CCl4-induced liver injury. Their livers also exhibited less histological changes and contained lower levels of liver-related metabolism enzymes compared with the livers of wild-type (WT) mice. Furthermore, the Rdhl3 1 mice had Rdh13 deficiency and thus their liver cells were protected from apoptosis, and the quantity of their proliferative cells became lower than that in WT after CC14 exposure. The ablation of Rdhl3 gene decreased the expression levels of thyroid hormone-inducible nuclear protein 14 (Spot14) and cytochrome P450 (Cyp2el) in the liver, especially after CC14 treatment for 48 h. These data suggested that the alleviated liver damage induced by CC14 in Rdh13^-/- mice was caused by Cyp2el enzymes, which promoted reductive CC14 metabolism by altering the status of thyroxine metabolism. | 18 | Transition metal embedded in nonmetal-doped T-carbon [110]: A superior synergistic trifunctional electrocatalyst for HER, OER and ORR显示文摘The design of efficient and low-cost multifunctional electrocatalysts for hydrogen evolution reaction(HER), oxygen evolution reaction(OER) and oxygen reduction reaction(ORR) is critical for the development of clean energy. Two-dimensional(2D) carbon-based nano-materials are becoming more and more popular in heterogeneous catalysis due to their cost-effective and multi-scale tunability as single-atom catalysis(SACs) substrates. In this paper, by using first-principles calculation, we designed and demonstrated a novel macropore T-carbon [110](TC) monolayer as 2D electrocatalyst substrate for HER/OER/ORR, and the synergistic modification of the transition metal and nonmetal atoms(TM-X) were investigated to enhance the multifunctional electrocatalytic performance. We screened out the Co embedded in N-doped TC(Co3@N-TC) from 30 TM@X-TC monolayers as a trifunctional electrocatalysts, which exhibits superior performance for HER/ORR/OER on both thermodynamic and kinetic scales, and with excellent thermal and electrochemical stability. Then, the TC monolayer is naturally macropore with a diameter of 5.36 A and exhibits excellent adsorption capacity for the intermediates and products of the redox reactions. Moreover, we revealed the origin of the electrocatalytic activity using the crystal orbital Hamilton population(COHP) and the molecular orbitals(MOs). The d orbital of Co3@N-TC is significantly hybridized with the p orbital of the intermediates, so that the lone electrons initially occupied in the antibonding state pair up and occupy the downward bonding state, allowing *OH to be appropriately adsorbed onto the TC monolayer.
