| 1 | Novel and potent inhibitors targeting DHODH are broad-spectrum antivirals against RNA viruses including newly-emerged coronavirus SARS-CoV-2显示文摘Emerging and re-emerging RNA viruses occasionally cause epidemics and pandemics worldwide,such as the on-going outbreak of the novel coronavirus SARS-CoV-2.Herein,we identified two potent inhibitors of human DHODH,S312 and S416,with favorable drug-likeness and pharmacokinetic profiles,which all showed broad-spectrum antiviral effects against various RNA viruses,including influenza A virus,Zika virus,Ebola virus,and particularly against SARS-CoV-2.Notably,S416 is reported to be the most potent inhibitor so far with an EC5o of 17 nmol/L and an SI value of 10,505.88 in infec-ted cells.Our results are the first to validate that DHODH is an attractive host target through high antiviral efficacy in vivo and low virus replication in DHODH knock-out cells.This work demonstrates that both S312/S416 and old drugs(Leflunomide/Teriflunomide)with dual actions of antiviral and immuno-regulation may have clinical potentials to cure SARS-CoV-2 or other RNA viruses circulating worldwide,no matter such viruses are mutated or not. | Rui Xiong Leike Zhang Shiliang Li Yuan Sun Minyi Ding Yong Wang Yongliang Zhao Yan Wu Weijuan Shang Xiaming Jiang Jiwei Shan Zihao Shen Yi Tong Liuxin Xu Yu Chen Yingle Liu Gang Zou Dimitri Lavillete Zhenjiang Zhao Rui Wang Lili Zhu Gengfu Xiao Ke Lan Honglin Li Ke Xu | 2020 | Protein & Cell2020,11,10: | 14 |
| 2 | “Exploring petroleum inside source kitchen”: Shale oil and gas in Sichuan Basin显示文摘The Sichuan Basin is rich in shale oil and gas resources,with favorable geological conditions that the other shale reservoirs in China cannot match.Thus,the basin is an ideal option for fully'exploring petroleum inside source kitchen'with respect to onshore shale oil and gas in China.This paper analyzes the characteristics of shale oil and gas resources in the United States and China,and points out that maturity plays an important role in controlling shale oil and gas composition.US shale oil and gas exhibit high proportions of light hydrocarbon and wet gas,whereas Chinese marine and transitional shale gas is mainly dry gas and continental shale oil is generally heavy.A comprehensive geological study of shale oil and gas in the Sichuan Basin reveals findings with respect to the following three aspects.First,there are multiple sets of organic-rich shale reservoirs of three types in the basin,such as the Cambrian Qiongzhusi Formation and Ordovician Wufeng Formation-Silurian Longmaxi Formation marine shale,Permian Longtan Formation transitional shale,Triassic Xujiahe Formation lake-swamp shale,and Jurassic lacustrine shale.Marine shale gas enrichment is mainly controlled by four elements:Deep-water shelf facies,moderate thermal evolution,calcium-rich and silicon-rich rock association,and closed roof/floor.Second,the'sweet section'is generally characterized by high total organic carbon,high gas content,large porosity,high brittle minerals content,high formation pressure,and the presence of lamellation/bedding and natural microfractures.Moreover,the'sweet area'is generally characterized by very thick organic-rich shale,moderate thermal evolution,good preservation conditions,and shallow burial depth,which are exemplified by the shale oil and gas in the Wufeng-Longmaxi Formation,Longtan Formation,and Daanzhai Member of the Ziliujing Formation.Third,the marine,transitional,and continental shale oil and gas resources in the Sichuan Basin account for 50%,25%,and 30%of the respective types of shale oil and gas geological resources in China,with great potential to become the cradle of the shale oil and gas industrial revolution in China.Following the'Conventional Daqing-Oil'(i.e.,the Daqing oilfield in the Songliao Basin)and the'Western Daqing-Oil&Gas'(i.e.,the Changqing oilfield in the Ordos Basin),the Southwest oil and gas field in the Sichuan Basin is expected to be built into a'Sichuan-Chongqing Daqing-Gas'in China. | Caineng ZOU Zhi YANG Shasha SUN Qun ZHAO Wenhua BAI Honglin LIU Songqi PAN Songtao WU Yilin YUAN | 2020 | Science China Earth Sciences2020,63,7: | 6 |

| 4 | Human umbilical cord mesenchymal stem cells for psoriasis:a phase 1/2a,single-arm study显示文摘Psoriasis is a common,chronic immune-mediated systemic disease that had no effective and durable treatment.Mesenchymal stem cells(MSCs)have immunomodulatory properties.Therefore,we performed a phase 1/2a,single-arm clinical trial to evaluate the safety and efficacy of human umbilical cord-derived MSCs(UMSCs)in the treatment of psoriasis and to preliminarily explore the possible mechanisms.Seventeen patients with psoriasis were enrolled and received UMsC infusions.Adverse events,laboratory parameters,PASl,and PGA were analyzed.We did not observe obvious side effects during the treatment and 6-month follow-up.A total of 47.1%(8/17)of the psoriasis patients had at least 40%improvement in the PASl score,and 17.6%(3/17)had no sign of disease or minimal disease based on the PGA score.And the efficiency was 25%(2/8)for males and 66.7%(6/9)for females.After UMSC transplantation(UMSCT),the frequencies of Tregs and CD4^(+)memory T cells were significantly increased,and the frequencies of T helper(Th)17 and CD4^(+)naive T cells were significantly decreased in peripheral blood(PB)of psoriasis patients.And all responders showed significant increases in Tregs and CD4^(+)memory T cells,and significant decreases in Th17 cells and serum IL-17 level after UMsCT.And baseline level of Tregs in responders were significantly lower than those in nonresponders.In conclusion,allogeneic UMSCT is safe and partially effective in psoriasis patients,and level of Tregs may be used as a potent biomarker to predict the clinical efficacy of UMSCT.Trial registration Clinical Trials NCT03765957. | Lamei Cheng Siqi Wang Cong Peng Xiao Zou Chao Yang Hua Mei Chuang Li Xian Su Na Xiao Qi Ouyang Mi Zhang Qiaolin Wang Yan Luo Minxue Shen Qun Qin Honglin Wang Wu Zhu Guangxiu Lu Ge Lin Yehong Kuang Xiang Chen | 2022 | Signal Transduction and Targeted Therapy2022,7,9: | 2 |
| 5 | Quantifying the carbon source of pedogenic calcite veins in weathered limestone: implications for the terrestrial carbon cycle显示文摘The continent is the second largest carbon sink on Earth’s surface.With the diversification of vascular land plants in the late Paleozoic,terrestrial organic carbon burial is represented by massive coal formation,while the development of soil profiles would account for both organic and inorganic carbon burial.As compared with soil organic carbon,inorganic carbon burial,collectively known as the soil carbonate,would have a greater impact on the long-term carbon cycle.Soil carbonate would have multiple carbon sources,including dissolution of host calcareous rocks,dissolved inorganic carbon from freshwater,and oxidation of organic matter,but the host calcareous rock dissolution would not cause atmospheric CO2drawdown.Thus,to evaluate the potential effect of soil carbonate formation on the atmospheric p CO2level,different carbon sources of soil carbonate should be quantitatively differentiated.In this study,we analyzed the carbon and magnesium isotopes of pedogenic calcite veins developed in a heavily weathered outcrop,consisting of limestone of the early Paleogene Guanzhuang Group in North China.Based on the C and Mg isotope data,we developed a numerical model to quantify the carbon source of calcite veins.The modeling results indicate that4–37 wt%of carbon in these calcite veins was derived from atmospheric CO2.The low contribution from atmospheric CO2might be attributed to the host limestone that might have diluted the atmospheric CO2sink.Nevertheless,taking this value into consideration,it is estimated that soil carbonate formation would lower 1 ppm atmospheric CO2within 2000 years,i.e.,soil carbonate alone would sequester all atmospheric CO2within 1 million years.Finally,our study suggests the C–Mg isotope system might be a better tool in quantifying the carbon source of soil carbonate. | Lin Zou Lin Dong Meng Ning Kangjun Huang Yongbo Peng Shujian Qin Honglin Yuan Bing Shen | 2019 | Acta Geochimica2019,38,4: | 2 |
| 6 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 10 | The process and mechanisms of metamorphism and deformation of hornblende-An example from the Hengshan Mountains, Shanxi Province显示文摘Hornblende's plastic and superplastic deformation mechanisms were studied on the basis of some new information obtained from amphibolite samples in the Hengshan Mountains (Hengshan, Shanxi, China).For this purpose, the samples were measured and analyzed by optical microscope (OM), electron probe microanalysis (EPMA), transmission electron microscope (TEM) and electron backscattered diffraction (EBSD) respectively.Because localized strong strain is more than 1000%, it is presumed that structural superplastic deformation was developed during the ductile shearing process.It is calculated that deformation occurred at 650-679℃ and 0.770-0.914 GPa, based on the study of plagioclase-hornblende geothermometry and geobarometry.TEM images show that new hornblende grains developed few dislocations and hornblende crystals exhibit straight grain boundaries.EBSD of strongly deformed hornblendes reveals a fabric dominated by a {100} <001> lattice preferred orientation (LPO), indicative of new grain recrystallization along the lineation direction (X axis).Thus superplastic deformation of hornblendes from the Hengshan Mountains was developed by mainly solid-state diffusive mass transfer and grain boundary sliding. | JI Mo HU Ling LIU JunLai SHEN Liang ZOU YunXin SONG HongLin MIAO PeiSen | 2012 | Science China Earth Sciences2012,55,12: | 0 |
| 11 | Value of TCD foaming Test in Screening Patent Foramen Ovale in Migraine Patients and Efficacy of PFO Occlusion显示文摘Objective:To explore the effect of TCD foaming test in screening patent foramen ovale in migraine patients,as well as in the treatment effect.Methods:From September 2019 to August 2020,236 patients with migraine and 362 patients with normal physical examination were treated in our hospital.According to the random number table method,60 patients with migraine were selected as the observation group,and 60 patients with normal physical examination were selected as the control group.48 cases of PFO were confirmed by TCD foaming test Among the patients with migraine,36 patients received interventional occlusion therapy.The therapeutic effect and VAS score of the two groups were analyzed.Results:Through TCD foaming test,the proportion of PFO in the observation group was 80%,and that in the control group was 13.33%.The proportion of PFO in the observation group was significantly higher than that in the control group,and the difference was statistically significant(χ^(2)=53.5714,P<0.01);The symptoms disappeared in 30 patients and improved in 4 patients;Among the 36 patients who received interventional occlusion therapy,the VAS score of patients before and after the operation was significantly changed,and the proportion of patients whose pain disappeared after the operation was 83.33%,which was significantly lower than that before the operation,and the difference was comparable(χ^(2)=51.4286,P<0.01).Conclusion:TCD foaming test for PFO screening has strong applicability,not only safe and convenient,will not cause trauma to patients,and through interventional occlusion treatment,can significantly improve the clinical symptoms of migraine patients and cure patients. | Lilan Du Xiaorong Guo Honglin Zhang Min Zhang Guangli Zou | 2021 | Proceedings of Anticancer Research2021,5,2: | 0 |
