| 4 | Gut Microbiota Dysbiosis Strengthens Kupffer Cell-mediated Hepatitis B Virus Persistence through Inducing Endotoxemia in Mice显示文摘Background and Aims:Change of gut microbiota com-position is associated with the outcome of hepatitis B virus(HBV)infection,yet the related mechanisms are not fully characterized.The objective of this study was to investigate the immune mechanism associated with HBV persistence induced by gut microbiota dysbiosis.Methods:C57BL/6 mice were sterilized for gut-microbiota by using an antibi-otic(ABX)mixture protocol,and were monitored for their serum endotoxin(lipopolysaccharide[LPS])levels.An HBV-replicating mouse model was established by performing HBV-expressing plasmid pAAV/HBV1.2 hydrodynamic injec-tion(HDI)with or without LPS,and was monitored for se-rum hepatitis B surface antigen,hepatitis B e antigen,HBV DNA,and cytokine levels.Kupffer cells(KCs)were purified from antibiotic-treated mice and HBV-replicating mice and analyzed for IL-10 production and T cell suppression ability.Results:ABX treatment resulted in increased serum LPS levels in mice.The KCs separated from both ABX-treated and LPS-treated HBV-replicating mice showed significantly increased IL-10 production and enhanced ability to suppress IFN-γproduction of TCR-activated T cells than the KCs sep-arated from their counterpart controls.HDI of pAAV/HBV1.2 in combination with LPS in mice led to a delayed HBV clear-ance and early elevation of serum IL-10 levels compared to pAAV/HBV1.2 HDI alone.Moreover,IL-10 function blockade or KC depletion led to accelerated HBV clearance in LPS-treated HBV-replicating mice.Conclusions:Our results suggest that dysbiosis of the gut microbiota in mice leads to endotoxemia,which induces KC IL-10 production and strengthens KC-mediated T cell suppression,and thus fa-cilitates HBV persistence. | Wenqing Zhou Jinzhuo Luo Xiaohong Xie Shangqing Yang Dan Zhu Hongming Huang Dongliang Yang Jia Liu | 2022 | Journal of Clinical and Translational Hepatology2022，10，1： | 4 |

| 17 | Anisotropic magnetoelastic response in the magnetic Weyl semimetal Co_(3)Sn_(2)S_(2)显示文摘Co_(3)Sn_(2)S_(2) is a recently identified magnetic Weyl semimetal in Shandite compounds. Upon cooling, Co_(3)Sn_(2)S_(2) undergoes a ferromagnetic transition with c-axis polarized moments(0.3 μ_(B)/Co) around T_(C)= 175 K, followed by another magnetic anomaly around T_(A)≈ 140 K. A large intrinsic anomalous Hall effect is observed in the magnetic state below TC with a maximum of anomalous Hall angle near T_(A). Here, we report an elastic neutron scattering on the crystalline lattice of Co_(3)Sn_(2)S_(2) in a magnetic field up to 10 T. A strongly anisotropic magnetoelastic response is observed, while only a slight enhancement of the Bragg peaks is observed when B//c. The in-plane magnetic field(B//ab) dramatically suppresses the Bragg peak intensity probably by tilting the moments and lattice toward the external field direction. The in-plane magnetoelastic response commences from T_(C), and as it is further strengthened below T_(A), it becomes nonmonotonic against the field between T_(A) and T_(C) because of the competition from another in-plane magnetic order. | 20 | Transcriptome and carotenoid profiling of different varieties of Coffea arabica provides insights into fruit color formation显示文摘The processability and ultimate quality of coffee(Coffea arabica)are determined by the composition of the matured fruits.The basis of genetic variation in coffee fruit quality could be explained by studying color formation during fruit maturation.Transcriptome profiling was conducted on matured fruits of four C.arabica varieties(orange colored fruits(ORF);purple colored fruits(PF);red colored fruits(RF)and yellow colored fruits(YF))to identify key color-regulating genes,biosynthesis pathways and transcription factors implicated in fruit color formation.A total of 39,938 genes were identified in the transcriptomes of the four C.arabica varieties.In all,2745,781 and 1224 differentially expressed genes(DEGs)were detected in YF_vs_PF,YF_vs_RF and YF_vs_ORF,respectively,with 1732 DEGs conserved among the three pairwise groups.Functional annotation of the DEGs led to the detection of 28 and 82 key genes involved in the biosynthesis of carotenoids and anthocyanins,respectively.Key transcription factors bHLH,MYB,NAC,MADS,and WRKY implicated in fruit color regulation were detected.The high expression levels of gene-LOC113688784(PSY),gene-LOC113730013(b-CHY),gene-LOC113728842(CCD7),gene-LOC113689681(NCED)and gene-LOC113729473(ABA2)in YF may have accounted for the yellow coloration.The differential expression of several anthocyanin and carotenoid-specific genes in the fruits substantially account for the purple(PF),red(RF),and orange(ORF)colorations.This study provides important insights into fruit color formation and variations in C.arabica and will help to develop coffee varieties with specific color and quality traits. | Faguang Hu Xiaofei Bi Hongming Liu Xingfei Fu Yanan Li Yang Yang Xiaofang Zhang Ruirui Wu Guiping Li Yulan Lv Jiaxiong Huang Xinping Luo Rui Shi | 2022 | Plant Diversity2022，44，3： | 0 |
