| 1 | miR-25 modulates triacylglycerol and lipid accumulation in goat mammary epithelial cells by repressing PGC-1beta显示文摘Background: The goat(Caprahircus) is one of the most important livestock animals. Goat milk fat is an important component in the nutritional quality of goat milk. Growing evidence points to the critical roles of microRNAs(miRNAs) in lipid metabolism.Results: Using a highly sensitive method of S-poly(T) plus for miRNAs detection, we analyze the expression patterns of 715 miRNAs in goat mammary gland tissues at different stages of lactation. We observed that miR-25 expression had an inverse relationship with milk production. Overexpression of miR-25 significantly repressed triacylglycerol synthesis and lipid droplet accumulation. To explore the regulatory mechanism of miR-25 in milk lipid metabolism,we analyzed its putative target genes with bioinformatics analysis followed by 3′-UTR assays. Peroxisome proliferative activated receptor gamma coactivator 1 beta(PGC-1 beta), a key regulator of lipogenics was identified as a direct target of miR-25 with three specific sites within its 3′-UTR. | 4 | Ample dietary fat reduced the risk of primary vesical calculi by inducing macrophages to engulf budding crystals in mice显示文摘Although primary vesical calculi is an ancient disease,the mechanism of calculi formation remains unclear.In this study,we established a novel primary vesical calculi model with D,L-choline tartrate in mice.Compared with commonly used melamine and ethylene glycol models,our model was the only approach that induced vesical calculi without causing kidney injury.Previous studies suggest that proteins in the daily diet are the main contributors to the prevention of vesical calculi,yet the effect of fat is overlooked.To assay the relationship of dietary fat with the formation of primary vesical calculi,D,L-choline tartrate-treated mice were fed a high-fat,low-fat,or normal-fat diet.Genetic changes in the mouse bladder were detected with transcriptome analysis.A high-fat diet remarkably reduced the morbidity of primary vesical calculi.Higher fatty acid levels in serum and urine were observed in the high-fat diet group,and more intact epithelia in bladder were observed in the same group compared with the normal-and low-fat diet groups,suggesting the protective effect of fatty acids on bladder epithelia to maintain its normal histological structure.Transcriptome analysis revealed that the macrophage differentiation-related gene C-X-C motif chemokine ligand 14(Cxcl14)was upregulated in the bladders of high-fat diet-fed mice compared with those of normal-or low-fat diet-fed mice,which was consistent with histological observations.The expression of CXCL14 significantly increased in the bladder in the high-fat diet group.CXCL14 enhanced the recruitment of macrophages to the crystal nucleus and induced the transformation of M2 macrophages,which led to phagocytosis of budding crystals and prevented accumulation of calculi.In human bladder epithelia(HCV-29)cells,high fatty acid supplementation significantly increased the expression of CXCL14.Dietary fat is essential for the maintenance of physiological functions of the bladder and for the prevention of primary vesical calculi,which provides new ideas for the reduction of morbidity of primary vesical calculi. | Huiling Chen Kaiqiang Hu Yaru Liang Yuqi Gao Chenye Zeng Kang Xu Xiaojin Shi Liyuan Li Yuemiao Yin Yi Qiao Ying Qiu Qingfei Liu Zhao Wang | 2022 | Acta Pharmaceutica Sinica B2022,12,2: | 1 |

BACKGROUND The purposes of prcsent study aimed at comparing the protecting effect of some antileukocytic drugs: chloroquine, Colchieine, Cyclophosphamid (CTX) ,Cyclosporin A, Multiglycoside tripteygil against cerebral ischemic injury in MCAO models in the rats. METHODS 130 SD rats with MCAO model randomly divided into one control group and 10 treatment groups. The observation items were as follows: (1) The infarction volume. (2) Brain edema. (3) Histopathoiogical evaluation. (4) The count of neurons and microglia cells in the infarction area. (5) Neurological dysfunction score. (6) The determination of serum lL-1 β and TNF- α levels. RESULTS Antileukocytic drugs can decrease the volume of infarction, brain edema, inflammatory reaction and neuron death of ischemic injury, the low dose of chloroquine plus colchicine plus CTX had better effects, and prolonged therapeutic time windows. DISCUSSION The accumulation of infiammatory cell and cytoking release from them can promote and increase neuron death in infarction area. | 6 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 7 | Applications of EMD to analyses of high-frequency beachface responses to Storm Bebinca in the Qing’an Bay,Guangdong Province,China显示文摘On average,five to six storms occur in the Qiongzhou Strait every year,causing significant damage to coastal geomorphology and several property losses.Tropical Storm Bebinca is the most unusual and complex storm event that has occurred in this region over the last 10 years.To detect the high-frequency beachface responses to the storm,a pressure sensor was deployed in the surf zone to record the free sea surface height,and the heights of grid pile points on the beachface were measured manually to determine beach elevation changes during this storm.Empirical Mode Decomposition and related analysis techniques were used to analyze the high-frequency topography and wave data.The results showed that:(1)the beachface response process occurred in three stages.The first stage was the rapid response stage,wherein the spring tide berm began to erode significantly,and the front edge of the beach berm reacted closely.The two beach sections resisted the harmful energy of the main storm.In the second stage,the beach slope increased after a large sediment loss on the beach berm and its front edge.To adapt to the storm energy,the beach at the low tide line began to erode,and the beach slope decreased.In the third stage,after the storm turned,the wave energy was significantly attenuated,and the beach berm eroded to resist the residual wave energy.The beachface began to oscillate and recover.(2)The main wave surface was the superimposed product of a few internal mode functions.Similar results were observed in beachface changes.High-frequency driving factors determine the local characteristics of beach evolution,and low-frequency driving factors determine the beach evolution trend.(3)The response of sediment to the storm was not a single sea-transportation,but a single-or two-way conversion driven by factors such as wave energy,swash flow,and secondary wave breaking.(4)TheΩ-RTR model is not completely applicable to beach states that undergo rapid changes during storms.Therefore,it is necessary to carry out further research on beach state identification during storms. | Taihuan Hu Zhiqiang Li Chunhua Zeng Gaocong Li Huiling Zhang | 2022 | Acta Oceanologica Sinica2022,41,5: | 0 |
| 8 | Influence of Nozzle Orifice Shape on the Atomization Process of Si3N4 in a Dry Granulation Process显示文摘In order to reveal the intrinsic fluid-dynamic mechanisms of a pressure-swirl nozzle used for Si3N4 dry granulation,and effectively predict its external spray characteristics,the dynamics of air-atomized liquid two-phase flow is analyzed using a VOF(Volume of Fraction)method together with the modified realizable k-εturbulence model.The influence of nozzle orifice shape on velocity distribution,pressure distribution is studied.The results show that the pressure difference in a convergent conical nozzle is the largest with a hollow air core being formed in the nozzle.The corresponding velocity of atomized liquid at nozzle orifice is the largest.Using a self-designed atomization experiment platform,the velocity and pressure of atomized liquid and the spray cone angle are measured for three nozzles with different orifice shapes.The micro-morphology of Si3N4 particles is also determined.These data confirm the correctness of numerical simulation.Considering atomization performance of the nozzle,the contraction conical nozzle is more suitable for the atomization of Si3N4 in practical production based on the dry granulation approach. | Dongling Yu Huiling Zhang Xu Zeng Dahai Liao Nanxing Wu | 2021 | Fluid Dynamics & Materials Processing2021,17,3: | 0 |
| 9 | Amphibian-derived peptide homodimer OA-GL17d promotes skin wound regeneration through the miR-663a/TGF-β1/Smad axis显示文摘Background:Amphibian-derived peptides exhibit considerable potential in the discovery and development of new therapeutic interventions for clinically challenging chronic skin wounds.MicroRNAs(miRNAs)are also considered promising targets for the development of effective therapies against skin wounds.However,further research in this field is anticipated.This study aims to identify and provide a new peptide drug candidate,as well as to explore the underlying miRNA mechanisms and possible miRNA drug target for skin wound healing.Methods:A combination of Edman degradation,mass spectrometry and cDNA cloning were adopted to determine the amino acid sequence of a peptide thatwas fractionated from the secretion of Odorrana andersonii frog skin using gel-filtration and reversed-phase high-performance liquid chromatography.The toxicity of the peptide was evaluated by Calcein-AM/propidium iodide(PI)double staining against human keratinocytes(HaCaT cells),hemolytic activity against mice blood cells and acute toxicity against mice.The stability of the peptide in plasma was also evaluated.The prohealing potency of the peptide was determined by MTS,scratch healing and a Transwell experiment against HaCaT cells,full-thickness injury wounds and scald wounds in the dorsal skin of mice.miRNA transcriptome sequencing analysis,enzyme-linked immunosorbent assay,real-time polymerase chain reaction and western blotting were performed to explore the molecular mechanisms.Results:A novel peptide homodimer(named OA-GL17d)that contains a disulfide bond between the 16th cysteine residue of the peptide monomer and the sequence‘GLFKWHPRCGEEQSMWT’was identified.Analysis showed that OA-GL17d exhibited no hemolytic activity or acute toxicity,but effectively promoted keratinocyte proliferation and migration and strongly stimulated the repair of full-thickness injury wounds and scald wounds in the dorsal skin of mice.Mechanistically,OA-GL17d decreased the level of miR-663a to increase the level of transforming growth factor-β1(TGF-β1)and activate the subsequent TGF-β1/Smad signaling pathway,thereby resulting in accelerated skin wound re-epithelialization and granular tissue formation.Conclusions:Our results suggest that OA-GL17d is a new peptide drug candidate for skin wound repair.This study emphasizes the importance of exogenous peptides as molecular probes for exploring competing endogenous RNA mechanisms and indicates that miR-663a may be an effective target for promoting skin repair. | Yue Zhang Ying Wang Lin Zeng Yixiang Liu Huiling Sun Shanshan Li Siyu Wang Longjun Shu Naixin Liu Saige Yin Junsong Wang Dan Ni Yutong Wu Ying Yang Li He Buliang Meng Xinwang Yang | 2022 | Burns & Trauma2022,10,1: | 0 |
| 10 | Near-field electrospun PCL fibers/GelMA hydrogel composite dressing with controlled deferoxamine-release ability and retiform surface for diabetic wound healing显示文摘Wound ulceration caused by diabetes is a typical chronic wound wherein healing the local tissue is difficult due to lack of blood vessels and tissue necrosis caused by the long-term accumulation of free radicals.Near-field electrospinning(NFES)is an innovative technology used to produce micro-nano-scaled,controllable sequencing fibers.In this study,we constructed a novel wound dressing based on the NFES polycaprolactone(PCL)fiber network and modified gelatin with methacrylic anhydride(GelMA)hydrogel to promote angiogenesis and the re-epithelialization of diabetic wounds.An angiogenic and antioxidant drug named deferoxamine(DFO)was encapsulated in a GelMA hydrogel to achieve a slow-release effect that is more suitable for chronic wounds.The cell adhesion experiment showed that the cells could attach to the fibers in the dressing group having a network of PCL fibers on the surface and grow along the direction of the fibers,which in turn,effectively regulates cell behavior from the physical structure.Additionally,the large pore size(~500μm)of the network allowed the cells to penetrate the pores and enter the surface of the hydrogel without being blocked out.Besides,the composite dressing had a notable effect on angiogenesis.Furthermore,antioxidation experiments confirmed that the DFO-loaded hydrogel exhibited antioxidant activity.Experimental animal models of diabetes showed that rats treated with the PCL-GelMA-DFO(PGD)hydrogel had faster ability of hemostasis,scab formation,and wound healing.In conclusion,the PGD hydrogel effectively promoted the repair of chronic wounds. | Huiling Zhong Jun Huang Moucheng Luo Yifei Fang Xinchen Zeng Jun Wu Jianhang Du | 2023 | Nano Research2023,16,1: | 0 |
