| 2 | Urinary donor-derived cell-free DNA as a non-invasive biomarker for BK polyomavirus-associated nephropathy显示文摘BK polyomavirus-associated nephropathy(BKPyVAN)is a common cause of allograft failure.However,differentiation between BKPyVAN and type I T cell-mediated rejection(TCMR)is challenging when simian virus 40(SV40)staining is negative,because of the similarities in histopathology.This study investigated whether donor-derived cell-free DNA(ddcfDNA)can be used to differentiate BKPyVAN.Target region capture sequencing was applied to detect the ddcfDNAs of 12 recipients with stable graft function,22 with type I TCMR,21 with proven BKPy VAN,and 5 with possible Py VAN.We found that urinary ddcfDNA levels were upregulated in recipients with graft injury,whereas plasma ddcfDNA levels were comparable for all groups.The median urinary concentrations and fractions of ddcfDNA in proven BKPyVAN recipients were significantly higher than those in type I TCMR recipients(10.4 vs.6.1 ng/mL,P<0.001 and 68.4%vs.55.3%,P=0.013,respectively).Urinary ddcfDNA fractions(not concentrations)were higher in the BKPyVAN-pure subgroup than in the BKPyVAN-rejection-like subgroup(81.30%vs.56.64%,P=0.025).With a cut-off value of 7.81 ng/m L,urinary ddcf DNA concentrations distinguished proven BKPyVAN from type I TCMR(area under the curve(AUC)=0.848,95%confidence interval(95%CI):0.734 to 0.963).These findings suggest that urinary ddcf DNA is a non-invasive biomarker which can reliably differentiate BKPy VAN from type I TCMR. | Jia SHEN Luying GUO Wenhua LEI Shuaihui LIU Pengpeng YAN Haitao LIU Jingyi ZHOU Qin ZHOU Feng LIU Tingya JIANG Huiping WANG Jianyong WU Jianghua CHEN Rending WANG | 2021 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,11: | 4 |
| 3 | An elevated preoperative serum calcium level is a significant predictor for positive peritoneal cytology in endometrial carcinoma显示文摘Objective:To evaluate preoperative serum calcium concentration and investigate the association between calcium level and positive peritoneal cytology in endometrial carcinoma(EC).Methods:A total of 510 patients who were diagnosed with EC and had surgery were initially enrolled in this study at Peking University People's Hospital between January 2012 and December 2016.Clinical characteristics and preoperative serum calcium,albumin,carbohydrate antigen(CA)125,CA19-9,carcinoembryonic antigen(CEA)were extracted from patient records and evaluated according to postoperative peritoneal cytology.Predictive factors were assessed with Cox univariate and multivariate analyses.Factors selected from multivariate analysis results were used to build a predictive model.Results:A total of 510 patients are identified in our database and 444 patients who fulfilled inclusion and exclusion criteria are included in this study.Univariate analysis revealed that ionized calcium concentration was closely related to positive peritoneal cytology,tumor grade and lymph-vascular space invasion(LVSI).Moreover,peritoneal cytology was significantly associated with hypertension,tubal ligation,serum CA125,CA19-9,CEA and ionized calcium level.Multivariate analysis revealed that albumin-adjusted calcium level,CA125 and tubal ligation were independent predictive factors of positive peritoneal cytology(P<0.05).A combination of ionized calcium level with the other two indexes yielded significantly great area under the curve(AUC=0.824).Conclusions:This study enhanced the value of preoperative ionized calcium level.We also identified several potential biomarkers to predict positive peritoneal cytology in EC patients before surgery. | Xingchen Li Yuan Cheng Yangyang Dong Boqiang Shen Xiao Yang Jiaqi Wang Jingyi Zhou Jianliu Wang | 2019 | Chinese Journal of Cancer Research2019,31,6: | 3 |

| 6 | Association between time in range and cancer mortality among patients with type 2 diabetes:a prospective cohort study显示文摘Background:Little was known about the association among time in range(TIR),time above range(TAR),time below range(TBR),and cancer mortality among patients with type 2 diabetes.We aimed to investigate the association among TIR,TAR,TBR,and the risk of cancer mortality among patients with type 2 diabetes.Methods:A total of 6225 patients with type 2 diabetes were prospectively recruited in Shanghai,China.TIR was measured with continuous glucose monitoring at baseline and was defined as the average percentage of time in the target glucose range during a 24 h period.Cox proportion hazard regression analysis was used to determine the association between TIR and the risk of cancer mortality.Results:During a mean follow-up of 7.10 years,we confirmed 237 death events related to cancer.The multivariable-adjusted hazard ratio(HR)for cancer mortality was 1.32(95%confidence interval[CI]:1.01-1.75)in patients with TIR≤70%compared with those with TIR>70%.When TIR was considered as a continuous variable,the multivariable-adjusted HR for cancer mortality associated with each 10%decrease in TIR was 1.07(95%CI:1.02-1.14).In the site-specific analysis,a significant association between TIR as a continuous variable and the risk of hepatocellular cancer was found(HR:1.24;95%CI:1.09-1.41).However,no relationship between hemoglobin A1c and cancer mortality was observed(HR:1.04;95%CI:0.97-1.10).Conclusions:The present study found an inverse association of TIR with the risk of cancer mortality among patients with type 2 diabetes.New evidence of TIR was added into the clinical practice that TIR may be an optimal target of glycemic control among patients with type 2 diabetes. | Yun Shen Chunfang Wang Yaxin Wang Jingyi Lu Lei Chen Lei Zhang Wei Lu Wei Zhu Gang Hu Tian Xia Jian Zhou | 2022 | Chinese Medical Journal2022,,3: | 1 |

| 8 | A dynamic risk-based early warning monitoring system for population-based management of cardiovascular disease显示文摘Risk prediction tools are crucial for population-based management of cardiovascular disease(CVD).However,most prediction models are currently used to assess long-term risk instead of the risk of short-term CVD onset.We developed a Dynamic Risk-based Early wAming Monitoring(DREAM)system using large-scale,real-time electronic health record data from 2010 to 2020 from the CHinese Electronic health Records Research in Yinzhou study.The dynamic risk scores were derived from a 1:5 matched nested case-control set comprising 70,470 individuals(11,745 CVD events)and then validated in a cohort of 81,205 individuals(5950 CVD events).The individuals were Chinese adults aged 40-79 years without a history of CVD at baseline.Eleven predictors related to vital signs,laboratory tests,and health service utilization were selected to establish the dynamic scores.The proposed scores were significantly associated with the subsequent CVD onset(adjusted odds ratio,1.21;95%confidence interval,1.20-1.23).The area under the receiver operating characteristic curves(AUCs)was 0.6010(0.5929-0.6092)and 0.6021(0.5937-0.6105)for the long-term 10-year CVD risk<10%and≥10%groups in the derivation set,respectively.In the long-term 10-year CVD risk>10%group in the validation set,the change in AUC in addition to the long-term risk was 0.0235(0.0155-0.0315).By increasing the risk threshold from 7 to 16 points,the proportion of true subsequent CVD cases among those given alerts increased from 40.61%to 85.31%.In terms of management efficiency,the number needed to manage per CVD case ranged from 2.46 to 1.17 using the risk scores.With the increasing popularity and integration of EHR systems with wearable technology,the DREAM scores can be incorporated into an early-warning system and applied in dynamic,real-time,EHR-based,automated management to support healthcare decision making for individuals,general practitioners,and policymakers. | Xiaofei Liu Qianqian Li Weiye Chen Peng Shen Yexiang Sun Qi Chen Jinguo Wu Jingyi Zhang Ping Lu Hongbo Lin Xun Tang Pei Gao | 2021 | Fundamental Research2021,1,5: | 1 |
| 9 | Identification of A-to-I RNA editing profiles and their clinical relevance in lung adenocarcinoma显示文摘Adenosine-to-inosine(A-to-I)RNA editing is a widespread posttranscriptional modification that has been shown to play an important role in tumorigenesis.Here,we evaluated a total of 19,316 RNA editing sites in the tissues of 80 lung adenocarcinoma(LUAD)patients from our Nanjing Lung Cancer Cohort(NJLCC)and 486 LUAD patients from the TCGA database.The global RNA editing level was significantly increased in tumor tissues and was highly heterogeneous across patients.The high RNA editing level in tumors was attributed to both RNA(ADAR1 expression)and DNA alterations(mutation load).Consensus clustering on RNA editing sites revealed a new molecular subtype(EC3)that was associated with the poorest prognosis of LUAD patients.Importantly,the new classification was independent of classic molecular subtypes based on gene expression or DNA methylation.We further proposed a simplified model including eight RNA editing sites to accurately distinguish the EC3 subtype in our patients.The model was further validated in the TCGA dataset and had an area under the curve(AUC)of the receiver operating characteristic curve of 0.93(95%CI:0.91-0.95).In addition,we found that LUAD cell lines with the EC3 subtype were sensitive to four chemotherapy drugs.These findings highlighted the importance of RNA editing events in the tumorigenesis of LUAD and provided insight into the application of RNA editing in the molecular subtyping and clinical treatment of cancer. | Cheng Wang Mingtao Huang Congcong Chen Yuancheng Li Na Qin Zijian Ma Jingyi Fan Linnan Gong Hui Zeng Liu Yang Xianfeng Xu Jun Zhou Juncheng Dai Guangfu Jin Zhibin Hu Hongxia Ma Fengwei Tan Hongbing Shen | 2022 | Science China(Life Sciences)2022,65,1: | 1 |

| 11 | How psychiatrists coordinate treatment for COVID-19: a retrospective study and experience from China显示文摘Background Patients with C0VID-19 are at high risk of developing mental health problems;however,the prevalence and management of mental disorders and how psychiatrists coordinate the treatment are unclear.Aims We aimed to investigate the mental health problems of patients infected with C0VID-19 and to identify the role of psychiatrists in the clinical treatment team during the pandemic.We also share the experience of psychiatric consultations of patients with COVID-19 in Shanghai,China.Methods We analysed data from the psychiatric medical records of 329 patients with COVID-19 in the Shanghai Public Health Clinical Center from 20 January to 8 March 2020.We collected information including sociodemographic characteristics,whether patients received psychiatric consultation,mental health symptoms,psychiatric diagnoses,psychiatric treatments and severity level of COVID-19.Results Psychiatric consultations were received by 84(25.5%)patients with COVID-19.The most common symptoms of mental health problems were sleep disorders(75%),anxiety(58.3%)and depressive symptoms(11.9%).The psychiatric consultation rate was highest in critically ill patients(69.2%),with affective symptoms or disturbed behaviour as their main mental health problems.Psychiatric diagnoses for patients who received consultation included acute stress reaction(39.3%),sleep disorders(33.3%),anxiety(15.5%),depression(7.1%)and delirium(4.8%).In terms of psychiatric treatments,86.9%of patients who received psychiatric consultation were treated with psychotropic medications,including non-benzodiazepine sedative-hypnotic agents(54.8%),antidepressants(26.2%),benzodiazepines(22.6%)and antipsychotics(10.7%).Among the 76 patients who were discharged from the hospital,79%had recovered from mental health problems and were not prescribed any psychotropic medications.The symptoms of the remaining 21%of patients had improved and they were prescribed medications to continue the treatment.Conclusions This is the first study to report psychiatric consultations for patients with COVID-19.Our study indicated that a considerable proportion of patients with COVID-19,especially critically ill cases,experienced mental health problems.Given the remarkable effect of psychiatric treatments,we recommend that psychiatrists be timely and actively involved in the treatment of COVID-19. | Ling Yue Jingyi Wang Mingliang Ju Yi Zhu Lianghu Chen Lin Shi Bohai Shi Jun Chen Yifeng Shen | 2020 | General Psychiatry2020,33,4: | 1 |
| 12 | Climate Warming Mitigation from Nationally Determined Contributions显示文摘Individual countries are requested to submit nationally determined contributions(NDCs)to alleviate global warming in the Paris Agreement.However,the global climate effects and regional contributions are not explicitly considered in the countries’decision-making process.In this study,we evaluate the global temperature slowdown of the NDC scenario(ΔT=0.6°C)and attribute the global temperature slowdown to certain regions of the world with a compact earth system model.Considering reductions in CO_(2),CH_(4),N_(2)O,BC,and SO_(2),the R5OECD(the Organization for Economic Co-operation and Development in 1990)and R5ASIA(Asian countries)are the top two contributors to global warming mitigation,accounting for 39.3%and 36.8%,respectively.R5LAM(Latin America and the Caribbean)and R5MAF(the Middle East and Africa)followed behind,with contributions of 11.5%and 8.9%,respectively.The remaining 3.5%is attributed to R5REF(the Reforming Economies).Carbon Dioxide emission reduction is the decisive factor of regional contributions,but not the only one.Other greenhouse gases are also important,especially for R5MAF.The contribution of short-lived aerosols is small but significant,notably SO_(2)reduction in R5ASIA.We argue that additional species beyond CO_(2)need to be considered,including short-lived pollutants,when planning a route to mitigate climate change.It needs to be emphasized that there is still a gap to achieve the Paris Agreement 2-degree target with current NDC efforts,let alone the ambitious 1.5-degree target.All countries need to pursue stricter reduction policies for a more sustainable world. | Bo FU Jingyi LI Thomas GASSER Philippe CIAIS Shilong PIAO Shu TAO Guofeng SHEN Yuqin LAI Luchao HAN Bengang LI | 2022 | Advances in Atmospheric Sciences2022,39,8: | 1 |

| 14 | Heterostructured NiS_(2)@SnS_(2) hollow spheres as superior high-rate and durable anodes for sodium-ion batteries显示文摘Tin-based sulfides have attracted increasing attention as anodes for sodium-ion batteries(SIBs) owing to their high theoretical capacity;however, the poor rate capability and inferior cycling stability caused by the low electrical conductivity, sluggish kinetics and drastic volume variations during cycling have greatly hampered their practical applications. Herein, heterostructured NiS_(2)@SnS_(2) hybrid spheres were delicately designed and constructed by anchoring interconnected SnS_(2) nanosheets on metalorganic frameworks(MOFs)-derived Ni S_(2) hollow spheres coupled with N-doped carbon skeleton through facile solvothermal and sulfurization/carbonization processes. The unique hollow heterostructure with highly conductive carbon matrix can effectively facilitate the charge transfer kinetics and ensure the desired buffer space while endowing more active sites and enhanced structural integrity, as demonstrated by the experimental and density functional theory(DFT) results. Benefitting from these merits, the NiS_(2)@SnS_(2) hybrid composite displays a high reversible capacity of 820 m Ah g^(-1) after 250 cycles at 1 A g^(-1), and retains a value of 673 m Ah g^(-1)after 1,300 cycles at 5 A g^(-1), manifesting the excellent high-rate and durable sodium storage behaviors when applied in SIBs. | 19 | Hepatic Zbtb18 (Zinc Finger and BTB Domain Containing 18) alleviates hepatic steatohepatitis via FXR (Farnesoid X Receptor)显示文摘A lasting imbalance between fatty acid synthesis and consumption leads to non-alcoholic fatty liver disease(NAFLD),coupled with hepatitis and insulin resistance.Yet the details of the underlying mechanisms are not fully understood.Here,we unraveled that the expression of the transcription factor Zbtb18 is markedly decreased in the livers of both patients and murine models of NAFLD.Hepatic Zbtb18 knockout promoted NAFLD features like impaired energy expenditure and fatty acid oxidation(FAO),and induced insulin resistance.Conversely,hepatic Zbtb18 overexpression alleviated hepato-steatosis,insulin resistance,and hyperglycemia in mice fed on a high-fat diet(HFD)or in diabetic mice.Notably,in vitro and in vivo mechanistic studies revealed that Zbtb18 transcriptional activation of Farnesoid X receptor(FXR)mediated FAO and Clathrin Heavy Chain(CLTC)protein hinders NLRP3 inflammasome activity.This key mechanism by which hepatocyte’s Zbtb18 expression alleviates NAFLD and consequent liver fibrosis was further verified by FXR’s deletion and forced expression in mice and cultured mouse primary hepatocytes(MPHs).Moreover,CLTC deletion significantly abrogated the hepatic Zbtb18 overexpression-driven inhibition of NLRP3 inflammasome activity in macrophages.Altogether,Zbtb18 transcriptionally activates the FXR-mediated FAO and CLTC expression,which inhibits NLRP3 inflammasome’s activity alleviating inflammatory stress and insulin resistance,representing an attractive remedy for hepatic steatosis and fibrosis. | Lei Zhang Jiabing Chen Xiaoying Yang Chuangpeng Shen Jiawen Huang Dong Zhang Naihua Liu Chaonan Liu Yadi Zhong Yingjian Chen Kaijia Tang Jingyi Guo Tianqi Cui Siwei Duan Jiayu Li Shangyi Huang Huafeng Pan Huabing Zhang Xiaoqiang Tang Yongsheng Chang Yong Gao | 2024 | Signal Transduction and Targeted Therapy2024,9,2: | 0 |
| 20 | Spy chemistry enables stable protein immobilization on iron oxide nanoparticles with enhanced magnetic properties显示文摘Iron oxide nanoparticles(IONPs)modified with functional proteins hold great promise in the biomedical field.However,conventional protein modification strategies,such as adsorption and covalent coupling,are either unstable or nonspecific,or may result in the changes of protein structure and ultimately the loss of protein activity.Modification of active proteins on small-sized IONPs with a particle size of less than 30 nm is especially difficult due to their high surface energy.Herein,we developed a universal modifica-tion method based on Spy chemistry for rapid and stable protein immobilization on small-sized IONPs,which only requires the presence of active groups on the surface of nanoparticles that can couple with SpyCatcher.In short,the SpyCatcher peptides were first coated on the surface of IONPs by cross-linking with activated groups,and then the SpyTag peptide fused with a model protein(enhanced green fluo-rescent protein,EGFP)was engineered(SpyTag-EGFP)and directly coupled to SpyCatcher-modified IONPs by self-assembly,which is spontaneous and robust while avoiding the effect of chemical reactions on functional protein activity.The obtained EGFP-functionalized IONPs exhibited enhanced and stable green fluorescence and improved magnetic properties.In addition,the cell internalization efficiency of EGFP-functionalized IONPs was significantly increased as compared to unmodified IONPs,providing an ideal solution for efficient cell labeling and tracking.In conclusion,here we report a rapid and easy strategy for EGFP immobilization on IONPs based on Spy chemistry,which could be further adapted to other functional proteins in the future.SpyCatcher-modified IONPs and SpyTag-X(arbitrary functional fusion proteins)hold great potential to be applied as a versatile platform for protein immobilization on IONPs and enable its multifunctional application in the future. | Chu Shi Jingyi Sheng Guangxiang Si Yan Li Yan Shen Ning Gu | 2023 | Journal of Materials Science & Technology2023,,30: | 0 |
