2A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 5Effects of mixture ratio on anaerobic co-digestion with fruit and vegetable waste and food waste of China显示文摘The biochemical methane potentials for typical fruit and vegetable waste (FVW) and food waste (FW) from a northern China city were investigated,which were 0.30,0.56 m 3 CH 4/kgVS (volatile solids) with biodegradabilities of 59.3% and 83.6%,respectively.Individual anaerobic digestion testes of FVW and FW were conducted at the organic loading rate (OLR) of 3 kg VS/(m 3 ·day) using a lab-scale continuous stirred-tank reactor at 35℃.FVW could be digested stably with the biogas production rate of 2.17 m 3/(m 3 ·day) and methane production yield of 0.42 m 3 CH 4/kg VS.However,anaerobic digestion process for FW was failed due to acids accumulation.The effects of FVW:FW ratio on co-digestion stability and performance were further investigated at the same OLR.At FVW and FW mixing ratios of 2:1 and 1:1,the performance and operation of the digester were maintained stable,with no accumulation of volatile fatty acids (VFA) and ammonia.Changing the feed to a higher FW content in a ratio of FVW to FW 1:2,resulted in an increase in VFAs concentration to 1100-1200mg/L,and the methanogenesis was slightly inhibited.At the optimum mixture ratio 1:1 for co-digestion of FVW with FW,the methane production yield was 0.49 m 3 CH 4/kg VS,and the volatile solids and soluble chemical oxygen demand (sCOD) removal efficiencies were 74.9% and 96.1%,respectively.Jia Lin Jiane Zuo Lili Gan Peng Li Fenglin Liu Kaijun Wang Lei Chen Hainan Gan 2011Journal of Environmental Sciences2011,23,8:36
6Multivariate analysis of surface water quality in the Three Gorges area of China and implications for water management显示文摘Multivariate statistical techniques,cluster analysis,non-parametric tests,and factor analysis were applied to analyze a water quality dataset including 13 parameters at 37 sites of the Three Gorges area,China,from 2003–2008 to investigate spatio-temporal variations and identify potential pollution sources.Using cluster analysis,the twelve months of the year were classified into three periods of lowflow (LF),normal-flow (NF),and high-flow (HF);and the 37 monitoring sites were divided into low pollution (LP),moderate pollution (MP),and high pollution (HP).Dissolved oxygen (DO),potassium permanganate index (COD Mn ),and ammonia-nitrogen (NH 4 +-N) were identified as significant variables affecting temporal and spatial variations by non-parametric tests.Factor analysis identified that the major pollutants in the HP region were organic matters and nutrients during NF,heavy metals during LF,and petroleum during HF.In the MP region,the identified pollutants primarily included organic matter and heavy metals year-around,while in the LP region,organic pollution was significant during both NF and HF,and nutrient and heavy metal levels were high during both LF and HF.The main sources of pollution came from domestic wastewater and agricultural activities and runoff;however,they contributed differently to each region in regards to pollution levels.For the HP region,inputs from wastewater treatment plants were significant;but for MP and LP regions,water pollution was more likely from the combined effects of agriculture,domestic wastewater,and chemical industry.These results provide fundamental information for developing better water pollution control strategies for the Three Gorges area.Jian Zhao Guo Fu Kun Lei Yanwu Li 2011Journal of Environmental Sciences2011,23,9:25
7Calycosin improves cognitive function in a transgenic mouse model of Alzheimer's disease by activating the protein kinase C pathway显示文摘The major pathological changes in Alzheimer's disease are beta amyloid deposits and cognitive impairment. Calycosin is a typical phytoestrogen derived from radix astragali that binds to estrogen receptors to produce estrogen-like effects. Radix astragali Calycosin has been shown to relieve cognitive impairment induced by diabetes mellitus, suggesting calycosin may improve the cognitive function of Alzheimer's disease patients. The protein kinase C pathway is upstream of the mitogen-activated protein kinase pathway and exerts a neuroprotective effect by regulating Alzheimer's disease-related beta amyloid degradation. We hypothesized that calycosin improves the cognitive function of a transgenic mouse model of Alzheimer's disease by activating the protein kinase C pathway. Various doses of calycosin(10, 20 and 40 mg/kg) were intraperitoneally injected into APP/PS1 transgenic mice that model Alzheimer's disease. Calycosin diminished hippocampal beta amyloid, Tau protein, interleukin-1 beta, tumor necrosis factor-alpha, acetylcholinesterase and malondialdehyde levels in a dose-dependent manner, and increased acetylcholine and glutathione activities. The administration of a protein kinase C inhibitor, calphostin C, abolished the neuroprotective effects of calycosin including improving cognitive ability, and anti-oxidative and anti-inflammatory effects. 8Xinfeng capsule improves pulmonary function in ankylosing spondylitis patients via NF-κB-iNOS-NO signaling pathway显示文摘OBJECTIVE: To study changes in the nuclear factor-κB p65(NF-κB p65)-inducible nitric oxide synthase(i NOS)-nitric oxide(NO) signaling pathway and the effects of Xinfeng capsules(XFC) in patients with ankylosing spondylitis(AS)METHODS: One hundred twenty patients with AS were randomly divided into an XFC group and a Salazopyrin group. Sixty health subjects were included as a normal control group. In the two treatment groups, pulmonary functional parameters,forced vital capacity(FVC), forced expiratory volume in 1 second(FEV1), maximal voluntary ventilation(MVV), peak expiratory flow(PEF), forced expiratory flow at 25% of forced vital capacity(FEF25),forced expiratory flow at 50% of forced vital capacity(FEF50), and forced expiratory flow at 75% of forced vital capacity(FEF75) were determined. Enzyme linked immunosorbent assays were used for detection of the serum oxidative stress indexes,NF-κB p65, i NOS, NO, reactive oxygen species(ROS), reactive nitrogen species(RNS), malondialdehyde(MDA), superoxide dismutase(SOD), catalase(CAT), total antioxidative capacity(TAOC) and interleukin-4(IL-4), IL-10, IL-1β, and tumor necrosis factor-α(TNF-α) contents. Westergren's method was used for determination of erythrocyte sedimentation rate(ESR). High-sensitivity C-reactive protein(Hs-CRP) was detected with a 7060 full-automatic biochemical analyzer(Hitachi, Japan).RESULTS: The clinical therapeutic effect in the XFC group was significantly superior to that in the Salazopyrin group(P<0.01). Compared with the normal control group, FEV1, MVV, PEF, FEF50, FEF75, SOD, CAT,TAOC, IL-4, IL-10 were significantly lower, and NF-κB p65, i NOS, NO, ROS, RNS, MDA, IL-1β, TNF-α, ESR,and Hs-CRP significantly higher in patients with AS(P<0.01 or P<0.05). Compared with before treatment, FEV1, MVV, PEF, FEF50, FEF75, SOD, CAT, TAOC,IL-4, and IL-10 were significantly increased, and NF-κB p65, i NOS, NO, ROS, RNS, MDA, IL-1β, TNF-α,ESR, CRP, visual analog scales(VAS), Bath ankylosing spondylitis disease active index, Bath ankylosing spondylitis functional index, and Bath ankylosing spondylitis global index significantly decreased in the two treatment groups after treatment(P<0.01 or P<0.05), with significant differences between the XFC and Salazopyrin groups(P<0.01 or P<0.05). Spearman correlation analysis indicated that FEV1, MVV, PEF, FEF50, and FEF75 were positively correlated with SOD, CAT, TAOC, IL-4, and IL-10, and were negatively correlated with NF-κB p65, i NOS,NO, ROS, RNS, MDA, IL-1β, TNF-α, ESR, and CRP.CONCLUSION: Patients with AS have local pathologic changes in the spinal cord and other joints.They also have decreased pulmonary function,which is negatively correlated with the NF-κB-i NOS-NO signaling pathway, oxidative indexes, and inflammatory factors. XFC improves rigidity and pain in spinal joints and other symptoms, laboratory indexes, and pulmonary function. 9Effects of molasses on the fermentation characteristics of mixed silage prepared with rice straw, local vegetable by-products and alfalfa in Southeast China显示文摘This experiment was conducted to study the effect of molasses on the fermentation characteristics of mixed silage ensiled rice straw and vegetable by-products with alfalfa.Mixture(202 g kg^(–1) dry matter(DM))consisting of rice straw,broccoli residue and alfalfa at the ratio of 5:4:1 was ensiled with three experimental treatments:(1)no additives(control);(2)molasses at 2.5%(M1);(3)molasses at 5%(M2)on a fresh matter basis of mixture,respectively.All treatments were packed into laboratory-scale silos,and three silos per treatment were sampled on days 1,3,5,14 and 30.The result showed that the p H value of all mixed silages decreased gradually with the time of ensiling except for the control silage,in which a significant increase(P<0.05)on day 30 occurred.The lactic acid content increased gradually with the time of ensiling and reached the highest value on day 14,and a marked decrease(P<0.05)was found in the control silage on day 30.The changes of acetic acid content showed similar pattern with lactic acid content.A trace amount of propionic and butyric acid contents were found in the three mixed silages during the fermentation period.Comparing to the control,M1 and M2 treatments improved the fermentation quality of mixed silages as indicated by higher(P<0.05)lactic acid contents and lower(P<0.05)p H and ammonia-N contents.The Flieg points also showed that M1 and M2 silages were well preserved,whereas the control silage had a bad quality.Overall,the findings of this study suggested that adding molasses could improve fermentation quality of mixed silage,and M1 was more suitable for practical application.WANG Jian CHEN Lei YUAN Xian-jun GUO Gang LI Jun-feng BAI Yun-feng SHAO Tao 2017Journal of Integrative Agriculture2017,16,3:24
10Relationship between hs-CRP, proMMP-1, TIMP-1 and coronary plaque morphology: intravascular ultrasound study显示文摘Background Rupture of unstable plaque with subsequent thrombus formation is the common pathophy- siological substrate of the acute coronary syndrome (ACS). It is of potential significance to explore the blood indexes predicting plaque characteristics. Little studies have focused on this field. Therefore we investigated the relationship between hypersensitive C-reactive protein (hs-CRP), pro-matrix metalloproteinase-1 (proMMP-1), tissue inhibitors of matrix metalloproteinase-1 (TIMP-1) and coronary plaque morphology. Methods Intravascular ultrasound (IVUS) examination was done in 152 patients with confirmed coronary heart disease before percutaneous coronary intervention from February 2003 to July 2005. Plasma samples of arterial blood were collected prior to the procedure. The level of hs-CRP, proMMP-1 and TIMP-1 were respectively measured by enzyme-linked immunosorbent assay (ELISA). Results Unstable and ruptured plaque were found more frequently in patients with acute myocardial infarction and unstable angina. External elastic membrane cross-sectional area (EEM CSA), plaque area, lipid pool area and plaque burden were significantly larger in ruptured and unstable plaque group. Positive remolding, thinner fabric-cap, smaller minimal lumen cross-sectional area (MLA), dissection and thrombus were significantly more frequent in ruptured and unstable plaque group. The levels of plasma hs-CRP, proMMP-1 and TIMP-1 were higher in ruptured plaque group. hs-CRP>8.94 mg/L was used to predict ruptured plaque with a ROC curve area of 0.76 [95% confidence interval (CI), 67.0%-85.8%], sensitivity of 71.8%, specificity of 77.0% and accuracy of 69.2% (P<0.01), similarly for proMMP-1>0.12 ng/ml with a ROC curve area of 0.69 [95% CI, 58.2%-80.2%], sensitivity of 69.2%, specificity of 75.2% and accuracy of 66.2% (P<0.01), and TIMP-1>83.45 ng/ml with a ROC curve area of 0.67 [95% CI, 56.2%-78.3%], sensitivity of 66.7%, specificity of 61.9% and accuracy of 66.2% (P<0.01). Conclusion The plaque characteristics correlate with the clinical presentation. 15Effects of Xinfeng capsule on pulmonary function based on treg-mediated notch pathway in a rat model of adjuvant arthritis显示文摘OBJECTIVE:To observe the impact of xinfeng xapsule(XFC) on pulmonary function in a rat model of adjuvant arthritis(AA) and to investigate the mechanism of action.METHODS:Forty rats were randomly divided into four groups of ten:normal control(NC);model control(MC);tripterygium glycosides tablet(TPT);and xinfeng capsule(XFC).Except for the NC group,AA was induced in all rats by intracutaneous injection of 0.1 mL Freund's complete adjuvant in the right paw on the 19th day.NC and MC groups were given(0.9%) physiological saline.The TPT and XFC groups were given TPT(10 mg/kg) and XFC(1.2 g/kg),respectively.Thirty days after administration,changes in paw edema(E),the arthritis index(AI),pulmonary function,levels of regulatory T-cells(Treg),ultrastructure of lung tissue,and expression of Notch receptors and ligands in lung tissue were observed.RESULTS:In the MC group,E and the AI were increased and pulmonary function significantly decreased;the structure of alveolar type-II cells was damaged;ratios of Treg in peripheral blood were reduced;and expression of Notch receptors such as Notch3 and Notch4 and ligands such as Delta1 in lung tissue were significantly increased whereas expression of Notch1,Jagged1 and Jagged2 were significantly decreased.After intervention with XFC,E and the AI were decreased;pulmonary function was enhanced;the structure of alveolar type-II cells was improved;and expression of Treg,Notch1,Jagged1,Jagged2 was elevated,whereas that of Notch3,Notch4 and Delta1 was reduced.CONCLUSION:XFC can not only inhibit E and the AI and improve joint symptoms,it can also improve pulmonary function and reduce inflammation in lung tissue.These actions could be carried out through increases in the expression of Treg,Notch receptors(Notch1) and ligands(Jagged1,Jagged2),and reductions in the expression of Notch3,Notch4 and Delta1.These phenomena would reduce the deposition of immune complexes and the inflammatory response in lung tissue,thereby improving joint symptoms and pulmonary function.Lei Wan Jian Liu Chuanbing Huang Yuan Wang Xi Shen Wandong Zhang Guizheng Wang Haixia Fan Yao Ge Ruilian Chen Yunxiang Cao Ruikai Zong Li Lei 2012Journal of Traditional Chinese Medicine2012,32,3:17
16Molecular Epidemiological Characteristics of Streptococcus pyogenes Strains Involved in an Outbreak of Scarlet Fever in China,2011显示文摘Objective To investigate molecular characterization of streptococcus pyogenes isolates involved in an outbreak of scarlet fever in China in 2011.Methods Seventy-four Streptococcal pyogenes involved in an outbreak of scarlet fever were isolated from pediatric patients in the areas with high incidence in China from May to August of 2011.Emm genotyping,pulsed-field gel electrophoresis (PFGE),superantigen (SAg) genes and antimicrobial susceptibility profiling were analyzed for these isolates.Results A total of 4 different emm types were identified.Emm12 was the most prevalent type which contained four predominating PFGE patterns corresponding to four different virulence and superantigen profiles.Emm12 (79.7%) and emml (14.9%) accounted for approximately 94%of all the isolates.The speA gene was all negative in emm12 isolates and positive in emml isolates.All strains were resistant to erythromycin,and 89.4%of them were resistant to erythromycin,tracycline,and clindamycin simultaneously.Conclusion Several highly diversified clones with a high macrolide resistance rate comprise a predominant proportion of circulating strains,though no new emm type was found in this outbreak.The data provide a baseline for further surveillance of scarlet fever,which may contribute to the explanation of the outbreak and development of a GAS vaccine in China.YOU Yuan Hai SONG Yan Yan YAN Xiao Mei WANG Hai Bin ZHANG Meng Han TAO Xiao Xia Li Lei Lei ZHANG Yu Xin JIANG Xi Hong ZHANG Bing Hua ZHOU Hao XIAO Di JIN Lian Mei FENG Zi Jian LUO Feng Ji ZHANG Jian Zhong 2013Biomedical and Environmental Sciences2013,26,11:15
17Effects of viral infection and microbial diversity on patients with sepsis:A retrospective study based on metagenomic next-generation sequencing显示文摘BACKGROUND: The study aims to investigate the performance of a metagenomic next-generationsequencing (NGS)-based diagnostic technique for the identifi cation of potential bacterial and viral infectionsand eff ects of concomitant viral infection on the survival rate of intensive care unit (ICU) sepsis patients.METHODS: A total of 74 ICU patients with sepsis who were admitted to our institution from February1, 2018 to June 30, 2019 were enrolled. Separate blood samples were collected from patients for bloodcultures and metagenomic NGS when the patients’ body temperature was higher than 38 °C. Patients’demographic data, including gender, age, ICU duration, ICU scores, and laboratory results, were recorded.The correlations between pathogen types and sepsis severity and survival rate were evaluated.RESULTS: NGS produced higher positive results (105 of 118;88.98%) than blood cultures(18 of 118;15.25%) over the whole study period. Concomitant viral infection correlated closelywith sepsis severity and had the negative effect on the survival of patients with sepsis. However,correlation analysis indicated that the bacterial variety did not correlate with the severity of sepsis.CONCLUSIONS: Concurrent viral load correlates closely with the severity of sepsis and thesurvival rate of the ICU sepsis patients. 18Microwave-assisted extraction of Shenfu coal and its macromolecule structure显示文摘Coals consist of some molecules trapped within an organic matrix from which some organic compounds can be extracted by solvents. The Soxhlet technique has been widely used for extracting organic compounds. Microwave heating methods may be successfully applied in the field of coal science. Acetone extraction yields and the chemical composition of the extract were investigated using a typical Chinese coal, Shenfu coal, with microwave-assisted extraction. The acetone extract and residue were analyzed by GC/MS and carbon-13 nuclear magnetic resonance spectroscopy respectively. The carbon spectra were converted into several numerical parameters, fa, Ha, Xb, which indicate the difference in macromolecular structure between Shenfu coal and its residue.
