1FLEXIBLE MAGNETIC WHEEL TYPE INTELLIGENT WELDING ROBOT FOR SPHERICAL TANK显示文摘An intelligent welding robot for spherical tank's all-position multi-layer welds is developed. Based on the dynamics analyzing and simulation testing, a flexible magnetic wheel mechanism is created as the robot's walking carriage. It makes the robot directly attracted to the surface of the spherical tank so as to realize the all-position walking and welding without rail. At the same time, a CCD real-time tracing system is developed for the robot to repeatedly trace the all-position and multi-layer seams. 2High-Resolution Genome-wide Association Study Identifies Genomic Regions and Candidate Genes for Important Agronomic Traits in Wheat显示文摘Wheat(Triticum aestivum)is a major staple food crop worldwide.Genetic dissection of important agronomic traits is essential for continuous improvement of wheat yield to meet the demand of the world's growing population.We conducted a large-scale genome-wide association study(GWAS)using a panel of 768 wheat cultivars that were genotyped with 327609 single-nucleotide polymorphisms generated by genotyping-by-sequencing and detected 395 quantitative trait loci(QTLs)for 12 traits under 7 environments.Among them,273 QTLs were delimited to≤1.0-Mb intervals and 7 of them are either known genes(Rht-D,Vrn-B1,and Vrn-D1)that have been cloned or known QTLs(TaGA2ox8,APO1,TaSus1-7B,and Rht12)that were previously mapped.Eight putative candidate genes were identified for three QTLs that enhance spike seed setting and grain size using gene expression data and were validated in three bi-parental populations.Protein sequence analysis identified 33 putative wheat orthologs that have high identity with rice genes in QTLs affecting similar traits.Large r^2 values for additive effects observed among the QTLs for most traits indicated that the phenotypes of these identified QTLs were highly predictable.Results from this study demonstrated that significantly increasing GWAS population size and marker density greatly improves detection and identification of candidate genes underlying a QTL,solidifying the foundation for large-scale QTL fine mapping,candidate gene validation,and developing functional markers for genomics-based breeding in wheat.Yunlong Pang Chunxia Liu Danfeng Wang Paul St.Amand Amy Bernardo Wenhui Li Fang He Linzhi Li Liming Wang Xiufang Yuan Lei Dong Yu Su Huirui Zhang Meng Zhao Yunlong Liangi Hongze Jia Xitong Shen Yue Lu Hongming Jiang Yuye Wu Anfei Li Honggang Wang Lingrang Kong Guihua Bai Shubing Liu 2020Molecular Plant2020,13,9:8
3Coagulation factors VII, IX and X are effective antibacterial proteins against drug-resistant Gram-negative bacteria显示文摘Infections caused by drug-resistant“superbugs”pose an urgent public health threat due to the lack of effective drugs;however, certain mammalian proteins with intrinsic antibacterial activity might be underappreciated. Here, we reveal an antibacterial property against Gram-negative bacteria for factors VII, IX and X, three proteins with well-established roles in initiation of the coagulation cascade. These factors exert antibacterial function via their light chains (LCs). Unlike many antibacterial agents that target cell metabolism or the cytoplasmic membrane, the LCs act by hydrolyzing the major components of bacterial outer membrane, lipopolysaccharides, which are crucial for the survival of Gram-negative bacteria. The LC of factor VII exhibits in vitro efficacy towards all Gram-negative bacteria tested, including extensively drug-resistant (XDR) pathogens, at nanomolar concentrations. It is also highly effective in combating XDR Pseudomonas aeruginosa and Acinetobacter baumannii infections in vivo. Through decoding a unique mechanism whereby factors VII, IX and X behave as antimicrobial proteins, this study advances our understanding of the coagulation system in host defense, and suggests that these factors may participate in the pathogenesis of coagulation disorder-related diseases such as sepsis via their dual functions in blood coagulation and resistance to infection. 4A Teosinte-derived Allele of a MYB Transcription Repressor Confers Multiple Disease Resistance in Maize.显示文摘Natural alleles controlling multiple disease resistances (MDR) are valuable for crop breeding. However, only one MDR gene have been cloned in maize, and molecular mechanisms of MDR are not clear. By map-based cloning, we have cloned a teosinte-derived allele of a resistance gene, Mexicana lesion mimic 1 (ZmMM1), which has a lesion mimic phenotype and confers resistance to northern leaf blight (NLB), gray leaf spot (GLS) and southern corn rust (SCR). Strong MDR conferred by the teosinte allele is linked with the polymorphisms in the 3' untranslated region of the ZmMM1 gene that cause increased accumulation of ZmMM1 protein. ZmMM1 acts as a transcription repressor and negatively regulates transcription of specific target genes including ZmMM1-target gene 3 (ZmMT3), which functions as a negative regulator of plant immunity and associated cell death. 6Homeobox B8 Targets Sterile Alpha Motif Domain-Containing Protein 9 and Drives Glioma Progression显示文摘Gliomas are the most commonly occurring tumors of the central nervous system. Glioblastoma multiforme (GBM) is the most malignant and aggressive brain cancer in adults. Further understanding of the mechanisms underlying the aggressive nature of GBM is urgently needed. Here we identified homeobox B8(HOXB8), a member of the homeobox family, as a crucial contributor to the aggressiveness of GBM. Data mining of publicly accessible RNA sequence datasets and our patient cohorts confirmed a higher expression of HOXB8 in the tumor tissue of GBM patients, and a strong positive correlation between the expression level and pathological grading of tumors and a negative correlation between the expression level and the overall survival rate. We next showed that HOXB8 promotes the proliferation and migration of glioblastoma cells and is crucial for the activation of the PI3K/AKT pathway and expression of epithelial–mesenchymal transition-related genes, possibly through direct binding to the promoter of SAMD9 (Sterile Alpha Motif Domain-Containing Protein 9) and activating its transcription. 9Low-valence ion addition induced more compact passive films on nickel-copper nano-coatings显示文摘Ni-Cu nano-coatings were prepared by pulsed electroplating technique in the baths containing various amount of boric acid. Their microstructure, morphologies and corrosion resistance were characterized in detail. The addition of boric acid strongly influences on the microstructure of the Ni-Cu coatings.The coating with a grain size of 130 nm, obtained from the bath containing 35 g L^-1 boric acid, shows the highest corrosion resistance. This is attributed to the low-valence Cu ion(Cu^+) additions in nickel oxide, which could significantly decrease the oxygen ion vacancy density in the passive film to form a more compact passive film. 14The slope creep law for a soft rock in an open-pit mine in the Gobi region of Xinjiang, China显示文摘The lithology of the strata in the Gobi region of the Xinjiang autonomous region of China is mainly composed of mudstone,silty mudstone,and other soft rocks.Because of the low strength of the rock mass and the serious effects of physical weathering in this area,the slope stability in open-pit mines is poor,and creep deformation and instability can readily occur.Taking the Dananhu No.2 open-pit mine as a typical example,the creep test of a mudstone sample under different stress levels was studied.Then,based on a bottom friction experiment and a FLAC3D numerical simulation,the deformation and failure processes of the slope were analyzed.The stress–displacement curve and the displacement–time curve for the monitoring points were plotted to obtain the relationship between the stress and displacement for the slope of the soft rock.The results showed that the long-term strength of the mudstone was between 8.0 and 8.8 MPa,and that stable creep occurred when the slope was under low stress.The potential failure mode for this type of slope is that the front edge creeps along the weak layer and then a crack is formed at the trailing edge of the slope.When the crack penetrates the weak layer,cutting bedding and bedding sliding occur.The deformation process of the stable creep slope includes an initial deformation stage,an initial creep stage,a constant velocity creep stage and a deceleration creep stage.Hongze Zhao Yu Tian Qingyong Guo Meijie Li Jianwen Wu 2020International Journal of Coal Science & Technology2020,7,2:1

20Translation and rotation of particles in different flow pattern areas of a silo显示文摘The present paper reports the results obtained for translational and rotational velocity profiles of spherical particles for the mixed flow in a conical silo.The discrete element method(DEM)based on Hertz-Mindlin(no slip)with RVD rolling friction contact model is used for simulations.Opposite correlations are found between translational and rotational velocities in different flow areas of the silo.In particular,the abrasion caused by rotation is dominant in the funnel flow area.In addition,increase of the mass flow rate of silo can effectively reduce the abrasion induced by rotation.This highlights that understanding of dynamic characteristics of particles is helpful for optimization of silos and reduction of granular material abrasion.Ran Li Quan Chen Hongze Li Wenzheng Xiu Vladimir Zivkovic Hui Yang 2023Particuology2023,,2:0
