| 1 | Paclitaxel formulation with stable sustained-release behavior and its biological safety evaluation显示文摘Biological safety and stable sustained-release of the drug are two crucial issues involved in the formulation of paclitaxel.Focusing on these issues, by using the FDA approved polylactide as carrier material, soybean lecithin as surfactant and maltodextrin as thickener, paclitaxel loaded PLA microspheres were simply prepared by solvent evaporation, thus guaranteeing the biological safety. The introduction of maltodextrin as a thickener aided to a stable sustained-release of paclitaxel. Surface morphology, particle size, drug loading rate, encapsulation efficiency and in vitro drug release behavior were investigated.Biological safety evaluations such as acute toxicity, allergies, hemolysis, skin stimulation and genotoxicity test were also carried out. | 3 | Single-cell RNA-sequencing reveals the dynamic process and novel markers in porcine spermatogenesis显示文摘Background:Spermatogenesis is the process by which male gametes are formed from spermatogonial stem cells and it is essential for the reliable transmission of genetic information between generations.To date,the dynamic transcriptional changes of defined populations of male germ cells in pigs have not been reported.Results:To characterize the atlas of porcine spermatogenesis,we profiled the transcriptomes of~16,966 testicular cells from a 150-day-old pig testis through single-cell RNA-sequencing(scRNA-seq).The scRNA-seq analysis identified spermatogonia,spermatocytes,spermatids and three somatic cell types in porcine testes.The functional enrichment analysis demonstrated that these cell types played diverse roles in porcine spermatogenesis.The accuracy of the defined porcine germ cell types was further validated by comparing the data from scRNA-seq with those from bulk RNA-seq.Since we delineated four distinct spermatogonial subsets,we further identified CD99 and PODXL2 as novel cell surface markers for undifferentiated and differentiating spermatogonia,respectively.Conclusions:The present study has for the first time analyzed the transcriptome of male germ cells and somatic cells in porcine testes through scRNA-seq.Four subsets of spermatogonia were identified and two novel cell surface markers were discovered,which would be helpful for studies on spermatogonial differentiation in pigs.The datasets offer valuable information on porcine spermatogenesis,and pave the way for identification of key molecular markers involved in development of male germ cells. | Lingkai Zhang Fuyuan Li Peipei Lei Ming Guo Ruifang Liu Ling Wang Taiyong Yu Yinghua Lv Tao Zhang Wenxian Zeng Hongzhao Lu Yi Zheng | 2022 | Journal of Animal Science and Biotechnology2022,13,2: | 2 |

| 7 | Transcriptome-wide Dynamics of m^(6)A mRNA Methylation During Porcine Spermatogenesis显示文摘Spermatogenesis is a continual process that occurs in the testes,in which diploid spermatogonial stem cells(SSCs)differentiate and generate haploid spermatozoa.This highly efficient and intricate process is orchestrated at multiple levels.N^(6)-methyladenosine(m^(6)A),an epigenetic modification prevalent in mRNAs,is implicated in the transcriptional regulation during spermatogenesis.However,the dynamics of m^(6)A modification in non-rodent mammalian species remains unclear.Here,we systematically investigated the profile and role of m^(6)A during spermatogenesis in pigs.By analyzing the transcriptomic distribution of m^(6)A in spermatogonia,spermatocytes,and round spermatids,we identified a globally conserved m^(6)A pattern between porcine and murine genes with spermatogenic function.We found that m^(6)A was enriched in a group of genes that specifically encode the metabolic enzymes and regulators.In addition,transcriptomes in porcine male germ cells could be subjected to the m^(6)A modification.Our data show that m^(6)A plays the regulatory roles during spermatogenesis in pigs,which is similar to that in mice.Illustrations of this point are three genes(SETDB1,FOXO1,and FOXO3)that are crucial to the determination of the fate of SSCs.To the best of our knowledge,this study for the first time uncovers the expression profile and role of m^(6)A during spermatogenesis in large animals and provides insights into the intricate transcriptional regulation underlying the lifelong male fertility in non-rodent mammalian species. | Zidong Liu Xiaoxu Chen Pengfei Zhang Fuyuan Li Lingkai Zhang Xueliang Li Tao Huang Yi Zheng Taiyong Yu Tao Zhang Wenxian Zeng Hongzhao Lu Yinghua Lv | 2023 | Genomics, Proteomics & Bioinformatics2023,21,4: | 1 |
| 8 | Excitatory effect of histamine on neuronal activity of rat cerebellar fastigial nucleus in vitro显示文摘The cerebellar fastigial nucleus (FN) holds an important role in motor control and body balance. Previous studies have revealed that the nucleus is innervated by direct hypothalamocerebellar histaminergic fibers. However, the functional role of histaminergic projection in cerebellar FN has never been established. In this study, we investigated the effect of histamine on neuronal firing of cerebellar FN by using slice preparations. Sixty-five FN cells were recorded from 47 cerebellar slices, and a vast majority of the cells responded to histamine stimulation with an excitatory response (58/65, 89.2%). Perfusing slices with low-Ca2+/high-Mg2+ medium did not block the histamine-induced excitation (n=10), supporting a direct postsynaptic action of histamine on the cells. Furthermore, the excitatory effect of histamine on FN neurons was not blocked by selective histamine H1 receptor antagonist triprolidine (n=15) or chlorpheniramine (n=10), but was effectively suppressed by ranitidine (n=15), a highly selective histamine H2 receptor antagonist. On the other hand, highly selective histamine H2 receptor agonist dimaprit (n=20) instead of histamine H1 receptor agonist 2-pyridylethylamine (n=16) mimicked the ex- citatory effect of histamine on FN neurons. The dimaprit-induced FN neuronal excitation was effectively antagonized by selective histamine H2 receptor antagonist ranitidine (n=13) but not influenced by se- lective histamine H1 receptor antagonist triprolidine (n=15). | 10 | 3D Printed Multi-material Medical Phantoms for Needle-tissue Interaction Modelling of Heterogeneous Structures显示文摘The fabrication of multi-material medical phantoms with both patient-specificity and realistic mechanical properties is of great importance for the development of surgical planning and medical training.In this work,a 3D multi-material printing system for medical phantom manufacturing was developed.Rigid and elastomeric materials are firstly combined in such application for an accurate tactile feedback.The phantom is designed with multiple layers,where silicone ink,Thermoplastic Polyurethane(TPU),and Acrylonitrile Butadiene Styrene(ABS)were chosen as printing materials for skin,soft tissue,and bone,respectively.Then,the printed phantoms were utilized for the investigation of needle-phantom interaction by needle insertion experiments.The mechanical needle-phantom interaction was characterized by skin-soft tissue interfacial puncture force,puncture depth,and number of insertion force peaks.The experiments demonstrated that the manufacturing conditions,i.e.the silicone grease ratio,interfacial thickness and the infill rate,played effective roles in regulating mechanical needle-phantom interaction.Moreover,the influences of material properties,including interfacial thickness and ultimate stress,on needle-phantom interaction were studied by finite element simulation.Also,a patient-specific forearm phantom was printed,where the anatomical features were acquired from Computed Tomography(CT)data.This study provided a potential manufacturing method for multi-material medical phantoms with tunable mechanical properties and offered guidelines for better phantom design. | Jun Yin Manqi Li Guangli Dai Hongzhao Zhou Liang Ma Yixiong Zheng | 2021 | Journal of Bionic Engineering2021,18,2: | 1 |

| 12 | Profiling of miRNAs in porcine Sertoli cells显示文摘Background: Sertoli cells(SCs) create a specialized environment to support and dictate spermatogenesis.MicroRNAs(miRNAs), a kind of ~ 22 nt small noncoding RNAs, have been reported to be highly abundant in mouse SCs and play critical roles in spermatogenesis. However, the miRNAs of porcine SCs remain largely unknown.Methods: We isolated porcine SCs and conducted small RNA sequencing. By comparing miRNAs in germ cells, we systematically analyzed the miRNA expression pattern of porcine SCs. We screened the highly enriched SC miRNAs and predicted their functions by Gene Ontology analysis. The dual luciferase assay was used to elucidate the regulation of tumor necrosis factor receptor(TNFR)-associated factor 3(TRAF3) by ssc-miR-149.Results: The analysis showed that 18 miRNAs were highly expressed in SCs and 15 miRNAs were highly expressed in germ cells. These miRNAs were predicted to mediate SC and germ cell functions. | 20 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment.
