3Responses of root growth of Alhagi sparsifolia Shap. (Fabaceae) to different simulated groundwater depths in the southern fringe of the Taklimakan Desert, China显示文摘Alhagi sparsifolia Shap. (Fabaceae) is a spiny, perennial herb. The species grows in the salinized, arid regions in North China. This study investigated the response characteristics of the root growth and the distribution of one-year-old A. sparsifolia seedlings to different groundwater depths in controlled plots. The ecological adaptability of the root systems of A. sparsifolia seedlings was examined using the artificial digging method. Results showed that: (1) A. sparsifolia seedlings adapted to an increase in groundwater depth mainly through increasing the penetration depth and growth rate of vertical roots. The vertical roots grew rapidly when soil moisture content reached 3%-9%, but slowly when soil moisture content was 13%-20%. The vertical roots stopped growing when soil moisture content reached 30% (the critical soil moisture point). (2) The morphological plasticity of roots is an important strategy used by A. sparsifolia seedlings to obtain water and adapt to dry soil conditions. When the groundwater table was shallow, horizontal roots quickly expanded and tillering increased in order to compete for light resources, whereas when the groundwater table was deeper, vertical roots developed quickly to exploit space in the deeper soil layers. (3) The decrease in groundwater depth was probably responsible for the root distribution in the shallow soil layers. Root biomass and surface area both decreased with soil depth. One strategy of A. sparsifolia seedlings in dealing with the increase in groundwater depth is to increase root biomass in the deep soil layers. The relationship between the root growth/distribution of A. sparsifolia and the depth of groundwater table can be used as guidance for harvesting A. sparsifolia biomass and managing water resources for forage grasses. 6Xa7, a new executor R gene that confers durable and broad-spectrum resistance to bacterial blight disease in rice显示文摘Bacterial blight(BB)is a globally devastating rice disease caused by Xanthomonas oryzae pv.oryzae(Xoo).The use of disease resistance(R)genes in rice breeding is an effective and economical strategy for the control of this disease.Nevertheless,a majority of R genes lack durable resistance for long-term use under global warming conditions.Here,we report the isolation of a novel executor R gene,Xa7,that confers extremely durable,broad-spectrum,and heat-tolerant resistance to Xoo.The expression of Xa7 was induced by incompatible Xoo strains that secreted the transcription activator-like effector(TALE)AvrXa7 or PthXo3,which recognized effector binding elements(EBEs)in the Xa7 promoter.Furthermore,Xa7 induction was faster and stronger under high temperatures.Overexpression of Xa7 or co-transformation of Xa7 with avrXa7 triggered a hypersensitive response in plants.Constitutive expression of Xa7 activated a defense response in the absence of Xoo but inhibited the growth of transgenic rice plants.In addition,analysis of over 3000 rice varieties showed that the Xa7 locuswas found primarily in the indica and aus subgroups.A variation consisting of an 11-bp insertion and a base substitution(G to T)was found in EBEAvrXa7 in the tested varieties,resulting in a loss of Xa7 BB resistance.Through a decade of effort,we have identified an important BB resistance gene and characterized its distinctive interaction with Xoo strains;these findings will greatly facilitate research on the molecular mechanism of Xa7-mediated resistance and promote the use of this valuable gene in breeding.Xifeng Chen Pengcheng Liu Le Mei Xiaoling He Long Chen Hui Liu Shurong Shen Zhandong Ji Xixi Zheng Yuchen Zhang Zhenyu Gao Dali Zeng Qian Qian Bojun Ma 2021Plant Communications2021,2,3:9

8EMBRYONIC FACTOR 19 Encodes a Pentatricopeptide Repeat Protein that is Essential for the Initiation of Zygotic Embryogenesis in Arabidopsis显示文摘Early embryogenesis is the most fundamental developmental process in biology.Screening of ethyl methanesulfonate(EMS)-mutagenized populations of Arabidopsis thaliana led to the identification of a zygote-lethal mutant embryonic factor 19(fac19)in which embryo development was arrested at the elongated zygote to octant stage.The number of endosperm nuclei decreased significantly in fac19 embryos.Genetic analysis showed fac19 was caused by a single recessive mutation with typical mendelian segregation,suggesting equal maternal and paternal contributions of FAC19 towards zygotic embryogenesis.Positional cloning showed that FAC19 encodes a putative mitochondrial protein with 16 conserved pentatricopeptide repeat(PPR)motifs.The fac19 mutation caused a conversion from hydrophilic serine located in a previously unknown domain to hydrophobic leucine.Crosses between FAC19/fac19 and the T-DNA insertion mutants in the same gene failed to complement the fac19 defects,confirming the identity of the gene.This study revealed the critical importance of a PPR protein-mediated mitochondrial function in early embryogenesis.Dali Yu Li Jiang Huaqin Gong Chun-Ming Liu 2012Journal of Integrative Plant Biology2012,54,1:8
9Characterization and fine mapping of an early senescence mutant (es-t) in Oryza sativa L.显示文摘An es-t (early senescence-temporary) mutant, produced by ethylene methylsulfonate treatment of strain Nipponbare, was identified in rice. The leaves of es-t appeared yellow at the seedling stage, and had decreased chlorophyll content. Rust spots were found during growth in es-t, especially at the leaf margin and tip. The plants showed a typical early-senescence phenotype at the milky stage. The leaf surface of es-t appeared smoother than wild-type leaves under a scanning electron microscope, because the leaves lack siliceous protuberances around the stoma. Chloroplasts grow abnormally and are filled with many starch grains in es-t. Paraffin section analysis showed that the development of the sclerenchyma cells and vascular bundles were also abnormal in es-t. Genetic analysis indicated that es-t was controlled by a recessive gene, which was finely mapped to a 42-kb interval on chromosome 5. 10Development of nutritious rice with high zinc/selenium and low cadmium in grains through QTL pyramiding显示文摘Enriching zinc(Zn) and selenium(Se) levels,while reducing cadmium(Cd) concentration in rice grains is of great benefit for human diet and health.Large natural variations in grain Zn, Se, and Cd concentrations in different rice accessions enable Zn/Sebiofortification and Cd-minimization through molecular breeding. Here, we report the development of new elite varieties by pyramiding major quantitative trait loci(QTLs) that significantly contribute to high Zn/Se and low Cd accumulation in grains. A chromosome segment substitution line CSSLGCC7 with the PA64s-derived GCC7 allele in the 93-11 background, exhibited steadily higher Mn and lower Cd concentrations in grains than those of 93-11. This elite chromosome segment substitution line(CSSL) was used as the core breeding material to cross with CSSLs harboring other major QTLs for essential mineral elements, especially CSSLGZC6 for grain Zn concentration and CSSLGSC5 for grain Se concentration. The CSSLGCC7+GZC6 and CSSLGCC7+GSC5 exhibited lower Cd concentration with higher Zn and Se concentrations in grains, respectively. 12Quantitative Trait Loci Mapping of Flag-leaf Ligule Length in Rice and Alignment with ZmLG1 Gene显示文摘A doubled haploid(DH) population,which consists of 120 lines derived from anther culture of a typical indica and japonica hybrid 'CJ06'/'TN1',was used in this study.Ligule lengths of flag leaf were investigated for quantitative trait loci(QTL) mapping using the DH population.Five QTLs(qLL-2,qLL-4,qLL-6,qLL-10 and qLL-12) controlling the ligule length(LL) were detected on chromosomes 2,4,6,10 and 12,with the variances explained 11.4%,13.6%,27.8%,22.1% and 11.0%,respectively.Using four known genes of ZmGL1,ZmGL2,ZmGL3 and ZmGL4 in maize from the MaizeGDB,their homologs in rice were aligned and integrated into the existing simple sequence repeats linkage map by in silico mapping.A ZmLG1 homolog gene,OsLG1 encoding a squamosa promoter binding protein,was located between the markers RM255 and RM280,which is just identical to the interval of qLL-4 on the long arm of chromosome 4.The results are beneficial to dissection of the ligule molecular mechanism and the study of cereal evolution.Dali Zeng Jiang Hu Guojun Dong Jian Liu Longjun Zeng Guangheng Zhang Longbiao Guo Yihua Zhou Qian Qian 2009Journal of Integrative Plant Biology2009,51,4:6

15Generation of obese rat model by transcription activator-like effector nucleases targeting the leptin receptor gene显示文摘The laboratory rat is a valuable mammalian model organism for basic research and drug discovery. Here we demonstrate an efficient methodology by applying transcription activator-like effector nucleases(TALENs) technology to generate Leptin receptor(Lepr) knockout rats on the Sprague Dawley(SD) genetic background. Through direct injection of in vitro transcribed m RNA of TALEN pairs into SD rat zygotes, somatic mutations were induced in two of three resulting pups. One of the founders carrying bi-allelic mutation exhibited early onset of obesity and infertility. The other founder carried a chimeric mutation which was efficiently transmitted to the progenies. Through phenotyping of the resulting three lines of rats bearing distinct mutations in the Lepr locus, we found that the strains with a frame-shifted or premature stop codon mutation led to obesity and metabolic disorders. However, no obvious defect was observed in a strain with an in-frame 57 bp deletion in the extracellular domain of Lepr. This suggests the deleted amino acids do not significantly affect Lepr structure and function. This is the first report of generating the Lepr mutant obese rat model in SD strain through a reverse genetic approach. 16Ecosystem service delivery in Karst landscapes: anthropogenic perturbation and recovery显示文摘Covering extensive parts of China, Karst landscapes are exceptional because rapid and intensive land use change has caused severe ecosystem degradation within only the last 50 years. The twentieth century intensification in food production through agriculture has led to a rapid deterioration of soil quality, evidenced in reduced crop production and rapid loss of soil. In many areas, a tipping point appears to have been passed as basement rock is exposed and 'rocky desertification' dominates. Through the establishment of the 'Soil processes and ecological services in the karst critical zone of SW China'(SPECTRA)Critical Zone Observatory(CZO) we will endevaour to understand the fundmental processes involved in soil production and erosion, and investigate the integrated geophysical-geochemical-ecological responses of the CZ to perturbations. The CZ spans a gradient from undisturbed natural vegetation through human perturbed landscapes.We seek to understand the importance of heterogeneity insurface and below-ground morphology and flow pathways in determining the spatial distribution of key stocks(soil,C, vegetation, etc.) and their control on ecosystem service delivery. We will assess the extent to which the highly heterogeneous critical zone resources can be restored to enable sustainable delivery of ecosystem services. 18An echo detection algorithm for underwater continuous wave active detection显示文摘The model of linear frequency modulation continuous wave(LFMCW)applied in underwater detection and the method for the detection of echo signal and the estimation of target parameters were studied.By analyzing the heterodyne signal,an algorithm with the structure of heterodyne-Fractional Fourier Transform(FRFT)was proposed.To reduce the computation ofsearching targets in a two-dimensional FRFT result.the heterodyne signal would be processed by FRFT at a specific order,after Radon-Ambiguity Transform(RAT)was applied toc cstimate the sweep rate of the signal.Simulations proved that the algorithm can eliminate the coupling phenomenon of distance and velocity of LFMCW,and estimate targets'parameters accurately.The lake trial results showed that the processing gain of LFMCW processed by the algorithm in this paper was 13 dB better than that of the LFM processed by matched filter.The research results indicated that the algorithm applied in LFMCW underwater detection was feasible and effective,and it could estimate targets'parameters accurately and obtain a good detection performance.LIU Dali LIU Yuntao CAI Huizhi 2014Chinese Journal of Acoustics2014,33,1:2
19Integrating photocatalytic reduction of CO2 with selective oxidation of tetrahydroisoquinoline over InP–In2O3 Z-scheme p-n junction显示文摘The development of a facile strategy to construct stable hierarchal porous heterogeneous photocatalysts remains a great challenge for efficient CO2 reduction.Additionally,hole-trapping sacrificial agents(e.g.,triethanolamine,triethylamine,and methanol)are mostly necessary,which produce useless chemicals,and thus cause costs/environmental concerns.Therefore,utilizing oxidation ability of holes to develop an alternative photooxidation reaction to produce value-added chemicals,especially coupled with CO2 photoreduction,is highly desirable.Here,an in situ partial phosphating method of In2O3 is reported for synthesizing In P–In2O3 p-n junction.A highly selective photooxidation of tetrahydroisoquinoline(THIQ)into value-added dihydroisoquinoline(DHIQ)is to replace the hole driven oxidation of typical sacrificial agents.Meanwhile,the photoelectrons of In P–In2O3 p-n junction can induce the efficient photoreduction of CO2 to CO with high selectivity and stability.The evolution rates of DHIQ and CO are 2 and 3.8 times higher than those of the corresponding In2O3 n-type precursor,respectively.In situ irradiated X-ray photoelectron spectroscopy and electron spin resonance are utilized to confirm that the direct Z-scheme mechanism of In P–In2O3 p-n junction accelerate the efficient separation of photocarriers.Bohang Zhao Yi Huang Dali Liu Yifu Yu Bin Zhang 2020Science China Chemistry2020,63,1:2
