| 3 | Lighting up plants with near-infrared fluorescence probes显示文摘Fluorescence imaging is a non-invasive and highly sensitive bioimaging technique that has shown remarkable strides in plant science. It enables real-time monitoring and analysis of biological and pathological processes in plants by labeling specific molecular or cellular structures with fluorescent probes. However, tissue scattering and phytochrome interference have been obstacles for conventional fluorescence imaging of plants in the ultraviolet and visible spectrum, resulting in unsatisfactory imaging quality. Fortunately, advances in near-infrared(NIR) fluorescence imaging technology(650-900 nm) offer superior spatial-temporal resolution and reduced tissue scattering, which is sure to improve plant imaging quality. In this review, we summarize recent progress in the development of NIR fluorescence imaging probes and their applications for in vivo plant imaging and the identification of plant-related biomolecules. | 6 | Extracellular vesicles derived from human dental mesenchymal stem cells stimulated with low-intensity pulsed ultrasound alleviate inflammation-induced bone loss in a mouse model of periodontitis显示文摘Extracellular vesicles(EVs)derived from mesenchymal stem cells(MSCs)have emerged as a new mode of intercellular crosstalk and are responsible for many of the thera-peutic effects of MSCs.To promote the application of MSC-EVs,recent studies have focused on the manipulation of MSCs to improve the production of EVs and EV-mediated activities.The current paper details an optimization method using non-invasive low-intensity pulsed ul-trasound(LIPUS)as the stimulation for improving oral MSC-EV production and effectiveness.Stem cells from apical papilla(SCAP),a type of oral mesenchymal stem cell,displayed inten-sity-dependent pro-osteogenic and anti-inflammatory responses to LIPUS without significant cytotoxicity or apoptosis.The stimuli increased the secretion of EVs by promoting the expres-sion of neutral sphingomyelinases in SCAP.In addition,EVs from LIPUS-induced SCAP exhibited stronger efficacy in promoting the osteogenic differentiation and anti-inflammation of peri-odontal ligament cells in vitro and alleviating oral inflammatory bone loss in vivo.In addition,LIPUS stimulation affected the physical characteristics and miRNA cargo of SCAP-EVs.Further investigations indicated that miR-935 is an important mediator of the pro-osteogenic and anti-inflammatory capabilities of LIPUS-induced SCAP-EVs.Taken together,these findings demonstrate that LIPUS is a simple and effective physical method to optimize SCAP-EV produc-tion and efficacy. | Tingwei Zhang Ziqi Chen Mengyuan Zhu Xuan Jing Xiaohui Xu Xulei Yuan Mengjiao Zhou Yanan Zhang Miao Lu Duanjing Chen Shihan Xu Jinlin Song | 2023 | Genes & Diseases2023,10,4: | 0 |

| 8 | An effector of Erysiphe necator translocates to chloroplasts and plasma membrane to suppress host immunity in grapevine显示文摘The powdery mildew(Erysiphe necator)is a prevalent pathogen hampering grapevine growth in the vineyard.An arsenal of candidate secreted effector proteins(CSEPs)was encoded in the E.necator genome,but it is largely unclear what role CSEPs plays during the E.necator infection.In the present study,we identified a secreted effector CSEP080 of E.necator,which was located in plant chloroplasts and plasma membrane.Transient expressing CSEP080 promotes plant photosynthesis and inhibits INF1-induced cell death in tobacco leaves.We found that CSEP080 was a necessary effector for the E.necator pathogenicity,which interacted with grapevine chloroplast protein VviB6f(cytochrome b6-f complex iron–sulfur subunit),affecting plant photosynthesis.Transient silencing VviB6f increased the plant hydrogen peroxide production,and the plant resistance to powdery mildew.In addition,CSEP080 manipulated the VviPE(pectinesterase)to promote pectin degradation.Our results demonstrated the molecular mechanisms that an effector of E.necator translocates to host chloroplasts and plasma membrane,which suppresses with the grapevine immunity system by targeting the chloroplast protein VviB6f to suppress hydrogen peroxide accumulation and manipulating VviPE to promote pectin degradation. | Bo Mu Zhaolin Teng Ruixin Tang Mengjiao Lu Jinfu Chen Xiangnan Xu Ying-Qiang Wen | 2023 | Horticulture Research2023,10,9: | 0 |
| 9 | Systemic antibiotics increase microbiota pathogenicity and oral bone loss显示文摘Periodontitis is the most widespread oral disease and is closely related to the oral microbiota.The oral microbiota is adversely affected by some pharmacologic treatments.Systemic antibiotics are widely used for infectious diseases but can lead to gut dysbiosis,causing negative effects on the human body.Whether systemic antibiotic-induced gut dysbiosis can affect the oral microbiota or even periodontitis has not yet been addressed.In this research,mice were exposed to drinking water containing a cocktail of four antibiotics to explore how systemic antibiotics affect microbiota pathogenicity and oral bone loss.The results demonstrated,for the first time,that gut dysbiosis caused by long-term use of antibiotics can disturb the oral microbiota and aggravate periodontitis.Moreover,the expression of cytokines related to Th17 was increased while transcription factors and cytokines related to Treg were decreased in the periodontal tissue.Fecal microbiota transplantation with normal mice feces restored the gut microbiota and barrier,decreased the pathogenicity of the oral microbiota,reversed the Th17/Treg imbalance in periodontal tissue,and alleviated alveolar bone loss.This study highlights the potential adverse effects of long-term systemic antibiotics-induced gut dysbiosis on the oral microbiota and periodontitis.A Th17/Treg imbalance might be related to this relationship.Importantly,these results reveal that the periodontal condition of patients should be assessed regularly when using systemic antibiotics in clinical practice. | Xulei Yuan Fuyuan Zhou He Wang Xinxin Xu Shihan Xu Chuangwei Zhang Yanan Zhang Miao Lu Yang Zhang Mengjiao Zhou Han Li Ximu Zhang Tingwei Zhang Jinlin Song | 2023 | International Journal of Oral Science2023,15,1: | 0 |
| 10 | Magnetic field distribution simulation and performance experiment of a magnetic seed-metering device based on the combined magnetic system显示文摘The magnetic seed-metering method is one of the universal approaches for plug tray seeding.Conventional electromagnetic and permanent-magnetic seed-metering devices either cannot supply a desirable magnitude of magnetic force or need a seed-clearing mechanism.Thus the objective of this research was to develop a combined magnetic system(CMS)seed-metering device for generating higher magnetic forces than the electromagnetic ones without using any auxiliary seed-clearing mechanisms.Firstly,the CMS component was designed and its magnetic field distributions in both attractive and clearing states were obtained by finite element method simulations.Secondly,based on the CMS components,a magnetic seed-metering device was developed and validated by a prototype experiment.The simulation results displayed that along the axial direction of the magnetic head,the maximum magnetic flux density in the clearing state was 21.03%of that in the attractive state.In addition,along the radial direction,the proportion was 24.16%.Concerning the spatial magnetic flux density distribution,the magnetic flux density on the seeding planes of CMS components(approximately 60 mT)was higher than that of transitional space between two CMS components(nearly 0 mT).As for the seeding performance experiment,when the rotational speed of the roller was 21 r/min and the exciting current was 0.15 A,the highest single rate was acquired(90.20%).In the same condition,the reseeding rate was 5.88%and the miss-seeding rate was 3.92%.The results suggest that the magnetic field distribution and seeding performance of the developed magnetic seed-metering device are acceptable.Therefore,the developed magnetic seed-metering device can be used in practical plug tray seeding processes. | Wei Liu Jianping Hu Mengjiao Yao Jun Zhao Imran Ali Lakhiar Chuantong Lu Haoran Pan Wei Wang | 2021 | International Journal of Agricultural and Biological Engineering2021,14,1: | 0 |
| 11 | Tuning osteoporotic macrophage responses to favour regeneration by Cu-bearing titanium alloy in Porphyromonas gingivalis lipopolysaccharide-induced microenvironments显示文摘Guided bone regeneration in inflammatory microenvironments of osteoporotic patients with large alve-olar bone defects remains a great challenge.Macrophages are necessary for alveolar bone regeneration via their polarization and paracrine actions.Our previous studies showed that Cu-bearing Ti6AI4V alloys are capable of regulating macrophage responses.When considering the complexity of oral microenvir-onments,the influences of Cu-bearing Ti6AI4V alloys on osteoporotic macrophages in infectious microenvironments are worthy of further investigations.In this study,we fabricated Ti6AI4V-Cu alloy by selective laser melting technology and used Porphyromonas gingivalis lipopolysaccharide(P.g-LPS)to imitate oral pathogenic bacterial infections.Then,we evaluated the impacts of T6AI4V-Cu on osteoporotic macrophages in infectious microenvironments.Our results indicated that Ti6AI4V-Cu not only inhibited the P.g-LPS-induced M1 polarization and pro-inflammatory cytokine production of osteoporotic macrophages but also shifted polarization towards the pro-regenerative M2 phenotype and remarkably promoted antinflammatory cytokine release.In addition,T6AI4V-Cu effectively promoted the actity of COMMD1 to potentially repress NF-κB-mediated transcription.It is concluded that the Cubearing Ti6AI4V alloy results in ameliorated osteoporotic macrophage responses to create a favourable microenvironment under infectious conditions,which holds promise to develop a GBR-barrier membrane for alveolar bone regeneration of osteoporosis patients. | Xiongcheng Xu Yanjin Lu Ling Zhou Mengjiao He Jin Zhuo Quan Zhong Kai Luo Jinxin Lin | 2021 | Regenerative Biomaterials2021,8,1: | 0 |
