| 1 | Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite. | Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie | 2021 | Cell Research2021,31,7: | 12 |
| 2 | The expression and binding properties of the rice WRKY68 protein in the Xa21-mediated resistance response to Xanthomonas oryzae pv. Oryzae显示文摘Plant WRKY transcription factors are involved in various physiological processes, including biotic and abiotic stress responses, as well as developmental processes. In this study, the expression patterns of the WRKY68 protein during interactions between rice 4021 containing the bacterial blight resistance gene Xa21 and Xanthomonas oryzae pv. oryzae(Xoo) were investigated. A possible modified form of the WRKY68 protein appeared in the Xa21-mediated disease resistance response, and its expression levels were similar in compatible and incompatible responses, but differed significantly from that of the mock control treatment, suggesting that WRKY68 may be involved in the bacterial blight response in rice. To further understand WRKY68's roles in the resistance signaling pathway, WRKY68 recombinant protein was expressed in Escherichia coli and a microscale thermophoresis analysis was performed to investigate the interactions between WRKY68 and cis-elements in crucial pathogenesis-related(PR) genes. The results showed that the WRKY68 protein binds to W-boxes in the PR1 b promoter region, with an apparent dissociation constant of 25 nmol L–1, while the binding between WRKY68 and PR10 a was W-box independent. The results suggested that a possible modified form of the WRKY68 protein was induced during the interaction between rice and Xoo, which then regulated the activity of the downstream PR genes by binding with the W-boxes in the PR1 b gene's promoter region. | 4 | Effect of Rotational Speed on the Stability of Two Rotating Side-by-side Circular Cylinders at Low Reynolds Number显示文摘Flow around two rotating side-by-side circular cylinders of equal diameter D is numerically studied at the Reynolds number 40≤ Re ≤200 and various rotation rate θ_i. The incoming flow is assumed to be two-dimensional laminar flow. The governing equations are the incompressible Navier-Stokes equations and solved by the finite volume method(FVM). The ratio of the center-to-center spacing to the cylinder diameter is T/D=2. The objective of the present work is to investigate the effect of rotational speed and Reynolds number on the stability of the flow.The simulation results are compared with the experimental data and a good agreement is achieved. The stability of the flow is analyzed by using the energy gradient theory, which produces the energy gradient function K to identify the region where the flow is the most prone to be destabilized and the degree of the destabilization. Numerical results reveal that K is the most significant at the separated shear layers of the cylinder pair. | 9 | Planting uniformity performance of motor-driven maize precision seeding systems显示文摘Low accuracy planting uniformity affects yield.Seed meter type and forward speed typically interfere with the planting uniformity accuracy of motor-driven seeding systems.Two types of maize precision planters equipped with motor-driven planting systems are investigated in this study to ascertain the rule of planting uniformity in both simulated and field speeds.The simulated speed increases from 5 to 12 km/h at a 1 km/h interval in a laboratory environment.The test results show that the quality of feed index(QTFI)of the two planters decreased by 16.79%and 9.88%.This is primarily attributed to the increase in the miss index(MISS)by 11.62%and 9.70%,respectively.The field speed was set to four levels from 5 to 12 km/h in a field environment.The plant spacing scatter distribution results were analyzed,and the results of the two planters indicated that the average positive difference of the two planters linearly increased with the forward speed,and the negative difference of the two planters did not exhibit a linear correlation.The number of positive moving average points was 2.49 times greater than that of the negative moving average points of the finger pick-up maize precision planter,and 4.49 times in the air-suction maize precision planter.The results indicated that the increase of the positive difference of plant spacing is the major effect factor in the field planting uniformity of the two motor-driven maize precision planters.In addition,the plant spacing corresponded to the distribution frequency of the two planters in field was close to the target seed spacing of 25 cm with a max coefficient of variation(CV)of 21.55%and 20.66%,respectively,and those plant spacing values corresponded to max distribution frequency of the two planters at the four level field speeds were(24.69±0.63)cm and(25.63±0.32)cm,respectively.However,the multiples index(MUL)changed randomly affected by the increasing speed.The research results provide a direction for the optimization design of motor-driven maize precision planters. | Shuo Yang Changyuan Zhai Yuanyuan Gao Hanjie Dou Xueguan Zhao Yakai He Xiu Wang | 2022 | International Journal of Agricultural and Biological Engineering2022,15,5: | 0 |

The nervous system secretes multiple neuropeptides affecting the nervous system as well as immune cells. The precise impact of the neuropeptide CGRP on NLRP3 inflammasome activation is still unclear. Here, we show that CGRP negatively regulates the antibacterial process of host cells. CGRP prevents NLRP3 inflammasome activation and reduces mature IL-1β secretion. Following NLRP3 inflammasome stimulation that triggers endosome leakage, CGRP internalized to endosomal compartments is released into the cell cytosol. Cytosolic CGRP binds directly to NLRP3 and dismantles the NLRP3-NEK7 complex, which is crucial for NLRP3 inflammasome activation. CGRP administration exacerbates bacterial infection, while the treatment with a CGRP antagonist has the opposite effect. | 13 | Magnetic field induced synthesis of (Ni, Zn)Fe_(2)O_(4) spinel nanorod for enhanced alkaline hydrogen evolution reaction显示文摘Recently, the introduction of external fields (light, thermal, magnetism, etc.) during electrocatalysis reactions gradually becomes a new strategy to modulate the catalytic activities. In this work, an external magnetic field was innovatively employed for the synthesis progress of (Ni, Zn)Fe_(2)O_(4) spinel oxide (M-(Ni, Zn)Fe_(2)O_(4)). Results indicated the magnetic field (≤250 mT) would affect the morphology of catalyst due to the existing Fe ions, inducing the M-(Ni, Zn)Fe_(2)O_(4) nanosphere particles to be uniform and coral-like nanorods. In addition, the electronic structure of the catalyst was regulated by the valence state of Fe, changing the bonding of metal to oxygen atoms in different spinel sites. The results manifested that the M-(Ni, Zn)Fe_(2)O_(4) requires a lower overpotential of only 67 mV to deliver 10 mA cm−2 for hydrogen evolution reaction (HER) in alkaline electrolyte. Moreover, M-(Ni, Zn)Fe_(2)O_(4) respectively as the anode and cathode electrode for the overall water splitting, the catalysis system requires a cell voltage of only 1.76 V to gain a current density of 50 mA cm−2, combining with an excellent discharging stability after 10 h.
