Hydrolyzed seawater pearl tablet modulates the immunity via attenuating Th1/Th2 imbalance in an immunosuppressed mouse model显示文摘OBJECTIVE: To investigate whether Hydrolyzed Seawater Pearl tablet(HSPT) could modulate the Th1/Th2 imbalance in an immunosuppressed mouse model with Th1 to Th2 shift induced by Cyclosporine A(Cs A) which can be used in the clinical treatment of Th2 to Th1 shift diseases, and explore the possible mechanism for the adjuvant therapeutic efficacy of HSPT on recurrent respiratory infections(RRI) and acquired immune deficiency syndrome(AIDS).METHODS: The mice were randomly divided into six groups of five animals each, namely normal group, model group, lentinan polysaccharide tablet(LPT) group and three HPST treated groups. HPST treated groups were administered with HPST(0.51,1.02, 2.04 g/kg) via intragastric gavage(i.g) for 30 consecutive days. LPT used as reference drug for positive control, LPT group was administered with LPT(8.2 mg/kg) for 30 consecutive days. Normal group and model group were received distilled water. The animals in model group, LPT group and HPST treated groups were injected intraperitoneally with Cs A(50 mg/kg) to establish the immunosuppressed mice model with Th1 to Th2 shift on the20 th, 22 nd and 24 th day, one hour after the administration of the respective treatment. Animals were sacrificed one hour after the last administration to collect blood and splenic tissue. The proportion of T cells including CD8+ and CD4+ T cells, Th1 and Th2 in peripheral blood of experimental mice were measured by flow cytometric. The protein level in serum and m RNA level in splenic tissue of experimental mice for interleukin(IL)-2, IL-12, interferon-γ(IFN-γ), IL-4, IL-6, IL-10 and IL-13 were measured by enzyme linked immunosorbent assay and fluorescence quantitative polymerase chain reaction respectively.RESULTS: HSPT elevated the proportion of T cells including both CD8+ and CD4+ T cells, in which the proportion of Th1 and Th2 cells increased, while the ratio of Th1/Th2 cells decreased in peripheral blood of the immunosuppressed mouse model with Th1 to Th2 shift induced by Cs A. Furthermore,HSPT elevated both protein and m RNA level of Th1-type cytokines IL-2 and IFN-γ, while had no significant effect on protein and m RNA level of Th1-type cytokine IL-12 and Th2-type cytokines IL-4, IL-6, IL-10, IL-13 in mouse model.CONCLUSION: Our findings suggest that HSPT can increase proportion of T cells including both CD8+and CD4+T cells and induce Th2 to Th1 shift in both cells and cytokines, which probably was the mechanism to account for the adjuvant therapeutic efficacy of HSPT on RRI and AIDS. | LAN Taijin LUO Xuefei MO Mingyue CHEN Zhenxing LUO Fei HAN Siyin LIU Peng LIANG Zhongxiu ZHANG Ting LI Tianyi YAN Qiangqiang LIN Yong LIN Jiang | 2021 | Journal of Traditional Chinese Medicine2021,41,3: | 6

A comprehensive analysis of the Bencao(herbal)small RNA Atlas reveals novel RNA therapeutics for treating human diseases显示文摘Cross-kingdom herbal mi RNA was first reported in 2012.Using a modified herbal extraction protocol,we obtained 73,677,287sequences by RNA-seq from 245 traditional Chinese Medicine(TCM),of which 20,758,257 were unique sequences. We constructed a Bencao(herbal)small RNA(s RNA)Atlas(http://gffzz4a3adbfa122d4a47h5k6vvcv5u9vw6u09.ffgz.tsg.suse.edu.cn),annotated the sequences by sequence-based clustering, and created a nomenclature system for Bencao s RNAs.The profiles of 21,757 mi RNAs in the Atlas were highly consistent with those of plant mi RNAs in mi RBase.Using software tools, our results demonstrated that all human genes might be regulated by s RNAs from the Bencao s RNA Atlas, part of the predicted human target genes were experimentally validated, suggesting that Bencao s RNAs might be one of the main bioactive components of herbal medicines. We established roadmaps for oligonucleotide drugs development and optimization of TCM prescriptions. Moreover, the decoctosome,a lipo-nano particle consisting of 0.5%–2.5%of the decoction, demonstrated potent medical effects. We propose a Bencao(herbal)Index, including small-molecule compounds(SM),protein peptides(P),nucleic acid(N),non-nucleic and non-proteinogenic large-molecule compounds(LM)and elements from Mendeleev's periodic table(E),to quantitatively measure the medical effects of botanic medicine. The Bencao s RNA Atlas is a resource for developing gene-targeting oligonucleotide drugs and optimizing botanical medicine, and may provide potential remedies for the theory and practice of one medicine. | Yinghao Cao Yexuan Lin Na Sun Xinyi Du Yixin Dong Song Mei Xingyu Deng Xiaobei Li Shaoting Guo Kegong Tang Jiaqi Liu Xiangyu Qiao Dandan Zhao Yuhao Qin Cong Zhang Tianyi Xin Xiaohu Shi Congzhao Zhou Tao Dong De-an Guo Benedikt MKessler Dong Xu Jingyuan Song Fengming Huang Xiaoyue Wang Chengyu Jiang | 2023 | Science China(Life Sciences)2023,66,10: | 2
Behavioral responses of blue sheep(Pseudois nayaur) to nonlethal human recreational disturbance显示文摘Nonlethal human recreational activities have been confirmed to have negative effect on wild animals in a number of ways, including changes in behaviors, avoidance of suitable habitats and declines in breeding success.Studies on the anti-disturbance mechanism of wild animals to human disturbance can provide valuable knowledge to the management of wild animals and the evolutionary mechanisms of behavioral adaptation to their habitats. To evaluate how blue sheep(Pseudois nayaur) would react to nonlethal human recreational disturbance,we studied their anti-disturbance strategy towards human disturbance in Suyukou National Forest Park(SNFP),Helan Mountains,Yinchuan of Ningxia Hui Autonomous Region.Using multinomial logistic regression models(MLRMs),we sought to answer two questions:(1) which kind of human recreational behavior would evoke the most serious anti-disturbance behaviors in blue sheep;and(2) how would various ecological factors influence the anti-disturbance strategy of blue sheep to human recreational disturbance? We collected 10 habitat and population variables and evaluated three kinds of reaction of blue sheep-no response, vigilance and flight.A total of 921 observations qualified to enter MLRMs.We found that habitat type(HT),gender(GEN),head direction(HD),visibility index(VI),and disturbance source(DS) were the five variables that significantly influenced the intensity of reactions of blue sheep.Blue sheep were more alert to tourists than to vehicles, and roads were the habitat type that caused the most intensive reaction of alertness where human disturbance was the highest.Females were more vigilant than males.Blue sheep might feel safer when staying in open habitat, and taking a front head direction provided them with the highest vigilance.Based on these results,we present suggestions to SNFP for the management of ecotourism and blue sheep conservation. | JIANG TianYi WANG XiaoMing DING YouZhong LIU ZhenSheng WANG ZhengHuan | 2013 | Chinese Science Bulletin2013,58,17: | 2
Nanomedicine potentiates mild photothermal therapy for tumor ablation显示文摘The booming photothermal therapy(PTT)has achieved great progress in non-invasive oncotherapy, and paves a novel way for clinical oncotherapy.Of note, mild temperature PTT(mPTT)of 42–45°C could avoid treatment bottleneck of the traditional PTT,including nonspecific injury to normal tissues, vasculature and host antitumor immunity. However, cancer cells can resist mPTT via heat shock response and autophagy, thus leading to insufficient mPTT monotherapy to ablate tumor. To overcome the deficient antitumor efficacy caused by thermo-resistance of cancer cells and mono mPTT,synergistic therapies towards cancer cells have been conducted with mPTT.This review summarizes the recent advances in nanomedicine-potentiated mPTT for cancer treatment, including strategies for enhanced single-mode mPTT and mPTT plus synergistic therapies. Moreover, challenges and prospects for clinical translation of nanomedicine-potentiated mPTT are discussed. | Zijun Jiang Tianyi Li Hao Cheng Feng Zhang Xiaoyu Yang Shihao Wang Jianping Zhou Yang Ding | 2021 | Asian Journal of Pharmaceutical Sciences2021,16,6: | 2
Overexpression of LPCAT1 enhances endometrial cancer stemness and metastasis by changing lipid components and activating TGF-β/Smad2/3 signaling pathway显示文摘The incidence of endometrial cancer(EC)increases annually and tends to occur in younger women.A particularly important relationship exists between EC and metabolic disorders. As one of the most important components of lipid metabolism, phospholipids play an indispensable role in metabolic balance.LPCAT1 is a key enzyme regulating phospholipid metabolism. In this study,we perform further investigations to seek mechanistic insight of LPCAT1 in EC.Our results demonstrate that silencing of LPCAT1 inhibits the growth of endometrial cancer, while overexpression of LPCAT1 results in enhanced stemness and metastasis in endometrial cancer cell lines.Meanwhile, the contents of various phospholipids including phosphatidylethanolamine(PE),phosphatidylcholine(PC),and triglyceride(TG)change significantly after overexpression of LPCAT1.In addition, through RNA-sequencing and western blot analysis,we observe that the TGF-β/Smad2/3 signaling pathway is of great importance in the tumor-promoting function of LPCAT1.LPCAT1 promotes the expressions of stem cell-related transcription factors and epithelialmesenchymal transition(EMT)related proteins through the TGF-β/Smad2/3 signaling pathway. Moreover,we find that TSI-01,which can inhibit the activity of LPCAT1,is able to restrain the proliferation of EC cell lines and promote cell apoptosis.Collectively,we demonstrate that LPCAT1 enhances the stemness and metastasis of EC by activating the TGF-β/Smad2/3 signaling pathway and that TSI-01 may have potential use for the treatment of EC. | Tianyi Zhao Rui Sun Xiaohong Ma Lina Wei Yixin Hou Kun Song Jie Jiang | 2022 | Acta Biochimica et Biophysica Sinica2022,54,7: | 2
Activation of the bitter taste sensor TRPM5 prevents high saltinduced cardiovascular dysfunction显示文摘High salt intake is a known risk factor of cardiovascular diseases. Our recent study demonstrated that long-term high salt intake impairs transient receptor potential channel M5(TRPM5)-mediated aversion to high salt concentrations, consequently promoting high salt intake and hypertension;however, it remains unknown whether TRPM5 activation ameliorates cardiovascular dysfunction. Herein we found that bitter melon extract(BME) and cucurbitacin E(CuE), a major compound in BME, lowered high salt-induced hypertension. Long-term BME intake significantly enhanced the aversion to high salt concentrations by upregulating TRPM5 expression and function, eventually decreasing excessive salt consumption in mice. Moreover, dietary BME ameliorated high salt-induced cardiovascular dysfunction and angiotensin II-induced hypertension in vivo. The mechanistic evidence demonstrated that dietary BME inhibited high salt-induced RhoA/Rho kinase pathway overactivation, leading to reduced phosphorylation levels of myosin light chain kinase and myosin phosphatase targeting subunit 1. Furthermore, CuE inhibited vasoconstriction by attenuating L-type Ca^(2+) channel-induced Ca^(2+) influx in vascular smooth muscle cells. To summarize, our findings indicate that dietary BME has a beneficial role in antagonizing excessive salt consumption and thus appears promising for the prevention of high salt-induced cardiovascular dysfunction. | Hao Wu Yuanting Cui Chengkang He Peng Gao Qiang Li Hexuan Zhang Yanli Jiang Yingru Hu Xiao Wei Zongshi Lu Tianyi Ma Daoyan Liu Zhiming Zhu | 2020 | Science China(Life Sciences)2020,63,11: | 2

Amphiphilic Pd@micro-organohydrogels with controlled wettability for enhancing gas-liquid-solid triphasic catalytic performance显示文摘The wettability of catalyst plays an important role in regulating catalytic performance in heterogenous catalysis because the microenvironment around the catalytic sites directly determines the mass transfer process of reactants.Inspired by gas trapped on the surface of subaquatic spiders, amphiphilic micro-organohydrogels with tunable surface wettabilities were developed by anchoring various alkane chains onto a poly(2-(dimethylamino)ethyl methacrylate)(p(DMAEMA))hydrophilic microgel network.Palladium nanoparticles(Pd NPs)were encapsulated in amphiphilic microgels(amphiphilic Pd@M)to catalyze hydrogenation reaction, achieving higher activities than pristine monohydrophilic Pd@M composite. The underwater oleophilicity and aerophilicity of Pd@M composites were quantified by oil/gas adhesion measurements and computational simulations. The higher amphiphilic catalytic activities are attributed to the formation of a gas-oil-solid reaction interface on the catalyst surfaces, allowing rapid transport of H2 and organic substrates through water to the Pd catalytic sites.Additionally, amphiphilic Pd@M composites also exhibit more superior catalytic performance in multi-substrates reaction. | Minghui Zhang Tianyi Zhao Cunming Yu Qian Liu Guangyan Wang Hui Yang Ming Yang Lei Jiang Mingjie Liu | 2022 | Nano Research2022,15,1: | 2
Mirror-enhanced scanning light-field microscopy for long-term high-speed 3D imaging with isotropic resolution显示文摘Various biological behaviors can only be observed in 3D at high speed over the long term with low phototoxicity.Light-field microscopy(LFM)provides an elegant compact solution to record 3D information in a tomographic manner simultaneously, which can facilitate high photon efficiency. However,LFM still suffers from the missing-cone problem, leading to degraded axial resolution and ringing effects after deconvolution. Here,we propose a mirrorenhanced scanning LFM(MiSLFM)to achieve long-term high-speed 3D imaging at super-resolved axial resolution with a single objective,by fully exploiting the extended depth of field of LFM with a tilted mirror placed below samples. To establish the unique capabilities of MiSLFM,we performed extensive experiments,we observed various organelle interactions and intercellular interactions in different types of photosensitive cells under extremely low light conditions. Moreover,we demonstrated that superior axial resolution facilitates more robust blood cell tracking in zebrafish larvae at high speed. | Bo Xiong Tianyi Zhu Yuhan Xiang Xiaopeng Li Jinqiang Yu Zheng Jiang Yihan Niu Dong Jiang Xu Zhang Lu Fang Jiamin Wu Qionghai Dai | 2021 | Light(Science & Applications)2021,10,12: | 1
Interwoven nickel(II)-dimethylglyoxime nanowires in 3D nickel foam for dendrite-free lithium deposition显示文摘Metal skeletons, such as Nickel Foam(NF) has attracted worldwide interests as stable host for lithium metal anode because of its high stability, large specific surface area and high conductivity. However, most metal skeletons have lithophobic surface and uneven current distribution that result in sporadic lithium nucleation and uncontrolled dendrites growth.Herein,we describe a sequential immersing strategy to generate interwoven Nickel(Ⅱ)-dimethylglyoxime(Ni-DMG) nanowires at NF to obtain composite skeleton(NDNF),which can be used as an stable host for Li metal storage. The Ni-DMG has proved effective to realize uniform lithium nucleation and dendrite-free lithium deposition.Combing with the three dimensional(3 D) hierarchical porous structure, the composite host shows a significantly improved coulombic efficiency(CE) than pristine commercial nickel foam. Moreover, the corresponding Li‖Li symmetrical cells can run more than 700 h with low voltage hysteresis 22 mV at 1.0 mA/cm^(2),and Li@NDNF‖LiFePO;full-cell exhibits a high capacity retention of 82.03% at 1.0 C during 630 cycles. These results proved the effectiveness of metal-organic complexes in governing Li metal growth and can be employed as a new strategy for dendrite-free Li metal anode and safe Li metal batteries(LMBs). | Tianyi Zhou Yanlu Mu Jianyang Wu Bing Zhong Chengkai Yang Qian Wang Wen Liu Henghui Zhou Peng Jiang | 2022 | Chinese Chemical Letters2022,33,4: | 1

Bifunctional LiI additive for poly(ethylene oxide)electrolyte with high ionic conductivity and stable interfacial chemistry显示文摘The development of polymer-based solid-state batteries is severely limited by the low ionic conductivity of solid polymer electrolyte and the instable interface between polymer electrolyte and Li-metal anode. In this work, lithium iodide(LiI)as a bifunctional additive was introduced into the poly(ethylene oxide)(PEO)-based electrolyte to improve the ionic conductivity and to construct a stable interphase at the Li/PEO interface.I-anions offer a strong electrostatic interaction with hydrogen atoms on PEO chains(HPEO)and forming massive I–H bonds that cross-link PEO chains, decrease crystallinity of PEO,and thus improve Li~+interchain transport. In addition,LiI participates in the formation of an inorganic-rich interphase layer, which decreases the energy barrier of Li+transport across the interface and thus inhibits the growth of lithium dendrites. As a result, the composite PEO electrolyte with 2 wt%LiI(PEO-2 LiI)presents a very high ionic conductivity of 2.1×10^(-4) S cm-1 and a critical current density of 2.0 m A cm^(-2) at 45°C.Li symmetric cell with this PEO-2 LiI electrolyte exhibits a long-term cyclability over 600 h at 0.2 m A cm^(-2).Furthermore,solid-state LiFePO_(4) and LiNi_(0.8)Mn_(0.1)Co_(0.1)O_(2) batteries with the PEO-2 LiI electrolyte show an impressive electrochemical performance with outstanding cycling stability and rate capability at 45°C. | Haonan Wang Tianyi Hou Hang Cheng Bowen Jiang Henghui Xu Yunhui Huang | 2022 | Journal of Energy Chemistry2022,31,8: | 1
