1Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12
2Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. 3Short-term effects of biochar and gypsum on soil hydraulic properties and sodicity in a saline-alkali soil显示文摘Salt and sodicity of saline-alkali soil adversely affect the construction of ecological landscapes and negatively impact crop production.The reclamation potential of biochar(BC,wheat straw biochar applied at\%by weight),gypsum(G,0.4%by weight),and gypsum coupled with biochar(GBC)was examined in this laboratory-based study by evaluating their effects on a saline-alkali soil(silt loam)with no amendment as a control(CK).Saline ice and fresh water(simulated rainfall)were leached through soil columns to investigate changes in salt content,sodium adsorption ratio(SAR),alkalinity,and pH of the leachate and the soil.Results showed that saturated water content and field water capacity(FWC)significantly increased by 4.4%and 5.6%,respectively,in the BC treatment after a short incubation time.Co-application of biochar and gypsum(GBC)increased soil saturated hydraulic conductivity(Ks)by 58.4%,which was also significantly higher than the sole addition.Electrical conductivity(EC)of the leachate decreased sharply after saline ice leaching;subsequent freshwater leaching accelerated the removal of the rest of the salts,irrespective of the amendment application.However,the application of gypsum(G and GB)significantly enhanced the removal of exchangeable Na^+and reduced leachate SAR.After leaching,the soil salt content decreased significantly for all treatments.The application of gypsum resulted in a significantly lower soil pH,exchangeable sodium percentage(ESP),SAR,and alkalinity values than those recorded for the CK and BC treatments.These results demonstrated that the co-application of gypsum and biochar could improve saline-alkali soil hydraulic conductivity and decrease leaching-induced sodicity over a short period.Yue ZHANG Jingsong YANG Rongjiang YAO Xiangping WANG Wenping XIE 2020Pedosphere2020,30,5:6
4Temporal and Spatial Variability of Dissolved Organic Carbon Concentration in the Xijiang River, South China显示文摘A whole year analysis of riverine dissolved organic carbon (DOC) concentrations in the Xijiang River (XJR), South China, showed that the mean riverine DOC concentration (1.24 mg L-1) in the XJR was notably lower than the averaged value (5.75 mg L-1) of the global riverine DOC concentration in several major rivers. There is an inconspicuous monthly fluctuation of the DOC signal in the XJR, but on a semi-yearly time scale, however, the riverine DOC concentration had significant difference between hydrological seasons. The DOC level during the flood season (1.18 mg L-1) was less than that during the non-flood season (1.40 mg L-1). Owing to the concomitance of the flushing and dilution effects of the runoff during the high-water period, the variation of riverine DOC concentration with discharge in the XJR differed from that reported in many other major rivers. The DOC export flux above the city of Wuzhou was about 0.62× 106 g C km-2 yr-1. The DOC transported during the '056' Massive Flood period comprised 30.35% of the annual total, while the discharge accounted for 36.32% of the total annual flow. 14Wide measurement range and high sensitivity spongy MWCNT/polydimethylsiloxane pressure sensor based on a single-electrode enhanced triboelectric nanogenerator显示文摘Flexible pressure sensors have broad application prospects,such as human motion monitoring and personalized recognition.However,their applicability is limited by complex structures,low output performance,low sensitivity,and narrow measurement range.In this study,we report a single-electrode spongy triboelectric sensor(SSTS)mainly composed of spongy composite multi-walled carbon nanotubes/polydimethylsiloxane(MWCNT/PDMS)film and conductive fabric,which can simultaneously generate contact electrification and electrostatic induction coupling in a single-electrode contact-separation mode.The SSTS combines the triboelectric effect,properties of doping material,and spongy porous structure(soft sugar as a sacrificial template).An SSTS with an MWCNT content of 10 wt%and a porosity of 64%exhibits high sensitivity,a wide measurement range,and excellent linearity.It also displays two sensitivity regions(slopes):1.324 V/kPa from 1.5 to 28 kPa in the low-pressure range and 0.096 V/kPa from 28 to 316.5 kPa in the high-pressure range,with linearities of 0.980 and 0.979,respectively.Furthermore,the SSTS delivers a high-performance output and high stability,thus enhancing the monitoring of hand pressure changes,human movement,personalized spatial recognition,and other detection tasks.This new strategy for human motion monitoring shows great potential in the healthcare fields,sports rehabilitation,and human-computer interactions.XIE Xin HOU XiaoJuan QIAN Shuo HOU JianWei ZHANG Jie BI XiaoXue GENG WenPing MU JiLiang HE Jian CHOU XiuJian 2023Science China(Technological Sciences)2023,66,6:0
15Effects of Rice-Wheat Rotation and Afforestation on Microbial Biomass Carbon in Coastal Salt-Affected Soils of Eastern China显示文摘Rice-wheat rotation and poplar afforestation are two typical land use types in the coastal reclaimed flatlands of eastern China.This study investigated two rice-wheat rotation lands(one reclaimed from 1995 to 2004 and cultivated since 2005, RW1, and the other reclaimed from 1975 to 1995 and cultivated since 1996, RW2) and a poplar woodland(reclaimed from 1995 to 2004 and planted in2004, PW1) to determine the effects of land use types and years of cultivation on soil microbial biomass and mineralizable carbon(C) in this coastal salt-affected region. The results showed that the soil in PW1 remained highly salinized, whereas desalinization was observed in RW1. The total organic C(TOC) in the top soil of PW1 and RW1 did not show significant differences, whereas at a soil depth of 20–30 cm, the TOC of RW1 was approximately 40%–67% higher than that of PW1. The TOC of 0–30-cm soil in RW2 was approximately 37% higher than that in RW1. Microbial biomass C(MBC) and mineralizable C(MNC) exhibited the trend of RW2 > RW1 > PW1. Sufficient nutrition with more abundant C substrates resulted in higher MBC and MNC, and soil respiration rates were negatively correlated with C/N in RW1 and RW2. Nutrient deficiency and high salinity played key roles in limiting MBC in PW1. 16Control Effects of Different Agents on Tobacco Mosaic Virus Disease显示文摘[Objectives] This study was conducted to screen out suitable agents for controlling tobacco mosaic virus disease and the best control period in Zhangzhou tobacco area, providing a theoretical basis for the control of virus diseases, thereby improving the quality of flue-cured tobacco and the income of tobacco farmers. [Methods] The effects on tobacco mosaic virus disease under the interaction between different agents and different application periods were investigated. The incidence of tobacco mosaic virus disease was investigated, and its control effect was analyzed. [Results] Different agents and different application periods had different control effects on tobacco mosaic virus disease. The incidence of tobacco mosaic virus disease: At 30 and 45 d after transplanting, the incidences of A2B1 treatment were the lowest, at 0.85%, 1.71%, respectively;and at 60 d after transplanting, the incidence of A3B1 treatment was the lowest, only 10.68%. The control effect: At 30 and 45 d after transplanting, A2B1 treatment had better control effects, reaching 79.39% and 73.06%, respectively. [Conclusions] 3% hypersensitive protein sprayed at 1 d before transplanting and 7 and 15 d after transplanting achieved the best effect, followed by 10% ningnanmycin sprayed at 1 d before transplanting and 7 and 15 d after transplanting.
