1Simultaneous oxidation of NO,SO_2 and Hg^0 from flue gas by pulsed corona discharge显示文摘A process capable of simultaneously oxidizing NO,SO2,and Hg0 was proposed,using a high-voltage and short-duration positive pulsed corona discharge.By focusing on NO,SO2,and Hg0 oxidation effciencies,the influences of pulse peak voltage,pulse frequency,initial concentration,electrode number,residence time and water vapor addition were investigated.The results indicate that NO,SO2 and Hg0 oxidation effciencies depend primarily on the radicals(OH,HO2,O) and the active species(O3,H2O2,etc.) produced by the pulsed corona discharge.The NO,SO2 and Hg0 oxidation effciencies could be improved as pulse peak voltage,pulse frequency,electrode number and residence time increased,but they were reduced with increasing initial concentrations.By adding water vapor,the SO2 oxidation effciency was improved remarkably,while the NO oxidation effciency decreased slightly.In our experiments,the simultaneous NO,SO2,and Hg0 oxidation effciencies reached to 40%,98%,and 55% with the initial concentrations 479 mg/m3,1040 mg/m3,and 15.0 μg/m3,respectively.XU Fei,LUO Zhongyang,CAO Wei,WANG Peng,WEI Bo,GAO Xiang,FANG Mengxiang,CEN Kefa State key Laboratory of Clean Energy Utilization,Zhejiang University,Hangzhou 310027,China. 2009Journal of Environmental Sciences2009,21,3:22

3Gain-of-function mutant p53 in cancer progression and therapy显示文摘p53 is a key tumor suppressor,and loss of p53 function is frequently a prerequisite for cancer development.The p53 gene is the most frequently mutated gene in human cancers;p53 mutations occur in>50%of all human cancers and in almost every type of human cancers.Most of p53 mutations in cancers are missense mutations,which produce the full-length mutant p53(mutp53)protein with only one amino acid difference from wild-type p53 protein.In addition to loss of the tumor-suppressive function of wild-type p53,many mutp53 proteins acquire new oncogenic activities independently of wild-type p53 to promote cancer progression,termed gain-of-function(GOF).Mutp53 protein often accumulates to very high levels in cancer cells,which is critical for its GOF.Given the high mutation frequency of the p53 gene and the GOF activities of mutp53 in cancer,therapies targeting mutp53 have attracted great interest.Further understanding the mechanisms underlying mutp53 protein accumulation and GOF will help develop effective therapies treating human cancers containing mutp53.In this review,we summarize the recent advances in the studies on mutp53 regulation and GOF as well as therapies targeting mutp53 in human cancers.Cen Zhang Juan Liu Dandan Xu Tianliang Zhang Wenwei Hu Zhaohui Feng 2020Journal of Molecular Cell Biology2020,12,9:18
4Genome Sequences Provide Insights into the Reticulate Origin and Unique Traits of Woody Bamboos显示文摘Polyploidization is a major driver of speciation and its importance to plant evolution has been well recognized.Bamboos comprise one diploid herbaceous and three polyploid woody lineages,and are members of the only major subfamily in grasses that diversified in forests,with the woody members having a tree-like lignified culm.In this study,we generated four draft genome assemblies of major bamboo lineages with three different ploidy levels(diploid,tetraploid,and hexaploid).We also constructed a high-density genetic linkage map for a hexaploid species of bamboo,and used a linkage-map-based strategy for genome assembly and identification of subgenomes in polyploids.Further phylogenomic analyses using a large dataset of syntenic genes with expected copies based on ploidy levels revealed that woody bamboos originated subsequent to the divergence of the herbaceous bamboo lineage,and experienced complex reticulate evolution through three independent allopolyploid events involving four extinct diploid ancestors.A shared but distinct subgenome was identified in all polyploid forms,and the progenitor of this subgenome could have been critical in ancient polyploidizations and the origin of woody bamboos.Important genetic clues to the unique flowering behavior and woody trait in bamboos were also found.Taken together,our study provides significant insights into ancient reticulate evolution at the subgenome level in the absence of extant donor species,and offers a potential model scenario for broad-scale study of angiosperm origination by allopolyploidization.Zhen-Hua Guo Peng-Fei Ma Guo-Qian Yang Jin-Yong Hu Yun-Long Liu En-Hua Xia Mi-Cai Zhong Lei Zhao Gui-Ling Sun Yu-Xing Xu You-Jie Zhao Yi-Chi Zhang Yu-Xiao Zhang Xue-Mei Zhang Meng-Yuan Zhou Ying Guo Cen Guo Jing-Xia Liu Xia-Ying Ye Yun-Mei Chen Yang Yang Bin Han Choun-Sea Lin Ying Lu De-Zhu Li 2019Molecular Plant2019,12,10:13
5Hangzhou criteria as downstaging criteria in hepatocellular ca显示文摘Background:The downstaging of hepatocellular carcinoma(HCC)has been confirmed to benefit liver transplantation(LT)patients whose tumors are beyond the transplantation criteria.Milan criteria(MC),a tumor size and number-based assessment,is currently used as the endpoint in these patients.However,many studies believe that tumor biological behavior should be added to the evaluation criteria for downstaging efficacy.Hence,this study aimed to explore the feasibility of Hangzhou criteria(HC),which introduced tumor grading and alpha-fetoprotein in addition to tumor size and number,as an endpoint of downstaging.Methods:We performed a multicenter and retrospective study of 206 patients accepted locoregional therapy(LRT)as downstaging/bridge treatment prior to LT in three centers of China.Results:Recipients were divided into four groups:failed downstaging to the HC(group A,n=46),successful downstaging to the HC(group B,n=30),remained within the HC all the time(group C,n=113),and tumor progressed(group D,n=17).The 3-year HCC recurrence probabilities of groups B and C were not significantly different(10.3%vs.11.6%,P=0.87).The HCC recurrent rate was significantly higher in group A(52.3%)compared with that in group B/C(P<0.05).Seven patients(7/76,9.2%)whose tumor exceeded the the HC were successfully downstaged to the MC,and 39.5%(30/76)to the the HC.In group B,23 patients remained beyond the MC and their survivals were as well as those of patients within the MC.Conclusions:Compared to the MC,HC downstaging criteria can give more HCC patients access to LT and furthermore,the outcome of these patients is the same as those matching MC downstaging criteria.Hangzhou downstaging criteria therefore is applicable in clinical practice.Qi-Fan Zhan Sun-Bin Ling Yi-Nan Deng Qiao-Nan Shan Qian-Wei Ye Sheng-Jun Xu Guang-Jiang Jiang Di Lu Xu-Yong Wei Li Zhuang Wu Zhang Tian Shen Bei-Ni Cen Hai-Yang Xie Ji-Min Liu Jian Wu Shu-Sen Zheng Yang Yang Xiao Xu 2020Hepatobiliary & Pancreatic Diseases International2020,19,4:11

8Medium optimization for enhanced production of cytosine-substituted mildiomycin analogue (MIL-C) by Streptoverticillium rimofaciens ZJU 5119显示文摘Cytosine-substituted mildiomycin analogue (MIL-C) was produced effectively by supplementing cytosine into the culture of Streptoverticillium rimofaciens. In order to improve the yield of MIL-C,statistically-based experimental designs were applied to optimize the fermentation medium for S. rimofaciens ZJU 5119. Fifteen culture conditions were examined for their significances on MIL-C production using Plackett-Burman design. The Plackett-Burman design and one-variable-at-a-time design indicated that glucose and rice meal as the complex carbon sources,and peanut cake meal and NH4NO3 as the complex nitrogen sources were beneficial for MIL-C production in S. rimofaciens ZJU 5119. The results of further central composition design (CCD) showed that the optimal concentration of glucose,rice meal and peanut cake meal were 18.7 g/L,64.8 g/L and 65.1 g/L,respectively. By using this optimal fermentation medium,the MIL-C concentration was increased up to 1336.5 mg/L,an ap-proximate 3.8-fold improvement over the previous concentration (350.0 mg/L) with un-optimized medium. 11LES investigation of swirl intensity effect on unconfined turbulent swirling premixed flame显示文摘A finite reaction rate model is presented as a closure of large eddy simulation(LES) to numerically study an open premixed methane/air swirling flame. The resultant model is firstly validated by comparing with reported data and then employed to investigate the effect of swirling intensity on flow field, flame characteristics and combustion instability of the swirling flame. Three different swirl numbers are considered. The LES results show that as swirling intensity increases, the vortex entrainment and micro-mixing are enhanced, leading to more lean equivalent ratios at flame front; consequently, higher swirling number causes lower flame temperatures and slower CO oxidization; for all simulated swirl numbers,flame fronts are completely located out of the recirculation zones and anchored at the inner surface of the annular swirling steams; swirl number has a crucial effect on swirling flame extension toward radial and tangential dimensions and then significantly affects streamwise flame length, which is a great influencing factor on combustion instability; vortex-induced disturbance on flame in streamwise plays a critical role in combustion instability.Zhihua Wang Yanwei Xu Yu L Zhijun Zhou Junhu Zhou Kefa Cen 2014Chinese Science Bulletin2014,59,33:6
12Peripheral Lymphocyte Subsets as a Marker of Parkinson's Disease in a Chinese Population显示文摘In this study, we conducted a clinical analysis of lymphocyte subtypes in 268 patients with Parkinson's disease(PD) to assess their clinical impact as a potential marker of advanced PD in Chinese patients. The participants comprised 268 sporadic PD patients and 268 healthy controls. The numbers of natural killer(NK) cells and CD3+, CD3+CD4+, CD3+CD8+, and CD19+ lymphocytes from peripheral blood were determined by immunostaining and flow cytometric analysis and the percentages of these CD+ T cells were calculated. The ratio of regulatory T(Treg)/helper T 17(Th17) lymphocytes from 64 PD patients and 46 controls was determined by flow cytometric analysis.The results showed that the percentage of NK cells was higher in advanced PD patients than in controls(22.92% ±10.08% versus 19.76% ± 10.09%, P = 0.006), while CD3+ T cells are decreased(62.93% ± 9.27% versus65.75% ± 9.13%, P = 0.005). The percentage of CD19+B cells in male patients was lower(P = 0.021) than in female patients, whereas NK cells were increased(P \ 0.0001). The scores on the Unified Parkinson's Disease Rating Scale(UPDRS) and the Non-Motor Symptoms Scale in late-onset PD patients were significantly higher than those in earlyonset patients(P = 0.024 and P = 0.007, respectively). The percentage of CD19+ B cells in patients with UPDRS scores[24 was lower than in those with scores \24(10.17% ±4.19% versus 12.22% ± 5.39%, P = 0.009). In addition, the Treg/Th17 ratio in female patients was higher than that in female controls(13.88 ± 6.32 versus 9.94 ± 4.06, P =0.042). 14Metformin activates chaperone-mediated autophagy and improves disease pathologies in an Alzheimer disease mouse model显示文摘Chaperone-mediated autophagy(CMA)is a lysosome-dependent selective degradation pathway implicated in the pathogenesis of cancer and neurodegenerative diseases.However,the mechanisms that regulate CMA are not fully understood.Here,using unbiased drug screening approaches,we discover Metformin,a drug that is commonly the first medication prescribed for type 2 diabetes,can induce CMA.We delineate the mechanism of CMA induction by Metformin to be via activation of TAK1-IKKα/β signaling that leads to phosphorylation of Ser85 of the key mediator of CMA,Hsc70,and its activation.Notably,we find that amyloid-beta precursor protein(APP)is a CMA substrate and that it binds to Hsc70 in an IKKα/β-dependent manner.The inhibition of CMA-mediated degradation of APP enhances its cytotoxicity.Importantly,we find that in the APP/PS1 mouse model of Alzheimer's disease(AD),activation of CMA by Hsc70 overexpression or Metformin potently reduces the accumulated brain Aβplaque levels and reverses the molecular and behavioral AD phenotypes.Our study elucidates a novel mechanism of CMA regulation via Metformin-TAK1-IKKα/β-Hsc70 signaling and suggests Metformin as a new activator of CMA for diseases,such as AD,where such therapeutic intervention could be beneficial.Xiaoyan Xu Yaqin Sun Xufeng Cen Bing Shan Qingwei Zhao Tingxue Xie Zhe Wang Tingjun Hou Yu Xue Mengmeng Zhang Di Peng Qiming Sun Cong Yi Ayaz Najafov Hongguang Xia 2021Protein & Cell2021,12,10:5
15BMSC-derived exosomes ameliorate sulfur mustard-induced acute lung injury by regulating the GPRC5A-YAP axis显示文摘Sulfur mustard(SM)is a highly toxic chemical warfare agent that causes acute lung injury(ALI)and/or acute respiratory distress syndrome(ARDS).There are no effective therapeutic treatments or antidotes available currently to counteract its toxic effects.Our previous study shows that bone marrow-derived mesenchymal stromal cells(BMSCs)could exert therapeutic effects against SM-induced lung injury.In this study,we explored the therapeutic potential of BMSC-derived exosomes(BMSC-Exs)against ALI and the underlying mechanisms.ALI was induced in mice by injection of SM(30 mg/kg,sc)at their medial and dorsal surfaces.BMSC-Exs(20μg/kg in 200μL PBS,iv)were injected for a 5-day period after SM exposure.We showed that BMSC-Exs administration caused a protective effect against pulmonary edema.Using a lung epithelial cell barrier model,BMSC-Exs(10,20,40μg)dose-dependently inhibited SM-induced cell apoptosis and promoted the recovery of epithelial barrier function by facilitating the expression and relocalization of junction proteins(E-cadherin,claudin-1,occludin,and ZO-1).We further demonstrated that BMSC-Exs protected against apoptosis and promoted the restoration of barrier function against SM through upregulating G protein-coupled receptor family C group 5 type A(GPRC5A),a retinoic acid target gene predominately expressed in the epithelial cells of the lung.Knockdown of GPRC5A reduced the antiapoptotic and barrier regeneration abilities of BMSC-Exs and diminished their therapeutic effects in vitro and in vivo.BMSC-Exs-caused upregulation of GPRC5A promoted the expression of Bcl-2 and junction proteins via regulating the YAP pathway.In summary,BMSC-Exs treatment exerts protective effects against SM-induced ALI by promoting alveolar epithelial barrier repair and may be an alternative approach to stem cell-based therapy.Guan-chao Mao Chu-chu Gong Zhen Wang Ming-xue Sun Zhi-peng Pei Wen-qi Meng Jin-feng Cen Xiao-wen He Ying Lu Qing-qiang Xu Kai Xiao 2021Acta Pharmacologica Sinica2021,42,12:4
16Association between serumα-L-fucosidase andnon-alcoholic fatty liver disease:Cross-sectional study显示文摘AIM: To explore the association between serum α-Lfucosidase(Af U) and non-alcoholic fatty liver disease(NAf LD).METHODS: A total of 16473 individuals(9456 men and 7017 women) were included in the current study, who presented for a health examination at the first Affiliated hospital of Zhejiang University School of medicine in 2014. The baseline characteristics of the cohort were compared by NAf LD status. Linear regression analysis and stepwise multiple regression analysis were applied to assess the risk factors for NAf LD. Receiver operating characteristic curve was used to determine the sensitivity and specificity of Af U in the diagnosis of NAf LD.RESULTS: The prevalence rates of NAf LD and metabolic syndrome(met S) were 38.0% and 25.4%, respectively. The NAf LD group had significantly higher Af U levels than the non-NAf LD group(28.7 ± 7.9 U/L vs 26.0 ± 7.3 U/L, P < 0.001) and the prevalence rate of NAf LD increased with progressively higher serum Af U levels. Af U was positively correlated with met S and its five components: central obesity, hypertriglyceridemia, low high-density lipoprotein cholesterol, and elevated blood pressure and fasting glucose. Stepwise multiple logistic regression analysis showed that Af U was associated with an increased risk of NAf LD(OR = 1.009, 95%CI: 1.003-1.014, P < 0.001). The best cut-off value of Af U for the diagnosis of NAf LD was 27.5 U/L. The area under the curve(diagnostic efficacy index) was 0.606. The sensitivity and specificity were 54.6% and 61.8%, respectively. 17C–C motif chemokine ligand 16 inhibits the progression of liver cirrhosis via inactivating hepatic stellate cells显示文摘Background:Liver cirrhosis results from many forms of chronic damage,characterized by accumulation of extracellular matrix.The present study aimed to explore a potential non-invasive biomarker and its mechanism in the progression of liver cirrhosis.Methods:Gene Expression Omnibus(GEO)dataset(GSE15654,n=216)was analyzed to screen genes associated with progression of liver cirrhosis.A total of 181 plasma samples,including healthy control(HC,n=20),chronic hepatitis B(CHB,n=77)and HBV-related liver cirrhosis(LC,n=84),were enrolled for validation.In vitro and in vivo experiments were employed for the mechanistic investigation.Results:GEO dataset analysis showed that relatively low mRNA-expression of C–C motif chemokine ligand 16(CCL16)was associated with elevated Child-Pugh score(P=0.034)and worse prognosis(P=0.025).Plasma CCL16 level decreased in a stepwise pattern,with a median concentration of 10.29,6.57 and 4.47 ng/mL in the HC,CHB and LC groups,respectively(P<0.001).Low plasma CCL16 was significantly related to hepatic dysfunction both in the CHB and LC groups(P<0.05).Combination of CCL16 and ALT showed improved distinguishing capability for LC compared to either alone.In vitro,CCL16 expression was downregulated by lipopolysaccharide and hypoxia.Overexpression of CCL16 from human normal liver cell line(LO2)reduced the extracellular matrix associated proteins(Col1 and Col4)in human hepatic stellate cell line(LX-2).In vivo,the pathological feature of cirrhosis was alleviated by the hepatocytespecific expression of CCL16.Conclusions:CCL16 could be a feasible plasma marker to predict the occurrence and progression of liver cirrhosis.CCL16 might impact liver cirrhosis through inactivating hepatic stellate cells.Jian-Yong Zhuo Di Lu Zu-Yuan Lin Bei-Ni Cen Xu-Yong Wei Hai-Yang Xie Shu-Sen Zheng Xiao Xu 2020Hepatobiliary & Pancreatic Diseases International2020,19,5:3
