| 1 | Improved Brassica rapa reference genome by single-molecule sequencing and chromosome conformation capture technologies显示文摘Brassica rapa comprises several important cultivated vegetables and oil crops.Current reference genome assemblies of Brassica rapa are quite fragmented and not highly contiguous,thereby limiting extensive genetic and genomic analyses.Here,we report an improved assembly of the B.rapa genome(v3.0)using single-molecule sequencing,optical mapping,and chromosome conformation capture technologies(Hi-C).Relative to the previous reference genomes,our assembly features a contig N50 size of 1.45 Mb,representing a~30-fold improvement.We also identified a new event that occurred in the B.rapa genome~1.2 million years ago,when a long terminal repeat retrotransposon(LTR-RT)expanded.Further analysis refined the relationship of genome blocks and accurately located the centromeres in the B.rapa genome.The B.rapa genome v3.0 will serve as an important community resource for future genetic and genomic studies in B.rapa.This resource will facilitate breeding efforts in B.rapa,as well as comparative genomic analysis with other Brassica species. | Lei Zhang Xu Cai Jian Wu Min Liu Stefan Grob Feng Cheng Jianli Liang Chengcheng Cai Zhiyuan Liu Bo Liu Fan Wang Song Li Fuyan Liu Xuming Li Lin Cheng Wencai Yang Mai-he Li Ueli Grossniklaus Hongkun Zheng Xiaowu Wang | 2018 | Horticulture Research2018,5,1: | 10 |

| 3 | Whole-Genome Resequencing of a Worldwide Collection of Rapeseed Accessions Reveals the Genetic Basis of Ecotype Divergence显示文摘Rapeseed (Brassica napus),an important oilseed crop,has adapted to diverse climate zones and latitudes by forming three main ecotype groups,namely winter,semiwinter,and spring types. However,genetic variations underlying the divergence of these ecotypes are largely unknown. Here,we report the global pattern of genetic polymorphisms in rapeseed determined by resequencing a worldwide collection of 991 germplasm accessions.A total of 5.56 and 5.53 million singlenucleotide polymorphisms (SNPs)as Well as 1.86 and 1.92 million InDels were identified by mapping reads to the reference genomes of 'Darmor-bzh'and 'Tapidor,'respectively.We generated a map of allelic drift paths that shows splits and mixtures of the main populations,and revealed an asymmetric evolution of the two subgenomes of B.napus by calculating the genetic diversity and linkage disequilibrium parameters.Selective-sweep analysis revealed genetic changes in genes orthologous to those regulating various aspects of plant development and response to stresses.A genome-wide association study identified SNPs in the promoter regions of FLOWERING LOCUS T and FLOWERING LOCUS C orthologs that corresponded to the different rapeseed ecotype groups. | 5 | Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). | 6 | A Plant Lectin Receptor-like Kinase Phosphorylates the Bacterial Effector AvrPtoB to Dampen Its Virulence in Arabidopsis显示文摘Plasma membrane-localized receptor-like kinases(RLKs)perceive conserved pathogen-associated molecular pattems(PAMPs)in plants,leading to PAMP-triggered immunity(PTI).TheArabidopsis thaliana lectin RLK LecRK-Ⅸ.2 has been shown to regulate the bacterial flagellin-derived peptide flg22-induced PTI.Here,we discover that Pseudomonas syringae effector AvrPtoB targets LecRK-Ⅸ.2 for degradation,which subsequently suppresses LecRK-Ⅸ.2-mediated PTI and disease resistance.However,LecRK-Ⅸ.2 can interact with and phosphorylate AvrPtoB at serine site 335(S335).AvrPtoB self-associates in vitro and in vivo,and the association appears to be essential for its E3 ligase activity in ubiquitinating substrate in plants.Phosphorylation of S335 disrupts the self-association and as a result,phosphomimetic AvrPtoBS335D cannot ubiquitinate LecRK-Ⅸ.2 efficiently,leading to the compromised virulence of AvrPtoB in suppressing PTI responses.fig22 enhances AvrPtoB S335 phosphorylation by inducing the expression and activating of LecRK-Ⅸ.2.Our study demonstrates that host RLKs can modify pathogen effectors to dampen their virulence and undermine their ability in suppressing PTI. | Ning Xu Xuming Luo Wei Wu Yingying Xing Yingbo Liang Yanzhi Liu Huasong Zou Hai-Lei Wei Jun Liu | 2020 | Molecular Plant2020,13,10: | 5 |
| 7 | Aberrant seed development in Litchi chinensis is associated with the impaired expression of cell wall invertase genes显示文摘Cell wall invertase(CWIN)are known to play important roles in seed development.However,most reports to date have focused on a single gene family member,and have mainly investigated CWIN functions during the filling stage of seed development.In this study,we found significant lower levels of CWIN protein and activity associated with seed abortion in the Litchi chinensis cultivar“Nuomici.”We identified five litchi CWIN genes and observed that the expression of LcCWIN5 was limited to the flower tissues and decreased sharply with fruit development.Silencing of LcCWIN5 expression before 28 DAA(cell division stage)resulted in perturbed liquid endosperm development,smaller seeds,and higher seed abortion rate,while silencing after 28 DAA(filling stage)had no effect on seed development.In contrast,LcCWIN2 was mostly expressed in the funicle and seed coat,and increased with fruit development.Decreased LcCWIN2 expression and CWIN activity during early seed filling coincided with smaller seeds in the cultivar“Feizixiao.”Silencing of LcCWIN2 caused a reduction in the seed size without inducing seed abortion.We propose that CWIN activity in seed maternal tissues during cell division stage is likely due to LcCWIN5 expression,which regulates early seed development.On the other hand,CWIN activity during the filling stage is due to the expression of LcCWIN2,which may promote carbon import by creating a sucrose gradient.Comparable LcCWIN5 expression,but much lower CWIN activity,detected in the funicle of“Nuomici”is consistent with post-translational regulation. | Jieqiong Zhang Zichen Wu Fuchu Hu Lian Liu Xuming Huang Jietang Zhao Huicong Wang | 2018 | Horticulture Research2018,5,1: | 4 |

| 9 | Kongsheng Zhenzhong pill's effect on the learning and memory ability and its neuroprotective effects in vascular dementia rats显示文摘Clinical reports have demonstrated that the Kongsheng Zhenzhong pill (KSZZP),a classical prescription deriving from Valuable Prescription for Emergencies,has good therapeutic effects on vascular dementia.However,the mechanisms that mediate its effects remain unclear.In this study,the expression of N-methyl-D-aspartate receptor 1 mRNA,the content of nitric oxide,and the concentration of calcium in neurons was determined with in situ hybridization,spectrophotometry and flow cytometry,respectively.In addition,the expressions of N-methyl-D-aspartate receptor 1,nerve growth factor protein,and glial cell line-derived neurotrophic factor protein were detected with immunohistochemistry.We found that KSZZP could significantly decrease the expression of N-methyl-D-aspartate receptor 1 mRNA and protein,the content of nitric oxide,and the concentration of calcium in neurons.KSZZP also increased the expression of nerve growth factor and glial cell line-derived neurotrophic factor protein in the hippocampus CA1 region and in the cerebral cortex.Morris water maze and passive avoidance tests verified that KSZZP ameliorated the cognitive impairments of vascular dementia rats.Moreover,the KSZZP-induced improvements in the cognitive functions of vascular dementia rats were correlated with both inhibition of N-methyl-D-aspartate-induced excitable neurotoxicity and elevation of neurotrophic factor expression. | Xuming Ji Huayun Yu Bin Ouyang Guowei Liu Zhichun Wu Heng Liu Fang Du | 2011 | Neural Regeneration Research2011,6,30: | 3 |
| 10 | Attaining the liquid helium temperature with a compact pulse tube cryocooler for space applications显示文摘Recent breakthroughs in space science have motivated space exploration programs in many countries including China. Cryocoolers, which provide the mandatory low-temperature environment for many sensitive yet delicate space detectors, are crucial for the proper functioning of various systems. One benchmark for the cryocooler performance is attaining the liquid helium temperature. However, even with complex configurations and multiple driving sources, only a few cryocoolers to date can achieve this goal. Here we report a high-frequency pulse tube cryocooler(HPTC) driven by a single non-oil-lubrication compressor which is capable of reaching the liquid helium temperature while offering other advantages such as high compactness, excellent reliability and high efficiency. The HPTC obtains a no-load temperature of 4.4 K, which is the first realization of cooling below the4 He critical point with a gas-coupled two-stage arrangement. The prototype can provide a cooling power of 87 mW at 8 K, and 5.2 mW at 5 K with a 425 W input electric power, showing leading-level efficiency. Moreover, we demonstrate the ability of the cryocooler to simultaneously provide cooling power at different temperature levels to meet different requirements. | 14 | A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. | 15 | Highly sensitive infrared polarized photodetector enabled by out-ofplane PSN architecture composing of p-MoTe_(2),semimetal-MoTe_(2)and n-SnSe_(2)显示文摘Leveraging the unique physical properties,two-dimensional(2D)materials have circumvented the disadvantages of conventional epitaxial semiconductors and held great promise for potential optoelectronic applications.So far,two main detector architectures including photodiode based on a van der Waals P-N junction or Schottky junction and phototransistor based on individual 2D materials or hybrids have been well developed.However,a trade-off between responsivity and speed always exists in those technologies thus hindering the overall performance improvement.Here,we propose a new device concept by sandwiching the 2D anisotropic semimetal between p-type and n-type semiconductors in the out-of-plane direction,called PSN architecture,realizing the improvement of each parameter including broad spectral coverage,fast speed,high sensitivity,power-free and polarization-sensitive.We stack the p-type 2H-MoTe_(2),Weyl semimetal 1T-MoTe_(2)and n-type SnSe_(2)layer-by-layer constructing vertical sandwich structure where the top and bottom layers contribute to the internal built-in electric field,the intermediate layer can facilitate the exciton dissociation and act as infrared polarized light sensitizers.As a result,this PSN device exhibits broadband photo-response from 405 to 1,550 nm without external bias supply.At optical communication band(1,310 nm),operating at self-driven mode and room temperature,the responsivity and detectivity can reach up to 64.2 mA·W^(–1)and 2.2×10^(11)Jones,respectively,along with fast speed on the order of millisecond.Moreover,the device simultaneously exhibits exceptional detection capability for infrared polarized light,demonstrating the anisotropic photocurrent ratio of 1.55 at 1,310 nm and 2.02 at 1,550 nm,which is attributed to the strong in-plane optical anisotropy of middle 1T-MoTe_(2)layer.This work develops a new photodetector scheme with novel PSN architecture toward broadband,self-power,polarized light sensing and imaging modules. | Yiming Sun Jingxian Xiong Xuming Wu Wei Gao Nengjie Huo Jingbo Li | 2022 | Nano Research2022,15,6: | 1 |

| 20 | A Plasmonic Mass Spectrometry Approach for Detection of Small Nutrients and Toxins显示文摘Nutriology relies on advanced analytical tools to study the molecular compositions of food and provide key information on sample quality/safety. Small nutrients detection is challenging due to the high diversity and broad dynamic range of molecules in food samples, and a further issue is to track low abundance toxins. Herein, we developed a novel plasmonic matrix-assisted laser desorption/ionization mass spectrometry(MALDI MS)approach to detect small nutrients and toxins in complex biological emulsion samples. Silver nanoshells(SiO_2@-Ag) with optimized structures were used as matrices andachieved direct analysis of ~ 6 n L of human breast milk without any enrichment or separation. We performed identification and quantitation of small nutrients and toxins with limit-of-detection down to 0.4 pmol(for melamine) and reaction time shortened to minutes, which is superior to the conventional biochemical method currently in use.
