4Successful Production of an All-Female Common Carp (Cyprinus carpio L.) Population Using cyp17a1-Deficient Neomale Carp显示文摘Due to sexual dimorphism in the growth of certain cultured fish species,the production of monosex fishes is desirable for the aquaculture industry.Nowadays,the most widely practiced technique available for the mass production of monosex fish populations is sex steroid-induced sex reversal.Here,a novel strategy for the successful production of all-female(AF)common carp(Cyprinus carpio L.),to take advantage of the sexual dimorphism in growth documented in this species,has been developed using genetic engineering via single gene-targeting manipulation without any exogenous hormone treatments.Male and female heterozygous cyp17a1-deficient common carp were first obtained using the clustered regularly interspaced short palindromic repeats/CRISPR-associated endonuclease 9(CRISPR/Cas9)technique.An all-male phenotype for homozygous cyp17a1-deficient carp,regardless of the individuals’sexdetermination genotypes(XY or XX),has been observed.A male-specific DNA marker newly identified in our laboratory was used to screen the neomale carp population with the XX genotype from the cyp17a1-deficient carp.These neomale carp develop a normal testis structure with normal spermatogenesis and sperm capacity.The neomale common carp were then mated with wild-type(WT)females(cyp17a1^(+/+)XX genotype)using artificial fertilization.All the AF offspring sample fish from the neomale-WT female mating were confirmed as having the cyp17a^(1+/-)XX genotype,and normal development of gonads to ovaries was observed in 100.00%of this group at eight months post-fertilization(mpf).A total of 1000 carp fingerlings,500 from the WT male and female and 500 from the neomale and WT female mating,were mixed and reared in the same pond.The average body weight of cyp17a1^(+/-)XX females was higher by 6.60%(8 mpf)and 32.66%(12 mpf)than that of the control common carp.Our study demonstrates the first successful production of a monosex teleost population with the advantages of sexual dimorphism in growth using genetic manipulation targeting a single locus.Gang Zhai Tingting Shu Kuangxin Chen Qiyong Lou Jingyi Jia Jianfei Huang Chuang Shi Xia Jin Jiangyan He Donghuo Jiang Xueqiao Qian Wei Hu Zhan Yin 2022Engineering2022,8,1:7

6Characteristics of distribution of Mycobacterium tuberculosis lineages in China显示文摘The genotyping methods of Mycobacterium tuberculosis would dramatically improve our understanding of the molecular epidemiology of tuberculosis. 3,929 isolates, from a National Survey of Drug-Resistant Tuberculosis in 2007 in China, were successfully genotyped by large sequence polymorphisms and 15 loci variable number tandem repeats. We found that 2,905(2,905/3,929, 73.9%) cases belonged to Lineage 2, dominated in the east and central regions, 975 cases(975/3,929, 24.8%) were Lineage 4, highly prevailed in the west regions, and 36 and 13 cases were Lineage 3 and Lineage 1, respectively. We also explored the associations between lineages(Lineage 2 vs. Lineage 4) and clinical characteristics by logistic regression. For Lineage 2, the risk factors were Han-ethnicity population and fever. However, for Lineage 4, they were occupation(farmer), and degree of education(non-literate). 7Virus-induced Phytoene Desaturase(PDS) Gene Silencing Using Tobacco Rattle Virus in Lilium × formolongi显示文摘Gene function analysis is challenging for Lilium due to the lack of an efficient method for stable genetic transformation. Virus-induced gene silencing(VIGS) is an attractive tool for determining gene function in plants. This study reported that Tobacco rattle virus(TRV)-based VIGS of a PHYTOENE DESATURASE(PDS) ortholog(LhPDS) can be achieved in Lilium × formolongi seedlings, with a survival rate of 92%, using the inoculation method of rubbing plus injection. Compared with untreated and mock-treated seedlings, the photobleached leaf phenotype of silenced plants significantly correlates with down-regulation of endogenous LhPDS(P ≤ 0.05). In addition, the silencing phenomenon can be observed in the growing points of plants, indicating that systemic viral infection was achieved using the protocol. The results indicate that TRV-based VIGS can be used to characterize gene function in Lilium × formolongi. 8Warranty Claims Forecasting for New Products Sold with a Two-Dimensional Warranty显示文摘Warranty claims forecasting plays an increasingly important role not only for preparing financial plans but also for optimizing warranty policy and improving after-sale services.In the case of new products,an important feature is that the new generation of products often has a close connection with the previous generations of products it replaces.Thus,the warranty claims data of the previous generations of products can be used for extracting reliability information of new products.In this context,we propose a warranty claims forecasting model considering usage rate for new products sold with a two-dimensional warranty.The accelerate failure time model is introduced to investigate the effect of usage rate on product degradation.The non-homogeneous Poisson process is used to model failure counts of repairable products and the constrained maximum likelihood estimation method is used to estimate model parameters.The results of data experiments based on both simulation and real data collected from an automobile manufacturer in China show that the proposed model considering the varying usage rate outperforms the traditional models in forecasting the number of warranty claims.Anshu Dai Zhaomin Zhang Pengwen Hou Jingyi Yue Shuguang He Zhen He 2019Journal of Systems Science and Systems Engineering2019,28,6:4
9Potential Impact of Tonga Volcano Eruption on Global Mean Surface Air Temperature显示文摘The undersea volcano,located in the South Pacific island nation of Tonga,violently erupted from 14 to 15 January 2022.The Tonga volcano eruption has aroused extensive discussion in the climate change field.Some climatologists believe that this event will cause little effect on global climate change while others insist that it will trigger“the year without a summer”as the Tambora eruption did in 1815.How will the Tonga volcano eruption affect global climate change?Based on the indices of past volcanic eruptions and the eruption data of El Chichón volcano in 1982,we use a simplified radiation equilibrium model to quantify the stratospheric aerosol radiative forcing and the change in global mean surface air temperature(Ts)caused by the Tonga volcano eruption.The results show that the global average Ts will decrease by about 0.0315-0.1118℃in the next 1-2 years.The Tonga eruption will slightly slow down the global warming in a short period of time,but it will not change the global warming trend in the long term.In addition,we propose a generalized approach for estimating the impact of future volcanic eruption on global mean T_(s).Hua ZHANG Fei WANG Jian LI Yihong DUAN Congwen ZHU Jingyi HE 2022Journal of Meteorological Research2022,36,1:4

11Ultrafine SnSSe/multilayer graphene nanosheet nanocomposite as a high-performance anode material for potassium-ion half/full batteries显示文摘Layer-structured Shsse attracts much attention as an anode material for potassium storage due to its la rge theoretical capacity.Unfortunately,their practical application is severely restrained by the dramatic volumetric variation of SnSSe.Herein,we synthesize ultrafine SnSSe/multilayer graphene nanosheet(SnSSe/MGS) by a vacuum solid-phase reaction and subsequent ball milling.Owing to the strong synergistic effect between the two components,the obtained SnSSe/MGS nanocomposite exhibits a high reversible capacity(423 mAh g^(-1) at 100 mA g^(-1)),excellent rate property(218 mAh g^(-1) at 5 A g^(-1)),and stable cycling performance(271 mAh g^(-1) after 500 cycles at 100 mA g^(-1)) in potassium-ion half batteries.Moreover,the full cell assembled by the SnSSe/MGS anode and the potassiated 3,4,9,10-perylene-tetracar boxylic aciddianhydride cathode shows excellent electrochemical performance between 0.2 and 3.3 V(209 mAh g^(-1) at 50 mA g^(-1) after 100 cycles).The presented two-step synthesis strategy of SnSSe/MGS may also provide ideas to craft other alloy-type anode materials.Zuyue Yi Jingyi Xu Zhenhua Xu Min Zhang Yanan He Jianchun Bao Xiaosi Zhou 2021Journal of Energy Chemistry2021,30,9:3
12Ecological principle meets cancer treatment: treating children with acute myeloid leukemia with low-dose chemotherapy显示文摘Standard chemotherapy regimens for remission induction of pediatric acute myeloid leukemia(AML) are associated with significant morbidity and mortality. We performed a cohort study to determine the impact of reducing the intensity of remission induction chemotherapy on the outcomes of selected children with AML treated with a low-dose induction regimen plus granulocyte colony stimulating factor(G-CSF)(low-dose chemotherapy(LDC)/G-CSF). Complete response(CR) after two induction courses was attained in 87.0%(40/46) of patients receiving LDC/G-CSF. Post-remission therapy was offered to all patients, and included standard consolidation and/or stem cell transplantation. During the study period, an additional 94 consecutive children with AML treated with standard chemotherapy(SDC) for induction(80/94(85.1%) of the patients attained CR after induction Ⅱ, P = 0.953) and post-remission. In this non-randomized study, there were no significant differences in 4-year event-free(67.4 vs. 70.7%;P = 0.99)and overall(70.3 vs. 74.6%, P = 0.69) survival in the LDC/G-CSF and SDC cohorts, respectively. After the first course of induction, recovery of white blood cell(WBC) and platelet counts were significantly faster in patients receiving LDC/G-CSF than in those receiving SDC(11.5 vs. 18.5 d for WBCs(P < 0.001);15.5 vs. 22.0 d for platelets(P < 0.001)). To examine the quality of molecular response, targeted deep sequencing was performed. Of 137 mutations detected at diagnosis in 20 children who attained hematological CR after two courses of LDC/G-CSF(n = 9) or SDC(n = 11), all of the mutations were below the reference value(variant allelic frequency <2.5%) after two courses, irrespective of the treatment group. In conclusion, children with AML receiving LDC/G-CSF appear to have similar outcomes and mutation clearance levels, but significantly lower toxicity than those receiving SDC. 14A review on perfluoroalkyl acids studies:Environmental behaviors,toxic effects,and ecological and health risks显示文摘Perfluoroalkyl acids(PFAAs)are emerging persistent organic pollutants that are globally distributed in the environment.In the present review,the occurrence of PFAAs and their behavior in aquatic ecosystem were summarized,and the health and ecological risk assessment and the multimedia fate simulation were investigated.PFAAs are most likely to exist in the aqueous phase,and PFAAs in atmosphere are also able to enter water bodies through diffusion and wet and dry deposition and eventually become widely distributed in various environmental media.The air-solid partition is considered to be one of the major factors in the long-distance transportation of the pollutants.The pKa values and organic carbon fraction of the sediment could influence the partition of PFAAs between water and sediment.Otherwise,PFAAs have teratogenic,mutagenic and other toxic effects and they could be accumulated by biota,and magnified through trophic level.The ecological and health risks of PFOA and PFOS were assessment.In order to explore the partition mechanism and reduce the uncertainty of the simulation of the transport,transformation and fate,the experimental methods on physicochemical properties of PFAAs should be developed.Moreover,further studies on toxicities of PFAAs are necessary for health and ecological risk assessment.Wenxiu Liu Jingyi Wu Wei He Fuliu Xu 2019Ecosystem Health and Sustainability2019,5,1:3
15Trade-offs/Synergies in Land-use Function Changes in Central China from 2000 to 2015显示文摘To solve the problems caused by irrational land-use, studying the functions of land-use, its changing characteristics, and the relationship between each land-use function will be beneficial for achieving sustainable land development. In this research, we constructed an evaluation framework of multiple land-use functions(LUFs) based on sustainable land-use theory. Specifically,, we classified the multiple LUFs into three types: agricultural production function(APF), living function(LVF), and ecological service function(ESF).We then spatialized the economic and social data, and implemented the InVEST(Integrated Valuation of Ecosystem Services and Tradeoffs) model and RUSLE(Revised Universal Soil Loss Equation) model to evaluate each sub-LUF(crop production, aquatic production, woodlands production, livestock production, living space, life quality, water supply, soil conservation, climate regulation, biological conservation) in central China in 2000 and again in 2015. Moreover, by analyzing the changes to LUFs and the relationships between each LUF change, we were able to discern patterns of LUF change in central China. The results show that: (1)42.12% of total territory in the study area increased their APF from 2000 to 2015, while 43.41% of the lands increased their ESF yet only 8.98% of the lands increased their LVF;(2) in Hubei and Hunan, there was more land with an increase of APF than in Anhui or Jiangxi. The APF in Jiangxi exhibited the greatest decline over time period, the LVF increased more in the provincial capital cities than in other regions, and the ESF expanded more in Jiangxi than in the other provinces;and (3) the changes in APF were significantly and positively correlated with changes in LVF. 16High performance liquid chromatographic separation of eight drugs collected in Chinese Pharmacopoeia 2010 on amylose ramification chiral stationary phase显示文摘The enantiomers separation of ei ght pharmaceutical racemates collected in Chinese Pharmacopoeia 2010(Ch.P2010),including nitrendipine,felodipine,omeprazole,praziquantel,sulpiride,clenbuterol hydrochloride,verapamil hydrochloride and chlorphenamine maleate,was performed on chiral stationary phase of amylose ramification by high performance liquid chromatography(HPLC)on Chiralpak AD-H column and Chiralpak AS-H column with the mobile phase consisted of isopropanol and n-hexane.The detection wavelength and the flow rate were set at 254 nm and 0.7 mL/min,respectively.The effects of proportion of organic additives,alcohol displacer and temperature on the separation were investigated.The results indicated that eight chiral drugs were separated on chiral stationary phase of amylase ramification in normal phase chroma tographic system.The chromatographic retention and resolution of enantiomers were adjusted by factors,including the changes of the concentration of alcohol displacer in mobile phase,organic alkaline modifier and column temperature.It was shown that the resolution was improved with reducing concentration of alcohol displacer.When the concentration of organic alkaline modifier was 0.2%,the resolution and the peak shape were fairly good.Most racemates mentioned above had the best resolution at column temperature of 25℃.The best temperature should be kept unchanged in the process of separation so as to obtain stable separation results.Yan Wang Ying Zhou Chao Ma Beibei Yang Ru Feng Yiying Zhang Jie Fu Wenjing Chen Yupeng Sun Jingyi Ma Qiming Zhang Yulin Deng Yukui Zhang Wenyi He 2012Acta Pharmaceutica Sinica B2012,2,5:3

18Photochemistry of hypocrellin derivatives under anaerobic conditions显示文摘To improve the red absorption and solubility of hypocrellin, we have synthesized a series of hypocrellin B derivatives. The pnotochemistry of these new compounds in anaerobic media has been investigated by using electronic paramagnetic resonance (EPR) and spectrophotometric methods. The semiquinone anion radicals can be produced by self-electron transfer on irradiation, with the formation efficiency and EPR hyperfine structures of the semiquinone anion radicals dependent on the structures of the derivatives. When an electron donor is present, the electron transfer from electron donor to hypocrellin B derivatives enhanced the production of the corresponding semiquinone anion radical; in addition, the semiquinone anion radical and hydroquinone can be detected spectrophotometrically. 19Circumferential Propagation of Tip Leakage Flow Unsteadiness for a Low-Speed Axial Compressor显示文摘Full-annulus three-dimensional unsteady numerical simulations were conducted for a low-speed isolated axialcompressor rotor, intending to identify the behavior of self-induced unsteady tip leakage flow within multi-bladepassages. There is a critical mass flow rate near stall point, below it, the self-induced unsteadiness of tip leakageflow can propagate circumferentially and thus initiates two circumferential waves. Otherwise, the self-inducedunsteady tip leakage flow oscillates synchronously in each single blade passage. The major findings are: 1) whilethe self-induced unsteadiness of tip leakage flow is a single-passage phenomenon, there exist phase shifts amongblade passages in multi-passage environments then evolving into the first short length wave propagating at abouttwo times of rotor rotation speed after the transient period ends; and 2) the time traces of the pseudo sensors locatedon the rotor blade tips reveal another much longer length-scale wave modulated with the first wave due tophase shift propagating at about half of rotor rotation speed.
