12018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 4Washed microbiota transplantation vs.manual fecal microbiota transplantation:clinical findings,animal studies and in vitro screening显示文摘Fecal microbiota transplantation(FMT)by manual preparation has been applied to treat diseases for thousands of years.However,this method still endures safety risks and challenges the psychological endurance and acceptance of doctors,patients and donors.Population evidence showed the washed microbiota preparation with microfiltration based on an automatic purification system followed by repeated centrifugation plus suspension for three times significantly reduced FMT-related adverse events.This washing preparation makes delivering a precise dose of the enriched microbiota feasible,instead of using the weight of stool.Intraperitoneal injection in mice with the fecal microbiota supernatant obtained after repeated centrifugation plus suspension for three times induced less toxic reaction than that by the first centrifugation following the microfiltration.The toxic reactions that include death,the change in the level of peripheral white blood cells,and the proliferation of germinal center in secondary lymphoid follicles in spleen were noted.The metagenomic next-generation sequencing(NGS)indicated the increasing types and amount of viruses could be washed out during the washing process.Metabolomics analysis indicated metabolites with pro-inflammatory effects in the fecal microbiota supernatant such as leukotriene B4,corticosterone,and prostaglandin G2 could be removed by repeated washing.Near-infrared absorption spectroscopy could be served as a rapid detection method to control the quality of the washingprocess.In conclusion,this study for the first time provides evidence linking clinical findings and animal experiments to support that washed microbiota transplantation(WMT)is safer,more precise and more quality-controllable than the crude FMT by manual.Ting Zhang Gaochen Lu Zhe Zhao Yafei Liu Quan Shen Pan Li Yaoyao Chen Haoran Yin Huiquan Wang Cicilia Marcella Bota Cui Lei Cheng Guozhong Ji Faming Zhang 2020Protein & Cell2020,11,4:44
5Roles of Chemokine Receptor 4 (CXCR4) and Chemokine Ligand 12 (CXCL12) in Metastasis of Hepatocellular Carcinoma Cells显示文摘Chemokines are involved in human hepatocellular carcinoma (HCC) carcinogenesis. However, the exact mechanism of chemokines in HCC carcinogenesis remains unknown. Here we investigated the roles of chemokine receptor 4 (CXCR4) and chemokine ligand 12 (CXCL12) in the metastasis of HCC. We found that the expression levels of CXCR4 mRNA in HCC tissues, MHCC97 cells, and HUVEC cells were 2.52 ± 1.13, 2.34 ± 1.16 and 1.63 ± 1.26, respectively and that the CXCR4 protein levels were 1.38 ± 0.13, 1.96 ± 0.32 and 1.86 ± 0.21, respectively. In contrast, CXCR4 was not detected in normal hepatic tissues. In 78 HCC patients, we also found that the concentration of CXCL12 in cancerous ascitic fluid was 783-8,364 pg/ml and that CXCL12 mRNA level in HCC metastasis portal lymph nodes was 1.21 ± 0.87 but undetectable in normal hepatic tissues. Finally we discovered that recombinant human CXCL12 could induce MHCC97 cells and HUVEC cells to migrate with chemotactic indexes (CI) of 3.9 ± 1.1 and 4.1 ± 1.6, respectively. Cancerous ascitic fluid could also induce the migration of MHCC97 cells with a CI of 1.9 ± 0.8. 6Main controlling factors and enrichment area evaluation of shale gas of the Lower Paleozoic marine strata in south China显示文摘The Lower Paleozoic shale in south China has a very high maturity and experienced strong tectonic deformation. This character is quite different from the North America shale and has inhibited the shale gas evaluation and exploration in this area. The present paper reports a comprehensive investigation of maturity, reservoir properties,fluid pressure, gas content, preservation conditions, and other relevant aspects of the Lower Paleozoic shale from the Sichuan Basin and its surrounding areas. It is found that within the main maturity range(2.5 %3.5 %) is considered a high risk for shale gas exploration. Taking the southern area of the Sichuan Basin and the southeastern area of Chongqing as examples of uplifted/folded and faulted/folded areas, respectively, geological models of shale gas content and loss were proposed.For the uplifted/folded area with a simple tectonic deformation, the shale system(with a depth >2000 m) has largely retained overpressure during uplifting without a great loss of gas, and an industrial shale gas potential is generally possible. However, for the faulted/folded area with a strong tectonic deformation, the sealing condition of the shalesystem was usually destroyed to a certain degree with a great loss of free gas, which decreased the pressure coefficient and resulted in a low production capacity. 7Relationship between hs-CRP, proMMP-1, TIMP-1 and coronary plaque morphology: intravascular ultrasound study显示文摘Background Rupture of unstable plaque with subsequent thrombus formation is the common pathophy- siological substrate of the acute coronary syndrome (ACS). It is of potential significance to explore the blood indexes predicting plaque characteristics. Little studies have focused on this field. Therefore we investigated the relationship between hypersensitive C-reactive protein (hs-CRP), pro-matrix metalloproteinase-1 (proMMP-1), tissue inhibitors of matrix metalloproteinase-1 (TIMP-1) and coronary plaque morphology. Methods Intravascular ultrasound (IVUS) examination was done in 152 patients with confirmed coronary heart disease before percutaneous coronary intervention from February 2003 to July 2005. Plasma samples of arterial blood were collected prior to the procedure. The level of hs-CRP, proMMP-1 and TIMP-1 were respectively measured by enzyme-linked immunosorbent assay (ELISA). Results Unstable and ruptured plaque were found more frequently in patients with acute myocardial infarction and unstable angina. External elastic membrane cross-sectional area (EEM CSA), plaque area, lipid pool area and plaque burden were significantly larger in ruptured and unstable plaque group. Positive remolding, thinner fabric-cap, smaller minimal lumen cross-sectional area (MLA), dissection and thrombus were significantly more frequent in ruptured and unstable plaque group. The levels of plasma hs-CRP, proMMP-1 and TIMP-1 were higher in ruptured plaque group. hs-CRP>8.94 mg/L was used to predict ruptured plaque with a ROC curve area of 0.76 [95% confidence interval (CI), 67.0%-85.8%], sensitivity of 71.8%, specificity of 77.0% and accuracy of 69.2% (P<0.01), similarly for proMMP-1>0.12 ng/ml with a ROC curve area of 0.69 [95% CI, 58.2%-80.2%], sensitivity of 69.2%, specificity of 75.2% and accuracy of 66.2% (P<0.01), and TIMP-1>83.45 ng/ml with a ROC curve area of 0.67 [95% CI, 56.2%-78.3%], sensitivity of 66.7%, specificity of 61.9% and accuracy of 66.2% (P<0.01). Conclusion The plaque characteristics correlate with the clinical presentation. 8The role of KDR in the interactions between human gastric carcinoma cell and vascular endothelial cell显示文摘AIM:To study the interactions between human gastriccarcinoma cell (HGCC) and human vascular endothelialcell (HVEC), and the role of KDR in these interactions.METHODS:Antisense oligodexynucleotide(ASODN)specific to KDR gene was devised and added to theculture medium of HGCC and HVEC. After the action ofASODN, the proliferation of two cells was measured byMTT method.The role of KDR in regulating theproliferation of two kinds of cells was known throughobserving the effect of ASODN on them. The conditionedmediums (CMs) of HGCC and HVEC were prepared. TheCM of one kind of cell was added acting on the otherkind of cell, then the cell proliferation was measuredby MTT. After the action of ASODN or CM, the cellularexpression of KDR gene was detected with in situhybridization(ISH) for mRNA level and withimmunohistochemical staining for protein level. ABC-ELISA was used to detect/NEGF in the CMs of two cells.RESULTS: KDR ASODN could specifically inhibit theproliferation of HGCC and HVEC significantiy. The growthinhibitory rate amounted to 55.35 % and 54.83 %,respectively (P<0.01). HGCC and HVEC could secret acertain level of hVEGF(92.06±1.69 ng/L, 77.70±8.04ng/L). The CM of HGCC could significantiy stimulate thegrowth(2.70±0.01 times) and KDR gene expression ofHVEC(P<0.01) while the CM of HVEC could significantiyinhibit the growth(52.97±0.01%) and KDR geneexpression of HGCC (P <0.01).CONCLUSION: KDR plays a key role in regulating theproliferation of HGCC and HVEC. 11YAP promotes osteogenesis and suppresses adipogenic differentiation by regulatingβ-catenin signaling显示文摘YAP(yes-associated protein) is a transcriptional factor that is negatively regulated by Hippo pathway, a conserved pathway for the development and size control of multiple organs. The exact function of YAP in bone homeostasis remains controversial. Here we provide evidence for YAP's function in promoting osteogenesis, suppressing adipogenesis, and thus maintaining bone homeostasis.YAP is selectively expressed in osteoblast(OB)-lineage cells. Conditionally knocking out Yap in the OB lineage in mice reduces cell proliferation and OB differentiation and increases adipocyte formation, resulting in a trabecular bone loss. Mechanistically, YAP interacts with β-catenin and is necessary for maintenance of nuclear β-catenin level and Wnt/β-catenin signaling. Expression of β-catenin in YAP-deficient BMSCs(bone marrow stromal cells) diminishes the osteogenesis deficit. 16Bone marrow stromal cells as a therapeutic treatment for ischemic stroke显示文摘Cerebral ischemia remains the most frequent cause of death and quality-of-life impairments due to neurological deficits, and accounts for the majority of total healthcare costs. However, treatments for cerebral ischemia are limited. Over the last decade, bone marrow stromal cell(BMSC) therapy has emerged as a particularly appealing option, as it is possible to help patients even when initiated days or even weeks after the ischemic insult. BMSCs are a class of multipotent, self-renewing cells that give rise to differentiated progeny when implanted into appropriate tissues. Therapeutic effects of BMSC treatment for ischemic stroke, including sensory and motor recovery, have been reported in pre-clinical studies and clinical trials. In this article, we review the recent progress in BMSC-based therapy for ischemic stroke, focusing on the route of delivery and pre-processing of BMSCs. Selecting an optimal delivery route is of particular importance. The ideal approach, as well as the least risky, for translational applications still requires further identification. 20Xuebijing injection alleviates liver injury by inhibiting secretory function of Kupffer cells in heat stroke rats显示文摘OBJECTIVE:To evaluate the effects of Xuebijing(XBJ) injection in heat stroke(HS) rats and to investigate the mechanisms underlying these effects.METHODS:Sixty anesthetized rats were randomized into three groups and intravenously injected twice daily for 3 days with 4 mL XBJ(XBJ group) or phosphate buffered saline(HS and Sham groups) per kg body weight.HS was initiated in the HS and XBJ groups by placing rats in a simulated climate chamber(ambient temperature 40 ℃,humidity 60%).Rectal temperature,aterial pressure,and heart rate were monitored and recorded.Time to HS onset and survival were determined,and serum concentrations of tumor necrosis factor(TNF)-α,inRESULTS:Time to HS onset and survival were significantly longer in the XBJ than in the HS group.Moreover,the concentrations of TNF-α,IL-1β,IL-6,ALT and AST were lower and liver injury was milder in the XBJ than in the HS group.Heat-stress induced structural changes in KCs and hepatic cells were more severe in the HS than in the XBJ group and the concentrations of TNF-α,IL-β and IL-6 secreted by KCs were lower in the XBJ than in the HS group.CONCLUSION:XBJ can alleviate HS-induced systemic inflammatory response syndrome and liver injury in rats,and improve outcomes.These protective effects may be due to the ability of XBJ to inhibit cytokine secretion by KCs.Yi Chen Huasheng Tong Xingqin Zhang Liqun Tang Zhiguo Pan Zhifeng Liu Pengkai Duan Lei Su 2013Journal of Traditional Chinese Medicine2013,33,2:14
