| 1 | Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021,41,11: | 29 |

| 3 | Association of Dietary Pattern during Pregnancy and Gestational Diabetes Mellitus: A Prospective Cohort Study in Northern China显示文摘Objective To examine the association of maternal dietary patterns during pregnancy with gestational diabetes mellitus(GDM) in northern China.Methods The dietary intakes of pregnant women were recorded twice by 24‐hour dietary recalls for three days prior to having been diagnosed with GDM, at 5‐15 and 24‐28 gestational weeks, respectively.GDM was diagnosed, and serum glycosylated hemoglobin(HbA1c) was measured at 24‐28 weeks.Dietary patterns were assessed by factor analysis. The association of the dietary pattern with GDM and HbA1c was examined by multiple logistic models.Results Of 753 participants, 64(8.5%) were diagnosed with GDM. Four dietary patterns were identified: Western pattern(dairy, baked/fried food and white meat), traditional pattern(light‐colored vegetables, fine grain, red meat and tubers), mixed pattern(edible fungi, shrimp/shellfish and red meat)and prudent pattern(dark‐colored vegetables and deep‐sea fish). Compared with the prudent pattern,both the Western pattern and the traditional pattern were associated with an increased risk of GDM(a OR = 4.40, 95% CI: 1.58‐12.22; a OR = 4.88, 95% CI: 1.79‐13.32) and a high level of HbA1c(a OR = 12.37,95% CI: 1.47‐103.91; a OR = 26.23, 95% CI: 2.54‐270.74). | 4 | Grape Seed Proanthocyanidin Extract Alleviates Arsenic-induced Oxidative Reproductive Toxicity in Male Mice显示文摘Objective To determine the ability of grape seed proanthocyanidin extract(GSPE) in alleviating arsenic-induced reproductive toxicity. Methods Sixty male Kunming mice received the following treatments by gavage: normal saline solution(control); arsenic trioxide(ATO; 4 mg/kg); GSPE(400 mg/kg); ATO+GSPE(100 mg/kg); ATO+GSPE(200 mg/kg) and ATO+GSPE(400 mg/kg). Thereafter, the mice were sacrificed and weighed, and the testis was examined for pathological changes. Nuclear factor(erythroid-derived 2)-like 2(Nrf2), heme oxygenase 1(HO1), glutathione S-transferase(GST), NAD(P)H dehydrogenase, and quinone 1(NQO1) expression in the testis was detected by real-time PCR. Superoxide dismutase(SOD), glutathione(GSH), total antioxidative capability(T-AOC), malondialdehyde(MDA), 8-hydroxydeoxyguanosine(8-OHd G), and reproductive indexes were analyzed. Results ATO-treated mice showed a significantly decreased sperm count and testis somatic index and activity levels of SOD, GSH, and T-AOC than control group. Compared to the ATO-treated group, ATO +GSPE group showed recovery of the measured parameters. Mice treated with ATO+high-dose GSPE showed the highest level of m RNA expression of Nrf2, HO, NQO1, and GST. | 5 | A comparative study of natural and experimental nano-sized grinding grain textures in rocks显示文摘In three shallow-level fault systems in eastern China, nano-sized (30―100 nm) grinding grain textures are found in the thin-shelled rheological layer representing frictional-viscous high strain field. The surface layer near slipping plane is composed of stacks of grinding grains with high sphericity and uniform nano-sized diameter, while the underlying layer appears as a mixture of irregular ones with diverse diameters. Rock deformation experiments indicate that the grinding grain texture is a common phenomenon in shearing fractures, suggesting a potential transition from sliding friction to rolling one during the rock deformation process. | 6 | Association between Chinese Medicine Therapy and Survival Outcomes in Postoperative Patients with NSCLC: A Multicenter, Prospective, Cohort Study显示文摘Objective:To evaluate the association between Chinese medicine(CM)therapy and disease-free survival(DFS)outcomes in postoperative patients with non-small cell lung cancer(NSCLC).Methods:This multiple-center prospective cohort study was conducted in 13 medical centers in China.Patients with stage I,II,or IDA NSCLC who had undergone radical resection and received conventional postoperative treatment according to the National Comprehensive Cancer Network(NCCN)guidelines were recruited.The recruited patients were divided into a CM treatment group and a control group according to their wishes.Patients in the CM treatment group received continuous CM therapy for more than 6 months or until disease progression.Patients in the control group received CM therapy for less than 1 month.Follow-up was 8nducted over 3 years.The primary outcome was DFS,with recurrence/metastasis rates as a secondary outcome.Results:Between May 2013 and August 2016,503 patients were enrolled into the cohort;266 were classified in the CM treatment group and 237 in the control group.Adjusting for covariates,high exposure to CM was associated with better DFS[hazard ratio(HR)=0.417,95%confidential interval(Cl):0.307-0.567)].A longer duration of CM therapy(6-12 months,12-18 months,>24 months)was associated with lower recurrence and metastasis rates(HR=0.225,0.119 and 0.083,respectively).In a subgroup exploratory analysis,CM therapy was also a protective factor of cancer recurrence and metastasis in both stage n-MA(HR=0.50,95%Cl:0.37-0.67)and stage IDA NSCLC postoperative patients(HR=0.48,95%Cl:0.33-0.71),DFS was even longer among CM treatment group patients.Conclusions:Lon ger duration of CM therapy could be 8nsidered a protective factor of cancer recurrence and metastasis.CM treatment is associated with improving survival outcomes of postoperative NSCLC patients in China. | WANG Xue-qian ZHANG Ying HOU Wei WANG Ying-tian ZHENG Jia-bin LI Jie LIN Li-zhu JIANG Yi-lan WANG Shen-yu XIE Ying ZHANG Hong-liang SHU Qi-jin LI Ping WANG Wei YOU Jian-liang LI Ge LIU Jie FAN Hui-ting ZHANG Mei-ying LIN Hong-sheng | 2019 | Chinese Journal of Integrative Medicine2019,25,11: | 12 |

| 11 | Protective Effects of Quercetin on Cadmium-induced Cytotoxicity in Primary Cultures of Rat Proximal Tubular Cells显示文摘Objective To investigate the protective effects of quercetin on cadmium-induced cytotoxicity in primary cultures of rat proximal tubular(rPT)cells.Methods Primary cultures of rPT cells undergoing exponential growth were incubated with 1.0 μg/mL quercetin and/or cadmium(2.5,5.0 μmol/L),in a serum-free medium at 37 ℃ at different time intervals.Commercial kits were used and flow cytometric analyses were performed on rPT cell cultures to assay apoptosis and oxidative stress.Results Exposure of rPT cells to cadmium acetate(2.5,5.0 μmol/L)induced a decrease in cell viability,caused an increase in apoptotic rate and apoptotic morphological changes.Simultaneously,elevation of intracellular reactive oxygen species,malondialdehyde and calcium levels,depletion of mitochondrial membrane potential and intracellular glutathione,and inhibition of Na +,K +-ATPase,Ca 2+-ATPase,glutathione peroxidase(GSH-Px),catalase(CAT),and superoxide dismutase(SOD)activities were revealed during the cadmium exposure of rPT cells.However,simultaneous supplementation with 1 μg/mL quercetin protected rPT cells against cadmium-induced cytotoxicity through inhibiting apoptosis,attenuating lipid peroxidation,renewing mitochondrial function and elevating the intracellular antioxidants(non-enzymatic and enzymic)levels.Conclusion The present study has suggested that quercetin,as a widely distributed dietary antioxidant,contributes potentially to prevent cadmium-induced cytotoxicity in rPT cells. | WANG Lin LIN Shu Qian HE Yuan Long LIU Gang WANG Zhen Yong | 2013 | Biomedical and Environmental Sciences2013,26,4: | 8 |

| 13 | Irinotecan plus S-1 versus S-1 in patients with previously treated recurrent or metastatic esophageal cancer(ESWN 01):a prospective randomized,multicenter,open-labeled phase 3 trial显示文摘Background:The benefit of systemic treatments in esophageal squamous cell carcinoma(ESCC)which has pro-gressed after chemotherapy is still uncertain and optimal regimens based on randomized trials have not yet been established.We aimed to compare the efficacy of irinotecan plus S-1 with S-1 monotherapy in recurrent or metastatic ESCC patients who had resistance to platinum-or taxane-based chemotherapy.Methods:We conducted a prospective randomized,multicenter,open-label,phase 3 trial in 15 centers across China.Eligible patients were adults with histologically confirmed recurrent or metastatic ESCC,and were randomly assigned(ratio,1:1)to receive either irinotecan plus S-1(intravenous infusion of irinotecan[160 mg/m2]on day 1 and oral S-1[80-120 mg]on days 1-10,repeated every 14 days)or oral S-1 monotherapy(80-120 mg/day on days 1-14,repeated every 21 days)using a central computerized minimization procedure.The primary endpoint was progression-free survival(PFS).Results:Between December 23,2014 and July 25,2016,we screened 148 patients and randomly assigned 123 patients to receive either irinotecan plus S-1 regimen(n=61)or S-1 monotherapy(n=62).After a median follow-up of 29.2 months(95%confidence interval[CI]17.5-40.9 months),the median PFS was significantly longer in the irinotecan plus S-1 group than in the S-1 monotherapy group(3.8 months[95%CI 2.9-4.3 months]vs.1.7 months[95%CI 1.4-2.7 months],hazard ratio=0.58,95%CI 0.38-0.86,P=0.006).The objective response rates were 24.6%in the irinotecan plus S-1 group and 9.7%in the S-1 monotherapy group(P=0.002).The patients in the irinotecan plus S-1 group presented with increased rates of grade 3-4 leukopenia(16.4%vs.0%),neutropenia(14.8%vs.1.6%),and nausea(4.9%vs.0%).No significant difference in grade 3-4 diarrhea and no treatment-related deaths were observed in both groups.Conclusions: The combination of irinotecan with S-1 was similarly tolerable but significantly prolonged PFS compared to S-1 monotherapy as a second- or third-line treatment in patients with recurrent or metastatic ESCC. | Jing Huang Binghe Xu Ying Liu Junxing Huang Ping Lu Yi Ba Lin Wu Yuxian Bai Shu Zhang Jifeng Feng Ying Cheng Jie Li Lu Wen Xianglin Yuan Changwu Ma Chunhong Hu Qingxia Fan Xi Wang | 2019 | Cancer Communications2019,39,1: | 8 |
| 14 | Identification of biomarkers for hepatocellular carcinoma by semiquantitative immunocytochemistry显示文摘AIM:To investigate the expression of key biomarkers in hepatoma cell lines,tumor cells from patients’blood samples,and tumor tissues.METHODS:We performed the biomarker tests in two steps.First,cells plated on coverslips were used to assess biomarkers,and fluorescence intensities were calculated using the NIH Image J software.The measured values were analyzed using the SPSS19.0 software to make comparisons among eight cell lines.Second,eighty-four individual samples were used to assess the biomarkers’expression.Negative enrichment of the blood samples was performed,and karyocytes were isolated and dropped onto pretreated glass slides for further analysis by immunofluorescence staining.Fluorescence intensities were compared among hepatocellular carcinoma(HCC)patients,chronic HBV-infected patients,and healthy controls following methods similar to those used for cell lines.The relationships between the expression of biomarkers and clinical pathological parameters were analyzed by Spearman rank correlation tests.In addition,we studied the distinct biomarkers’expression with three-dimensional laser confocal microscopy reconstructions,and Kaplan-Meier survival analysis was performed to understand the clinical significance of these biomarkers.RESULTS:Microscopic examination and fluorescence intensity calculations indicated that cytokeratin 8/18/19(CK)expression was significantly higher in six of the seven HCC cell lines examined than in the control cells,and the expression levels of asialoglycoprotein receptor(ASGPR)and glypican-3(GPC3)were higher in all seven HCC cell lines than in the control.Cells obtained from HCC patients’blood samples also displayed significantly higher expression levels of ASGPR,GPC3,and CK than cells from chronic HBV-infected patients or healthy controls;these proteins may be valuable surface biomarkers for identifying HCC circulating tumor cells isolated and enriched from the blood samples.The stem cell-like and epithelial-mesenchymal transition-related biomarkers could be detected on the karyocyte slides.ASGPR and GPC3 were expressed at high levels,and thus three-dimensional reconstructions were used to observe their expression in detail.This analysis indicated that GPC3 was localized in the cytoplasm and membrane,but that ASGPR had a polar localization.Survival analyses showed that expression of GPC3 and ASGPR is associated with a patient’s overall survival(OS).CONCLUSION:ASGPR,GPC3,and CK may be valuable HCC biomarkers for CTC detection;the expression of ASGPR and GPC3 might be helpful for understanding patients’OS. | Hong Mu Kai-Xuan Lin Hong Zhao Shu Xing Cong Li Fang Liu Hai-Zhen Lu Ze Zhang Yu-Lin Sun Xi-Yun Yan Jian-Qiang Cai Xiao-Hang Zhao | 2014 | World Journal of Gastroenterology2014,20,19: | 8 |

| 17 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 19 | Recent progress in studies layer in rock on the nano-sized particle shear planes显示文摘Since the 1990s, an ultramicro-particle texture in nano- or micron-sized scale has been continually found in shear planes of various rocks, and recently further progress was made in generic physical mechanisms. In this article, the characteristics of the nano-sized particle layer in rock shear planes (including widespread distribution, layering texture, non-linear genetic mechanism, and multiple functions) on an ultra micro-scale were briefly introduced and more information was obtained by analyzing cases from home and abroad. Our findings suggest that (1) there is a paragenetic relationship between the development of shear frictional- viscous and the formation of nano-sized particles, and (2) in the shear movement, partitioning, segregating, and layering in rocks are initiated by the sliding motion of the nano-sized particle layer.
