1Neural mechanism of gastric motility regulation by electroacupuncture at RN12 and BL21: A paraventricular hypothalamic nucleus-dorsal vagal complex-vagus nervegastric channel pathway显示文摘AIM: To study the neural mechanism by which electroacupuncture(EA) at RN12(Zhongwan) and BL21(Weishu) regulates gastric motility.METHODS: One hundred and forty-four adult Sprague Dawley rats were studied in four separate experiments. Intragastric pressure was measured using custommade rubber balloons, and extracellular neuron firing activity, which is sensitive to gastric distention in the dorsal vagal complex(DVC), was recorded by an electrophysiological technique. The expression levels of c-fos, motilin(MTL) and gastrin(GAS) in the paraventricular hypothalamic nucleus(PVN) were assayed by immunohistochemistry, and the expression levels of motilin receptor(MTL-R) and gastrin receptor(GAS-R) in both the PVN and the gastric antrum were assayed by western blotting.RESULTS: EA at RN12 + BL21(gastric Shu and Mu points), BL21(gastric Back-Shu point), RN12(gastric Front-Mu point), resulted in increased neuron-activating frequency in the DVC(2.08 ± 0.050, 1.17 ± 0.023, 1.55 ± 0.079 vs 0.75 ± 0.046, P < 0.001) compared with a model group. The expression of c-fos(36.24 ± 1.67, 29.41 ± 2.55, 31.79 ± 3.00 vs 5.73 ± 2.18, P < 0.001), MTL(22.48 ± 2.66, 20.76 ± 2.41, 19.17 ± 1.71 vs 11.68 ± 2.52, P < 0.001), GAS(24.99 ± 2.95, 21.69 ± 3.24, 23.03 ± 3.09 vs 12.53 ± 2.15, P < 0.001), MTL-R(1.39 ± 0.05, 1.22 ± 0.05, 1.17 ± 0.12 vs 0.84 ± 0.06, P < 0.001), and GAS-R(1.07 ± 0.07, 0.91 ± 0.06, 0.78 ± 0.05 vs 0.45 ± 0.04, P < 0.001) increased in the PVN after EA compared with the model group. The expression of MTL-R(1.46 ± 0.14, 1.26 ± 0.11, 0.99 ± 0.07 vs 0.65 ± 0.03, P < 0.001), and GAS-R(1.63 ± 0.11, 1.26 ± 0.16, 1.13 ± 0.02 vs 0.80 ± 0.11, P < 0.001) increased in the gastric antrum after EA compared with the model group. Damaging the PVN resulted in reduced intragastric pressure(13.67 ± 3.72 vs 4.27 ± 1.48, P < 0.001). These data demonstrate that the signals induced by EA stimulation of acupoints RN12 and BL21 are detectable in the DVC and the PVN, and increase the levels of gastrointestinal hormones and their receptors in the PVN and gastric antrum to regulate gastric motility. 3Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 6Extractive distillation: Advances in conceptual design, solvent selection,and separation strategies显示文摘Extractive distillation(ED) is one of the most promising approaches for the separation of the azeotropic or closeboiling mixtures in the chemical industry. The purpose of this paper is to provide a broad overview of the recent development of key aspects in the ED process involving conceptual design, solvent selection, and separation strategies. To obtain the minimum entrainer feed flow rate and reflux ratio for the ED process, the conceptual design of azeotropic mixture separation based on a topological analysis via thermodynamic feasibility insights involving residue curve maps, univolatility lines, and unidistribution curves is presented. The method is applicable to arbitrary multicomponent mixtures and allows direct screening of design alternatives. The determination of a suitable solvent is one of the key steps to ensure an effective and economical ED process. Candidate entrainers can be obtained from heuristics or literature studies while computer aided molecular design(CAMD) has superiority in efficiency and reliability. To achieve optimized extractive distillation systems, a brief review of evaluation method for both entrainer design and selection through CAMD is presented. Extractive distillation can be operated either in continuous extractive distillation(CED) or batch extractive distillation(BED), and both modes have been well-studied depending on the advantages in flexibility and low capital costs. To improve the energy efficiency, several configurations and technological alternatives can be used for both CED and BED depending on strategies and main azeotropic feeds. 7Tenofovir vs.entecavir on recurrence of hepatitis B virus-related hepatocellular carcinoma beyond Milan criteria after hepatectomy显示文摘Background:Hepatectomy for hepatocellular carcinoma(HCC)beyond the Milan criteria is shown to be beneficial.However,a high rate of post-operative HCC recurrence hinders the long-term survival of the patients.This study aimed to investigate and compare the impacts of tenofovir(TDF)and entecavir(ETV)on the recurrence of hepatitis B viral(HBV)-related HCC beyond the Milan criteria.Methods:Data pertaining to 1532 patients who underwent hepatectomy and received antiviral therapy between January 2014 and January 2019 were collected from five centers.Recurrence-free survival(RFS)analysis was performed using the Kaplan-Meier method.Cox proportional hazards regression analysis was performed to determine prognostic factors for HCC recurrence.Results:The analysis incorporates 595 HBV-related HCC patients.The overall 5-year RFS was 21.3%.Among them,533 and 62 patients received ETV and TDF treatment,respectively.The 1-,3-,and 5-year RFS rates were 46.3%,27.4%,and 19.6%,respectively,in the ETV group compared with 65.1%,41.8%,and 37.2%,respectively,in the TDF group(P<0.001).Multivariate analysis showed that TDF treatment(hazard ratio[HR]:0.604,P=0.005),cirrhosis(HR:1.557,P=0.004),tumor size(HR:1.037,P=0.008),microvascular invasion(MVI)(HR:1.403,P=0.002),portal vein tumor thrombus(PVTT)(HR:1.358,P=0.012),capsular invasion(HR:1.228,P=0.040),and creatinine levels(CREA)(HR:0.993,P=0.031)were statistically significant prognostic factors associated with RFS.Conclusions:Patients with HCC beyond the Milan criteria exhibited a high rate of HCC recurrence after hepatectomy.Compared to the ETV therapy,TDF administration significantly lowered the risk of HCC recurrence.Junyi Shen Weili Qi Junlong Dai Shusheng Leng Kangyi Jiang Yu Zhang Shun Ran Chuan Li Tianfu Wen 2022Chinese Medical Journal2022,,3:3

10Biomedical polymers: synthesis, properties, and applications显示文摘Biomedical polymers have been extensively developed for promising applications in a lot of biomedical fields, such as therapeutic medicine delivery, disease detection and diagnosis, biosensing, regenerative medicine, and disease treatment. In this review, we summarize the most recent advances in the synthesis and application of biomedical polymers, and discuss the comprehensive understanding of their property-function relationship for corresponding biomedical applications. In particular, a few burgeoning bioactive polymers, such as peptide/biomembrane/microorganism/cell-based biomedical polymers, are also introduced and highlighted as the emerging biomaterials for cancer precision therapy. Furthermore, the foreseeable challenges and outlook of the development of more efficient, healthier and safer biomedical polymers are discussed. 13Immunogenicity and protective efficacy of DHBV DNA vaccines expressing envelope and capsid fusion proteins in ducks delivered by attenuated Salmonella typhimurium显示文摘Duck hepatitis B virus(DHBV) shares many basic characteristics with hepatitis B virus(HBV) and is an attractive model for vaccine development. In this study, DHBV DNA vaccines were designed to express envelope and capsid fusion proteins to enhance the breadth of immune response in ducks. Attenuated Salmonella typhimurium(SL7207) was used as a carrier and adjuvant to boost the magnitude of immune response. Based on this strategy, novel DNA vaccines(SL7207-p VAX1-LC and SL7207-p VAX1-SC) were generated. Growth kinetics, genetic stabilities and relative transcription levels of the L, S and C genes introduced by these vaccine strains were measured before inoculation to guarantee safety and efficacy. The relative transcript levels of the CD4 and CD8 T genes and the antibody levels(Ig Y) in ducks receiving the vaccines were higher than those in single gene delivered groups. 18Diffusion of helium in FCT zircon显示文摘To gain a better quantitative understanding of zircon(U-Th)/He ages and evaluate the applicability of zircon(U-Th)/He dating, the diffusion characteristics, He diffusion kinetics, helium partial retention zone(HePRZ), closure temperature, and(U-Th)/He ages were investigated using high-precision laboratory step heating experiments based on the thermally activated diffusion process. The ln(D/a^2) in Fish Canyon Tuff(FCT) zircons determined from laboratory step heating experiments was negatively correlated with reciprocal temperature, as expected for thermally activated volume diffusion. The zircon activation energies ranged from 144 to 184 kJ mol^(-1) with a mean of 169±12 kJ mol^(-1). The closure temperatures ranged from 144 to 216°C(a cooling rate of 10°C Ma^(-1) and an effective grain radius of 38–60 μm) with an average of 176±18°C. The calculated closure temperature increased with increasing cooling rate, yielding an average zircon He closure temperature of ~136°C at a slow cooling rate of 0.1°C Ma^(-1), whereas the closure value was ~199°C at a cooling rate of 100°C Ma^(-1). The closure temperature increased with the equivalent spherical radius assuming a constant cooling rate. The He ages from FCT zircons were negligibly affected by grain size because of the rapid cooling. He preserved in the zircon was sensitive to temperature and holding time, and the temperature range for zircon HePRZ gradually decreased with increasing holding time. The(U-Th)/He ages from 26 FCT zircons yielded an algorithmic mean of 28.3±0.3 Ma(S.E.) and a geometric mean of 28.4±0.3 Ma(S.E.), consistent with the ages of 28.4±1.9 Ma reported by other laboratories. The FCT zircons were characterized by rapid cooling, young(U-Th)/He ages with good reproducibility, and low alpha doses. 19Real-time measurements of PM_(2.5),PM_(10-2.5),and BC in an urban street canyon显示文摘A continuous dichotomous beta gauge monitor was used to characterize the hourly content of PM_(2.5).PM_(10-2.5).and Black Carbon(BC)over a 12-month period in an urban street canyon of Hong Kong.Hourly vehicle counts for nine vehicle classes and meteorological data were also recorded.The average weekly cycles of PM_(2.5),PM_(10-2.5).and BC suggested that all species are related to traffic,with high concentrations on workdays and low concentrations over the weekends.PM_(2.5) exhibited two comparable concentrations at 10:00-11:00(63.4 μg/m^3) and 17:00-18:00(65.0 μg/m^3) local time(LT) during workdays,corresponding to the hours when the numbers of diesel-fueled and gasoline-fueled vehicles were at their maximum levels:3179 and 2907 h^(-1),respectively.BC is emitted mainly by diesel-fueled vehicles and this showed the highest concentration(31.2 μg/m^3) during the midday period(10:00-11:00 LT) on workdays.A poor correlation was found between PM_(2.5) concentration and wind speed(R = 0.51,P-value> 0.001).In contrast,the concentration of PM_(10-2.5) was found to depend upon wind speed and it increased with obvious statistical significance as wind speed increased(R = 0.98,P-value< 0.0001).Yan Cheng Shun Cheng Lee Yuan Gao Long Cui Wenjing Deng Junji Cao Zhenxing Shen Jian Sun 2015Particuology2015,13,3:1
