| 1 | Crustal velocity structure in the Emeishan large igneous province and evidence of the Permian mantle plume activity显示文摘The Emeishan large igneous province(ELIP) in SW China is interpreted to be associated with an ancient mantle plume. Most of the constraints on the role of mantle plume in the generation of the Emeishan flood basalts were provided by geological and geochemical methods, but the geophysical investigation is very limited. In order to better understand the deep structure and features of ELIP, we have studied the crustal velocity structure using the data acquired from the Lijiang-Panzhihua-Qingzhen wide-angle seismic profile. This profile crosses the three sub-zones of the ELIP(the inner, intermediate, and outer zones), divided based on the differential erosion and uplift of the Maokou limestone. The results provided by the active source seismic experiment demonstrate:(1) The average depth of the crystalline basement along the profile is about 2 km.(2) The middle crust in the Inner Zone is characterized by high-velocity anomalies, with the average velocity of 6.2-6.6 km/s, which is about 0.1– 0.2 km/s higher than the normal one. The velocity of the lower crust in the inner zone is 6.9-7.2 km/s, higher than those observed in the intermediate and outer zones(6.7-7.0 km/s). Relatively low velocity anomalies appear in the upper, middle and lower crusts near the junction of the inner zone and intermediate zone, probably due to the effect of the Xiaojiang fault(XJF).(3) The average velocity of the crust is comparatively low on both sides of XJF, especially on the east side, and the average velocity of the consolidated continental crust is also low there. This may suggest that the XJF extends at least down to 40 km deep, even beyond through the crust.(4) The depth to the Moho discontinuity decrease gradually from 47-53 km in the inner zone, via 42-50 km in the intermediate zone to 38-42 km in the outer zone. In the inner zone, the Moho uplifts locally and the(consolidated) crust is characterized by high-velocity anomalies, which are likely related to intensive magma intrusion and underplating associated with melting of plume head. | 2 | Functionalization of small black phosphorus nanoparticles for targeted imaging and photothermal therapy of cancer显示文摘Black phosphorus(BP) nanomaterials have attracted extensive attention due to their unique physical,chemical, and biological properties. In this study, small BP nanoparticles were synthesized and modified with dextran and poly(ethyleneimine) for functionalization with folic acid and cyanine 7. The functionalized BP nanoparticles exhibit excellent biocompatibility, stability, and near infrared optical properties for targeted imaging of tumors through photoacoustic imaging and near-infrared fluorescence imaging.They also display high photothermal conversion efficiency for photothermal therapy of cancer. | 5 | Functional assembly of root-associated microbial consortia improves nutrient efficiency and yield in soybean显示文摘Root-associated microbes are critical for plant growth and nutrient acquisition. However, scant information exists on optimizing communities of beneficial root-associated microbes or the mechanisms underlying their interactions with host plants. In this report, we demonstrate that rootassociated microbes are critical influencers of host plant growth and nutrient acquisition. Three synthetic communities(SynComs) were constructed based on functional screening of 1,893 microbial strains isolated from root-associated compartments of soybean plants. Functional assemblage of SynComs promoted significant plant growth and nutrient acquisition under both N/P nutrient deficiency and sufficiency conditions.Field trials further revealed that application of SynComs stably and significantly promoted plant growth, facilitated N and P acquisition, and subsequently increased soybean yield. Among the tested communities, SynCom1 exhibited the greatest promotion effect, with yield increases of up to 36.1% observed in two field sites. Further RNA-seq implied that SynCom application systemically regulates N and P signaling networks at the transcriptional level, which leads to increased representation of important growth pathways, especially those related to auxin responses. | 6 | Identification of HSC/MPP expansion units in fetal liver by single-cell spatiotemporal transcriptomics显示文摘Limited knowledge of cellular and molecular mechanisms underlying hematopoietic stem cell and multipotent progenitor(HSC/MPP)expansion within their native niche has impeded the application of stem cell-based therapies for hematological malignancies.Here,we constructed a spatiotemporal transcriptome map of mouse fetal liver(FL)as a platform for hypothesis generation and subsequent experimental validation of novel regulatory mechanisms.Single-cell transcriptomics revealed three transcriptionally heterogeneous HSC/MPP subsets,among which a CD93-enriched subset exhibited enhanced stem cell properties.Moreover,by employing integrative analysis of single-cell and spatial transcriptomics,we identified novel HSC/MPP‘pocket-like’units(HSC PLUS),composed of niche cells(hepatoblasts,stromal cells,endothelial cells,and macrophages)and enriched with growth factors.Unexpectedly,macrophages showed an 11-fold enrichment in the HSC PLUS.Functionally,macrophage-HSC/MPP co-culture assay and candidate molecule testing,respectively,validated the supportive role of macrophages and growth factors(MDK,PTN,and IGFBP5)in HSC/MPP expansion.Finally,cross-species analysis and functional validation showed conserved cell-cell interactions and expansion mechanisms but divergent transcriptome signatures between mouse and human FL HSCs/MPPs.Taken together,these results provide an essential resource for understanding HSC/MPP development in FL,and novel insight into functional HSC/MPP expansion ex vivo. | Suwei Gao Qiang Shi Yifan Zhang Guixian Liang Zhixin Kang Baofeng Huang Dongyuan Ma Lu Wang Jianwei Jiao Xiangdong Fang Cheng-Ran Xu Longqi Liu Xun Xu Berthold Gottgens Cheng Li Feng Liu | 2022 | Cell Research2022,32,1: | 4 |

| 9 | Transsphenoidal microsurgical treatment of infradiaphragmatic craniopharyngioma显示文摘Objective Typically,the transcranial approach has been used for the treatment of craniopharyngiomas with suprasellar extension,whereas the transsphenoidal approach has been used mostly for infradiaphragmatic craniopharyngioma.Total resection of craniopharyngioma can reduce the recurrence rate,especially in young children,but it may lead to severe complications.Therefore,any benefit of the degree of resection must be weighed against the risk of complications by the surgeons.The purpose of this study was to explore the therapeutic outcome after transsphenoidal microsurgical treatment of infradiaphragmatic craniopharyngioma and share our experiences.Methods Between January 2003 and June 2013,30 patients with infradiaphragmatic craniopharyngioma underwent transsphenoidal microsurgical resection in our hospital.The neurological,visual,and endocrine functions,and extent of resection were analyzed retrospectively.Recurrence or growth of residual tumor tissue during follow-up was assessed using magnetic resonance imaging(MRI).Results Total resection was achieved in 25 patients(83.3%),subtotal resection was achieved in 4 patients(13.3%),and partial resection was achieved in 1 patient(3.4%).There were no perioperative deaths.Cerebrospinal fluid(CSF) leakage occurred in 6 patients,and among them,2 required surgical repair of the sella.New-onset postoperative diabetes insipidus(DI) developed in 8 patients.Vision and visual fields were improved at different levels in 13 out of 16 patients who had sight impediments before treatment.Tumor recurrence and regrowth was observed in 2 patients;1 patient underwent transsphenoidal reoperation,the condition of the other patient who had undergone several craniotomies grew worse over the 6-month follow-up period.Conclusion Transsphenoidal surgery is an ideal choice in treating infradiaphragmatic craniopharyngioma.The transsphenoidal approach,which preserves pituitary function and avoids damage to the hypothalamic structures and optic nerve,is associated with fewer complications than the transcranial approach and a low mortality rate. | Ting Lei Baofeng Wang Juan Chen Yu Xu Kai Shu Wei Sun Shaozheng Liu Xiaopeng Li | 2016 | Oncology and Translational Medicine2016,2,5: | 2 |
| 10 | Dasatinib inhibits proliferation of liver cancer cells,but activation of Akt/mTOR compromises dasatinib as a cancer drug显示文摘Dasatinib is a multi-target protein tyrosine kinase inhibitor.Due to its potent inhibition of Src,Abl,the platelet-derived growth factor receptor(PDGFR)family kinases,and other oncogenic kinases,it has been investigated as a targeted therapy for a broad spectrum of cancer types.However,its efficacy has not been significantly extended beyond leukemia.The mechanism of resistance to dasatinib in a wide array of cancers is not clear.In the present study,we investigated the effect of dasatinib on hepatocellular carcinoma cell growth and explored the underlying mechanisms.Our results showed that dasatinib potently inhibited the proliferation of SNU-449 cells,but not that of other cell lines,such as SK-Hep-1,even though it inhibited the phosphorylation of Src on both negative and positive regulation sites in all these cells.Dasatinib activated the phosphoinositide-dependent protein kinase1(PDK1)/protein kinase B(Akt)/mammalian target of rapamycin(mTOR)signaling pathway in SK-Hep-1 cells,but not in SNU-449 cells.Blocking the Akt/mTOR signaling pathway strongly promoted the efficacy of dasatinib in SK-Hep-1 cells.In SNU-449 cells,dasatinib promoted apoptosis and the cleavage of caspase-3 and caspase-7,induced cell cycle arrest in the G1 phase,and inhibited the expression of Cyclin-dependent kinase(CDK4)/6/CyclinD1 complex.These findings demonstrate that dasatinib exerts its anti-proliferative effect on hepatocellular cell proliferation by blocking the Src family kinases;however,it causes Akt activation,which compromises dasatinib as an anti-cancer drug. | Chang Liu Xiaoxia Zhu Yuqi Jia Fenqing Chi Keru Qin Jinhong Pei Chan Zhang Xiuli Mu Hongwei Zhang Xiushan Dong Jun Xu Baofeng Yu | 2021 | Acta Biochimica et Biophysica Sinica2021,53,7: | 2 |

| 12 | Investigation of plateau basin crustal structures and thickening mechanisms in the northeastern margin of the Tibetan plateau显示文摘This paper uses deep seismic sounding (DSS) data to contrast and analyze the crustal structures of three plateau basins (Songpan-Garze, Qaidam, Longzhong) in the northeastern margin of the Qinghai-Xizang (Tibetan) plateau, as well as two stable cratonic basins (Ordos, Sichuan) in its peripheral areas. Plateau basin crustal structures, lithological variations and crustal thickening mechanisms were investigated. The results show that, compared to the peripheral stable cratonic basins, the crystalline crusts of plateau basins in the northeastern margin are up to 10-15 km thicker, and the relative medium velocity difference is about 5% less. The medium velocity change in crustal layers of plateau basin indicates that the upper crust undergoes brittle deformation, whereas the lower crust deforms plastically with low velocity. The middle crust shows a brittle-to-plastic transition zone in this region. Thickening in the lower crust (about 5-10 km), and rheological characteristics that show low- medium velocity (relatively reduced by 7%), suggest that crustal thickening mainly takes place in lower crust in the northeastern margin of the Tibetan plateau. The crust along the northeastern margin shows evidence of wholesale block movement, and crustal shortening and thickening seem to be the main deformation features of this region. The GPS data show that the block motion modes and crustal thickening in the Tibetan plateau is closely related to the peripheral tectonic stress field and motion direction of the Indian plate.
