1Chromosomal localization of the major ribosomal RNA genes in scallop Chlamys farreri显示文摘The chromosomes of Chlamys farreri were analyzed by means of silver staining and fluorescence in situ hybridization (FISH) with 18S—28S rDNA probe. Probe was made by PCR amplification of a DNA fragment containing internal transcribed spacers ITS1 between 18S and 5.8S ribosomal RNA gene, ITS2 between 5.8S and 28S ribosomal RNA gene and 5.8S rRNA gene, and labeled by PCR incorporation of bio-16-dUTP. FISH signals were located on the short arm of subtelocentric chromosome 10. After silver-staining, nucleolus organizer regions (NORs) could be observed on the telomere of the short arm of chromosome 10. 2Relationship Between 2:1 Mineral Structure and the Fixation and Release of Cations显示文摘The effects of different structures of 2:1 layer minerals, layer charge location, and changes of structure and charge during the weathering process on the fixation and release of interlayer cations are reviewed. It could be concluded that the fixation capacity is determined by the total amount of interlayer charge originating from both octahedral and tetrahedral sheets. The relationship between interlayer cation fixation and octahedral structure of the secondary minerals may be different from that of the primary minerals. 3Stock index adjustments, analyst coverage and institutional holdings: Evidence from China显示文摘Using 231 pairs of matched firms from 2009 to 2012 in Chinese stock market,we find that the stock index adjustment significantly affects the analyst coverage, which in addition to the stock index leads to more analyst coverage, while deletion from the stock index has no significant effect, indicating that stock index adjustment can significantly change the information environments of firms that are added to the index. An index adjustment also affects institutional holdings in consideration of new information(e.g., changes in fundamentals and information environments). 5Puromycin A, B and C, cryptic nucleosides identified from Streptomyces alboniger NRRL B-1832 by PPtase-based activation显示文摘Natural product discovery is pivot for drug development,however,this endeavor is often challenged by the wide inactivation or silence of natural products biosynthetic pathways.We recently developed a highly efficient approach to activate cryptic/silenced biosynthetic pathways through augmentation of the phosphopantetheinylation of carrier proteins.By applying this approach in the Streptomyces alboniger NRRL B-1832,we herein identified three cryptic nucleosides products,including one known puromycin A and two new derivatives(puromycin B and C).The biosynthesis of these products doesn't require the involvement of carrier protein,indicating the phosphopantetheinyl transferase(PPtase)indeed plays a fundamental regulatory role in metabolites biosynthesis.These results demonstrate that the PPtasebased approach have a much broader effective scope than the previously assumed carrier proteininvolving pathways,which will benefit future natural products discovery and biosynthetic studies.Xiaoli Yan Benyin Zhang Wenya Tian Qi Dai Xiaoqin Zheng Ke Hu Xinxin Liu Zixin Deng Xudong Qu 2018Synthetic and Systems Biotechnology2018,3,1:3

7Deficiency of two-pore segment channel 2 contributes to systemic lupus erythematosus via regulation of apoptosis and cell cycle显示文摘Background:Systemic lupus erythematosus(SLE)is a complex autoimmune disease,and the mechanism of SLE is yet to be fully elucidated.The aim of this study was to explore the role of two-pore segment channel 2(TPCN2)in SLE pathogenesis.Methods:Quantitative reverse transcription polymerase chain reaction(qRT-PCR)was used to detect the expression ofTPCN2 in SLE.We performed a loss-of-function assay by lentiviral construct in Jurkat and THP-1 cell.Knockdown ofTPCN2 were confirmed at the RNA level by qRT-PCR and protein level by Western blotting.Cell Count Kit-8 and flow cytometry were used to analyze the cell proliferation,apoptosis,and cell cycle ofTPCN2-deficient cells.In addition,gene expression profile ofTPCN2-deficient cells was analyzed by RNA sequencing(RNA-seq).Results:TPCN2 knockdown with short hairpin RNA(shRNA)-mediated lentiviruses inhibited cell proliferation,and induced apoptosis and cell-cycle arrest of G2/M phase in both Jurkat and THP-1 cells.We analyzed the transcriptome of knockdown-TPCN2-Jurkat cells,and screened the differential genes,which were enriched for the G2/M checkpoint,complement,and interleukin-6-Janus kinase-signal transducer and activator of transcription pathways,as well as changes in levels of forkhead box O,phosphatidylinositol 3-kinase/protein kinase B/mechanistic target of rapamycin,and T cell receptor pathways;moreover,TPCN2 significantly influenced cellular processes and biological regulation.Conclusion:TPCN2 might be a potential protective factor against SLE.Keke Li Jingkai Xu Ke Xue Ruixing Yu Chengxu Li Wenmin Fei Xiaoli Ning Yang Han Ziyi Wang Jun Shu Yong Cui 2022Chinese Medical Journal2022,,4:2

9Nickel single atom overcoordinated active sites to accelerate the electrochemical reaction kinetics for Li-S cathode显示文摘Lithium-sulfur(Li-S)batteries with high theoretical energy density are promising advanced energy storage devices.However,shuttling of dissolute lithium polysulfide(LiPSs)and sluggish conversion kinetics impede their applications.Herein,single nickel(Ni)atoms on two-dimensional(2D)nitrogen(N)-doped carbon with Ni-N_(4)-O overcoordinated structure(SANi-N_(4)-O/NC)are prepared and firstly used as a sulfur host of Li-S batteries.Due to the efficient polysulfides traps and highly LiPSs conversion effect of SANi-N_(4)-O/NC,the electrochemical performance of Li-S batteries obviously improved.The batteries can well operate even under high sulfur loading(5.8 mg cm^(-2))and lean electrolyte(6.1μL mg^(-1))condition.Meanwhile,density functional theory(DFT)calculations demonstrate that Ni single atom’s active sites decrease the energy barriers of conversion reactions from Li_(2)S_(8)to Li2S due to the strong interaction between SANi-N_(4)-O/NC and LiPSs.Thus,the kinetic conversion of LiPSs was accelerated and the shuttle effect is suppressed on SANi-N_(4)-O/NC host.This study provides a new design strategy for a 2D structure with single-atom overcoordinated active sites to facilitate the fast kinetic conversion of LiPSs for Li-S cathode.Juan Zhu Xinyue Wang Tian Ke Mingji Jia Biyu Jin Yuanyuan Li Qiwei Yang Lanhui Ren Yongyuan Ren Dangguo Cheng Jianguo Lu Xiang Gao Qinggang He Yang Hou Xiaoli Zhan Qinghua Zhang 2023Journal of Energy Chemistry2023,,3:2
10Sustainability of SARS-CoV-2 Induced Humoral Immune Responses in COVID-19 Patients from Hospitalization to Convalescence Over Six Months显示文摘Understanding the persistence of antibody in convalescent COVID-19 patients may help to answer the current major concerns such as the risk of reinfection,the protection period of vaccination and the possibility of building an active herd immunity.This retrospective cohort study included 172 COVID-19 patients who were hospitalized in Wuhan.A total of404 serum samples were obtained over six months from hospitalization to convalescence.Antibodies in the specimens were quantitatively analyzed by the capture chemiluminescence immunoassays(CLIA).All patients were positive for the anti-SARS-Co V-2 Ig M/Ig G at the onset of COVID-19 symptoms,and the Ig G antibody persisted in all the patients during the convalescence.However,only approximately 25%of patients can detect the Ig M antibodies,Ig M against N protein(NIg M)and receptor binding domain of S protein(RBD-Ig M)at the 27 th week.The titers of Ig M,N-Ig M and RBD-Ig M reduced to 16.7%,17.6%and 15.2%of their peak values respectively.In contrast,the titers of Ig G,N-Ig G and RBD-Ig G peaked at 4–5 th week and reduced to 85.9%,62.6%and 87.2%of their peak values respectively at the end of observation.Dynamic behavior of antibodies and their correlation in age,gender and severity groups were investigated.In general,the COVID-19 antibody was sustained at high levels for over six months in most of the convalescent patients.Only a few patients with antibody reducing to an undetectable level which needs further attention.The humoral immune response against SARS-Co V-2 infection in COVID-19 patients exhibits a typical dynamic of acquired immunity.Yang Zheng Qing Zhang Ashaq Ali Ke Li Nan Shao Xiaoli Zhou Zhiqin Ye Xiaomin Chen Shanshan Cao Jing Cui Juan Zhou Dianbing Wang Baidong Hou Min Li Mengmeng Cui Lihua Deng Xinyi Sun Qian Zhang Qinfang Yang Yong li Hui Wang Yake Lei Bo Yu Yegang Cheng Xiaolin Tong Dong Men Xian-En Zhang 2021Virologica Sinica2021,36,5:1
11Effect of Potassium on Ammonium Fixation and Adsorption of Vermiculite in Binary and Ternary Systems显示文摘Ammonium fixation and adsorption experiments were conducted to study the effect of potassium on ammonium fixation into interlayer of vermiculite and ammonium adsorption at the surface of vermiculite both in the binary (NH-4~(+)-Ca~(2+)) and ternary (NH-4~(+)-K~(+)-Ca~(2+)) systems. In the ammonium fixation experiment, 5 mmol NH-4~(+) kg~(-1) was added alone, or after, before or simultaneously with 5 mmol K~(+) kg~(-1) to the vermiculite (vermiculite:solution = 1:1), and the incubation was conducted for 3 days under constant 20℃. In the adsorption experiment, after addition of 10 mmol Ca~(2+) L~(-1) as matrix ions, either NH-4~(+) alone with a concentration series from 0.1 to 5.0 mmol NH-4~(+) L~(-1) (binary exchange system of NH-4~(+) and Ca~(2+) or NH-4~(+) together with 5 mmol K~(+) L~(-1) (ternary exchange system of NH-4 ~(+), K~(+) and Ca~(2+) was added to vermiculite at a vermiculite to solution ratio of l:10 for 24-h equilibrium. The results of the fixation experiment showed that the presence of K~(+) increased NH-4~(+) fixation for all the treatments, regardless of the addition orders. For ammonium adsorption a linear relationship between activity ratios of NH-4~(+) to Ca~(2+) and Mg~(2+) in the equilibrium solutions (ARNH-4) and increases of exchangeable NH-4~(+) at the surface of vermiculite after 24- h equilibrium (△Ex-NH-4) was shown for almost the whole concentration ranges tested. Compared with the binary system, the slope of the curve of the ternary system was steeper, indicating a more intensive adsorption of NH-4~(+) in the presence of K~(+). 12Plastic supported platinum modified nickel electrode and its high electrocatalytic activity for sodium borohydride electrooxidation显示文摘A novel plastic/multi-walled carbon nanotube(MWNTs)-nickel(Ni)-platinum(Pt) electrode(PMNP) is prepared by chemical-reducing Pt onto the surface of Ni film covered plastic/MWNTs(PM) substrate. The MWNTs are adhered by a piece of commercial double faced adhesive tape on the surface of plastic paper and the Ni film is prepared by a simple electrodeposition method. The morphology and phase structure of the PMNP electrode are characterized by scanning electron microscopy,transmission electron microscope and X-ray diffractometer. The catalytic activity of the PMNP electrode for Na BH4 electrooxidation is investigated by means of cyclic voltammetry and chronoamperometry. The catalyst combines tightly with the plastic paper and exhibits a good stability. MWNTs serve as both conductive material and hydrogen storage material and the Ni film and Pt are employed as electrochemical catalysts. The PMNP electrode exhibits a high electrocatalytic performance and the oxidation current density reaches to 10.76 A/(mg·cm) in 0.1 mol/dm3 Na BH4at0 V,which is much higher than those in the previous reports. 15Effects of restoration modes on the spatial distribution of soil physical properties after land consolidation: a multifractal analysis显示文摘Soil physical properties(SPP)are considered to be important indices that reflect soil structure,hydrological conditions and soil quality.It is of substantial interest to study the spatial distribution of SPP owing to the high spatial variability caused by land consolidation under various land restoration modes in excavated farmland in the loess hilly area of China.In our study,three land restoration modes were selected including natural restoration land(NR),alfalfa land(AL)and maize land(ML).Soil texture composition,including the contents of clay,silt and sand,field capacity(FC),saturated conductivity(Ks)and bulk density(BD)were determined using a multifractal analysis.SPP were found to possess variable characteristics,although land consolidation destroyed the soil structure and decreased the spatial autocorrelation.Furthermore,SPP varied with land restoration and could be illustrated by the multifractal parameters of D1,ΔD,ΔαandΔf in different modes of land restoration.Owing to multiple compaction from large machinery in the surface soil,soil particles were fine-grained and increased the spatial variability in soil texture composition under all the land restoration modes.Plough numbers and vegetative root characteristics had the most significant impacts on the improvement in SPP,which resulted in the best spatial distribution characteristics of SPP found in ML compared with those in AL and NR.In addition,compared with ML,Δαvalues of NR and AL were 4.9-and 3.0-fold that of FC,respectively,andΔαvalues of NR and AL were 2.3-and 1.5-fold higher than those of Ks,respectively.These results indicate that SPP can be rapidly improved by increasing plough numbers and planting vegetation types after land consolidation.Thus,we conclude that ML is an optimal land restoration mode that results in favorable conditions to rapidly improve SPP.KE Zengming LIU Xiaoli MA Lihui TU Wen FENG Zhe JIAO Feng WANG Zhanli 2021Journal of Arid Land2021,13,12:0
16Status of transjugular intrahepatic portosystemic shunt for portal hypertension in China:A national survey analysis显示文摘Aim:The transjugular intrahepatic portosystemic shunt(TIPS)procedure has been performed in China for 30 years,but our understanding of the status and difficulties associated with the use of TIPS for portal hypertension is incomplete.This study aims to investigate the current situation of the use of TIPS for portal hypertension,which should aid the development of TIPS in China.Methods:This study involved a questionnaire survey,initiated by the China Portal Hypertension Alliance(CHESS)to investigate the use of TIPS for portal hypertension in China.Questionnaires were released on the Internet between August 16,2022 and September 16,2022.Detailed information was collected from 545 hospitals contracted by the CHESS in China,which performed TIPS surgery in 2021.Results:The 545 hospitals covers the majority of hospitals in China that conduct TIPS intervention.In the survey,a total of 13,527 TIPS interventions were carried out with 94.1%(498/545)of them had a system of routine follow‐up after TIPS.The majority(42.6%[232/545])believed that the main indication of TIPS was the control of acute bleeding.Overall,48.1%(262/545)carried out early or pre‐emptive TIPS,53.0%(289/545)carried out TIPS for cavernous transformation of the portal vein,and 81.0%(441/545)chose routine embolization of collateral circulation during operation.Most used coils(97.0%[475/490])and biological glue(72.0%[353/490])as embolic materials,and 78.5%(428/545)routinely performed intraoperative portal pressure gradient measurements.When selecting TIPS stents,57.1%(311/545)chose viator‐specific stents,and 57.2%(312/545)chose conventional anticoagulation after TIPS.Conventional anticoagulation after TIPS is over a time interval of 3–6 months(55.4%[173/312]).The limitation of TIPS surgery is mainly its cost(72.3%[394/545])and insufficient understanding of the procedure by doctors in other related departments(77.4%[422/545]).Most of the hospital teams accept the use of domestic instruments in TIPS surgery(92.7%[505/545]).Conclusions:This survey shows that TIPS is an essential part of the treatment of portal hypertension in China.TIPS is widely used in Chinese patients with portal hypertension,and this survey presents a more comprehensive picture of the options and difficulties encountered in the implementation of TIPS.In the future,it is necessary to promote TIPS technology and further standardize surgical indications,routine operations,and instrument application.Haibo Shao Xinwei Han Jianbo Zhao Junhui Sun Zhuting Fang Bin Xiong Xiaoli Zhu Weixin Ren Min Yuan Shiping Yu Meng Niu Weifu Lv Xueqiang Zhang Chunqing Zhang Lei Li Xuefeng Luo Yusheng Song Yilong Ma Tong Dang Hua Xiang Yun Jin Hui Xue Guiyun Jin Xiao Li Jiarui Li Shi Zhou Changlu Yu Song He Lei Yu Hongmei Zu Jun Ma Yanming Lei Ke Xu Xiaolong Qi 2023Portal Hypertension & Cirrhosis2023,2,1:0
17Advances in Mathematical Modeling of NFκB Signal Transduction in Mammals显示文摘NFκB is a basic transcription factor of cell signal transduction in mammals. Mathematical modeling can be used to simulate NFκB signal transduction and investigate the molecular mechanism. The initial IκB-NFκB signal transduction model emphasized the effects of negative feedback on NFκB temporal dynamics and gene expression control,while subsequent studies are concentrated on other feedback loops,input-output behaviors of modules,crosslinking of several NFκB activation paths and NFκB oscillation. Computer technology will continue to contribute to studies of NFκB signaling pathway. 18A Systematic Approach for Designing Analytical Dynamics and Servo Control of Constrained Mechanical Systems显示文摘A systematic approach for designing analytical dynamics and servo control of constrained mechanical systems is proposed. Fundamental equation of constrained mechanical systems is first obtained according to Udwadia-Kalaba approach which is applicable to holonomic and nonholonomic constrained systems no matter whether they satisfy the D’Alember’s principle.The performance specifications are modeled as servo constraints.Constraint-following servo control is used to realize the servo constraints. For this inverse dynamics control problem, the determination of control inputs is based on the Moore-Penrose generalized inverse to complete the specified motion. Secondorder constraints are used in the dynamics and servo control.Constraint violation suppression methods can be adopted to eliminate constraint drift in the numerical simulation. Furthermore,this proposed approach is applicable to not only fully actuated but also underactuated and redundantly actuated mechanical systems. 20miR-204 ameliorates osteoarthritis pain by inhibiting SP1-LRP1 signaling and blocking neuro-cartilage interaction显示文摘Osteoarthritis(OA)is a painful degenerative joint disease and is the leading cause of chronic disability among elderly individuals.To improve the quality of life for patients with OA,the primary goal for OA treatment is to relieve the pain.During OA progression,nerve ingrowth was observed in synovial tissue and articular cartilage.These abnormal neonatal nerves act as nociceptors to detect OA pain signals.The molecular mechanisms for transmitting OA pain in the joint tissues to the central nerve system(CNS)is currently unknown.MicroRNA miR-204 has been demonstrated to maintain the homeostasis of joint tissues and have chondro-protective effect on OA pathogenesis.However,the role of miR-204 in OA pain has not been determined.In this study,we investigated interactions between chondrocytes and neural cells and evaluated the effect and mechanism of miR-204 delivered by exosome in the treatment of OA pain in an experimental OA mouse model.Our findings demonstrated that miR-204 could protect OA pain by inhibition of SP1-LDL Receptor Related Protein 1(LRP1)signaling and blocking neuro-cartilage interaction in the joint.Our studies defined novel molecular targets for the treatment of OA pain.Ke Lu Qingyun Wang Liuzhi Hao Guizheng Wei Tingyu Wang William WLu Guozhi Xiao Liping Tong Xiaoli Zhao Di Chen 2023Bioactive Materials2023,,8:0
