2TaSCL14,a Novel Wheat(Triticum aestivum L.)GRAS Gene,Regulates Plant Growth,Photosynthesis,Tolerance to Photooxidative Stress,and Senescence显示文摘Rates of photosynthesis,tolerance to photooxidative stress,and senescence are all important physiological factors that affect plant development and thus agricultural productivity.GRAS proteins play essential roles in plant growth and development as well as in plant responses to biotic and abiotic stresses.So far few GRAS genes in wheat(Triticum aestivum L.)have been characterized.A previous transcriptome analysis indicated that the expression of a GRAS gene(TaSCL14) was induced by high-light stress in Xiaoyan 54(XY54),a common wheat cultivar with strong tolerance to high-light stress.In this study,TaSCL14 gene was isolated from XY54 and mapped on chromosome 4A.TaSCL14 was expressed in various wheat organs,with high levels in stems and roots.Our results confirmed that TaSCL14 expression was indeed responsive to high-light stress.Barley stripe mosaic virus(BSMV)-based virus-induced gene silencing(VIGS)of TaSCL14 in wheat was performed to help characterize its potential functions.Silencing of TaSCL14 resulted in inhibited plant growth,decreased photosynthetic capacity,and reduced tolerance to photooxidative stress.In addition,silencing of TaSCL14 in wheat promoted leaf senescence induced by darkness.These results suggest that TaSCL14 may act as a multifunctional regulator involved in plant growth,photosynthesis,tolerance to photooxidative stress,and senescence.Kunmei Chen Hongwei Li Yaofeng Chen Qi Zheng Bin Li Zhensheng Li 2015Journal of Genetics and Genomics2015,42,1:11

6Comparative study of estimation methods for genomic breeding values显示文摘Estimation of genomic breeding values is important in genomic selection. Bayesian and BLUP methods are the main techniques employed. In this study,we conducted a comparative study of Bayes A, Bayes B,Bayes Cp and GBLUP methods in simulated data and real data of Chinese Holstein cattle. Results showed that, in simulated data, the accuracies of all methods were all similarly elevated with the increase of reference population size, but they made different responses to the changes of marker number or QTL number. In real data of Chinese Holstein cattle, Bayes A generated the highest accuracy almost for all six traits, and GBLUP performed as well as Bayes A for the traits of milk yield, fat yield and protein yield, while for the trait of fat percentage, protein percentage and somatic cell score, three Bayesian methods showed superior to GBLUP. Comprehensively analyzing above results, it can be speculated that accuracies of the three Bayesian methods are not only influenced by the absolute value of QTL number or marker number, but may also be influenced by the ratio of QTL number to marker number. 8End End-to to-End Chinese Entity Recognition Based on BERT BERT-BiLSTM BiLSTM-ATT ATT-CRF显示文摘Traditional named entity recognition methods need professional domain knowl-edge and a large amount of human participation to extract features,as well as the Chinese named entity recognition method based on a neural network model,which brings the prob-lem that vector representation is too singular in the process of character vector representa-tion.To solve the above problem,we propose a Chinese named entity recognition method based on the BERT-BiLSTM-ATT-CRF model.Firstly,we use the bidirectional encoder representations from transformers(BERT)pre-training language model to obtain the se-mantic vector of the word according to the context information of the word;Secondly,the word vectors trained by BERT are input into the bidirectional long-term and short-term memory network embedded with attention mechanism(BiLSTM-ATT)to capture the most important semantic information in the sentence;Finally,the conditional random field(CRF)is used to learn the dependence between adjacent tags to obtain the global optimal sentence level tag sequence.The experimental results show that the proposed model achieves state-of-the-art performance on both Microsoft Research Asia(MSRA)corpus and people’s daily corpus,with F1 values of 94.77% and 95.97% respectively.LI Daiyi TU Yaofeng ZHOU Xiangsheng ZHANG Yangming MA Zongmin 2022ZTE Communications2022,20,S01:1

15Exploring the stage-specific roles of Tcf-1 in T cell development and malignancy at single-cell resolution显示文摘Tcf-1(encoded by Tcf7)not only plays critical roles in promoting T cell development and differentiation but also has been identified as a tumor suppressor involved in preventing T cell malignancy.However,the comprehensive mechanisms of Tcf-1 involved in T cell transformation remain poorly understood.In this study,Tcf7^(fl/fl)mice were crossed with Vav-cre,Lck-cre,or Cd4-cre mice to delete Tcf-1 conditionally at the beginning of the HSC,DN2-DN3,or DP stage,respectively.The defective T cell development phenotypes became gradually less severe as the deletion stage became more advanced in distinct mouse models.Interestingly,consistent with Tcf7^(-/-)mice,Tcf^(fl/fl)Vav-cre mice developed aggressive T cell lymphoma within 45 weeks,but no tumors were generated in Tcf7^(fl/fl)Lck-cre or Tcf^(fl/fl)CdA-cre mice.Single-cell RNA-seq(ScRNA-seq)indicated that ablation of Tcf-1 at distinct phases can subdivide DN1 cells into three clusters(C1,C2,and C3)and DN2-DN3 cells into three clusters(C4,C5,and C6).Moreover,Tcf-1 deficiency redirects bifurcation among divergent cell fates,and clusters C1 and C4 exhibit high potential for leukemic transformation.Mechanistically,we found that Tcf-1 directly binds and mediates chromatin accessibility for both typical T cell regulators and proto-oncogenes#including Myb,Mycn,Runxl,and Lyl1 in the DN1 phase and Lefb ld2,Dtxb Fyn,Bclllb,and Zfp36l2 in the DN2-DN3 phase.The aberrant expression of these genes due to Tcf-1 deficiency in very early T cells contributes to subsequent tumorigenesis.Thus,we demonstrated that Tcf-1 plays stage-specific roles in regulating early thymocyte development and transformation,providing new insights and evidence for clinical trials on T-ALL leukemia.Fang Wang Zhihong Qi Yingpengao Guotao Yu Tao Feng Tianyan Zhao Hai-Hui Xue Yaofeng Zhao Peng Jiang Li Bao Shuyang Yu 2021Cellular & Molecular Immunology2021,18,3:0
16Genome‑wide DNA methylation and transcriptome analyses reveal the key gene for wool type variation in sheep显示文摘Background Wool fibers are valuable materials for textile industry.Typical wool fibers are divided into medullated and non-medullated types,with the former generated from primary wool follicles and the latter by either primary or secondary wool follicles.The medullated wool is a common wool type in the ancestors of fine wool sheep before breeding.The fine wool sheep have a non-medullated coat.However,the critical period determining the type of wool follicles is the embryonic stage,which limits the phenotypic observation and variant contrast,making both selection and studies of wool type variation fairly difficult.Results During the breeding of a modern fine(MF)wool sheep population with multiple-ovulation and embryo transfer technique,we serendipitously discovered lambs with ancestral-like coarse(ALC)wool.Whole-genome rese-quencing confirmed ALC wool lambs as a variant type from the MF wool population.We mapped the significantly associated methylation locus on chromosome 4 by using whole genome bisulfite sequencing signals,and in turn identified the SOSTDC1 gene as exons hypermethylated in ALC wool lambs compare to their half/full sibling MF wool lambs.Transcriptome sequencing found that SOSTDC1 was expressed dozens of times more in ALC wool lamb skin than that of MF and was at the top of all differentially expressed genes.An analogy with the transcriptome of coarse/fine wool breeds revealed that differentially expressed genes and enriched pathways at postnatal lamb stage in ALC/MF were highly similar to those at the embryonic stage in the former.Further experiments validated that the SOSTDC1 gene was specifically highly expressed in the nucleus of the dermal papilla of primary wool follicles.Conclusion In this study,we conducted genome-wide differential methylation site association analysis on differen-tial wool type trait,and located the only CpG locus that strongly associated with primary wool follicle development.Combined with transcriptome analysis,SOSTDC1 was identified as the only gene at this locus that was specifically overexpressed in the primary wool follicle stem cells of ALC wool lamb skin.The discovery of this key gene and its epigenetic regulation contributes to understanding the domestication and breeding of fine wool sheep.Jiankui Wang Guoying Hua Ganxian Cai Yuhao Ma Xue Yang Letian Zhang Rui Li Jianbin Liu Qing Ma Keliang Wu Yaofeng Zhao Xuemei Deng 2023Journal of Animal Science and Biotechnology2023,14,6:0
17Immunoglobulin genes and diversity: what we have learned from domestic animals显示文摘This review focuses on the diversity of immunoglobulin (Ig) genes and Ig isotypes that are expressed in domestic animals. Four livestock species-cattle, sheep, pigs, and horses-express a full range of Ig heavy chains (IgHs), including μ, δ, γ, ε, and α. Two poultry species (chickens and ducks) express three IgH isotypes, μ, υ, and α, but not δ. The κ and λ light chains are both utilized in the four livestock species, but only the λ chain is expressed in poultry. 20Expression of recombinant human butyrylcholinesterase in the milk of transgenic mice显示文摘Butyrylcholinesterase(BCHE)is a natural bioscavenger that protects humans against organophosphate toxicity.Due to the limited yield of human BCHE(hBCHE)when purifying from human plasma,it is necessary to find an alternative method to produce this protein.One potential method is to produce transgenic livestock that make modified milk containing high concentration of hBCHE.In this study,we cloned the hBCHEgene into a human lactoferrin(hLF)bacterial artificial chromosome(BAC)construct to make a hLFhBCHE BAC construct.Subsequently,we injected the BAC construct into pronuclei of mouse fertilized embryos and generated transgenic mice.Expression analysis showed that recombinant hBCHE(rhBCHE)was expressed efficiently in the mammary gland of the transgenic mice and the concentration of rhBCHE in the milk of individual mice ranged from 7612 to 15928 mg·L^(–1).Protein function tests showed that rhBCHE has the same enzymatic activity as the native hBCHE.Our results pave the way for making transgenic livestock to produce large quantities of rhBCHE.Dan LU Shengzhe SHANG Shen LIU Ying WU Fangfang WU Tan TAN Qiuyan LI Yunping DAI Xiaoxiang HU Yaofeng ZHAO Ning LI 2014Frontiers of Agricultural Science and Engineering2014,1,3:0
