Immunogenicity analysis following human immunodeficiency virus recombinant DNA and recombinant vaccinia virus Tian Tan prime-boost immunization显示文摘This study assessed and compared the immunogenicity of various immunization strategies in mice using combinations of recombinant DNA(pCCMp24) and recombinant attenuated vaccinia virus Tian Tan(rddVTT-CCMp24).Intramuscular immunization was performed on days 0(prime) and 21(boost).The immunogenicity of the vaccine schedules was determined by measuring human immunodeficiency virus(HIV)-specific binding antibody levels and cytokine(interleukin-2 and interleukin-4) concentrations in peripheral blood, analyzing lymphocyte proliferation capacity against HIV epitopes and CD4 + /CD8 + cell ratio, and monitoring interferon-gamma levels at different times post-immunization.The results showed that pCCMp24,rddVTT-CCMp24 and their prime-boost immunization induced humoral and cellular immune responses. The pCCMp24/rddVTT-CCMp24 immunization strategy increased CD8 + T cells and induced more IFN-γ-secreting cells compared with single-shot rDNA.The prime-boost immunization strategy also induced the generation of cellular immunological memory to HIV epitope peptides. These results demonstrated that prime-boost immunization with rDNA and rddVTT-CCMp24 had a tendency to induce greater cellular immune response than single-shot vaccinations, especially IFN-γ response, providing a basis for further studies. | LIU CunXia DU ShouWen LI Chang WANG YuHang WANG MaoPeng LI Yi YIN RongLan LI Xiao REN DaYong QIN YanQing REN JingQiang JIN NingYi | 2013 | Science China(Life Sciences)2013,56,6: | 7
Concrete durability under different circumstances based on multi-factor effects显示文摘Concrete durability has become a hot research field in civil engineering. Concrete structures suffer salt-erosion damage to different degrees in the semi-arid region of North China. The environmental condition is one of the important factors affecting the durability of concrete constructions. To realize fully the interaction between various environmental factors, this paper researched concrete durability in the salt environment under combined actions(immersing, freeze–thaw cycles, and wet–dry cycles). According to the laboratory test data, the concrete-durability degradation law under coupling-effect factors was investigated. The results show that concrete's compressive strength decreases with the increase of salt concentration and immersion time. No matter what the environmental conditions were, the compressive strength-loss ratio increased with the test time. The compressive strength-test results indicate that sodium sulfate has the strongest corrosive effect on concrete durability, followed by calcium chloride, with sodium chloride having the weakest corrosion. Compared with the other two environmental factors, the wet–dry cycle is the key factor affecting concrete durability. Therefore, in engineering practice, the influence of environment conditions on the strength and durability of concrete should be taken into full consideration, especially in the wet–dry environment with salt conditions. | Feng Ming ChengCheng Du YuHang Liu XiangYang Shi DongQing Li | 2017 | Research in Cold and Arid Regions2017,9,4: | 6
Trace Amounts of Triple-Functional Additives Enable Reversible Aqueous Zinc-Ion Batteries from a Comprehensive Perspective显示文摘Although their cost-effectiveness and intrinsic safety, aqueous zinc-ion batteries suffer from notorious side reactions including hydrogen evolution reaction,Zn corrosion and passivation, and Zn dendrite formation on the anode.Despite numerous strategies to alleviate these side reactions have been demonstrated, they can only provide limited performance improvement from a single aspect.Herein,a triple-functional additive with trace amounts, ammonium hydroxide, was demonstrated to comprehensively protect zinc anodes. The results show that the shift of electrolyte pH from 4.1 to 5.2 lowers the HER potential and encourages the in situ formation of a uniform ZHS-based solid electrolyte interphase on Zn anodes. Moreover, cationic NH^(4+)can preferentially adsorb on the Zn anode surface to shield the“tip effect”and homogenize the electric field.Benefitting from this comprehensive protection,dendrite-free Zn deposition and highly reversible Zn plating/stripping behaviors were realized.Besides, improved electrochemical performances can also be achieved in Zn//MnO_(2)full cells by taking the advantages of this triple-functional additive. This work provides a new strategy for stabilizing Zn anodes from a comprehensive perspective. | Ruwei Chen Wei Zhang Quanbo Huang Chaohong Guan Wei Zong Yuhang Dai Zijuan Du Zhenyu Zhang Jianwei Li Fei Guo Xuan Gao Haobo Dong Jiexin Zhu Xiaohui Wang Guanjie He | 2023 | Nano-Micro Letters2023,15,6: | 2
Large extracellular vesicles secreted by human iPSC-derived MSCs ameliorate tendinopathy via regulating macrophage heterogeneity显示文摘Tendinopathy is a common musculoskeletal disorder which results in chronic pain and reduced performance. The therapeutic effect of stem cell derived-small extracellular vesicles(sEVs)for tendinopathy has been validated in recent years. However, whether large extracellular vesicles(lEVs),another subset of extracellular vesicles, possesses the ability for the improvement of tendinopathy remains unknown. Here,we showed that lEVs secreted from iPSC-derived MSCs(iMSC-lEVs)significantly mitigated pain derived from tendinopathy in rats.Immuno-histochemical analysis showed that iMSC-lEVs regulated the heterogeneity of infiltrated macrophages and several inflammatory cytokines in rat tendon tissue.Meanwhile,in vitro experiments revealed that the M1 pro-inflammatory macrophages were repolarized towards M2 anti-inflammatory macrophages by iMSC-lEVs,and this effect was mediated by regulating p38 MAPK pathway. Moreover, liquid chromatography-tandem mass spectrometry analysis identified 2208 proteins encapsulated in iMSC-lEVs,including 134 new-found proteins beyond current Vesiclepedia database.By bioinformatics and Western blot analyses,we showed that DUSP2 and DUSP3,the negative regulator of p38 phosphorylation, were enriched in iMSC-lEVs and could be transported to macrophages.Further, the immunomodulatory effect of iMSC-lEVs on macrophages was validated in explant tendon tissue from tendinopathy patients.Taken together, our results demonstrate that iMSC-lEVs could reduce inflammation in tendinopathy by regulating macrophage heterogeneity, which is mediated via the p38 MAPK pathway by delivery of DUSP2 and DUSP3,and might be a promising candidate for tendinopathy therapy. | Teng Ye Zhengsheng Chen Jieyuan Zhang Lei Luo Renzhi Gao Liangzhi Gong Yuhang Du Zongping Xie Bizeng Zhao Qing Li Yang Wang | 2023 | Bioactive Materials2023,,3: | 2

Stability Mechanisms of Gas Bubbles Subject to Ultrasonic Field during the Rectified Diffusion in Liquids显示文摘The spherical oscillation of a gas bubble in liquids is important to growth in liquids during rectified diffusions( e. g., the onset of the sonoluminescence and the enhancement of sonochemical reactions). The present paper numerically shows stability maps( divided into four zones),in which gas bubbles maintain the linearly spherical oscillation without nonlinear disturbance of rectified diffusions within a large range of bubble radius. The critical pressures of spherical and diffusional oscillations are two decisive indexes determining the stability status. Specifically, the stability boundaries and influential factors( including acoustic parameters and gas concentration in liquids) were discussed and analyzed. The results show that the variations of gas concentration and acoustic parameters dramatically changed the stable status of the gas bubbles. The gas bubble maintained stable status when external parameters and gas concentration were set between the two critical values properly. The cases of high-frequency and low-frequency limits were also introduced at the end of the whole paper. | Yuhang Gao Xiaoze Du | 2019 | Journal of Harbin Institute of Technology(New Series)2019,26,1: | 1
Present and future of functionalized Cu current collectors for stabilizing lithium metal anodes显示文摘Li metal has been recognized as the most promising anode materials for next-generation high-energy-density batteries, however, the inherent issues of dendrite growth and huge volume fluctuations upon Li plating/stripping normally result in fast capacity fading and safety concerns.Functionalized Cu current collectors have so far exhibited significant regulatory effects on stabilizing Li metal anodes(LMAs),and hold a great practical potential owing to their easy fabrication,low-cost and good compatibility with the existing battery technology. In this review,a comprehensive overview of Cu-based current collectors, including planar modified Cu foil,3D architectured Cu foil and nanostructured 3D Cu substrates, for Li metal batteries is provided.Particularly, the design principles and strategies of functionalized Cu current collectors associated with their functionalities in optimizing Li plating/stripping behaviors are discussed. Finally, the critical issues where there is incomplete understanding and the future research directions of Cu current collectors in practical LMAs are also prospected. This review may shed light on the critical understanding of current collector engineering for high-energy-density Li metal batteries. | Yuhang Liu Yifan Li Jinmeng Sun Zhuzhu Du Xiaoqi Hu Jingxuan Bi Chuntai Liu Wei Ai Qingyu Yan | 2023 | Nano Research Energy2023,2,1: | 1
Wave propagation characteristics in frozen saturated soil显示文摘Ultrasonic detection technology is of great significance in the detection and evaluation of physical and mechanical properties of frozen soil, but wave propagation characteristics in frozen soil are unclear. Based on the three-phase composition of frozen saturated soil and the mixture theory, considering Bishop's effective stress formula, the wave propagation equations are establish for frozen saturated soil. In wave propagation, an entropy inequality was introduced to describe the coupling of different phases. The analytic expressions of propagation velocity and attenuation law of waves in frozen soil are obtained, and wave propagation characteristics in frozen saturated soil are discussed. Results show that four types of waves(i.e., P1, P2, P3 and S) are found in frozen saturated soil and all four wave types are dissipative waves, in which the attenuation of P3 is the maximum. The velocity of four waves increases sharply at the excitation frequency range of 10~3–10~9 Hz,but the wave velocity at high-frequency and low-frequency is almost constant. When volume ice content increases, the wave propagation velocity of P1 and S decreases dramatically, and the velocity of P2 increases gradually, but P3 velocity increases first and then decreases to zero with increasing saturation. The attenuation coefficients of P1 and S waves begins to increase gradually when the volume ice content is about 0.4, P2 increases first and then decreases with an increase of volume ice content and P3 increases with the volume ice content and decreases rapidly from extreme to zero. | ChengCheng Du DongQing Li Feng Ming YuHang Liu XiangYang Shi | 2018 | Research in Cold and Arid Regions2018,10,2: | 0
A consecutive joint shear strength model considering the 3D roughness of real contact joint surface显示文摘A consecutive joint shear strength model for soft rock joints is proposed in this paper, which takes into account the degradation law of the actual contact three-dimensional(3D)roughness.The essence of the degradation of the maximum possible dilation angle is the degradation of the 3D average equivalent dip angle of the actual contact joint asperities.Firstly, models for calculating the maximum possible dilation angle at the initial and residual shear stress stages are proposed by analyzing the 3D joint morphology characteristics of the corresponding shear stages.Secondly, the variation law of the maximum possible dilation angle is quantified by studying the degradation law of the joint micro convex body.Based on the variation law of the maximum possible dilation angle, the maximum possible shear strength model is proposed.Furthermore,a method to calculate the shear stiffness degradation in the plastic stage is proposed.According to the maximum possible shear strength of rock joints, the shear stress-shear displacement prediction model of rock joints is obtained. The new model reveals that there is a close relationship between joint shear strength and actual contact joint roughness, and the degradation of shear strength after the peak is due to the degradation of actual contact joint roughness. | Liren Ban Zhigang Tao Weisheng Du Yuhang Hou | 2023 | International Journal of Mining Science and Technology2023,33,5: | 0
Radon CLF:A Novel Approach for Skew Detection Using Radon Transform显示文摘In the digital world,a wide range of handwritten and printed documents should be converted to digital format using a variety of tools, including mobile phones and scanners.Unfortunately, this is not an optimal procedure, and the entire document image might be degraded.Imperfect conversion effects due to noise, motion blur, and skew distortion can lead to significant impact on the accuracy and effectiveness of document image segmentation and analysis in Optical Character Recognition(OCR)systems. In Document Image Analysis Systems(DIAS),skew estimation of images is a crucial step.In this paper,a novel, fast, and reliable skew detection algorithm based on the Radon Transform and Curve Length Fitness Function(CLF),so-called Radon CLF,was proposed. The Radon CLF model aims to take advantage of the properties of Radon spaces. The Radon CLF explores the dominating angle more effectively for a 1D signal than it does for a 2D input image due to an innovative fitness function formulation for a projected signal of the Radon space.Several significant performance indicators, including Mean Square Error(MSE),Mean Absolute Error(MAE),Peak Signal-to-Noise Ratio(PSNR),Structural Similarity Measure(SSIM),Accuracy, and run-time,were taken into consideration when assessing the performance of our model. In addition,a new dataset named DSI5000 was constructed to assess the accuracy of the CLF model. Both two-dimensional image signal and the Radon space have been used in our simulations to compare the noise effect.Obtained results show that the proposed method is more effective than other approaches already in use, with an accuracy of roughly 99.87%and a run-time of 0.048(s).The introduced model is far more accurate and timeefficient than current approaches in detecting image skew. | Yuhang Chen Mahdi Bahaghighat Aghil Esmaeili Kelishomi Jingyi Du | 2023 | Computer Systems Science & Engineering2023,47,10: | 0
Reliability and validity of the hidden hunger assessment scale in China-revised for high school students显示文摘Objective:Hidden hunger remains a severe public health problem that affects millions of people worldwide. In China, challenges related to dietary imbalance and hidden hunger persist.Micronutrient inadequacy deserves more attention among adolescents, given its vital role in their growth and development;however, this problem appears to have been largely ignored.High school students,in particular, are often at a high risk of hidden hunger but have limited assessment tools available. Therefore, this study aims to revise the hidden hunger assessment scale for high school students(HHAS-HSS)in China and assess its reliability and validity. Methods:Based on a literature review, expert consultation,pre-experiment,and formal survey,a hidden hunger assessment scale was revised for high school students. The formal survey involved 9336 high school students in 11 of the 16 cities in Anhui Province,China, and 9038 valid questionnaires were collected and included in the analysis. The item analysis, internal consistency reliability,test-retest reliability, content validity, exploratory factor analysis, and confirmatory factor analysis of the HHAS-HSS were examined. Results:The HHAS-HSS included a total of 4 dimensions and 12 items:'vegetables and food diversity'(three items),'fruits and dairy products'(three items),'micronutrient-dense foods'(four items),and'health condition and eating habits'(two items). The results showed a Cronbach's alpha of 0.758,a split-half reliability of 0.829,and a test-retest reliability of o.793,indicating good internal consistency.Using the Bartlett's test and Kaiser-Meyer-Olkin test(KMO)to test the exploratory factor analysis presented a four-factor model of the HHAS-HSS,the KMO0 value was 0.820(P<0.001),which indicated the possibility for factor confirmatory factor analysis.Using the maximum variance rotation method, four factors were obtained, and the cumulative variance explained rate was 57.974%.Confirmatory factor analysis also supported the division of the scale into four dimensions, and the fitting indices were x^(2)=1417.656,x^(2)/df=29.534,goodness-of-fit index=0.974,adjusted goodnessof-fit index=0.958,parsimonious goodness-of-fit index=0.600,normed fit index=0.938,incremental fit index=0.940,Tucker-Lewis index=0.917,comparative fit index=0.939,and root mean square error of approximation=0.056.Except for x^(2)/df,all the indices reached the fitting standard, and the above results showed that the construct validity of the scale reached an acceptable level. Conclusions:The HHAS-HSS has good validity and reliability for Chinese high school students. It is a convenient self-report measure of hidden hunger risk. | Ning Zhang Minao Wang Yuchen Zhang Huake Cao Yang Yang Yuhang Shi Yang Pei Feixiang Yang Yinan Du | 2023 | Global Health Journal2023,7,2: | 0
Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector. Some of JUNO's features make it an excellent location for^8B solar neutrino measurements, such as its low-energy threshold, high energy resolution compared with water Cherenkov detectors, and much larger target mass compared with previous liquid scintillator detectors. In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable, assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition, approximately 60,000 signal and 30,000 background events are expected. This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter, which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle. Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better, depending on the central value, and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 13 | GATA2^(−/−) human ESCs undergo attenuatedendothelial to hematopoietic transition andthereafter granulocyte commitment显示文摘Background: Hematopoiesis is a progressive process collectively controlled by an elaborate network of transcriptionfactors (TFs). Among these TFs, GATA2 has been implicated to be critical for regulating multiple steps of hematopoiesisin mouse models. However, whether similar function of GATA2 is conserved in human hematopoiesis, especially duringearly embryonic development stage, is largely unknown. Results: To examine the role of GATA2 in human background, we generated homozygous GATA2 knockout humanembryonic stem cells (GATA2^(−/−) hESCs) and analyzed their blood differentiation potential. Our results demonstratedthat GATA2^(−/−) hESCs displayed attenuated generation of CD34^(+)CD43^(+) hematopoietic progenitor cells (HPCs), due tothe impairment of endothelial to hematopoietic transition (EHT). Interestingly, GATA2^(−/−) hESCs retained the potentialto generate erythroblasts and macrophages, but never granulocytes. We further identified that SPI1 downregulationwas partially responsible for the defects of GATA2^(−/−) hESCs in generation of CD34^(+)CD43^(+) HPCs and granulocytes.Furthermore, we found that GATA2^(−/−) hESCs restored the granulocyte potential in the presence of Notch signaling. Conclusion: Our findings revealed the essential roles of GATA2 in EHT and granulocyte development throughregulating SPI1, and uncovered a role of Notch signaling in granulocyte generation during hematopoiesis modeled byhuman ESCs. | Ke Huang Juan Du Ning Ma Jiajun Liu Pengfei Wu Xiaoya Dong Minghui Meng Wenqian Wang Xin Chen Xi Shi Qianyu Chen Zhongzhou Yang Shubin Chen Jian Zhang Yuhang Li Wei Li Yi Zheng Jinglei Cai Peng Li Xiaofang Sun Jinyong Wang Duanqing Pei Guangjin Pan | 2015 | Cell Regeneration2015,4,1: | 0
Porous polyimide-based phase change materials with enhanced mechanical properties, thermal conductivity, and thermal shock resistance through precisely designed triple-functional layers显示文摘Numerous scenarios of direct contact between electronic components and skin appear in wearable electronic devices. As the “second skin” that lies next to the biological skin of the human body, flexible wearable devices need to be equipped with thermal protection. However, the use of flexible phase change materials(PCMs) for wearable devices remains a challenge due to their low thermal conductivity, weakened mechanical strength, and liquid leakage. Herein, we developed a multilayered polyimide(PI) composite film integrating stable latent heat absorption, high thermal conductivity, and enhanced mechanical strength. This single piece of material achieved more prominent hotspot protection than traditional foams. The in-plane thermal conductivity of the resultant substrate layer is up to 2.655 W/(m K), which provides a fast response and in-plane dissipation for heat flow. The deliberately arranged interlayer of the material significantly improved the tensile strength(37.6 MPa) of the composite film, representing 128.9% greater strength than that of a bilayer film without a dense layer. The top layer with abundant pores provides reversibly latent heat storing and releasing function after being well infiltrated with paraffin wax. The resulting synergistic effect between three functional layers shows superb heat-suppressing performance in the thickness direction in both thermal shockresistant tests and simulation results. Impressively, the maximum temperature drop reached 40.2℃ compared with the pure PI film. The heating time was delayed by 12 s, providing sufficient warning time for human emergency response. This strategy can potentially pave the way for the design and fabrication of multifunctional films for wearable electronics in thermal protection applications. | JIANG Miao XIAO Chao DING Xin WANG YanYan ZHANG Xian DU YuHang LI XiaoFei ZHENG Kang LIU XiangLan CHEN Lin TIAN XingYou | 2023 | Science China(Technological Sciences)2023,66,9: | 0
Advances in the Emerging Gradient Designs of Li Metal Hosts显示文摘Developing host has been recognized a potential countermeasure to circumvent the intrinsic drawbacks of Li metal anode(LMA),such as uncontrolled dendrite growth, unstable solid electrolyte interface, and infinite volume fluctuations. To realize proper Li accommodation, particularly bottom-up deposition of Li metal, gradient designs of host materials including lithiophilicity and/or conductivity have attracted a great deal of attention in recent years. However,a critical and specialized review on this quickly evolving topic is still absent. In this review,we attempt to comprehensively summarize and update the related advances in guiding Li nucleation and deposition.First, the fundamentals regarding Li deposition are discussed, with particular attention to the gradient design principles of host materials.Correspondingly, the progress of creating diferent gradients in terms of lithiophilicity, conductivity, and their hybrid is systematically reviewed. Finally, future challenges and perspective on the gradient design of advanced hosts towards practical LMAs are provided, which would provide a useful guidance for future studies. | Wanqing Guan Xiaoqi Hu Yuhang Liu Jinmeng Sun Chen He Zhuzhu Du Jingxuan Bi Ke Wang Wei Ai | 2022 | Research2022,,4: | 0
GmTOC1b negatively regulates resistance to Soybean mosaic virus显示文摘Soybean(Glycine max)is a major oil and feed crop worldwide.Soybean mosaic virus(SMV)is a globally occurring disease that severely reduces the yield and quality of soybean. Here,we characterized the role of the clock gene TIMING OF CAB EXPRESSION 1b(GmTOC1b)in the resistance of soybean to SMV.Homozygous Gmtoc1b mutants exhibited increased tolerance to SMV strain SC3 due to the activation of programmed cell death triggered by a hypersensitive response.Transcriptome deep sequencing and RT-qPCR analysis suggested that GmTOC1b likely regulates the expression of target genes involved in the salicylic acid(SA)signaling pathway.GmTOC1b binds to the promoter of GmWRKY40,which encodes a protein that activates the expression of SA-mediated defense-related genes. Moreover,we revealed that the GmTOC1bH1 haplotype, which confers increased tolerance to SMV,was artificially selected in improved cultivars from the Northern and Huang-Huai regions of China. Our results therefore identify a previously unknown SMV resistance component that could be deployed in the molecular breeding of soybean to enhance SMV resistance. | Yuhang Zhang Haiping Du Tiantian Zhao Chunmei Liao Tu Feng Jun Qin Baohui Liu Fanjiang Kong Zhijun Che Liyu Chen | 2023 | The Crop Journal2023,11,6: | 0
Design and Implementation of a Network Performance Analysis System for Space Network Emulation Platforms显示文摘Emulation platforms are critical for evaluation and verification in the research of networking technologies and protocols for space networks(SN).High fidelity emulating technologies have been extensively studied for SN in earlier work, while little emphasis has been placed on the performance evaluation part.In this paper, the design of a network performance analysis architecture is presented, with which high-speed network traffic can be captured and indexed, and the performance of the emulated SN can be well analyzed and evaluated. This architecture comprises three components, namely capture layer, storage layer and analysis layer.Analytic Hierarchy Process(AHP)and several analysis methods are adopted to evaluate the network performance comprehensively. In the implementation of the proposed architecture, configuration optimization and parallel processing are applied to handle large amount of high-speed network traffic. Finally, experiment results through the analysis system exhibits the effectiveness of the proposed architecture. | Dongxu Hou Fa Chen Chengpeng Kuang Yuhang Du Qi Zhang Kanglian Zhao Wenfeng Li Sidan Du Yuan Fang | 2022 | China Communications2022,19,9: | 0

Transmembrane domain of IFITM3 is responsible for its interaction with influenza virus HA_(2) subunit显示文摘Interferon-inducible transmembrane protein 3(IFITM3)inhibits influenza virus infection by blocking viral membrane fusion, but the exact mechanism remains elusive. Here,we investigated the function and key region of IFITM3 in blocking influenza virus entry mediated by hemagglutinin(HA).The restriction of IFITM3 on HAmediated viral entry was confirmed by pseudovirus harboring HA protein from H5 and H7 influenza viruses.Subcellular co-localization and immunocoprecipitation analyses revealed that IFITM3 partially co-located with the full-length HA protein and could directly interact with HA_(2) subunit but not HA_(1) subunit of H5 and H7 virus.Truncated analyses showed that the transmembrane domain of the IFITM3 and HA_(2) subunit might play an important role in their interaction. Finally, this interaction of IFITM3 was also verified with HA_(2) subunits from other subtypes of influenza A virus and influenza B virus.Overall, our data demonstrate for the first time a direct interaction between IFITM3 and influenza HA protein via the transmembrane domain, providing a new perspective for further exploring the biological significance of IFITM3 restriction on influenza virus infection or HA-mediated antagonism or escape. | Wang Xu Yuhang Wang Letian Li Xiaoyun Qu Quan Liu Tiyuan Li Shipin Wu Ming Liao Ningyi Jin Shouwen Du Chang Li | 2022 | Virologica Sinica2022,37,5: | 0
Investigation on the physical mechanism of cavity percentage dependent shear strength for rock joints considering the real contact joint surface显示文摘To explain the effect of joint roughness on joint peak shear strength(JPSS)and investigate the effect of different contact states of joint surface on JPSS,we try to clarify the physical mechanism of the effect of joint cavity percentage(JCP)on JPSS from the perspective of the three-dimensional(3D)distribution characteristics of the actual contact joint surface, and propose a JPSS model considering the JCP.Shear tests for red sandstone joints with three different surface morphologies and three different JCPs were performed under constant normal load condition.Based on test fitting results, the reduction effect of the JCP on JPSS is investigated, and a JPSS model for cavity-containing joints is obtained. However, the above model only considers the influence of JCP by fitting test data, and does not reveal the physical mechanism of JCP affecting the JPSS.Based on the peak dilation angle model for consideration of the actual contact joint morphology, and the influence of JCP on the roughness of the actual contact joint surface,a theoretical model of the JPSS considering the JCP is proposed. The derivation process does not depend on the test fitting, but is entirely based on the joint mechanical law,and its physical significance is clear. It is proposed that the essence of the influence of the JCP on JPSS is that the JCP first affects the normal stress of the actual contact joints, further affects the roughness of actual contact joints, and then affects the shear strength. | Liren Ban Zefan Wang Weisheng Du Yuhang Hou Chengzhi Qi Jin Yu | 2023 | Rock Mechanics Bulletin2023,2,4: | 0
