1Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. 3Novel nanometer scaffolds regulate the biological behaviors of neural stem cells显示文摘Ideal tissue-engineered scaffold materials regulate proliferation, apoptosis and differentiation of cells seeded on them by regulating gene expression. In this study, aligned and randomly oriented collagen nanofiber scaffolds were prepared using electronic spinning technology. Their diameters and appearance reached the standards of tissue-engineered nanometer scaffolds. The nanofiber scaffolds were characterized by a high swelling ratio, high porosity and good mechanical properties. The proliferation of spinal cord-derived neural stem cells on novel nanofiber scaffolds was obviously enhanced. The proportions of cells in the S and G 2 /M phases noticeably increased. Moreover, the proliferation rate of neural stem cells on the aligned collagen nanofiber scaffolds was high. The expression levels of cyclin D1 and cyclin-dependent kinase 2 were increased. Bcl-2 expression was significantly increased, but Bax and caspase-3 gene expressions were obviously decreased. There was no significant difference in the differentiation of neural stem cells into neurons on aligned and randomly oriented collagen nanofiber scaffolds. These results indicate that novel nanofiber scaffolds could promote the proliferation of spinal cord-derived neural stem cells and inhibit apoptosis without inducing differentiation. 4A synthetical methodology for identifying pr ior ity pollutants in reclaimed water based on meta-analysis显示文摘Wastewater reclamation and reuse is an increasing global project,while the reclamation treatment on wastewater does not completely remove all pollutants in water.The residual pollutants in reclaimed water would cause potential risk on human health and ecosystem safety during the long-term use.It is impossible to analyze and control all pollutants one by one in practice,therefore,identification and control of priority pollutants will be efficient strategy to ensure the safe use of reclaimed water.An integrated three-step methodology for identifying priority pollutants in reclaimed water was proposed in this study.First,a comprehensive literature survey on the occurrence of pollutants in reclaimed water was conducted,and a dataset D PR for pollutants occurrence in reclaimed water was established,containing 1,113 pollutants.Second,611 chemicals that had been recommended as hazardous pollutants for various water bodies in previous literatures were summarized,and a dataset D HP for hazardous pollutants in water was obtained.Third,meta-analysis on these two datasets(D PR and D HP)was performed,a new dataset D HPR for hazardous pollutants in reclaimed water was established,including 265 candidates.Finally,59 substances out of dataset D HPR were identified as priority pollutants for reclaimed water based on their recommendation frequency.It is expected that this synthetical methodology will provide powerful support for scientific evaluating and managing water pollution and ensuring safe use of reclaimed water.Feipeng Wang Dongbin Wei Miao Chen Shuang Peng Qiaorong Guo Xinyi Zhang Jun Liu Yuguo Du 2022Journal of Environmental Sciences2022,34,2:2

6The Investigation on Single Event Function Failure for DC/DC Converters with Three Single Terminal Topological Structures显示文摘Heavy ion radiation experiments have been done to DC/DC converters with different topological structures for space applications. The test results were analyzed about the function failure of three topological structures caused by single event effects. The relationship between the function failure and the input supply voltage, the output load current and the topological structure of the module were discussed. Based on the analysis of the variation relationship among the source/drain terminal voltage of MOSFETs and the input voltage and the output load, the sensitivity factors associated with the function failure caused by single event effects were discussed. 7Late Mesozoic tectonostratigraphic division and correlation of the Bohai Bay basin: Implications for the Yanshanian Orogeny显示文摘We present the results of Mesozoic sequences of the Bohai Bay basin in North China, based mainly on geochronology and interpretations of seismic profiles and logging data. Five tectono-stratigraphic sequences are defined: Lower-Middle Triassic, Lower-Middle Jurassic, Upper Jurassic, Lower Cretaceous and Upper Cretaceous. Based on an analysis of detrital zircons, the clastic rocks recorded two intervals of Jurassic magmatic activity, during 180–175 and 160–152 Ma, which can be correlated to the Nandaling and Tiaojishan Formations in the Yanshan area, respectively. However, since Jurassic volcanic rocks have not yet been found in the Bohai Bay basin, we speculate that these two stages of activity were mainly concentrated around the periphery of the North China Craton(NCC) rather than within. Based on an analysis of zircons from volcanic rocks and pyroclastics, the early Cretaceous magmatism in the Bohai Bay basin can be divided into two stages, 125–120 and 110–100 Ma,which can be correlated to magmatism in the eastern part of the NCC. The zircon ages indicate an absence of volcanic activity during the late Tuchengzi and Zhangjiakou periods which may correlate to the uplift of the Bohai Bay basin in the late Late Jurassic. Comparison of the development of Mesozoic basins and sedimentary strata in the central-eastern part of Yanshan tectonic belt and the the Bohai Bay basin indicates that the two areas are generally comparable, but with substantial differences.The central-eastern part of the Yanshan structural belt lacks Early-Middle Triassic strata, and the Bohai Bay basin lacks late Jurassic-early Cretaceous strata. Based on research results from late Mesozoic sedimentary structures in the central and eastern parts of the Yanshan tectonic belt, we infer that episode A of the Yanshanian Orogeny was weak in the Bohai Bay basin and its periphery, while episode B of the Yanshanian Orogeny had a strong influence on the Bohai Bay basin and its periphery. 8Microstructure Features with Ultrafined and Purified Grains for Si-Mn Hot-rolling TRIP Steel显示文摘The advanced hot-rolled transformation induced plasticity (TRIP) steel with high tensile strength of 775 MPa,high elongation of 29% and low ductile-brittle transmission temperature less than -80°C has been developed inlaboratory based on the common silicon and manganese chemical composition. 9A micro-kinetic model of enhanced foam stability under artificial seismic wave显示文摘To get a deeper understanding on the synergistic enhancement effect of low frequency artificial seismic wave on foam stability,a micro-kinetic model of enhanced foam stability under low frequency artificial seismic wave is established based on a vertical liquid film drainage model and elastic wave theory.The model is solved by non-dimensional transformation of the high order partial differential equations and a compound solution of implicit and explicit differences and is verified to be accurate.The foam film thickness,surfactant concentration distribution and drainage velocity under the action of low frequency artificial seismic wave are quantitatively analyzed.The research shows that low-frequency vibration can reduce the difference between the maximum and minimum concentrations of surfactant in the foam liquid film at the later stage of drainage,enhance the effect of Marangoni effect,and improve the stability of the foam liquid film.When the vibration frequency is close to the natural frequency of the foam liquid film,the vibration effect is the best,and the best vibration frequency is about 50 Hz.The higher the vibration acceleration,the faster the recovery rate of surfactant concentration in the foam liquid film is.The higher the vibration acceleration,the stronger the ability of Marangoni effect to delay the drainage of foam liquid film and the better the foam stability is.It is not the higher the vibration acceleration,the better.The best vibration acceleration is about 0.5 times of gravity acceleration.Reasonable vibration parameters would greatly enhance the effect of Marangoni effect.The smaller the initial concentration of surfactant,the better the vibration works in enhancing Marangoni effect.LIU Jing XIA Junyong LIU Xi WU Feipeng PU Chunsheng 2021Petroleum Exploration and Development2021,48,1:0
10Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 11A Machine-Learning Approach for the Prediction of Fly-Ash Concrete Strength显示文摘The composite exciter and the CaO to Na_(2)SO_(4) dosing ratios are known to have a strong impact on the mechanical strength offly-ash concrete.In the present study a hybrid approach relying on experiments and a machine-learn-ing technique has been used to tackle this problem.The tests have shown that the optimal admixture of CaO and Na_(2)SO_(4) alone is 8%.The best 3D mechanical strength offly-ash concrete is achieved at 8%of the compound activator;If the 28-day mechanical strength is considered,then,the best performances are obtained at 4%of the compound activator.Moreover,the 3D mechanical strength offly-ash concrete is better when the dosing ratio of CaO to Na_(2)SO_(4) in the compound activator is 1:1;the maximum strength offly-ash concrete at 28-day can be achieved for a 1:1 ratio of CaO to Na_(2)SO_(4) by considering a 4%compound activator.In this case,the compressive andflexural strengths are 260 MPa and 53.6 MPa,respectively;the mechanical strength offly-ash concrete at 28-day can be improved by a 4:1 ratio of CaO to Na_(2)SO_(4) by considering 8%and 12%compound excitants.It is shown that the predictions based on the aforementioned machine-learning approach are accurate and reliable.Shanqing Shao Aimin Gong Ran Wang Xiaoshuang Chen Jing Xu Fulai Wang Feipeng Liu 2023Fluid Dynamics & Materials Processing2023,19,12:0
12Microscopic Analysis of Cementitious Sand and Gravel Damming Materia显示文摘The mechanical properties of cementitious sand and gravel damming material have been experimentally determined by means of microscopic SEM(Scanning Electron Microscopy)image analysis.The results show that the combination of fly ash and water can fill the voids in cemented sand and gravel test blocks because of the presence of hydrated calcium silicate and other substances;thereby,the compactness and mechanical properties of these materials can be greatly improved.For every 10 kg/m^(3) increase in the amount of cementitious material,the density increases by about 2%,and the water content decreases by 0.2%.The amount of cementitious material used in the sand and gravel in these tests was 80-110 kg/m^(3),the water-binder ratio was 1-1.50.Moreover,the splitting tensile strength was 1/10 of the compressive strength,and the maximum strength was 7.42 MPa at 90 d.The optimal mix ratio has been found to be 50 kg of cement,60 kg of fly ash and 120 kg of water(C50F60W120).The related dry density was 2.6 g/cm^(3),the water content was 6%,and the water-binder ratio was 1.09.Ran Wang Aimin Gong Shanqing Shao Baoli Qu Jing Xu Fulai Wang Feipeng Liu 2024Fluid Dynamics & Materials Processing2024,20,4:0

14Transition metal-catalyzed conversion of aldehydes to ketones显示文摘Ketones are one of the most important classes of organic compounds, and widely present in various pharmacological compounds, biologically active molecules and functional materials. Over the past few decades, transition metal-catalyzed conversion of aldehydes has been found to be a powerful method.With the continuous development in recent years, it has become an efficient and uncomplicated strategy for constructing ketones. 1520μm Li metal modified with phosphate rich polymer-inorganic interphase applied in commercial carbonate electrolyte显示文摘Li metal batteries are supposed to reach real application in order to fulfill the high-energy density requirement of energy storage system.Unfortunately,the commonly used carbonate electrolyte react with pristine Li,which result in short lifetime of lithium metal battery.To alleviate the side reactions of Li metal with liquid electrolyte,here we propose a phosphate rich polymer-inorganic layer as an interphase.Due to the inert properties of lithium phosphate derived from LiPO_(2)F_(2)and poly-ether,the side-reaction of carbonate solvent are prevented.As a result,lithium metal anode sustains for 800 cycles in symmetrical cell test under 1 m A cm^(-2).Even under strict condition(20μm Li,capacity ratio N/P=2.3,electrolyte/active material=3μL mg^(-1)),coin cell test still runs stable for 150 cycles with high Coulombic efficiency.Furthermore,both LiFePO_(4)and LiNi_(0.8)Co_(0.1)Mn_(0.1)O_(2)pouch cell under 5μL m A^(-1)h^(-1)condition also exhibit good stability at 0.5 C and 2 C rate.With this approach,high-energy and high-power Li metal batteries are approaching to real application in the near future.Lin Lin Wei Lu Feipeng Zhao Siru Chen Jia Liu Haiming Xie Yulong Liu 2023Journal of Energy Chemistry2023,,1:0
16Tribological and mechanical behaviours of resin-based friction materials based on microcrack filling显示文摘To enhance the friction performance of resin-based friction materials,five types of specimens with different polymer ether ketone(PEEK)contents were fabricated and their physiomechanical behaviours were tested and,their tribological properties were investigated using a JF150F-II constant-speed tester.It was found that the addition of PEEK had a positive influence on the properties of the friction materials,and sample FM-3(the shorthand of‘Friction Materials-3’,containing 2 wt%PEEK)exhibited improved friction performance with a fade ratio and recovery ratio of 8.6%and 101.1%respectively.Among all samples,FM-4(the shorthand of‘Friction Materials-4’,con-taining 3 wt%PEEK)had the lowest specific wear rate with a value of 0.622×10^(−7)cm^(3)(N⋅m)^(−1)at 350℃.The PEEK can fill the microcracks in the composite at a high tem-perature and can also cover the hard abrasive particles to prevent them from directly damaging the composite.The findings from this study afford a foundation for studies to further improve the properties of resin-based friction materials.Lekai Li Guixiong Gao Jin Tong Jian Zhuang Wei Song Yunhai Ma Guoqin Liu Feipeng Cao Shengwang Yuan Qifeng Zhang 2023Biosurface and Biotribology2023,9,1:0
17Experimental characterization and mechanism of hydraulic pulsation waves driving microscopic residual oil显示文摘To clarify microscopic mechanisms of residual oil displacement by hydraulic pulsation wave,microscopic visualization experiments of hydraulic pulsation wave driving residual oil were carried out by using the microscopic visualization device of pulsating water drive.For the four types of residual oil left in the reservoir after water flooding,i.e.membrane,column,cluster,and blind end residual oils,hydraulic pulsation waves broke the micro-equilibrium of the interface by disturbing the oil-water interface,so that the injected water invaded into and contacted with the remaining oil in small pores and blind holes,and the remaining oil was pushed or stripped to the mainstream channel by deformation superposition effect and then carried out by the injected water.In the displacement,the pulsation frequency mainly affected the cluster and blind end remaining oil,and the hydraulic pulsation wave with a frequency of about 1 Hz had the best effect in improving the recovery.The pulsation amplitude value mainly affected the membrane and column residual oil,and the larger the amplitude value,the more remaining oil the hydraulic pulsation wave would displace.The presence of low intensity continuous flow pressure and holding pressure end pressure promoted the concentration of pulsating energy and greatly improve the recovery of cluster residual oil.The rise in temperature made the hydraulic pulsation wave work better in displacing remaining oil,improving the efficiency of oil flooding.WU Feipeng LI Na YANG Wei CHEN Jiahao DING Bujie XIA Lei LIU Jing WANG Cong WANG Lushan 2022Petroleum Exploration and Development2022,49,6:0
