| 1 | Clinical and biochemical indexes from 2019-nCoV infected patients linked to viral loads and lung injury显示文摘The outbreak of the 2019-nCoV infection began in December 2019 in Wuhan,Hubei province,and rapidly spread to many provinces in China as well as other countries.Here we report the epidemiological,clinical,laboratory,and radiological characteristics,as well as potential biomarkers for predicting disease severity in 2019-nCoV-infected patients in Shenzhen,China.All 12 cases of the 2019-nCoV-infected patients developed pneumonia and half of them developed acute respiratory distress syndrome (ARDS).The most common laboratory abnormalities were hypoalbuminemia,lymphopenia,decreased percentage of lymphocytes (LYM) and neutrophils (NEU),elevated C-reactive protein (CRP) and lactate dehydrogenase (LDH),and decreased CD8 count.The viral load of 2019-nCoV detected from patient respiratory tracts was positively linked to lung disease severity.ALB,LYM,LYM (%),LDH,NEU (%),and CRP were highly correlated to the acute lung injury.Age,viral load,lung injury score,and blood biochemistry indexes,albumin (ALB),CRP,LDH,LYM (%),LYM,and NEU (%),may be predictors of disease severity.Moreover,the Angiotensin Ⅱlevel in the plasma sample from 2019-nCoV infected patients was markedly elevated and linearly associated to viral load and lung injury.Our results suggest a number of potential diagnosis biomarkers and angiotensin receptor blocker (ARB) drugs for potential repurposing treatment of 2019-nCoV infection. | Yingxia Liu Yang Yang Cong Zhang Fengming Huang Fuxiang Wang Jing Yuan Zhaoqin Wang Jinxiu Li Jianming Li Cheng Feng Zheng Zhang Lifei Wang Ling Peng Li Chen Yuhao Qin Dandan Zhao Shuguang Tan Lu Yin Jun Xu Congzhao Zhou Chengyu Jiang Lei Liu | 2020 | Science China(Life Sciences)2020,63,3: | 157 |
| 2 | 3-N-butylphthalide improves neuronal morphology after chronic cerebral ischemia显示文摘3-N-butylphthalide is an effective drug for acute ischemic stroke. However, its effects on chronic cerebral ischemia-induced neuronal injury remain poorly understood. Therefore, this study ligated bilateral carotid arteries in 15-month-old rats to simulate chronic cerebral ischemia in aged humans. Aged rats were then intragastrically administered 3-n-butylphthalide. 3-N-butylphthalide administration improved the neuronal morphology in the cerebral cortex and hippocampus of rats with chronic cerebral ischemia, increased choline acetyltransferase activity, and decreased malondialdehyde and amyloid beta levels, and greatly improved cognitive function. | 3 | Millennial-centennial Scales Climate Changes of Holocene Indicated by Magnetic Susceptibility of High-resolution Section in Salawusu River Valley, China显示文摘The upmost segment (Holocene series) of the Milanggouwan stratigraphic section (MGS1) in the Salawusu River valley shows 11 sedimentary cycles of dune sands and fluvio-lacustrine facies, or dune sands and paleosols. The analysis of the magnetic susceptibility of this segment suggests that there are 11 magnetic susceptibility cycles with the value alternating from low to high, in which the layers of the dune sands correspond to the lower value of the magnetic susceptibility and the layers of fluvio-lacustrine facies and paleosols correspond to the higher peaks. The study reveals that the low and high magnetic susceptibility values indicate the climate dominated by cold-arid winter monsoon and warm-humid summer monsoon of East Asia, respectively, and the study area has experienced at least 22 times of millennial-centennial scales climate alternation from the cold-arid to the warm-humid during the Holocene. In terms of the time and the climate nature, the variations basically correspond to those of the North Atlantic and some records of cold-warm changes in China as well. | 4 | Is urban development an urban river killer? A case study of Yongding Diversion Channel in Beijing, China显示文摘The high population and concrete environment alter urban areas by changing temperature, rainfall runoff, and water resource utilization activities. This study was conducted to investigate the water quality features of the Yongding Diversion Channel in Beijing, China, and its relationship with rainfall and urban development. Monthly water quality data were obtained from April to October of 2004 at monitoring sites of Sanjiadian, Gaojing, Luodaozhuang, and Yuyuangtan. The monthly water quality grades from 2007 to 2011 were also investigated and compared with those of other rivers. Dissolved oxygen and pH showed greater decreases after one or two moderate rainfall events than several light rainfall events. The potassium permanganate index(CODMn), ammonia nitrogen(NH3-N) and total phosphorus(TP) increased more after several light rainfall events than after one or two moderate or heavy rainfall events. Pollutant concentrations(CODMn, NH3-N, TP) in downstream regions showed greater changes than those in upstream areas after heavy rainfall events. Intense human activities around the channel greatly influenced the water quality of the channel in rainy season because of runoff pollution; however, heavy rainfall had a strong dilution effect on the pollutant concentrations in rivers. Overall, urban development has obviously deteriorated the water quality of the Yongding Diversion Channel as indicated by an increase in the water quality index from 3.22 in 2008 to 4.55 in2010. | 6 | Garlic-derived compound S-allylmercaptocysteine inhibits hepatocarcinogenesis through targeting LRP6/Wnt pathway显示文摘Whether and how garlic-derived S-allylmercaptocysteine(SAMC) inhibits hepatocellular carcinoma(HCC) is largely unknown. In the current study, the role of low-density lipoprotein receptor(LDLR)-related protein 6(LRP6) in HCC progression and the anti-HCC mechanism of SAMC was examined in clinical sample, cell model and xenograft/orthotopic mouse models. We demonstrated that SAMC inhibited cell proliferation and tumorigenesis, while induced apoptosis of human HCC cells without influencing normal hepatocytes. SAMC directly interacted with Wnt-pathway co-receptor LRP6 on the cell membrane. LRP6 was frequently over-expressed in the tumor tissue of human HCC patients(66.7% of 48 patients) and its overexpression only correlated with the over-expression of β-catenin, but not with age, gender, tumor size, stage and metastasis. Deficiency or over-expression of LRP6 in hepatoma cells could partly mimic or counteract the anti-tumor properties of SAMC, respectively. In vivo administration of SAMC significantly suppressed the growth of Huh-7 xenograft/orthotopic HCC tumor without causing undesirable side effects. In addition, stable down-regulation of LRP6 in Huh-7 facilitated the anti-HCC effects of SAMC. In conclusion, LRP6 can be a potential therapeutic target of HCC. | 7 | Changes in nitrogen budget and potential risk to the environment over 20 years(1990–2010) in the agroecosystems of the Haihe Basin, China显示文摘The nitrogen balance can serve as an indicator of the risk to the environment of nitrogen loss from agricultural land. To investigate the temporal and spatial changes in agricultural nitrogen application and its potential threat to the environment of the Haihe Basin in China, we used a database of county-level agricultural statistics to calculate agricultural nitrogen input, output,surplus intensity, and use efficiency. Chemical fertilizer nitrogen input increased by 51.7% from1990 to 2000 and by 37.2% from 2000 to 2010, concomitant with increasing crop yields.Simultaneously, the nitrogen surplus intensity increased by 53.5% from 1990 to 2000 and by16.5% from 2000 to 2010, presenting a continuously increased environmental risk. Nitrogen use efficiency decreased from 0.46 in 1990 to 0.42 in 2000 and remained constant at 0.42 in 2010,partly due to fertilizer composition and type improvement. This level indicates that more than half of nitrogen inputs are lost in agroecosystems. Our results suggest that although the improvement in fertilizer composition and types has partially offset the decrease in nitrogen use efficiency, the environmental risk has still increased gradually over the past 20 years, along with the increase in crop yields and nitrogen application. | 9 | Preparation of CeO_2-Modified Mg(Al)O-Supported Pt–Cu Alloy Catalysts Derived from Hydrotalcite-Like Precursors and Their Catalytic Behavior for Direct Dehydrogenation of Propane显示文摘A series of PtCuCeMgAl quintuple hydrotalcite-like compounds with different Ce contents were synthesized by one-pot method. After calcining and reduction, CeO_2-modified Mg(Al)O-supported Pt–Cu alloy catalysts were obtained. To understand the effect of Cu and Ce, the structure and physico-chemistry properties of the catalysts were characterized and analyzed, and the catalytic behaviors were investigated in a direct dehydrogenation of propane to propene. The results show that the Pt^(4+), Cu^(2+), and Ce^(3+) ions can be incorporated into the brucite-like layers and the Ce content significantly affects the interaction strength between Pt and Cu and the dehydrogenation performance of propane. Under the reaction conditions, the highest propane conversion(45%) with 89% selectivity to propene and a 40% propene yield were achieved with a 0.3 wt% Ce-modified PtCu/Mg(Al)O catalyst. | 13 | Development of a reverse transcription quantitative polymerase chain reaction-based assay for broad coverage detection of African and Asian Zika virus lineages显示文摘The Zika virus(ZIKV) is an arbovirus that has spread rapidly worldwide within recent times. There is accumulating evidence that associates ZIKV infections with Guillain-Barré Syndrome(GBS) and microcephaly in humans. The ZIKV is genetically diverse and can be separated into Asian and African lineages. A rapid, sensitive, and specific assay is needed for the detection of ZIKV across various pandemic regions. So far, the available primers and probes do not cover the genetic diversity and geographic distribution of all ZIKV strains. To this end, we have developed a one-step quantitative reverse transcription polymerase chain reaction(qRT-PCR) assay based on conserved sequences in the ZIKV envelope(E) gene. The detection limit of the assay was determined to be five RNA transcript copies and 2.94 × 10^(–3) 50% tissue culture infectious doses(TCID50) of live ZIKV per reaction. The assay was highly specific and able to detect five different ZIKV strains covering the Asian and African lineages without nonspecific amplification, when tested against other flaviviruses. The assay was also successful in testing for ZIKV in clinical samples. | 15 | Drug repositioning of disulfiram induces endometrioid epithelial ovarian cancer cell death via the both apoptosis and cuproptosis pathways显示文摘Various therapeutic strategies have been developed to overcome ovarian cancer.However,the prognoses resulting from these strategies are still unclear.In the present work,we screened 54 small molecule compounds approved by the FDA to identify novel agents that could inhibit the viability of human epithelial ovarian cancer cells.Among these,we identified disulfiram(DSF),an old alcohol-abuse drug,as a potential inducer of cell death in ovarian cancer.Mechanistically,DSF treatment significantly reduced the expression of the anti-apoptosis marker Bcell lymphoma/leukemia-2(Bcl-2)and increase the expression of the apoptotic molecules Bcl2 associated X(Bax)and cleaved caspase-3 to promote human epithelial ovarian cancer cell apoptosis.Furthermore,DSF is a newly identified effective copper ionophore,thus the combination of DSF and copper was used to reduce ovarian cancer viability than DSF single treatment.Combination treatment with DSF and copper also led to the reduced expression of ferredoxin 1 and loss of Fe-S cluster proteins(biomarkers of cuproptosis).In vivo,DSF and copper gluconate significantly decreased the tumor volume and increased the survival rate in a murine ovarian cancer xenograft model.Thus,the role of DSF revealed its potential for used as a viable therapeutic agent for the ovarian cancer. | YAPING GAN TING LIU WEIFENG FENG LIANG WANG LI LI YINGXIA NING | 2023 | Oncology Research2023,31,3: | 2 |
