| 1 | Multimodal Identification by Transcriptomics and Multiscale Bioassays of Active Components in Xuanfeibaidu Formula to Suppress Macrophage-Mediated Immune Response显示文摘Xuanfeibaidu Formula (XFBD) is a Chinese medicine used in the clinical treatment of coronavirus disease 2019 (COVID-19) patients. Although XFBD has exhibited significant therapeutic efficacy in clinical practice, its underlying pharmacological mechanism remains unclear. Here, we combine a comprehensive research approach that includes network pharmacology, transcriptomics, and bioassays in multiple model systems to investigate the pharmacological mechanism of XFBD and its bioactive substances. High-resolution mass spectrometry was combined with molecular networking to profile the major active substances in XFBD. A total of 104 compounds were identified or tentatively characterized, including flavonoids, terpenes, carboxylic acids, and other types of constituents. Based on the chemical composition of XFBD, a network pharmacology-based analysis identified inflammation-related pathways as primary targets. Thus, we examined the anti-inflammation activity of XFBD in a lipopolysaccharide-induced acute inflammation mice model. XFBD significantly alleviated pulmonary inflammation and decreased the level of serum proinflammatory cytokines. Transcriptomic profiling suggested that genes related to macrophage function were differently expressed after XFBD treatment. Consequently, the effects of XFBD on macrophage activation and mobilization were investigated in a macrophage cell line and a zebrafish wounding model. XFBD exerts strong inhibitory effects on both macrophage activation and migration. Moreover, through multimodal screening, we further identified the major components and compounds from the different herbs of XFBD that mediate its anti-inflammation function. Active components from XFBD, including Polygoni cuspidati Rhizoma, Phragmitis Rhizoma, and Citri grandis Exocarpium rubrum, were then found to strongly downregulate macrophage activation, and polydatin, isoliquiritin, and acteoside were identified as active compounds. Components of Artemisiae annuae Herba and Ephedrae Herba were found to substantially inhibit endogenous macrophage migration, while the presence of ephedrine, atractylenolide I, and kaempferol was attributed to these effects. | 2 | An ultra-robust fingerprinting method for quality assessment of traditional Chinese medicine using multiple reaction monitoring mass spectrometry显示文摘Chromatographic fingerprinting has been perceived as an essential tool for assessing quality and chemical equivalence of traditional Chinese medicine.However,this pattern-oriented approach still has some weak points in terms of chemical coverage and robustness.In this work,we proposed a multiple reaction monitoring(MRM)-based fingerprinting method in which approximately 100 constituents were simultaneously detected for quality assessment.The derivative MRM approach was employed to rapidly design MRM transitions independent of chemical standards,based on which the large-scale fingerprinting method was efficiently established.This approach was exemplified on QiShenYiQi Pill(QSYQ),a traditional Chinese medicine-derived drug product,and its robustness was systematically evaluated by four indices:clustering analysis by principal component analysis,similarity analysis by the congruence coefficient,the number of separated peaks,and the peak area proportion of separated peaks.Compared with conventional ultraviolet-based fingerprints,the MRM fingerprints provided not only better discriminatory capacity for the tested normal/abnormal QSYQ samples,but also higher robustness under different chromatographic conditions(i.e.,flow rate,apparent pH,column temperature,and column).The result also showed for such large-scale fingerprints including a large number of peaks,the angle cosine measure after min-max normalization was more suitable for setting a decision criterion than the unnormalized algorithm.This proof-of-concept application gives evidence that combining MRM technique with proper similarity analysis metrices can provide a highly sensitive,robust and comprehensive analytical approach for quality assessment of traditional Chinese medicine. | Zhenhao Li Xiaohui Zhang Jie Liao Xiaohui Fan Yiyu Cheng | 2021 | Journal of Pharmaceutical Analysis2021,11,1: | 4 |
| 3 | Segmental Translocation Contributed to the Origin of the Brassica S-locus显示文摘Self-incompatibility(SI),which has recurred during the evolution of plants,is one of the most important cross-pollination mating systems.Three S-loci have been reported in Brassicaceae,namely,Arabidopsis lyrata(Al),Brassica(Br),and Leavenworthia alabamica(La)S-loci.Here,through multi-genomic comparative analysis of 20 species,we revealed that the most ancient S-locus was formed prior to the divergence of Brassicaceae lineage I and II.Itwas retained and inherited by Arabidopsis,as the Al S-locus in Brassicaceae lineage I.Furthermore,we found that the Br S-locus,which has been widely used in the breeding of Brassica crops to generate hybrid seeds,was formed through segmental translocation(ST)in the hexaploid ancestor of Brassica in Brassicaceae lineage II.The Br S-locus was evolved through a ST from one of the triplicated ancestral S-locus paralogs in the Brassica hexaploidy ancestor,while the other two S-locus paralogs were lost.Together with the previous discovery that the La S-locus was formed through a secondary origin in Brassicaceae lineage I,we conclude the monophyletic origin of Al and Br S-loci and clarify the evolutionary route of S-loci in the Brassicaceae family.Our findings will contribute to evolutionary studies and breeding applications of the S-locus in Brassicaceae. | Yinan Cui Mu Zhuang Jian Wu Jisheng Liu Yiyue Zhang Lingkui Zhang Yile Huang Xu Cai Jianli Liang Kang Zhang Xiaowu Wang Feng Cheng | 2020 | Horticultural Plant Journal2020,6,3: | 2 |

| 12 | Macro-evolutionary patterns of East Asian opsariichthyin-xenocyprinin-cultrin fishes related to the formation of river and river-lake environments under monsoon climate显示文摘Although the East Asian monsoon is believed to be an important driver for the origin and evolution of the biotas in this region,the association has rarely been rigorously tested.Here,using phylogenetic comparative methods and analyses of key innovations of adaptive traits,we investigated evolutionary patterns of the East Asian cyprinid opsariichthyin-xenocyprinin-cultrin clade;the dominant species in lakes and rivers,which are the typical productions of monsoon climate.Our molecular phylogenetic analyses revealed the relationships as(Opsariichthystribe t(((Oxygaster-tribe t Aphyocypris-tribe)t Metzia-tribe)t(Paralaubuca-tribe t(Squaliobarbus-tribe t(Hypophthalmichthys-tribe t(xenocyprinins t cultrins)))))).Based on Bayesian relaxed-clock methods,we found that origins of the Squaliobarbus-and Hypophthalmichthys-tribes bearing the key adaptation of riverine spawning with drifting eggs occurred 22.5 and 21.1 Mya,respectively.These times are consistent with the Early Miocene origin of major rivers(e.g.,Yangtze River)and the appearance of the East Asian monsoon climate.Diversification of the cultrins,characterized by spawning of adhesive eggs as well as swimming and feeding in lentic waters,were estimated to have evolved 16.6 to 0.2 Mya.This period covers three phases of enhancement of the East Asian monsoon from the Middle Miocene to the Pliocene(around 15–13,10–7,and 3.5 Mya).The habitats of these cultrins and their evolution also suggest the appearance and development of a lake-river environment during those periods.Therefore,our results suggest close relationships between the evolution of these fishes and geological events in East Asia.Further,they provide knowledge that may help facilitate future conservation strategies such as the maintenance of high river flows during dam construction and operation. | Peilin Cheng Dan Yu Qiongying Tang Jinquan Yang Yiyu Chen Huanzhang Liu | 2022 | Water Biology and Security2022,1,2: | 1 |

It is well known that scutellarin has anti-infammatory and antioxidant physiological functions. In this study, we detected the effects of scutellarin on hepatocyte cell apoptosis in type 2 diabetes mellitus(T2DM) rats.Methods: Sprague Dawley(SD)(6–8 weeks, 160–180 g) rats were randomly divided into six groups: control, model, scutellarin low-dose, medium-dose, high-dose treatment, and rosiglitazone positive groups;with 10 SD rats in each group(n = 10). The changes of biochemical factors in serum were detected by automatic biochemical instrument, the pathological changes of liver tissue were detected by hematoxylin and eosin(HE) staining, the apoptosis of liver tissue and cells was detected by tissue staining and fow analyzer, and the expression of apoptosis-related factors were determined by q PCR, Western blot and immunohistochemistry in liver tissues or cells.Results: The results showed that scutellarin decreased the levels of fasting blood glucose, total cholesterol, triglyceride, and low-density lipoprotein and increased the levels of high-density lipoprotein.Meanwhile, scutellarin decreased the levels of alanine transaminase(ALT) and aspartate transaminase(AST) and improved liver function. | 18 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO | 20 | Application of Heterogeneous Multiple Feature Data Fusion to Multitarget Association and Tracking显示文摘In this paper we discuss a kind of multitarget tracking and association method based on the data fusion of heterogeneous multiple feature data gained by a sensor such as space state, signal amplitude, Doppler frequency and so on. In order to introduce quantitatively those heterogeneous multiple feature data which are possibly gailied by a sensor into the discussion of tracking and association problem, we define a correlation measure which we explain as the generalization of conventional association decision. In conventional Nearest Neighbor method, the decision function can take only two values, 1 or 0, to represent the decision of association or not association. In our method, correlation measure can take any real value from 0 to 1 to represent the extent of correlation. Considering the practical circumstances that some feature data might not be easily gained continuously, we introduce an effective factor to deal with these cases.
