| 2 | Characterization of the essential role of bone morphogenetic protein 9 (BMP9) in osteogenic differentiation of mesenchymal stem cells (MSCs) through RNA interference显示文摘Mesenchymal stem cells(MSCs)are multipotent stem cells and capable of differentiating into multiple cell types including osteoblastic,chondrogenic and adipogenic lineages.We previously identified BMP9 as one of the most potent BMPs that induce osteoblastic differentiation of MSCs although exact molecular mechanism through which BMP9 regulates osteogenic differentiation remains to be fully understood.Here,we seek to develop a recombinant adenovirus system to optimally silence mouse BMP9 and then characterize the important role of BMP9 in osteogenic differentiation of MSCs.Using two different siRNA bioinformatic prediction programs,we design five siRNAs targeting mouse BMP9(or simB9),which are expressed under the control of the converging H1 and U6 promoters in recombinant adenovirus vectors.We demonstrate that two of the five siRNAs,simB9-4 and simB9-7,exhibit the highest efficiency on silencing exogenous mouse BMP9 in MSCs.Furthermore,simB9-4 and simB9-7 act synergistically in inhibiting BMP9-induced expression of osteogenic markers,matrix mineralization and ectopic bone formation from MSCs.Thus,our findings demonstrate the important role of BMP9 in osteogenic differentiation of MSCs.The characterized simB9 siRNAs may be used as an important tool to investigate the molecular mechanism behind BMP9 osteogenic signaling.Our results also indicate that recombinant adenovirus-mediated expression of siRNAs is efficient and sustained,and thus may be used as an effective delivery vehicle of siRNA therapeutics. | Shujuan Yan Ruyi Zhang Ke Wu Jing Cui Shifeng Huang Xiaojuan Ji Liping An Chengfu Yuan Cheng Gong Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Bo Liu Rex C.Haydon Michael J.Lee Russell R.Reid Jennifer Moriatis Wolf Qiong Shi Hue H.Luu Tong-Chuan He Yaguang Weng | 2018 | Genes & Diseases2018,5,2: | 8 |

| 4 | Ischemic stroke susceptibility gene in a Northern Han Chinese population显示文摘Interleukin-18 gene promoter polymorphisms are potential risk factors for ischemic cerebrovascular disease, and the -607C allele may increase ischemic stroke risk in the Han Chinese population. In the present study, we recruited 291 patients with ischemic cerebrovascular disease from the Affiliated Hospital of Qingdao University Medical College, China, and 226 healthy controls. Both patients and controls were from the Han population in northern China. Immunoresonance scattering assays detected increased serum amyloid A protein, C-reactive protein, and interleukin-18 levels in ischemic cerebrovascular disease patients compared with healthy controls. Analysis of the -607C/A (rs1946518) polymorphism in the interleukin-18 gene promoter showed ischemic cerebrovascular disease patients exhibited increased frequencies of the CC genotype and C alleles than healthy controls. Genotype and allele frequencies of the interleukin-18 -137G/C (rs187238) polymorphism and the -13T/C (rs11024595) polymorphism in the 5'-flanking region of serum amyloid A, showed no significant difference between the two groups. Multivariate logistic regression analysis on the interleukin-18 promoter A/C genetic locus, for correction of age, gender, history of smoking, hypertension, diabetes mellitus, hypercholesteremia, and an ischemic stroke family history, showed ischemic cerebrovascular disease risk in individuals without the A allele (C homozygotes) was 2.2-fold greater than in A allele carriers. Overall, our findings suggest that the -13T/C (rs11024595) polymorphism in the 5′-flanking region of serum amyloid A has no correlation with ischemic cerebrovascular disease, but the C allele of the -607C/A (rs1946518) polymorphism in the interleukin-18 promoter is a high-risk factor for ischemic cerebrovascular disease in the Han population of northern China. | 6 | A near-infrared phosphorescent probe for F^- based on a cationic iridium(Ⅲ) complex with triarylboron moieties显示文摘In this work, a near-infrared (NIR) phosphorescent probe for F- based on a cationic Ir(Ⅲ) complex [Ir(Bpq)2(quqo)]PF6 (1) with dimesitylboryl (Mes2B) groups on the cyclometalated C^N ligands (Bpq) and 2-(quinolin-2-yl)quinoxaline (quqo) as N^N ligand was designed and synthesized. The excited state properties of 1 were investigated in detail using molecular orbital cal- culations and experimental methods. Upon excitation, complex 1 shows NIR phosphorescent emission around 680 nm. Inter- estingly, the complex can be excited with long wavelength around 610 nm. Such long-wavelength excitation can reduce the background emission interference and improve the signal-to-noise ratio. Furthermore, the selective binding between boron at- om and F- can give rise to the quenching of emission and realize the near-infrared phosphorescent sensing for F-. | 10 | An Old Story Retold: Loss of G1 Control Defines A Distinct Genomic Subtype of Esophageal Squamous Cell Carcinoma显示文摘Esophageal squamous cell carcinoma(ESCC) has a high mortality rate. To determine the molecular basis of ESCC development, this study sought to identify characteristic genome-wide alterations in ESCC, including exonic mutations and structural alterations. The clinical implications of these genetic alterations were also analyzed. Exome sequencing and verification were performed for nine pairs of ESCC and the matched blood samples, followed by validation with additional samples using Sanger sequencing. Whole-genome SNP arrays were employed to detect copy number alteration(CNA) and loss of heterozygosity(LOH) in 55 cases, including the nine ESCC samples subjected to exome sequencing. A total of 108 non-synonymous somatic mutations(NSSMs) in102 genes were verified in nine patients. The chromatin modification process was found to be enriched in our gene ontology(GO) analysis. Tumor genomes with TP53 mutations were significantly more unstable than those without TP53 mutations. In terms of the landscape of genomic alterations, deletion of 9p21.3 covering CDKN2A/2B(30.9%), amplification of 11q13.3 covering CCND1(30.9%), and TP53 point mutation(50.9%) occurred in two-thirds of the cases. These results suggest that the deregulation of the G1 phase during the cell cycle is a key event in ESCC.Furthermore, six minimal common regions were found to be significantly altered in ESCC samples and three of them, 9p21.3, 7p11.2, and 3p12.1, were associated with lymph node metastasis. | 15 | The Inhibitory Effects of PSS-Loaded Nanoparticles on the Dysfunction of Cardiac Microvascular Endothelia in Rats with Diabetic Cardiomyopathy显示文摘Propylene glycol alginate sodium sulfate-loaded nanoparticles(PSS-NP) has been shown potential to prevent the microvascular endothelial injuries caused by diabetic cardiomyopathy(DCM). In this study, we aimed to investigate the effects of PSS-NP on the dysfunction of cardiac microvascular endothelia in streptozotocin(STZ)-induced DCM rat model. Echocardiographic measurements showed a significant improvement of cardiac function in the PSS-NP-treated group. Our results revealed that the abnormalities of cardiac systolic and diastolic functions were suppressed by the treatments of prostaglandin E1(PGE1), low molecular weight heparin(LMWH), PSS and PSS-NP. Our comparison analysis indicated that PSS-NP showed the strongest inhibitory effects on microvascular endothelial injuries. Transmission electron microscopy analysis demonstrated that PSS-NP protected the cardiac microvascular endothelium and further improved endothelium dysfunction in DCM rats. ELISA and Western blot assays further showed a high efficiency of improving cardiac microvascular endothelial dysfunction with PSS-NP. Our results demonstrated that PSS-NP increased the protein expression of phosphatidylinositol 3-kinase(PI3K)-p85 and vascular endothelial growth factor(VEGF)-A, and the phosphorylation of protein kinase B(Akt) and endothelial nitric oxide synthase(eNOS), which were involved in the amelioration of cardiac microvascular endothelial dysfunction. | 16 | Lattice vibrational modes and phonon thermal conductivity of singlelayer GaGeTe显示文摘Exploring the vibrational properties of experimental or theoretical finding of new phase twodimensional(2D)materials is one key to expand their promising application.The GaGeTe monolayer with high carrier mobility,tunable band structure,and low cleavage energy attracts rapidly growing interests in electronics.However,their vibrational modes and phonon properties are not explored.Based on first-principles calculations within the framework of density functional perturbation theory,we systematically study the lattice vibrational modes as well as phonon thermal conductivity of single-layer GaGeTe,which could be exfoliated from the experimentally synthesized GaGeTe crystal.We find that the frequencies and intensities of A1g and Eg Raman modes,contributing to the main peaks in Raman spectra of monolayer GaGeTe,show distinct responses to different external strains.The calculated lattice thermal conductivity of GaGeTe is about 58 Wm-1 K1,which is comparable to that of MoS2(around 52 Wm1K1)and two orders of magnitude lower than that of graphene(2000 Wm1K1)at room temperature.This is mainly due to the partial couple for acoustic branches and low frequency optic phonon branches,moderate group velocity,strong anharmonic ZA phonon branch.Moreover,atom substitution at tellurium could be as an effective strategy to further decrease lattice thermal conductivity.Our findings fill in the gap about the thermal transport properties of GaGeTe monolayer and trigger for further experimental verification of the predicted properties. | Jingyu Li Peng-Fei Liu Chi Zhang Xiaobo Shi Shujuan Jiang Weizhen Chen Huabing Yin Bao-Tian Wang | 2020 | Journal of Materiomics2020,6,4: | 0 |

| 20 | Computational prediction of Mo_(2)@g-C_(6)N_(6) monolayer as an efficient electrocatalyst for N_(2) reduction显示文摘Electrocatalytic nitrogen reduction reaction(NRR)is an environmentally friendly method for sustainable ammonia synthesis under ambient conditions.Searching for efficient NRR electrocatalysts with high activity and selectivity is currently urgent but remains great challenge.Herein,we systematically investigate the NRR catalytic activities of single and double transition metal atoms(TM=Fe,Co,Ni and Mo)anchored on g-C_(6)N_(6) monolayers by performing first-principles calculation.Based on the stability,activity,and selectivity analysis,Mo_(2)@g-C_(6)N_(6) monolayer is screened out as the most promising candidate for NRR.Further exploration of the reaction mechanism demonstrates that the Mo dimer anchored on g-C_(6)N_(6) can sufficiently activate and efficiently reduce the inert nitrogen molecule to ammonia through a preferred distal pathway with a particularly low limiting potential of -0.06 V.In addition,we find that Mo_(2)@g-C_(6)N_(6) has excellent NRR selectivity over the competing hydrogen evolution reaction,with the Faradaic efficiency being 100%.Our work not only predicts a kind of ideal NRR electrocatalyst but also encouraging more experimental and theoretical efforts to develop novel double-atom catalysts(DACs)for NRR. | Jiajun Wang Mengyao Shi Guolin Yi Lu Wang Shulai Lei Ke Xu Shujuan Li Jianshuai Mu | 2022 | Chinese Chemical Letters2022,33,10: | 0 |
